3Artificial Selection in Domestication and Breeding Prevents Speciation in Rice显示文摘Speciation has long been regarded as an irreversible process once the reproductive barriers had been established.However,unlike in natural populations,artificial selection might either accelerate or prevent speciation processes in domesticated species.Asian cultivated rice is a target crop for both domestication and artificial breeding;it contains two subspecies of indica and japonica,which usually produce sterile inter-subspecific hybrids due to reproductive barriers.In this study,we constructed the evolutionary trajectory of a reproductive isolation system S5,which regulates fertility in indica-japonica hybrids via three adjacent genes,based on the data of 606 accessions including two cultivated and 11 wild rice species.Although hybrid sterility haplotypes at S5 lead to establishment of a killer-protector reproductive barrier,origin of wide-compatibility haplotypes by complex hybridization and recombination provides an opposing force to reproductive isolation and thus prevents speciation during domestication.Analysis in a diallel set of 209 crosses involving 21 parents showed that the wide-compatibility genotypes largely rescued fertility of indica-japonica hybrids,indicating that the wide-compatibility gene would enable gene flow to maintain species coherence.This counteracting system indicates that combined effects of natural evolution and artificial selection may result in reversible processes of speciation in rice,which may also have implications for genetic improvement of rice.Jiaming Mi Guangwei Li Conghao Xu Jiangyi Yang Huihui Yu Gongwei Wang Xianghua Li Jinghua Xiao Huazhi Song Qifa Zhang Yidan Ouyang 2020Molecular Plant2020,13,4:6
4Spatiotemporal variations of nitrate sources and dynamics in a typical agricultural riverine system under monsoon climate显示文摘Nitrogen pollution is a serious environmental issue in the Danjiangkou Reservoir region(DRR),the water source of the South-to-North Water Diversion Project of China.In this research,seasonal surveys and a bi-weekly time series survey were conducted in the Qihe River Basin,one of the most densely populated agricultural basins in the DRR.Hydrochemical compositions(NO3^-Cl^-),dual isotopes(δD-H2O,δ^18O-H2O,δ^15N-NO3^-,andδ^18O-NO3^-),and a Markov Chain Monte Carlo isotope mixing model were jointly applied to unravel the sources,migrations,and transformations of the nitrate(NO3^-)in the basin.It was revealed that the mixing between different sources was the main process controlling the isotopic compositions of the riverine NO3the upper-middle reaches.In contrast,denitrification occurred in the lower reaches.For the first time,the sources of NO3^-quantified at a basin scale in the DRR.Overall,the river transported 484.2 tons/year of NO3^-N to the reservoir,of which 32.6%,36.4%,28.0%,and 3.0%was from soil organic nitrogen,chemical fertilizer,residential sewage and atmospheric precipitation,respectively.The NO3^-N fluxes of the different sources were regulated by the monsoon climate and anthropogenic activities.For example,high precipitation and intense fertilization resulted in severe nonpoint source pollution.Denitrification thrived in soils and reservoirs in wet seasons.Temperature could regulate the migration,nitrification and denitrification processes.Based on the results,we suggest that the management strategies dealing with nitrogen pollution issue in the DRR should follow the specific spatiotemporal characteristics of NO3migration and transformation mechanisms.Hao Jiang Wenjing Liu Jiangyi Zhang Li Zhou Xiaode Zhou Ke Pan Tong Zhao Yuchen Wang Zhifang Xu 2020Journal of Environmental Sciences2020,32,7:5
5O,H,and Sr isotope evidence for origin and mixing processes of the Gudui geothermal system,Himalayas,China显示文摘Tho Gudui geothermal field records the highest temperature at equivalent borehole depths among the lainland hydrothermal systems in China's Mainland.Located about 150 km southeast of Lhasa City,the capital of Tibet,the Gudui geothermal field belongs to the Sangri-Cuona rift belt,also known as the Sangri-Cuona geothermal belt,and is representative of the non-volcanic geothermal systems in the Himalayas.In this study,oxygen-18 and deuterium isotope compositions as well as 87Sr/86Sr ratios of water samples collected from the Gudui geothermal field were characterized to understand the origin and mixing processes of the geothermal fluids at Gudui.Hydrogen and oxygen isotope plots show both,deep and shallow reservoirs in the Gudui geothermal field.Deep geothermal fluids are the mixing product of magmatic and infiltrating snow-melt water.Calculations show that the magma fluid component of the deep geothermal fluids account for about 21.10%-24.04%;magma fluids lay also be a contributing source of lithium.The linear relationship of the 87Sr/86Sr isotopic ratio versus the 1/Sr plot indicates that shallow geothermal fluids form from the mixing of deep geothermal fluids with cold groundwater.Using a binary mixing model with deep geothermal fluid and cold groundwater as two end-members,the nixing ratios of the latter in most surface hot springs samples were calculated to be between 5% and 10%.Combined with basic geological characteristics,hydrogen and oxygen isotope characteristics,strontium concentration,87Sr/(86)Sr ratios,and the binary mixing model,we infer the 6 th-Class Reservoirs Evolution Conceptual Model(6-CRECM) for the Gudui geothermal system.This model represents an idealized summary of the characteristics of the Gudui geothermal field based on our comprehensive understanding of the origin and mixing processes of the geothermal fluid in Gudui.This study may aid in identifying the geothermal and geochemical origin of the Gudui high-temperature hydrothermal systems in remote Tibet of China,whose potential for geothermal development and utilization is enormous and untapped.Chenguang Wang Mianping Zheng Xuefei Zhang Enyuan Xing Jiangyi Zhang Jianhong Ren Yuan Ling 2020Geoscience Frontiers2020,11,4:5
6Tropoelastin improves adhesion and migration of intra-articular injected infrapatellar fat pad MSCs and reduces osteoarthritis progression显示文摘Intra-articular injection of mesenchymal stem cells(MSCs)is a promising strategy for osteoarthritis(OA)treatment.However,more and more studies reveal that the injected MSCs have poor adhesion,migration,and survival in the joint cavity.A recent study shows that tropoelastin(TE)regulates adhesion,proliferation and phenotypic maintenance of MSCs as a soluble additive,indicating that TE could promote MSCs-homing in regenerative medicine.In this study,we used TE as injection medium,and compared it with classic media in MSCs intra-articular injection such as normal saline(NS),hyaluronic acid(HA),and platelet-rich plasma(PRP).We found that TE could effectively improve adhesion,migration,chondrogenic differentiation of infrapatellar fat pad MSCs(IPFP-MSCs)and enhance matrix synthesis of osteoarthritic chondrocytes(OACs)in indirect-coculture system.Moreover,TE could significantly enhance IPFP-MSCs adhesion via activation of integrin β1,ERK1/2 and vinculin(VCL)in vitro.In addition,intra-articular injection of TE-IPFP MSCs suspension resulted in a short-term increase in survival rate of IPFP-MSCs and better histology scores of rat joint tissues.Inhibition of integrin β1 or ERK1/2 attenuated the protective effect of TE-IPFP MSCs suspension in vivo.In conclusion,TE promotes performance of IPFP-MSCs and protects knee cartilage from damage in OA through enhancement of cell adhesion and activation of integrin β1/ERK/VCL pathway.Our findings may provide new insights in MSCs intra-articular injection for OA treatment.Junjun Yang Xin Wang Yahan Fan Xiongbo Song Jiangyi Wu Zhenlan Fu Tao Li Yang Huang ZheXiong Tang Shuo Meng Na Liu Jiajia Chen Pingju Liu Liu Yang Xiaoyuan Gong Cheng Chen 2022Bioactive Materials2022,7,4:4
7Neural mechanisms of perceptual grouping in human visual cortex显示文摘The current work examined neural substrates of perceptual grouping in human visual cortex using event-related potential (ERP) recording. Stimulus arrays consisted of local elements that were either evenly spaced (uniform stimuli) or grouped into columns or rows by prox-imity or color similarity (grouping stimuli). High-density ERPs were recorded while subjects identified orientations of perceptual groups in stimulus arrays that were presented randomly in one of the four quadrants of the visual field. Both uniform and grouping stimulus arrays elicited an early ERP component (C1), which peaked at about 70 ms after stimulus onset and changed its polarity as a function of stimulated elevations. Dipole modeling based on realistic- head boundary-element models revealed generators of the C1 component in the calcarine cortex. The C1 was modu-lated by perceptual grouping of local elements based on proximity, and this grouping effect was stronger in the upper than in the lower visual field. 16Histological observation on acellular nerve grafts co-cultured with Schwann cells for repairing defects of the sciatic nerve显示文摘BACKGROUND: Animal experiments and clinical studies about tissue engineering method applied to repair nerve injury mainly focus on seeking ideal artificial nerve grafts, nerve conduit and seed cells. Autologous nerve, allogeneic nerve and xenogeneic nerve are used to bridge nerve defects, it is one of the methods to promote the repair of nerve injury by culturing and growing Schwann cells, which can secrete various neurotrophic factor activities, in the grafts. OBJECTIVE: To observe the effect of acellular nerve grafts co-cultured with Schwann cells in repairing defects of sciatic nerve. DESIGN: An observational comparative study. SETTING: Tissue Engineering Laboratory of China Medical University. MATERIALS: The experiment was carried out in the Tissue Engineering Laboratory of China Medical University between April 2004 and April 2005. Forty neonatal Sprague-Dawley rats of 5-8 days (either males or females) and 24 male Wistar rats of 180-220 g were provided by the experimental animal center of China Medical University. METHODS: ① Culture of Schwann cells: The bilateral sciatic nerves and branchial plexus were isolated from the 40 neonatal SD rats. The sciatic nerves were enzymatically digested with collagenase and dispase, isolatd, purified and cultured with the method of speed-difference adhersion, and identified with the SABC immunohistochemical method. ② Model establishment: In vitro Schwann cells were microinjected into 10-mm long acellular nerve grafts repairing a surgically created gap in the rat sciatic nerve. According to the different grafted methods, the animals were randomly divided into three groups: autografts (n=8), acellular nerve grafts (n=8), or acellular nerve grafts with Schwann cells (n=8). ③ The regenerated nerve fiber number and average diameter of myeline sheath after culture were statistically anlayzed. MAIN OUTCOME MEASURES: ① The regenerated nerve ultrastructure, total number and density of myelinated nerve fibers, and the thickness of myeline sheath were observed under electron microscope. ② The images were processed with the Mias-1000 imaging analytical system to calculate the number of myelinated nerve fibers, and the thickness of myeline sheath. RESULTS: All the 24 Wistar rats were involved in the analysis of results. ① Results observed under transmission electron microscope: The regenerated myelinated nerve fibers in the group of acellular nerve grafts with Schwann cells were more even than those in the group of acellular nerve grafts, the number of myelinated nerve fibers and thickness of myelin sheath were close to those in the allografts group (P > 0.05), but significantly different from those in the group of acellular nerve grafts (P < 0.05). ② Results observed under scanning electron microscope: A great amount of Schwann cells with two polars were observed in the group of grafts with Schwann cells, the feature of cultured Schwann cells showed shoulder by shoulder, head to head. ③ The number of myelinated nerve fibers and thickness of myelin sheath analyzed by Mias-1000 imaging system in the group of acellular nerve grafts with Schwann cells were close to those in the autografts group (P > 0.05), but significantly different from those in the group of acellular nerve grafts (P < 0.05). CONCLUSION: Host axonal regeneration is significantly increased after implant of acellular nerve grafts. 17Probing R-parity Violating Interactions via pp^-→eμ+X Channel on Tevatron显示文摘We investigate the lepton flavor violation processes p(p) → eμ + X induced by R-parity violating interactions at the Tevatron hadron collider. The theoretical calculation and Monte Carlo simulation demonstrate that with a set of suitable cuts on experimental observables, one might be capable to reduce the standard model physical background to a controllable level so that the signals of R-parity violating interactions could be detected distinctively. Furthermore, clear sneutrino information can be abstracted from the purified event sample where other SUSY scalar quark 'pollution' is heavily suppressed. 19Annual Variation of Water Quality in Aquaculture Ponds for Apostichopus japonicus显示文摘By measuring water temperature,salinity,pH,dissolved oxygen,nitrite and ammonia nitrogen,the annual variation regularity of water quality in the aquaculture ponds for sea cucumbers Apostichopus japonicus in Dalian were studied.Results showed that the physicochemical indicators kept changing in the year,and the various ranges of water temperature,salinity,pH,dissolved oxygen,nitrite,and ammonia nitrogen were 6.38-27.27℃,29.20‰-33.78‰,7.86-8.38,5.19-13.36 mg/L,0.0037-0.0090 mg/L and 0.0171-0.0908 mg/L,respectively.Analysis indicated that there was an extremely significant negative correlation between water temperature and dissolved oxygen(P<0.01),an extremely significant positive correlation between water temperature and ammonia nitrogen(P<0.01),a significant negative correlation between salinity and nitrite(P<0.05),a significant negative correlation between dissolved oxygen and nitrite(P<0.05),and an extremely significant negative correlation between dissolved oxygen and ammonia nitrogen(P<0.01)in the aquaculture ponds for sea cucumber in Dalian.Research showed that the water quality of aquaculture ponds kept fluctuating all the time during the tested period,there were complicated and close relationships among physicochemical indicators,and a slight variation of external factors would bring consecutive reactions to different indicators in the water quality of aquaculture ponds.A reference for the production and management of sea cucumbers could be provided from the results.Yaoming LUO Yuan CHENG Duohui LI Zhiying GAO Luping WANG Jiangyi WU Dawei JIANG 2019Asian Agricultural Research2019,11,11:1
