| 4 | Cytotoxic effect of interleukin-8 in retinal ganglion cells and its possible mechanisms显示文摘AIM: To investigate the effect of interleukin-8(IL-8) on neural retinal ganglion cells(RGCs) and whether it can be alleviated by G31P. METHODS: RGC-5 cells were exposed to IL-8 with or without its specific receptor antagonist G31P for 24h, and the cell viability was assessed by Cell Counting Kit 8(CCK-8). Apoptosis was measured by examining nuclear morphology and quantifying with flow cytometry. Reverse transcription quantitative real-time polymerase chain reaction(RT-qP CR) and Western blot were used to investigate the expression of apoptosis-related genes. RESULTS: CCK-8 assay showed that IL-8 significantly inhibits the viability of RGC-5 cells in a dose-dependent manner. Cell apoptosis assays exhibited higher apoptotic rate in IL-8 treatment group compared to control group. We further found that IL-8 could promote Bax and caspase-3 expressions, but decrease the level of Bcl-2 in the aspect of m RNA and protein. | 5 | 雨养农业:中国半干旱地区雨水灌溉土地上的水资源综合管理系统显示文摘雨养农业(RHA)最早是由甘肃省科学家在十几年前开发的,它是适用于半干旱地区靠雨水灌溉土地的一种水资源综合管理系统。这个系统由三个主要部分组成:雨水收集系统、节水灌溉系统和高效农作物生产系统。它的主要功能是为农民们提供一种能够在用水受限制的环境下获得满足家庭生活和农业所需要的水的途径。在甘肃省和中国西北地区的其他省份,RHA的最初设置表明RHA有能力改进靠雨水灌溉的农业系统的表现,并且能够致力于解决土壤侵蚀等一些环境问题。RHA系统规模小、成本低,很客易在农民家庭推广应用。但是,为了能够取得成功,需要把RHA系统纳入到一个综合农业管理系统之中:即RHA管理必须与其他农业技术与管理实践结合在一起运用。此外,在大范围内推广使用RHA需要考虑一系列技术、农业水文、生态、社会、文化、经济和政治因素。特别是需要向农民提供培训和技术指导,以开发和传播更为有效和廉价的RHA技术方式来更新换代并设计开发出替代性的政策工具和社会规则以便于RHA在实践中的应用。 | Li Fengrui Seth Cook Gordon T. The SRM requires interdisciplinary efforts to finalize the pre-designed organs. Three-dimensional(3D) printing, as an additive manufacturing technology, has been widely used in various fields due to its high resolution and individuation. In SRM, with the assistance of 3D printing, cells and biomaterials could be precisely positioned to construct complicated tissues. This review summarizes the state of the SRM advances and focuses in particular on the 3D printing application in biofabrication. | 7 | 鞘内注射吗啡预处理激活大鼠δ、κ、μ受体致心肌保护显示文摘背景鞘内注射小剂量吗啡可产生与静脉注射吗啡相似的心肌保护作用,但鞘内注射吗啡预处理(IT—MPC)与缺血预处理(IPC)作用强度的差别还未知,IT-MPC是否由阿片类受体所介导也未阐明。因此,本实验比较IT—MPC与IPC作用强度差别并探讨阿片类受体的作用。方法80只SD大鼠麻醉后开胸,成功置入鞘内导管后随机分为13组(n=6—7)。IPC组接受3次5分钟缺血-5分钟再灌注的心肌IPC(阻闭左冠状动脉),随之给予30分钟缺血-2小时再灌注诱导心肌缺血再灌注(IR)损伤。IT-MPC组按剂量不同分为4个亚组(分别是0.03、0.3、3及30μg/kg),以3次5分钟注射-5分钟间停的模式平均注入鞘内,随之诱导IR损伤。静脉注射吗啡预处理组(IV-MPC)组经静脉给予吗啡300μg/kg,对照组鞘内给予10μl生理盐水。受体阻滞剂组选择在IT—MPC(3μg/kg)前分别给予选择性δ、κ、μ受体阻滞剂NTD、nor-BNI及CTOP,以评价阿片类受体亚型的作用。通过2,3,5-tripheny ltetrazolium染色,计算心肌梗死面积(IS),即梗死心肌占缺血危险区(AAR)的百分比。结果与对照组相比,鞘内注射0.3~30μg/kg吗啡降低心肌IS,3组IS/AAR分别为33%±10%(0.3μg/kg)、29%±10%(3μg/kg)、29%±16%(30μg/kg),对照组为53%±8%(P〈0.01)。IT-MPC降低心肌IS/AAR程度与IV-MPC(33%±6%,P=0.84)及IPC(22%±4%,P=0.41)相近。注射选择性受体阻滞剂后,由IT-MPC(3μg/kg)产生的心肌预处理效果降低(NTD+IT-MPC,50%±9%:nor-BNI+IT-MPc,43%±6%;CTOP+IT—MPC,53%±9%;与对照组相比,P=0.14)。结论IT—MPC产生与IPC及IV—MPC相近的心肌保护作用,其机制涉及到δ、κ、μ受体。 | Rui Li Gordon T. | 9 | In vivo evaluation of additively manufacturedmulti-layered scaffold for the repair of large osteochondral defects显示文摘The repair of osteochondral defects is one of the major clinical challenges in orthopaedics.Well-established osteochondral tissue engineering methods have shown promising results for the early treatment of small defects.However,less success has been achieved for the regeneration of large defects,which is mainly due to the mechanical environment of the joint and the heterogeneous nature of the tissue.In this study,we developed a multi-layered osteochondral scaffold to match the heterogeneous nature of osteochondral tissue by harnessing additive manufacturing technologies and combining the established art laser sintering and material extrusion techniques.The developed scaffold is based on a titanium and polylactic acid matrix-reinforced collagen“sandwich”composite system.The microstructure and mechanical properties of the scaffold were examined,and its safety and efficacy in the repair of large osteochondral defects were tested in an ovine condyle model.The 12-week in vivo evaluation period revealed extensive and significantly higher bone in-growth in the multi-layered scaffold compared with the collagen–HAp scaffold,and the achieved stable mechanical fixation provided strong support to the healing of the overlying cartilage,as demonstrated by hyaline-like cartilage formation.The histological examination showed that the regenerated cartilage in the multi-layer scaffold group was superior to that formed in the control group.Chondrogenic genes such as aggrecan and collagen-II were upregulated in the scaffold and were higher than those in the control group.The findings showed the safety and efficacy of the cell-free“translation-ready”osteochondral scaffold,which has the potential to be used in a one-step surgical procedure for the treatment of large osteochondral defects. | Maryam Tamaddon Gordon Blunn Rongwei Tan Pan Yang Xiaodan Sun Shen-Mao Chen Jiajun Luo Ziyu Liu Ling Wang Dichen Li Ricardo Donate Mario Monzón Chaozong Liu | 2022 | Bio-Design and Manufacturing2022,5,3: | 2 |
