| 2 | Numerical simulation for the coupling effect of local atmospheric circulations over the area of Beijing,Tianjin and Hebei Province显示文摘Using the model system MM5.V3 and multi-layer grid nesting technique, we have done a multi-scale numerical simulation over the area of Beijing, Tianjin and Hebei Province to analyze the temperature and wind field there and study its local circulations. The results show a coupling effect of Urban Heat Island Circulation (UHIC), Mountain Valley Breeze (MVB) and Sea Land Breeze (SLB) occurs in this area when the synoptic system is weak. The SLB can penetrate deep into the mainland for about 200 km when it is blooming. MVB can extend to south and cover almost the whole plain area in Beijing. Both MVB and SLB are diurnal periodical; meanwhile the phase of MVB drops behind that of SLB for about six hours. As a local circulation, the UHIC weakens the two circulations above, and it also has a diurnal period. As a result, the coupling effect of circulations reveals not only different features in spring-summer period and autumn-winter period in a year but also the difference between early morn- ing to noonday and afternoon to night in a day. We noted the diffusion of contamination over the area around Beijing, and found the steady presence of a transport routine of contamination over North-China throughout the year caused by the Coupling Effect mentioned above. | 8 | Targeting a novel inducible GPX4 alternative isoform to alleviate ferroptosis and treat metabolic-associated fatty liver disease显示文摘Metabolic-associated fatty liver disease(MAFLD),which is previously known as non-alcoholic fatty liver disease(NAFLD),represents a major health concern worldwide with limited therapy.Here,we provide evidence that ferroptosis,a novel form of regulated cell death characterized by iron-driven lipid peroxidation,was comprehensively activated in liver tissues from MAFLD patients.The canonical-GPX4(cGPX4),which is the most important negative controller of ferroptosis,is downregulated at protein but not mRNA level.Interestingly,a non-canonical GPX4 transcript-variant is induced(inducible-GPX4,iGPX4)in MAFLD condition.The high fat-fructose/sucrose diet(HFFD)and methionine/choline-deficient diet(MCD)-induced MAFLD pathologies,including hepatocellular ballooning,steatohepatitis andfibrosis,were attenuated and aggravated,respectively,in cGPX4-and iGPX4-knockin mice.cGPX4 and iGPX4 isoforms also displayed opposing effects on oxidative stress and ferroptosis in hepatocytes.Knockdown of iGPX4 by siRNA alleviated lipid stress,ferroptosis and cell injury.Mechanistically,the triggered iGPX4 interacts with cGPX4 to facilitate the transformation of cGPX4 from enzymatic-active monomer to enzymatic-inactive oligomers upon lipid stress,and thus promotes ferroptosis.Co-immunoprecipitation and nano LC–MS/MS analyses confirmed the interaction between iGPX4 and cGPX4.Our results reveal a detrimental role of non-canonical GPX4 isoform in ferroptosis,and indicate selectively targeting iGPX4 may be a promising therapeutic strategy for MAFLD. | Jie Tong Dongjie Li Hongbo Meng Diyang Sun Xiuting Lan Min Ni Jiawei Ma Feiyan Zeng Sijia Sun Jiangtao Fu Guoqiang Li Qingxin Ji Guoyan Zhang Qirui Shen Yuanyuan Wang Jiahui Zhu Yi Zhao Xujie Wang Yi Liu Shenxi Ouyang Chunquan Sheng Fuming Shen Pei Wang | 2022 | Acta Pharmaceutica Sinica B2022,12,9: | 3 |
| 9 | An enrichment model using regular health examination data for early detection of colorectal cancer显示文摘Objective: Challenges remain in current practices of colorectal cancer(CRC) screening, such as low compliance,low specificities and expensive cost. This study aimed to identify high-risk groups for CRC from the general population using regular health examination data.Methods: The study population consist of more than 7,000 CRC cases and more than 140,000 controls. Using regular health examination data, a model detecting CRC cases was derived by the classification and regression trees(CART) algorithm. Receiver operating characteristic(ROC) curve was applied to evaluate the performance of models. The robustness and generalization of the CART model were validated by independent datasets. In addition, the effectiveness of CART-based screening was compared with stool-based screening.Results: After data quality control, 4,647 CRC cases and 133,898 controls free of colorectal neoplasms were used for downstream analysis. The final CART model based on four biomarkers(age, albumin, hematocrit and percent lymphocytes) was constructed. In the test set, the area under ROC curve(AUC) of the CART model was 0.88 [95%confidence interval(95% CI), 0.87-0.90] for detecting CRC. At the cutoff yielding 99.0% specificity, this model’s sensitivity was 62.2%(95% CI, 58.1%-66.2%), thereby achieving a 63-fold enrichment of CRC cases. We validated the robustness of the method across subsets of test set with diverse CRC incidences, aging rates, genders ratio, distributions of tumor stages and locations, and data sources. Importantly, CART-based screening had the higher positive predictive value(1.6%) than fecal immunochemical test(0.3%).Conclusions: As an alternative approach for the early detection of CRC, this study provides a low-cost method using regular health examination data to identify high-risk individuals for CRC for further examinations. | 11 | Removal of F- and organic matter from coking wastewater by coupling dosing FeCl3 and AlCl3显示文摘Coagulation and precipitation is a widely applied method to remove F-from wastewater.In this work,the effect of coagulation on the removal of F-and organic matter from coking wastewater was studied using Al Cl3and Fe Cl3as compound coagulants.The removal rates of F-and organic matter under different coagulant doses and p H conditions were investigated.The results show that the highest removal rates of F-by Al Cl3and Fe Cl3are 94.4%and 25.4%,respectively;when the dosage is 10 mmol/L,the TOC removal rates of Fe Cl3and Al Cl3reach 20.4%and 34.7%,respectively.Therefore,the removal rate of F-by Al Cl3is higher than that of Fe Cl3,but the removal rate of organic matter by Fe Cl3is relatively higher.The addition of Ca2+can promote the removal of F-,but the removal rate of organic matter decreases.In addition,by investigating the effects of different p H and Fe–Al ratio on the removal rate,the removal effect of adding Fe Cl3and Al Cl3at the same time was discussed.The results show that the most suitable working condition for the removal of organic matter and F-is that the p H is 6.5 and the molar ratio of Al/Fe is 8:2.Overall,the removal mechanism of F-and organic matter in coking wastewater by Fe Cl3and Al Cl3was explored in this study.The experimental results can provide reference for the advanced treatment of coking wastewater. | Shuo Li Mengjie Liu Fuming Meng Xia Hu Wenzheng Yu | 2021 | Journal of Environmental Sciences2021,33,12: | 2 |
