1Acupuncture treatment of lung-spleen Qi deficiency in stable chronic obstructive pulmonary disease: a randomized, open-label, controlled trial显示文摘OBJECTIVE:To evaluate the effectiveness and safety of acupuncture that reinforces the spleen to strengthen the lung in patients with stable chronic obstructive pulmonary disease(COPD).METHODS:This was a randomized,open-controlled trial in which the acupuncturist and the participants were not blinded,but the outcome evalu-ators and data analysts were blinded.One-hundred-and-two patients with stable COPD were randomly divided into two groups in a 1∶1 ratio.The acupuncture group received 30-minute sessions of acupuncture therapy at the same acupoints three times weekly for 6 weeks in addition to routine conventional Western Medicine treatment;the control group received routine conventional Western Medicine treatment alone.The primary outcome was the Borg scale score,which was assessed immediately after the 6-minute walk test.The secondary outcomes were the 6-minute walk distance,lung function,and oxygen saturation.Measurements were obtained at baseline and after 6 weeks of treatment.RESULTS:After 6 weeks of treatment,the Borg scale score in the acupuncture group was significantly better than that in the control group(2.02±0.71 versus 5.01±0.34,P<0.05).Furthermore,the post-treatment improvements in the 6-minute walk distance,lung function,and oxygen saturation were significantly greater in the acupuncture group than in the control group,showing that the acupuncture group had better exercise tolerance.CONCLUSION:The findings suggest that acupuncture that aims to reinforce the spleen to strengthen the lung is a safe and effective adjuvant therapy that effectively improves the exercise capacity of patients with stable COPD.Li Yu Xiong Chan Zeng Yan Wei Hua Zhuang Guangtong Zhao Lihong Li Chenyi Li Lin Qin Erqi Chen Xiaoqin Fu Juanjuan 2019Journal of Traditional Chinese Medicine2019,39,6:13

4Genome and transcriptome of Papaver somniferum Chinese landrace CHM indicates that massive genome expansion contributes to high benzylisoquinoline alkaloid biosynthesis显示文摘Opium poppy(Papaver somniferum)is a source of morphine,codeine,and semisynthetic derivatives,including oxycodone and naltrexone.Here,we report the de novo assembly and genomic analysis of P.somniferum traditional landrace‘Chinese Herbal Medicine’.Variations between the 2.62 Gb CHM genome and that of the previously sequenced high noscapine 1(HN1)variety were also explored.Among 79,668 protein-coding genes,we functionally annotated 88.9%,compared to 68.8%reported in the HN1 genome.Gene family and 4DTv comparative analyses with three other Papaveraceae species revealed that opium poppy underwent two whole-genome duplication(WGD)events.The first of these,in ancestral Ranunculales,expanded gene families related to characteristic secondary metabolite production and disease resistance.The more recent species-specific WGD mediated by transposable elements resulted in massive genome expansion.Genes carrying structural variations and large-effect variants associated with agronomically different phenotypes between CHM and HN1 that were identified through our transcriptomic comparison of multiple organs and developmental stages can enable the development of new varieties.These genomic and transcriptomic analyses will provide a valuable resource that informs future basic and agricultural studies of the opium poppy.Li Pei Baishi Wang Jian Ye Xiaodi Hu Lihong Fu Kui Li Zhiyu Ni Zhenlong Wang Yujie Wei Luye Shi Ying Zhang Xue Bai Mengwan Jiang Shuhui Wang Chunling Ma Shujin Li Kaihui Liu Wanshui Li Bin Cong 2021Horticulture Research2021,8,1:4
5Effects of Root Zone Temperature on Growth and Photosynthetic Parameters of Grafted Cucumber显示文摘Taking Cucurbita maxima and Cucurbita moschata as root stocks,and‘Jinyou No 3'cucumber as scion,the effects of different root zone temperature conditions optimal temperature( CK)( 18-20 ℃),suboptimal temperature( 13-15 ℃) and low temperature( 8-10 ℃) on the growth and photosynthesis indexes were studied. The results showed that,compared with optimal temperature( CK),suboptimal temperature and low temperature produced a significant inhibition of growth on cucumbers. The plant height,stem diameter,leaf area,number of leaves and dry weight of aboveground part were all reduced,dry weight of underground part and root shoot ratio all increased,while the inhibition was more significant at low temperature. Low and suboptimal temperature conditions also reduced SPAD value,net photosynthetic rate,transpiration rate,intercellular CO_2 concentration and stomatal conductance of the grafted cucumber. 8Enriched endoplasmic reticulum-mitochondria interactions result in mitochondrial dysfunction and apoptosis in oocytes from obese mice显示文摘Background: Maternal obesity alters oocytes and subsequent fetal metabolism. An increasing number of studies have shown that the endoplasmic reticulums(ER) or mitochondria have important effects on oocyte quality, but there has been no study of the effect of mitochondria-associated ER membranes(MAMs) on oocyte quality. The present study was designed to assess whether the level of MAM and MAM-related proteins were different in oocytes from obese and control mice.Results: First, oocytes from mice with high-fat-diet(HFD)-induced obesity had higher levels(either greater numbers or a higher proportion for the same numbers) of MAM than oocytes from control mice. The abundance of MAM-related proteins in oocytes from obese mice was significantly greater at both the messenger RNA and protein levels, including inositol 1,4,5-trisphosphate receptor, type 1(IP3R1), inositol 1,4,5-trisphosphate receptor,type 2(IP3 R2) and phosphofurin acidic cluster sorting protein 2(PACS-2). Further, there was an increase in mitochondrial Ca^(2+)([Ca^(2+)]_m) which was associated with increased apoptosis and compromised cytoplasmic maturation in oocytes from obese mice. Down-regulation of MAM-related protein IP3R1 in oocytes from obese mice decreased [Ca^(2+)]_mand apoptosis and improved cytoplasmic maturation but did not reduce the overal MAM level. However, down-regulating MAM-related protein PACS-2 in oocytes from obese mice did reduce the level of MAM and [Ca^(2+)]_m, which decreased the rate of apoptosis and improved cytoplasmic maturation of oocytes from obese mice.Conclusions: It is possible that enriched MAM could increase [Ca^(2+)]_m, and this increase has been found to be associated with increased apoptosis and compromised cytoplasmic maturation in oocytes from obese mice. 10Comprehensive Humoral and Cellular Immune Responses to SARS-CoV-2 Variants in Diverse Chinese Population显示文摘The SARS-CoV-2 variants have been emerging and have made great challenges to current vaccine and pandemic control strategies.It is urgent to understand the current immune status of various Chinese populations given that the preexisting immunity has been established by national vaccination or ex posure to past variants.Using sera from 85 individuals(including 21 convalescents of natural infection,15 cases which suffered a breakthrough infection after being fully vaccinated,and 49 healthy vaccinees),we showed signifcantly enhanced neutralizing activities against SRAS-CoV-2 variants in convalescent sera,especially those who had been fully vaccinated.The neut ralizing antibodies against Omicron were detectable in 75%of convalescents and 4.9%of healthy vaccinees(p=0.006),with a GMT of 289.5,180.9-463.3,and 42.6,31.3-59,respetively.However,the neutralizing activities were weaker in young convalescents(aged<18y),with a detectable rate of 50%and a GMT of 46.4 against Omicron.We also examined and found no pan.sarbecovirus neutralizing activities in vaccinated SARS-CoV-I survivors.A booster dose could further increase the breadth and magnitude of neutralization against WT and variants of concern(VOCs)to different degrees.In addition,we showed that COVID-19-inactivated vaccines can elicit Omicron-specific T-cell responses.The positive rates of ELISpot reactions were 26.7%(4/15)and 43.8%(7/16)in the full vaccination group and the booster vaccination group,respectively,although without statistically significant difference.The neutralizing antibody titers declined while T-cell responses remain consistent over 6 months.These findings will inform the optimization of public health vaccination and intervention strategies to protect diverse populations against SARS-CoV-2 variants.Advances.Breakthrough infection significantly boosted neutralizing activities against SARS-CoV-2 variants a compared to booster immunization with inaclivated vaccine.Vaccine-induced vir us-specific T-cell immunity,on the other hand,may compensate for the shortall.Furthermore,the public health system should target the most vulnerable group due to a poorer protective serological response in both infected and vaccinated adolescents.Jiwei Li Jing Wu Qiuyue Long Yan'an Wu Xiaoyi Hu Yukun He Mingzheng Jiang Jia Li Lili Zhao Shuoqi Yang Xiaoyong Chen Minghui Wang Jianshi Zheng Fangfang Wu Ruiliang Wu Lihong Ren Liang Bu Houzhao Wang Ke Li Lijuan Fu Guojun Zhang Yali Zheng Zhancheng Gao 2022Research2022,,2:1

20Postoperative sequential chemotherapy and radiotherapy for locally advanced gastric cancer显示文摘Objective The aim of the study was to evaluate the role of postoperative sequential chemotherapy and radiotherapy in patients with locally advanced gastric cancer.Methods From January 2003 to December 2010, 146 gastric cancer patients at our institution(Department of Oncology, Tongji Hospital, Tongji Medical College, Huazhong University of Science and Technology, Wuhan, China) received postoperative sequential chemotherapy and radiotherapy after radical surgery. Radiotherapy was administered as a dose of 4500 cGy in 25 fractions. For patients with positive margins, the dose was raised to 5040 cGy in 28 fractions. Three cycles of m FOLFOX or PF(cisplatin, 5-fluorouracil) chemotherapy regimen were applied before and after radiotherapy. Three-and 5-year survival rates were analyzed; any adverse effects with respect to hematology, hepatic and renal function, or the gastrointestinal tract that occurred during the treatment were evaluated.Results This cohort consisted of non-metastatic patients: 104 men and 42 women with a median age of 51.0 years. The full course of sequential chemotherapy and radiotherapy(4500–5040 cGy) was completed by 129 patients(88.4%). Seventeen regional relapses(9.8%) and 46 distant relapses(23.8%) were recorded. Fifty patients(34.2%) died during follow-up. The 3-and 5-year overall survival rates(OS) were 60% and 54%, and disease-free survival rates(DFS) were 53% and 47%, respectively. There were no significant differences in survival rate with respect to age, sex, histopathology, N stage, site of the tumor, or margin status. Multivariate analysis showed that only the depth of tumor invasion(T stage) was an independent prognostic factor for OS(P = 0.009) and DFS(P = 0.006). The rates of grades 3 and 4 neutropenia and vomiting were 9.6% and 3.4%, respectively, during the treatment.Conclusion Postoperative sequential chemotherapy with an m FOLFOX or PF regimen and radiotherapy were found to be an effective means of treating advanced gastric cancer patients with T3–T4 disease.
