Vascular endothelial dysfunction and pharmacological treatment显示文摘The endothelium exerts multiple actions involving regulation of vascular permeability and tone, coagulation and fibrinolysis, inflammatory and immunological reactions and cell growth. Alterations of one or more such actions may cause vascular endothelial dysfunction. Different risk factors such as hypercholesterolemia, homocystinemia, hyperglycemia, hypertension, smo-king, inflammation, and aging contribute to the development of endothelial dysfunction. Mechanisms underlying endothelial dysfunction are multiple, including impaired endothelium-derived vasodilators, enhanced endothelium-derived vasoconstrictors, over production of reactive oxygen species and reactive nitrogen species, activation of inflammatory and immune reactions, and imbalance of coagulation and fibrinolysis. Endothelial dysfunction occurs in many cardiovascular diseases, which involves different mechanisms, depending on specific risk factors affecting the disease. Among these mechanisms, a reduction in nitric oxide(NO) bioavailability plays a central role in the development of endothelial dysfunction because NO exerts diverse physiological actions, including vasodilation, anti-inflammation, antiplatelet, antiproliferation and antimigration. Experimental and clinical studies have demonstrated that a variety of currently used or investigational drugs, such as angiotensin-converting enzyme inhibitors, angiotensin AT1 receptors blockers, angiotensin-(1-7), antioxidants, beta-blockers, calcium channel blockers, endothelial NO synthase enhancers, phosphodiesterase 5 inhibitors, sphingosine-1-phosphate and statins, exert endothelial protective effects. Due to the difference in mechanisms of action, these drugs need to be used according to specific mechanisms underlying endothelial dysfunction of the disease. | Jin Bo Su | 2015 | World Journal of Cardiology2015,7,11: | 24
Failure mechanism and stability control of a large section of very soft roadway surrounding rock shear slip显示文摘The measured data and simulation test phenomenon of surrounding rock deformation and failure at the project site indicate that shear failure which firstly occurs in surrounding rock, block slip and second shear failure are the root cause of deformation and damage of supporting structure of the surrounding rock at a large scale. We derived limit load of surrounding rock shear slip failure and reasonable support resistance of given load by means of shear slip line field theory, discussed the main factors which influence the limit load of surrounding rock. Shear slip line field and limit load of circular tunnel surrounding rock were obtained by means of physical simulation test, which agreed well with the theoretical analysis results. Based on the theoretical analysis and physical simulation test, the cause deformation and failure at large scale of Xinshanghai No. 1 coal mine big section ingate was analyzed, and the shear failure resistance and block slip in surrounding rock were proposed as the core technical supporting ideas. Proper range of supporting resistance which came from calculation was suggested. The support scheme which is mainly composed of large grouting anchor, sprayed anchor net support technique and full-face grille concrete finally ended the dilemma of repeated failure and mending of ingate and created critical conditions for smooth production in the coal mine. | Meng Bo Jing Hongwen Chen Kunfu Su Haijian | 2013 | International Journal of Mining Science and Technology2013,23,1: | 22

Collagen type Ⅱ suppresses articular chondrocyte hypertrophy and osteoarthritis progression by promoting integrin β1-SMAD1 interaction显示文摘Hypertrophic differentiation is not only the terminal process of endochondral ossification in the growth plate but is also an important pathological change in osteoarthritic cartilage.Collagen type II(COL2A1)was previously considered to be only a structural component of the cartilage matrix, but recently,it has been revealed to be an extracellular signaling molecule that can significantly suppress chondrocyte hypertrophy. However, the mechanisms by which COL2A1 regulates hypertrophic differentiation remain unclear. In our study,a Col2a1 p.Gly1170Ser mutant mouse model was constructed, and Col2a1 loss was demonstrated in homozygotes.Loss of Col2a1 was found to accelerate chondrocyte hypertrophy through the bone morphogenetic protein(BMP)-SMAD1 pathway.Upon interacting with COL2A1,integrinβ1(ITGB1),the major receptor for COL2A1,competed with BMP receptors for binding to SMAD1 and then inhibited SMAD1 activation and nuclear import.COL2A1 could also activate ITGB1-induced ERK1/2 phosphorylation and, through ERK1/2-SMAD1 interaction,it further repressed SMAD1 activation, thus inhibiting BMP-SMAD1-mediated chondrocyte hypertrophy.Moreover,COL2A1 expression was downregulated, while chondrocyte hypertrophic markers and BMP-SMAD1 signaling activity were upregulated in degenerative human articular cartilage. Our study reveals novel mechanisms for the inhibition of chondrocyte hypertrophy by COL2A1 and suggests that the degradation and decrease in COL2A1 might initiate and promote osteoarthritis progression. | Chengjie Lian Xudong Wang Xianjian Qiu Zizhao Wu Bo Gao Lei Liu Guoyan Liang Hang Zhou Xiaoming Yang Yan Peng Anjing Liang Caixia Xu Dongsheng Huang Peiqiang Su | 2019 | Bone Research2019,7,1: | 17

A model and experimental study of phosphate uptake kinetics in algae:Considering surface adsorption and P-stress显示文摘Phosphorus is an important limiting nutrient in many ecosystems.Consequently, there is increasing interest on phosphate uptake and algal growth due to the increasing frequency and magnitude of algal blooms induced by eutrophication. The co-existence of surface adsorbed and intracellular phosphorus pools indicate that phosphate uptake by phytoplankton is,to some extent,a two-stage kinetic process. However, almost all previous uptake models considered the internal uptake stage only and ignored the possible impact of surface adsorption. In this article,a two-stage kinetic uptake model considering both surface adsorption and P-stress on phosphate uptake by algae was constructed and compared to conventional one-stage models, based on experimental data on short-term uptake kinetics of a green algae S.quadricauda.Results indicated that with suitable parameters, the two-stage uptake model not only fit the experimental data better, but also gave more reasonable and realistic explanations to the phosphate uptake process. The results are meaningful as surface-adsorption of phosphate may affect the uptake process of phosphate and assist in understanding realistic phosphate uptake kinetics in phytoplankton. | Bo Yao Beidou Xi Chunming Hu Shouliang Huo Jing Su Hongliang Liu | 2011 | Journal of Environmental Sciences2011,23,2: | 15
Coal–rock interface detection on the basis of image texture features显示文摘Based on the stability and inequality of texture features between coal and rock, this study used the digital image analysis technique to propose a coal–rock interface detection method.By using gray level co-occurrence matrix,twenty-two texture features were extracted from the images of coal and rock.Data dimension of the feature space reduced to four by feature selection, which was according to a separability criterion based on inter-class mean difference and within-class scatter. The experimental results show that the optimized features were effective in improving the separability of the samples and reducing the time complexity of the algorithm. In the optimized low-dimensional feature space, the coal–rock classifer was set up using the fsher discriminant method.Using the 10-fold cross-validation technique, the performance of the classifer was evaluated, and an average recognition rate of 94.12%was obtained. The results of comparative experiments show that the identifcation performance of the proposed method was superior to the texture description method based on gray histogram and gradient histogram. | Sun Jiping Su Bo | 2013 | International Journal of Mining Science and Technology2013,23,5: | 15
Determination of trace level of perchlorate in Antarctic snow and ice by ion chromatography coupled with tandem mass spectrometry using an automated sample on-line preconcentration method显示文摘A new ion chromatography coupled with tandem mass spectrometry(IC-ESI-MS/MS) method, with automated sampling and on-line preconcentration, has been developed for the determination of perchlorate in Antarctic snow and ice at low part-per-trillion(ng/L) levels. To the best of our knowledge, this is the first time that an analytical method is used for the determination of perchlorate in Antarctic snow and ice.The IC-ESI-MS/MS instrumentation consisted of an ICS2000 ion chromatography(IC) system coupled to an API3200 electrospray tandem mass spectrometer(ESI-MS/MS).On-line preconcentration was realized through a six-port injector valve,a TAC-ULP1 concentrator column and an AS auto-sampler.Multiple reaction monitoring(MRM) mode was used to quantify the perchlorate anion. The transition of ^(35)Cl^(16)O_4^-(m/z 98.9) into ^(35)Cl^(16)O_3^-(m/z 82.9) was monitored for quantifying the main analyte, and the transition of ^(37)Cl^(16)O_4^-(m/z 100.9) into ^(37)Cl^(16)O_3^-(m/z 84.9) was monitored for examining a proper isotopic abundance ratio of ^(35)Cl to ^(37)Cl,which was used as a confirmation tool. The limit of detection(LOD) and limit of quantitation(LOQ) for the method was 0.2 ng/ L and 0.5 ng/L,respectively.And this new method exhibited acceptable accuracy and precision for samples at ng/L levels. All the tested snow and ice samples were found to contain measurable amount of perchlorate, ranging from 10 ng/L to 340 ng/L. | Su Jiang Yuan-Sheng Li Bo Sun | 2013 | Chinese Chemical Letters2013,24,4: | 15
Complement C3 activation regulates the production of tRNA-derived fragments Gly-tRFs and promotes alcohol-induced liver injury and steatosis显示文摘Complement is known to play a role in alcoholic fatty liver disease (AFLD), but the underlying mechanisms are poorly understood, thereby constraining the development of a rational approach for therapeutic intervention in the complement system. C3 deficiency has been shown to impart protective effects against ethanol-induced hepatic steatosis and inflammation. Here we demonstrate a protection effect in wild-type mice by treatment with CR2-Crry, a specific inhibitor of C3 activation. The expression of glycine transfer (t) RNA-derived fragments (Gly-tRFs) is upregulated in ethanol-fed mice and inhibition of Gly-tRFs in vivo decreases chronic ethanol feeding-induced hepatosteatosis without affecting inflammation. The expression of Gly-tRF was downregulated in C3-deficient or CR2-Crry-treated mice, but not in C5-deficient mice;Gly-tRF expression was restored by the C3 activation products C3a or Asp (C3a-des-Arg) via the regulation of CYP2E1. Transcriptome profiling of hepatic tissues showed that Gly-tRF inhibitors upregulate the expression of sirtuin1 (Sirt1) and subsequently affect downstream lipogenesis and β-oxidation pathways. Mechanistically, Gly-tRF interacts with AGO3 to downregulate Sirt1 expression via sequence complementarity in the 3′ UTR. Notably, the expression levels of C3d, CYP2E1 and Gly-tRF are upregulated, whereas Sirt1 is decreased in AFLD patients compared to healthy controls. Collectively, our findings suggest that C3 activation products contribute to hepatosteatosis by regulating the expression of Gly-tRF. Complement inhibition at the C3 activation step and treatment with Gly-tRF inhibitors may be potential and precise therapeutic approaches for AFLD. | Fudi Zhong Zhigao Hu Keqing Jiang Biao Lei Zhan Wu Guandou Yuan Hongliang Luo Chunqiang Dong Bo Tang Chaowen Zheng Shuai Yang Yonglian Zeng Zhenya Guo Shuiping Yu Huizhao Su Guo Zhang Xiaoqiang Qiu Stephen Tomlinson Songqing He | 2019 | Cell Research2019,29,7: | 14
Management of entecavir-resistant chronic hepatitis B with adefovir-based combination therapies显示文摘AIM: To evaluate the long-term efficacy adefovir(ADV)-based combination therapies in entecavir(ETV)-resistant chronic hepatitis B(CHB) patients. METHODS: F i fty CHB pat ient s wi t h genotypic resistance to ETV at 13 medical centers in South Korea were included for the analysis. All the patients received rescue therapy with the combination of ADV plus ETV(ADV/ETV,n = 23) or ADV plus lamivudine(LMV)(ADV/LMV,n = 27) for more than 12 mo. Patients were monitored at least every 3-4 mo during ADV-based combination therapy by clinical examination as well as biochemical and virological assessments. Hepatitis B virus(HBV) DNA levels were measured by realtime PCR and logarithmically transformed for analysis. Cumulative rates of virologic response(VR; HBV DNA < 20 IU/m L) were calculated using the Kaplan-Meier method, and the difference was determined by a logrank test. Multivariate logistic regression and Cox proportional hazards models were used to identify independent risk factors significantly associated with short-term and long-term VR,respectively.RESULTS: Baseline median HBV DNA levels were 5.53(2.81-7.63) log10 IU/m L. The most commonly observed ETV genotypic mutation sites were rt184 and rt202. Patients were treated for a median of 27(12-45) mo. Overall, cumulative VR rates at 6,12,24,and 36 mo were 26%,36%,45%,and 68%,respectively. Patients treated with the ADV/ETV combination showed higher cumulative VR rates(35%,43%,65%,and 76%,respectively) than those with the ADV/LAM combination(18%,30%,30%,and 62%,respectively; P = 0.048). In the multivariate analysis, low baseline HBV DNA levels(< 5.2 log10 IU/m L) and initial virologic response at 3 mo(IVR-3; HBV DNA < 3.3 log10 IU/m L after 3 mo) were independent predictive factors for VR. Patients with favorable predictors achieved cumulative VR rates up to 90% at 36 mo. During the same period, the cumulative incidence of virologic breakthrough was as low as 6% in patients with the both favorable predictors.CONCLUSION: If tenofovir is not available,ADV/ETV combination could be considered in ETV-resistant patients with low HBV DNA titers, and may becontinued if IVR-3 is achieved. | Hyoung Su Kim Hyung Joon Yim Myoung Kuk Jang Ji Won Park Sang Jun Suh Yeon Seok Seo Ji Hoon Kim Bo Hyun Kim Sang Jong Park Sae Hwan Lee Sang Gyune Kim Young Seok Kim Jung Il Lee Jin-Woo Lee In Hee Kim Tae Yeob Kim Jin-Wook Kim Sook-Hyang Jeong Young Kul Jung Hana Park Seong Gyu Hwang | 2015 | World Journal of Gastroenterology2015,21,38: | 13
Laser joining of CFRTP to titanium alloy via laser surface texturing显示文摘Grid pattern was textured on Ti-6 Al-4 V alloy(TC4)substrate surface by nanosecond laser system.Laser joining of carbon fiber reinforced thermoplastic composite(CFRTP)to TC4 joints were performed, and the effect of texture grid depth was investigated. The contact angle of molten CFRTP on textured TC4 surface was measured and the tensile-shear force was tested. The fracture surface and interface morphology were observed. The results indicated that the wettability of molten CFRTP on TC4 surface improved remarkably after laser textured TC4.Shear force of CFRTP/TC4 joints was increased by 156%after laser textured TC4 surface. When the depth of grid was deeper than 100μm,contact angle increased and incomplete filling of molten CFRTP in grid occurred, the shear force thus decreased gradually.Resin-carbon fibers mixture was adhered on the fracture surface of TC4,and the variation tendency of adhesion ratio was consistent with that of shear force.TC4 matrix was exfoliated from substrate and adhered at the fracture surface of CFRTP,indicating stronger mechanical interlocking occurred at the joining interface after laser textured TC4 surface.Beside mechanical interlocking, compound layer consisted of CTi_(0.42)V_(1.58)carburization phase was also confirmed at interface, suggesting that chemical bonding also occurred at the joining interface. | Caiwang TAN Jianhui SU Ziwei FENG Yifan LIU Bo CHEN Xiaoguo SONG | 2021 | Chinese Journal of Aeronautics2021,34,5: | 11
Differentially expressed proteins of gamma-ray irradiated mouse intestinal epithelial cells by two-dimensional electrophoresis and MALDI-TOF mass spectrometry显示文摘AIM: To identify the differentially expressed proteins involved in ionizing radiation in mice and to explore new ways for studying radiation-related proteins.METHODS: Bal B/c mice grouped as sham-irradiation, 3h and 72 h irradiation were exposed to 9.0Gy single dose of γ-irradiation. Intestinal epithelia were isolated from mice, and total proteins were extracted with urea containing solution. A series of methods were used, including twodimensional electrophoresis, PDQuest 2-DE software analysis, peptide mass fingerprinting based on matrixassisted laser desorption/ionization time of flight masss pectrometry (MALDI-TOF-MS) and SWISS-PROT database searching, to separate and identify the differential proteins.Western blotting and RT-PCR were used to validate the differentially expressed proteins.RESULTS: Mouse intestine was severely damaged by 9.0 Gy γ-irradiation. Image analysis of two-dimensional gels revealed that averages of 638±39, 566±32 and 591±29 protein spots were detected in 3 groups, respectively, and the majority of these protein spots were matched. About 360 protein spots were matched between normal group and 3 h irradiation group, and the correlation coefficient was 0.78 by correlation analysis of gels. Also 312 protein spots matched between normal group and 72 h irradiation group, and 282 protein spots between 3 h and 72 h irradiation groups. Twenty-eight differential protein spots were isolated from gels, digested with trypsin, and measured with MALDI-TOF-MS. A total of 25 spots yielded good spectra, and 19 spots matched known proteins after database searching. These proteins were mainly involved in anti-oxidation, metabolism, signal transduction, and protein post-translational processes. Western-blotting confirmed that enolase was up-regulated by γ-irradiation. Up-regulation of peroxiredoxin I was verified by applying RTPCR technique, but no change occurred in QSVC72.CONCLUSION: These differentially expressed proteins might play important roles when mouse intestine was severely injured by γ-irradiation. It is suggested that differential proteomic analysis may be a useful tool to study the proteins involved in radiation damage of mouse intestinal epithelia. | Bo Zhang Yong-Ping Su Guo-Ping Ai Xiao-Hong Liu Feng-Chao Wang Tian-Min Cheng Institute of Combined Injury of PLA,Third Military Medical University,Chongqing 400038,China | 2003 | World Journal of Gastroenterology2003,9,12: | 10
Emerging Trends for Microbiome Analysis: From Single-Cell Functional Imaging to Microbiome Big Data显示文摘Method development has always been and will continue to be a core driving force of microbiome science. In this perspective, we argue that in the next decade, method development in microbiome analysis will be driven by three key changes in both ways of thinking and technological platforms:(1) a shift from dissecting microbiota structure by sequencing to tracking microbiota state, function, and intercellular interaction via imaging;(2) a shift from interrogating a consortium or population of cells to probing individual cells;and(3) a shift from microbiome data analysis to microbiome data science. Some of the recent methoddevelopment efforts by Chinese microbiome scientists and their international collaborators that underlie these technological trends are highlighted here. It is our belief that the China Microbiome Initiative has the opportunity to deliver outstanding 'Made-in-China' tools to the international research community, by building an ambitious, competitive, and collaborative program at the forefront of method development for microbiome science. | Jian xu Bo Ma Xiaoquan Su Shi Huang Xin Xu Xuedong Zhou Wei Huang Rob Knight | 2017 | Engineering2017,3,1: | 9
Preliminary 3D printing of large inclined-shaped alumina ceramic parts by direct ink writing显示文摘Three dimensional(3D)printing technology by direct ink writing(DIW)is an innovative complex shaping technology, possessing advantages of flexibility in fabrication, high efficiency, low cost, and environmental-friendliness.Herein,3D printing of complex alumina ceramic parts via DIW using thermally induced solidification with carrageenan swelling was investigated. The rheological properties of the slurry under different thermally-induced modes were systematically studied. The solidification properties of thermally-induced pastes with varying contents of carrageenan were optimized. The experimental results showed that the optimized paste consisting of 0.4 wt%carrageenan could be rapidly solidified at about 55℃,which could print inclined-plane more than 60°in vertical without support, resulting in better homogeneity of the green body.A nearly pore-free structure was obtained after sintering at 1600℃ for 2 h. | Liangliang YANG Xiaojun ZENG Allah DITTA Bo FENG Lizhong SU Yue ZHANG | 2020 | Journal of Advanced Ceramics2020,9,3: | 9
Biodiversity-based development and evolution:the emerging research systems in model and non-model organisms显示文摘Evolutionary developmental biology,or Evo-Devo for short, has become an established field that, broadly speaking, seeks to understand how changes in development drive major transitions and innovation in organismal evolution. It does so via integrating the principles and methods of many subdisciplines of biology.Although we have gained unprecedented knowledge from the studies on model organisms in the past decades, many fundamental and crucially essential processes remain a mystery.Considering the tremendous biodiversity of our planet, the current model organisms seem insufficient for us to understand the evolutionary and physiological processes of life and its adaptation to exterior environments. The currently increasing genomic data and the recently available gene-editing tools make it possible to extend our studies to non-model organisms. In this review,we review the recent work on the regulatory signaling of developmental and regeneration processes, environmental adaptation, and evolutionary mechanisms using both the existing model animals such as zebrafish and Drosophila, and the emerging nonstandard model organisms including amphioxus,ascidian,ciliates,single-celled phytoplankton, and marine nematode. In addition, the challenging questions and new directions in these systems are outlined as well. | Long Zhao Feng Gao Shan Gao Yujun Liang Hongan Long Zhiyi Lv Ying Su Naihao Ye Liusuo Zhang Chengtian Zhao Xiaoyu Wang Weibo Song Shicui Zhang Bo Dong | 2021 | Science China(Life Sciences)2021,64,8: | 9
Designing of highly selective and high-temperature endurable RWGS heterogeneous catalysts: recent advances and the future directions显示文摘Reverse water gas shift(RWGS) reaction can be served as a pivotal stage of transitioning the abundant CO_2 resource into chemicals or hydrocarbon fuels, which is attractive for the CO_2 utilization and of eventually significance in enabling a rebuilt ecological system for unconventional fuels. This concept is appealing when the process is considered as a solution for the storage of renewable energy, which may also find a variety of potential end uses for the outer space exploration. However, a big challenge to this issue is the rational design of high temperature endurable RWGS catalysts with desirable CO product selectivity. In this work, we present a comprehensive overview of recent publications on this research topic, mainly focusing on the catalytic performance of RWGS reaction over three major kinds of heterogeneous catalysts, including supported metal catalysts, mixed oxide catalysts and transition metal carbides. The reaction thermodynamic analysis, kinetics and mechanisms are also described in detail. The present review attempts to provide a general guideline about the construction of well-performed heterogeneous catalysts for the RWGS reaction, as well as discussing the challenges and further prospects of this process. | Xiong Su Xiaoli Yang Bo Zhao Yanqiang Huang | 2017 | Journal of Energy Chemistry2017,26,5: | 9
