1Long Term Culture of Human Kidney Proximal Tubule Epithelial Cells Maintains Lineage Functions and Serves as an Ex vivo Model for Coronavirus Associated Kidney Injury显示文摘The mechanism of how SARS-CoV-2 causes severe multi-organ failure is largely unknown. Acute kidney injury(AKI) is one of the frequent organ damage in severe COVID-19 patients. Previous studies have shown that human renal tubule cells could be the potential host cells targeted by SARS-CoV-2. Traditional cancer cell lines or immortalized cell lines are genetically and phenotypically different from host cells. Animal models are widely used, but often fail to reflect a physiological and pathogenic status because of species tropisms. There is an unmet need for normal human epithelial cells for disease modeling. In this study, we successfully established long term cultures of normal human kidney proximal tubule epithelial cells(KPTECs) in 2 D and 3 D culture systems using conditional reprogramming(CR) and organoids techniques.These cells had the ability to differentiate and repair DNA damage, and showed no transforming property. Importantly, the CR KPTECs maintained lineage function with expression of specific transporters(SLC34 A3 and cubilin). They also expressed angiotensin-converting enzyme 2(ACE2), a receptor for SARS-CoV and SARS-CoV-2. In contrast, cancer cell line did not express endogenous SLC34 A3, cubilin and ACE2. Very interestingly, ACE2 expression was around twofold higher in 3 D organoids culture compared to that in 2 D CR culture condition. Pseudovirion assays demonstrated that SARS-CoV spike(S) protein was able to enter CR cells with luciferase reporter. 2Removal of roosters alters the domestic phenotype and microbial and genetic profile of hens显示文摘Hens are raised apart from roosters in modern poultry production, a substantial change from their natural social structure. We compared productivity, injuries, behavior, physiology, microbiome and transcriptome of hens housed with(R+) or without(R–)roosters to quantify the effects of this change in social structure. Hens were raised free-range from 70 to 280 days when 30 birds per treatment were assigned to battery cages until Day 315(R+C vs. R–C), while 30 birds per treatment remained in free-range pens(R+F vs. R–F). Response to a novel environment and object, behavioral time budgets, cecum microbiome, blood composition and transcriptomic sequencing of thigh muscle and spleen were analyzed. Hens housed without roosters showed better survival, consumed less food, produced more eggs and had better feed conversion. R+F hens clustered around the rooster and were less mobile in the novel environment and object tests. R+F hens displayed the richest microbiome, and the presence of roosters resulted in differentially expressed genes related to muscle development, cellular processes, environmental information processing and immune function. 4Dynamic changes of renal cortical blood perfusion before and after percutaneous transluminal renal artery stenting in patients with severe atherosclerotic renal artery stenosis显示文摘Background:This study aims to observe the dynamic changes of renal artery(RA)disease and cortical blood perfusion(CBP)evaluated by contrast-enhanced ultrasound(CEUS)after percutaneous transluminal renal artery stenting(PTRAS)in patients with severe atherosclerotic renal artery stenosis(ARAS)and to analyze the relationship between CBP and prognosis.Methods:This was a single-center retrospective cohort study.A total of 98 patients with unilateral severe ARAS after successful PTRAS in Beijing Hospital from September 2017 to September 2020 were included.According to renal glomerular filtration rate(GFR)detected by radionuclide imaging at 12 months after PTRAS,all patients were divided into the poor prognosis group(n=21,GFR decreased by≥20%compared with baseline)and the control group(n=77,GFR decreased by<20%or improved compared with baseline).Renal artery stenosis was diagnosed by digital subtraction angiography,and renal CBP was evaluated by CEUS using TomTec Imaging Systems(Germany)before PTRAS,at 6 months and 12 months after discharge.The receiver operating characteristic(ROC)curve with area under the curve(AUC)was used to analyze the predictive value of CBP parameters,including area under ascending curve(AUC1),area under the descending curve(AUC2),rising time(RT),time to peak intensity(TTP),maximum intensity(IMAX),and mean transit time(MTT)for poor prognosis.Results:Among the 98 patients,there were 52 males(53.1%),aged 55–74 years old,with an average age of 62.1±8.7 years,and an average artery stenosis of 82.3±12.9%.The poor prognosis group was associated with significantly increased incidence of diabetes(76.2%vs.41.6%),and lower levels of GFR of the stenotic kidney(21.8 mL/min vs.25.0 mL/min)and total GFR(57.6 mL/min vs.63.7 mL/min)(all P<0.05),compared with the control group(P<0.05).In addition,the rate of RA restenosis was significantly higher in the poor prognosis group than in the control group(9.5%vs.0,χ^(2)=9.462,P=0.002).Compared with the control group,the poor prognosis group was associated with significantly decreased baseline AUC1 and AUC2,and extended duration of TTP and MTT(P<0.05).At 6 months and 12 months of follow-up,patients in the control group were associated with markedly increased AUC1,AUC2,and IMAX,and shorter duration of RT and MTT(P<0.05).The ROC curve showed that the predictive values of AUC1,AUC2,RT,TTP,IMAX,and MTT for poor prognosis were 0.812(95%CI:0.698–0.945),0.752(95%CI:0.591–0.957),0.724(95%CI:0.569–0.961),0.720(95%CI:0.522–0.993),0.693(95%CI:0.507–0.947),and 0.786(95%CI:0.631–0.979),respectively.Conclusions:Preoperative renal CBP in severe ARAS patients with poor prognosis is significantly reduced,and does not show significant improvement after stent treatment over the first year of follow-up.The parameter AUC1 may be a good predictor for renal dysfunction after PTRAS in severe ARAS patients.Trial Registration:ChiCTR.org.cn,ChiCTR1800016252.Na Ma Yan Li Siyu Wang Mengpu Li Yongjun Li Hu Ai Hui Zhu Yang Wang Fajin Guo Junhong Ren 2022Chinese Medical Journal2022,,13:1

6Feasibility and physics potential of detecting ^(8)B solar neutrinos at JUNO显示文摘The Jiangmen Underground Neutrino Observatory(JUNO)features a 20 kt multi-purpose underground liquid scintillator sphere as its main detector.Some of JUNO's features make it an excellent location for^8B solar neutrino measurements,such as its low-energy threshold,high energy resolution compared with water Cherenkov detectors,and much larger target mass compared with previous liquid scintillator detectors.In this paper,we present a comprehensive assessment of JUNO's potential for detecting^8B solar neutrinos via the neutrino-electron elastic scattering process.A reduced 2 MeV threshold for the recoil electron energy is found to be achievable,assuming that the intrinsic radioactive background^(238)U and^(232)Th in the liquid scintillator can be controlled to 10^(-17)g/g.With ten years of data acquisition,approximately 60,000 signal and 30,000 background events are expected.This large sample will enable an examination of the distortion of the recoil electron spectrum that is dominated by the neutrino flavor transformation in the dense solar matter,which will shed new light on the inconsistency between the measured electron spectra and the predictions of the standard three-flavor neutrino oscillation framework.IfDelta m^(2)_(21)=4.8times10^(-5);(7.5times10^(-5))eV^(2),JUNO can provide evidence of neutrino oscillation in the Earth at approximately the 3sigma(2sigma)level by measuring the non-zero signal rate variation with respect to the solar zenith angle.Moreover,JUNO can simultaneously measureDelta m^2_(21)using^8B solar neutrinos to a precision of 20% or better,depending on the central value,and to sub-percent precision using reactor antineutrinos.A comparison of these two measurements from the same detector will help understand the current mild inconsistency between the value of Delta m^2_(21)reported by solar neutrino experiments and the KamLAND 7A review of membrane distillation enhancement via thermal management and molecular transport through nanomaterial-based membranes显示文摘Low-energy,high-efficiency desalination techniques are important because of the critical water-energy nexus.In particular,membrane distillation has great potential to harness low-grade and renewable energy sources.Composite membranes using micro-and nanomaterials have new capabilities and characteristics.This review focuses on the most recent developments and potential capabilities in membrane distillation systems from the perspective of nanomaterial enhancements,thermal management,and water transport.Self-heating nanomaterial membrane distillations using permeable composite membranes with significant photothermal or Joule-heating conversion capabilities enable macroscale thermal management and mitigate temperature polarization effects.Modifying the membrane structure and its interaction with water can accelerate water transport and evaporation,improving distillation at the microscopic level.This is expected to provide directions for the fabrication and manipulation of novel micro-and nanocomposite membranes for distillation processes at various levels,and enhance their applications.HUANG Jian HE YuRong HUI SiYu WANG MingHao TANG TianQi 2023Science China(Technological Sciences)2023,66,6:0
8Interface engineering of Fe-Sn-Co sulfide/oxyhydroxide heterostructural electrocatalyst for synergistic water splitting显示文摘To realize large-scale hydrogen production by electrolysis of water,it is essential to develop non-precious metal catalysts for oxygen evolution reaction(OER)and hydrogen evolution reaction(HER).Here,we fabricate Sn-,Fe-,and Co-based sulfide/oxyhydroxide heterostructural catalyst on nickel foam(FeSnCo_(0.2)S_(x)O_(y)/NF)by solvothermal method.The FeSnCo_(0.2)S_(x)O_(y)/NF requires low overpotentials of 48 and 186 mV at 10 mA·cm^(-2),respectively,for HER and OER.When it is assembled into an electrolytic cell as a bifunctional electrocatalyst,it only needs 1.54 V to reach 10 mA·cm^(-2),far better than IrO_(2)||Pt/C electrolyzer.The formation of sulfide/hydroxide heterostructural interfaces improves the electron transfer and reduces the reaction energy barrier,thus promoting the electrocatalytic processes.Siyu Chen Ting Zhang Jingyi Han Hui Qi Shihui Jiao Changmin Hou Jingqi Guan 2024Nano Research Energy2024,3,2:0
9REEP1 Preserves Motor Function in SOD1^(G93A) Mice by Improving Mitochondrial Function via Interaction with NDUFA4显示文摘A decline in the activities of oxidative phosphorylation(OXPHOS)complexes has been consistently reported in amyotrophic lateral sclerosis(ALS)patients and animal models of ALS,although the underlying molecular mechanisms are still elusive.Here,we report that receptor expression enhancing protein 1(REEP1)acts as an important regulator of complex IV assembly,which is pivotal to preserving motor neurons in SOD1^(G93A) mice.We found the expression of REEP1 was greatly reduced in transgenic SOD1^(G93A) mice with ALS.Moreover,forced expression of REEP1 in the spinal cord extended the lifespan,decelerated symptom progression,and improved the motor performance of SOD1^(G93A) mice.The neuromuscular synaptic loss,gliosis,and even motor neuron loss in SOD1^(G93A) mice were alleviated by increased REEP1 through augmentation of mitochondrial function.Mechanistically,REEP1 associates with NDUFA4,and plays an important role in preserving the integrity of mitochondrial complex IV.Our findings offer insights into the pathogenic mechanism of REEP1 deficiency in neurodegenerative diseases and suggest a new therapeutic target for ALS.Siyue Qin Pan You Hui Yu Bo Su 2023Neuroscience Bulletin2023,39,6:0
10Surface reconstruction and structural transformation of twodimensional Ni-Fe MOFs for oxygen evolution in seawater media显示文摘As a four-electron transfer reaction,oxygen evolution reaction(OER)is limited by large overpotential and slow kinetics.Here,we in-situ synthesized two-dimensional(2D)Ni-Fe metal-organic framework nanosheets on nickel foam(NixFe-TPA/NF,TPA=terephthalic acid)for oxygen evolution in alkaline and alkaline seawater electrolytes.In 1 M KOH,Ni3Fe-TPA/NF shows a low overpotential(η10)of 189 mV at 10 mA·cm^(-2) and an ultra-low overpotential of only 260 mV at 500 mA·cm^(-2).In alkaline seawater,Ni3Fe-TPA/NF still provides impressive OER performance,with anη10 of 265 mV.In-situ Raman characterization results show that the phase transition occurs during the OER,and Ni3FeOOH with more oxygen vacancies is in-situ formed,reducing the OER energy barrier.Density functional theory(DFT)reveals that the synergy between Ni and Fe reduces the energy barrier and accelerates the rate-determining step.In addition,the ultra-thin 2D sheet structure and the close combination of Ni3FeOOH and highly conductive NF support ensure the high catalytic OER activity.Therefore,the surface reconstruction and structural modification strategy can be used to design and prepare high-performance OER electrocatalysts for energy-related applications.Liyuan Xiao Xue Bai Jingyi Han Tianmi Tang Siyu Chen Hui Qi Changmin Hou Fuquan Bai Zhenlu Wang Jingqi Guan 2024Nano Research2024,17,4:0
11Saturation evaluation of microporous low resistivity carbonate oil pays in Rub Al Khali Basin in the Middle East显示文摘To solve the problem that it is difficult to identify carbonate low resistivity pays(LRPs) by conventional logging methods in the Rub Al Khali Basin, the Middle East, the variation of fluid distribution and rock conductivity during displacement were analyzed by displacement resistivity experiments simulating the process of reservoir formation and production, together with the data from thin sections, mercury injection and nuclear magnetic resonance experiments. In combination with geological understandings, the genetic mechanisms of LRPs were revealed, then the saturation interpretation model was selected, the variation laws and distribution range of the model parameters were defined, and finally an updated comprehensive saturation interpretation technique for the LRPs has been proposed. In the study area, the LRPs have resistivity values of less than 1 Ω·m, similar to or even slightly lower than that of the water layers. Geological research reveals that the LRPs were developed in low-energy depositional environment and their reservoir spaces are controlled by micro-scale pore throats, with an average radius of less than 0.7 μm, so they are typical microporous LRPs. Different from LRPs of sandstone and mudstone, they have less tortuous conductive paths than conventional reservoirs, and thus lower resistivity value under the same saturation. Archie’s formula is applicable to the saturation interpretation of LRPs with a cementation index value of 1.77-1.93 and a saturation index value of 1.82-2.03 that are 0.2-0.4 lower than conventional reservoirs respectively. By using interpretation parameters determined by classification statistics of petrophysical groups(PGs), oil saturations of the LRPs were calculated at bout 30%-50%,15% higher than the results by conventional methods, and basically consistent with the data of Dean Stark, RST, oil testing and production. 14Distribution characteristics of delta reservoirs reshaped by bottom currents:A case study from the second member of the Yinggehai Formation in the DF1-1 gas field,Yinggehai Basin,South China Sea显示文摘The Dongfang1-1 gas field(DF1-1)in the Yinggehai Basin is currently the largest offshore self-developed gas field in China and is rich in oil and gas resources.The second member of the Pliocene Yinggehai Formation(YGHF)is the main gas-producing formation and is composed of various sedimentary types;however,a clear understanding of the sedimentary types and development patterns is lacking.Here,typical lithofacies,logging facies and seismic facies types and characteristics of the YGHF are identified based on high-precision 3D seismic data combined with drilling,logging,analysis and testing data.Based on 3D seismic interpretation and attribute analysis,the origin of high-amplitude reflections is clarified,and the main types and evolution characteristics of sedimentary facies are identified.Taking gas formation upper II(IIU)as an example,the plane distribution of the delta front and bottom current channel is determined;finally,a comprehensive sedimentary model of the YGHF second member is established.This second member is a shallowly buried“bright spot”gas reservoir with weak compaction.The velocity of sandstone is slightly lower than that of mudstone,and the reflection has medium amplitude when there is no gas.The velocity of sandstone decreases considerably after gas accumulation,resulting in an increase in the wave impedance difference and high-amplitude(bright spot)reflection between sandstone and mudstone;the range of high amplitudes is consistent with that of gas-bearing traps.The distribution of gas reservoirs is obviously controlled by dome-shaped diapir structural traps,and diapir faults are channels through which natural gas from underlying Miocene source rocks can enter traps.The study area is a delta front deposit developed on a shallow sea shelf.The lithologies of the reservoir are mainly composed of very fine sand and coarse silt,and a variety of sedimentary structural types reflect a shallow sea delta environment;upward thickening funnel type,strong toothed bell type and toothed funnel type logging facies are developed.In total,4 stages of delta front sand bodies(corresponding to progradational reflection seismic facies)derived from the Red River and Blue River in Vietnam have developed in the second member of the YGHF;these sand bodies are dated to 1.5 Ma and correspond to four gas formations.During sedimentation,many bottom current channels(corresponding to channel fill seismic facies)formed,which interacted with the superposed progradational reflections.When the provenance supply was strong in the northwest,the area was dominated by a large set of delta front deposits.In the period of relative sea level rise,surface bottom currents parallel to the coastline were dominant,and undercutting erosion was obvious,forming multistage superimposed erosion troughs.Three large bottom current channels that developed in the late sedimentary period of gas formation IIU are the most typical.Shuo Chen Renhai Pu Huiqiong Li Hongjun Qu Tianyu Ji Siyu Su Yunwen Guan Hui Zhang 2022Acta Oceanologica Sinica2022,41,9:0
