3Ciliary parathyroid hormone signaling activates transforming growth factor-βto maintain intervertebral disc homeostasis during aging显示文摘Degenerative disc disease(DDD) is associated with intervertebral disc degeneration of spinal instability. Here, we report that the cilia of nucleus pulposus(NP) cells mediate mechanotransduction to maintain anabolic activity in the discs. We found that mechanical stress promotes transport of parathyroid hormone 1 receptor(PTH1 R) to the cilia and enhances parathyroid hormone(PTH) signaling in NP cells. PTH induces transcription of integrin α_vβ_6 to activate the transforming growth factor(TGF)-β-connective tissue growth factor(CCN2)-matrix proteins signaling cascade. Intermittent injection of PTH(iPTH) effectively attenuates disc degeneration of aged mice by direct signaling through NP cells, specifically improving intervertebral disc height and volume by increasing levels of TGF-β activity, CCN2, and aggrecan. PTH1 R is expressed in both mouse and human NP cells. Importantly,knockout PTH1 R or cilia in the NP cells results in significant disc degeneration and blunts the effect of PTH on attenuation of aged discs. 4Green fluorescent protein (GFP) transgenic pig produced by somatic cell nuclear transfer显示文摘Transgenic somatic cell nuclear transfer is a very promising route for producing transgenic farm ani-mals. Research on GFP transgenic pigs can provide useful information for breeding transgenic pigs, human disease models and human organ xenotransplantation. In this study, a liposomal transfection system was screened and transgenic embryos were reconstructed by nuclear transfer of GFP positive cells into enucleated in vitro matured oocytes. The development of reconstructed embryos both in vitro and in vivo was observed, and GFP expression was determined. The results showed that porcine fe-tal-derived fibroblast cells cultured with 4.0 μL/mL liposome and 1.6 μg/mL plasmid DNA for 6 h re-sulted in the highest transfection rate (3.6%). The percentage of GFP reconstructed embryos that de-veloped in vitro to the blastocyst stage was 10%. Of those the GFP positive percentage was 48%. Re-constructed transgenic embryos were transferred to 10 recipients. 5 of them were pregnant, and 3 de-livered 6 cloned piglets in which 4 piglets were transgenic for the GFP as verified by both GFP protein expression and GFP DNA sequence analysis. The percentage of reconstructed embryos that resulted in cloned piglets was 1.0%; while the percentage of piglets that were transgenic was 0.7%. 620 Years Advances in Preparation of Potassium Salts from Potassic Rocks:A Review显示文摘Long-term research on key techniques of clean utilization of potassic rocks from over twenty localities has been performed to settle the shortage of soluble potassium resources in China. The results show that K-feldspar as the principal mineral enriched in potassium could be decomposed in the media of Ca(OH)_2, NaOH, KOH-H_2O solution via hydrothermal treatment, into tobermorite, hydroxylcancrinite, and kalsilite respectively. By further processing, these compounds are feasible for being as slow-release carrier of potassium nitrate, extracting alumina, and preparing farm-oriented fertilizers of potassium sulphate and nitrate. Correspondingly, the filtrate is KOH,(Na, K)_2SiO_3, and K_2SiO_3 solution, from which potassium carbonate, sulphate, nitrate, and phosphate could be easily fabricated. As NaO H and KOH are recycled in the processing chains by causticizing sodium and/or potassium metasilicate solutions, the hydrothermal alkaline techniques as developed in this research have several advantages as lower consumption of disposable mineral resources and energy, maximized utilization of potassic mineral resources, as well as clean productions etc. Based on the approaches presented in this paper, the technical system of efficiently utilizing insoluble potassium resources has been established. 7Effect of river plume on phytoplankton community structure in Zhujiang River estuary显示文摘To examine the phytoplankton assemblages and the effect of diluted waters on them,a research cruise was conducted from July 19 to August 7,2015 in the Zhujiang(Pearl)River estuary in the northern South China Sea(21°N–23.5°N,111°E–117°E).Samples were collected from 65 stations including one for time-series sampling.A total of 212 phytoplankton taxa were identified from 61 genera belonging to 4 phyla.Among them,122 species identified from 42 genera of Bacillariophyta and 83 species from 15 genera of Pyrrophyta.Chain-forming diatoms dominated the phytoplankton community where Pseudonitzschia delicatissima,Guinardia striata,Thalassionema nitzschioides,and P.pungens comprised about 52%of the total abundance.However,higher cell abundances concentrated on both sides of the estuary,because of low salinity and high nutrients brought by diluted water.In addition,Canonical Correspondence Analysis revealed that salinity and dissolved inorganic nitrogen shaped the species composition in the study area.Furthermore,the Jaccard similarity index showed prevailing high similarity in the distribution of species in low-salinity diluted waters,and the Bray-Curtis similarity depicted distinguished grouping for phytoplankton assemblages along the salinity gradient.However,phytoplankton diel vertical cycles showed maximum abundance occurred at 2:00 am,which was mainly contributed by benthic phytoplankton species Pseudo-nitzschia spp.and T.nitzschioides.Qi ZHONG Bing XUE Md Abu NOMAN Yuqiu WEI Haijiao LIU Hongbin LIU Liping ZHENG Hongmei JING Jun SUN 2021Journal of Oceanology and Limnology2021,39,2:2

11Highly selective metal recovery from spent lithium-ion batteries through stoichiometric hydrogen ion replacement显示文摘Spent lithium-ion battery recycling has attracted significant attention because of its importance in regard to the environment and resource importance.Traditional hydrometallurgical methods usually leach all valuable metals and subsequently extract target meals to prepare corresponding materials.However,Li recovery in these processes requires lengthy operational procedures,and the recovery efficiency is low.In this research,we demonstrate a method to selectively recover lithium before the leaching of other elements by introducing a hydrothermal treatment.Approximately 90%of Li is leached from high-Ni layered oxide cathode powders,while consuming a nearly stoichiometric amount of hydrogen ions.With this selective recovery of Li,the transition metals remain as solid residue hydroxides or oxides.Furthermore,the extraction of Li is found to be highly dependent on the content of transition metals in the cathode materials.A high leaching selectivity of Li(>98%)and nearly 95%leaching efficiency of Li can be reached with LiNi_(0.8)Co_(0.1)Mn_(0.1)O_(2).In this case,both the energy and material consumption during the proposed Li recovery is significantly decreased compared to traditional methods;furthermore,the proposed method makes full use of H+to leach Li+.This research is expected to provide new understanding for selectively recovering metal from secondary resources.Weiguang Lv Xiaohong Zheng Li Li Hongbin Cao Yi Zhang Renjie Chen Hancheng Ou Fei Kang Zhi Sun 2021Frontiers of Chemical Science and Engineering2021,15,5:1

17Honeycomb-like polyaniline for flexible and folding all-solid-state supercapacitors显示文摘Porous polyaniline (PANI) was prepared through an efficient and costeffective method by polymerization of aniline in the NaCl solution at room temperature. The resulting PANI provided large surface area due to its highly porous structure and the intercrossed nanorod, resulting in good electrochemical performance. The porous PANI electrodes showed a high specific capacitance of 480 F·g^-1, 3 times greater than that of PANI without using the NaCl solution. We also make chemically crosslinked hydrogel film for hydrogel polymer electrolyte as well as the flexible supercapacitors (SCs) with PANI. The specific capacitance of the device was 234 F·g^-1 at the current density of 1 A·g^-1. 18Synthetic vitamin K analogs inhibit inflammation by targeting the NLRP3 inflammasome显示文摘Vitamin K refers to a group of structurally similar vitamins that are essential for proper blood coagulation,as well as bone and cardiovascular health.Previous studies have indicated that vitamin K may also have anti-inflammatory properties,although the underlying mechanisms of its anti-inflammatory effects remain unclear.The NLRP3 inflammasome is a multiprotein complex,and its activation leads to IL-1βand IL-18 secretion and contributes to the pathogenesis of various human inflammatory diseases.Here,we show that synthetic vitamins K3 and K4 are selective,potent inhibitors of the NLRP3 inflammasome and specifically block the interaction between NLRP3 and ASC,thereby inhibiting NLRP3 inflammasome assembly.Moreover,we show that treatment with vitamin K3 or K4 attenuates the severity of inflammation in a mouse model of peritonitis.Our results demonstrate that vitamins K3 and K4 exert their anti-inflammatory effects by inhibiting NLRP3 inflammasome activation and indicate that vitamin K supplementation may be a treatment option for NLRP3-associated inflammatory diseases.Xicui Zheng Yingting Hou Hongbin He Yun Chen Rongbin Zhou Xiaqiong Wang Tao Gong Wei Jiang 2021Cellular & Molecular Immunology2021,18,10:1

20Investigating the function of Akt by tet-off inducible expression system显示文摘A tet-off inducible cell line named BBT derived from BA/F3β cell line was constructed and the effect of this inducible expression system was significant when detected by tet-off responded luciferase reporter gene assay. Then tet-off responded Akt expression plasmid was transfected into BBT cells, and the stable cell lines were screened. The result of Northern blot showed that the expression of akt was significantly inducible. The clone with the best inducible effect was selected and named BBA for investigating the function of Akt. We found that Akt could significantly inhibit zinc-induced decrease of cell viability when assayed by MTT method.
