| 5 | Cellular senescence in musculoskeletal homeostasis,diseases,and regeneration显示文摘Emerging insights into cellular senescence highlight the relevance of senescence in musculoskeletal disorders,which represent the leading global cause of disability.Cellular senescence was initially described by Hayflick et al.in 1961 as an irreversible nondividing state in in vitro cell culture studies.We now know that cellular senescence can occur in vivo in response to various stressors as a heterogeneous and tissue-specific cell state with a secretome phenotype acquired after the initial growth arrest.In the past two decades,compelling evidence from preclinical models and human data show an accumulation of senescent cells in many components of the musculoskeletal system.Cellular senescence is therefore a defining feature of age-related musculoskeletal disorders,and targeted elimination of these cells has emerged recently as a promising therapeutic approach to ameliorate tissue damage and promote repair and regeneration of the skeleton and skeletal muscles.In this review,we summarize evidence of the role of senescent cells in the maintenance of bone homeostasis during childhood and their contribution to the pathogenesis of chronic musculoskeletal disorders,including osteoporosis,osteoarthritis,and sarcopenia.We highlight the diversity of the senescent cells in the microenvironment of bone,joint,and skeletal muscle tissue,as well as the mechanisms by which these senescent cells are involved in musculoskeletal diseases.In addition,we discuss how identifying and targeting senescent cells might positively affect pathologic progression and musculoskeletal system regeneration. | Mei Wan Elise F.Gray-Gaillard Jennifer H.Elisseeff | 2021 | Bone Research2021,9,4: | 7 |

METHODS:This case-control study includes 326 patients with CRC and 369 age-and gender-matched controls.Genotypes of the COX-2 polymorphisms -765G→C and-1195A→G were determined by polymerase chain reaction-based restriction fragment length polymorphism.COX-2 genotypes and haplotypes were analyzed and odds ratios with 95%confi- dence intervals were estimated by logistic regression. RESULTS:The-765GG genotype was associated with an increased risk of developing CRC(OR,1.45; 95%CI,1.03-2.04).No significant difference was observed in the genotype distribution of the-1195A→ G polymorphism between patients and controls.The GG/AC haplotype was present significantly less often in patients than in controls(OR 0.44;95%CI,0.22-0.85). When the AC,AG and GG haplotypes were investigated separately,the AC haplotype showed a tendency to be less frequent in patients than in controls(OR(AG/AC)0.78; 95%CI,0.57-1.06). | 9 | Effects of insulin and pathway inhibitors on the PI3K-Akt- mTOR phosphorylation profile in acute myeloid leukemia cells显示文摘The phosphatidylinositol 3-kinase(PI3K)-Akt-mechanistic target of rapamycin(mTOR)pathway is constitutively activated in human acute myeloid leukemia(AML)cells and is regarded as a possible therapeutic target.Insulin is an agonist of this pathway and a growth factor for AML cells.We characterized the effect of insulin on the phosphorylation of 10 mediators in the main track of the PI3K-Akt-mTOR pathway in AML cells from 76 consecutive patients.The overall results showed that insulin significantly increased the phosphorylation of all investigated mediators.However,insulin effects on the pathway activation profile varied among patients,and increased phosphorylation in all mediators was observed only in a minority of patients;in other patients,insulin had divergent effects.Global gene expression profiling and proteomic/phosphoproteomic comparisons suggested that AML cells from these two patient subsets differed with regard to AML cell differentiation,transcriptional regulation,RNA metabolism,and cellular metabolism.Strong insulin-induced phosphorylation was associated with weakened antiproliferative effects of metabolic inhibitors.PI3K,Akt,and mTOR inhibitors also caused divergent effects on the overall pathway phosphorylation profile in the presence of insulin,although PI3K and Akt inhibition caused a general reduction in Akt pT308 and 4EBP1 pT36/pT45 phosphorylation.For Akt inhibition,the phosphorylation of upstream mediators was generally increased or unaltered.In contrast,mTOR inhibition reduced mTOR pS2448 and S6 pS244 phosphorylation but increased Akt pT308 phosphorylation.In conclusion,the effects of both insulin and PI3K-Akt-mTOR inhibitors differ between AML patient subsets,and differences in insulin responsiveness are associated with differential susceptibility to metabolic targeting. | Ina Nepstad Kimberley Joanne Hatfield Ida Sofie Grønningsæter Elise Aasebø Maria Hernandez-Valladares Karen Marie Hagen Kristin Paulsen Rye Frode SBerven Frode Selheim Håkon Reikvam Øystein Bruserud | 2019 | Signal Transduction and Targeted Therapy2019,4,1: | 4 |

Indeed, despite potent immunosuppression, chronic rejection is still a real problem in transplantation. The humoral response is an important mediator of chronic rejection, and numerous strategies have been developed to target either B cells or plasma cells. However, the use of anti-CD20 therapy has highlighted the beneficial role of subpopulation of B cells, termed regulatory B cells. These cells have been characterized mainly in mice models of auto-immune diseases but emerging literature suggests their role in graft tolerance in transplantation. Regulatory B cells seem to be induced following inflammation to restrain excessive response. Different phenotypes of regulatory B cells have been described and are functional at various differentiation steps from immature to plasma cells. These cells act by multiple mechanisms such as secretion of immuno-suppressive cytokines interleukin-10(IL-10) or IL-35, cytotoxicity, expression of inhibitory receptors or by secretion of non-inflammatory antibodies. | 12 | Characterization of two rat models of cystic fibrosis—KO and F508del CFTR—Generated by Crispr-Cas9显示文摘Background: Genetically engineered animals are essential for gaining a proper understanding of the disease mechanisms of cystic fibrosis(CF). The rat is a relevant laboratory model for CF because of its zootechnical capacity, size, and airway characteristics, including the presence of submucosal glands.Methods: We describe the generation of a CF rat model(F508 del) homozygous for the p.Phe508 del mutation in the transmembrane conductance regulator(Cftr) gene. This model was compared to new Cftr-/-rats(CFTR KO). Target organs in CF were examined by histological staining of tissue sections and tooth enamel was quantified by micro-computed tomography. The activity of CFTR was evaluated by nasal potential difference(NPD) and short-circuit current measurements. The effect of VX-809 and VX-770 was analyzed on nasal epithelial primary cell cultures from F508 del rats.Results: Both newborn F508 del and Knock out(KO) animals developed intestinal obstruction that could be partly compensated by special diet combined with an osmotic laxative. The two rat models exhibited CF phenotypic anomalies such as vas deferens agenesis and tooth enamel defects. Histology of the intestine, pancreas, liver, and lungs was normal. Absence of CFTR function in KO rats was confirmed ex vivo by short-circuit current measurements on colon mucosae and in vivo by NPD, whereas residual CFTR activity was observed in F508 del rats. | 16 | Vegetation Composition and Structure of Some NeotropicaMountain Grasslands in Brazil显示文摘The description and understanding of plant communities is fundamental for the implementation of conservation or restoration programs, especially when these communities are highly threatened and need to be restored.Campos rupestres, some Neotropical mountain grasslands located in central Brazil and part of the Cerrado biome(covering 2 million km2) host unique plant communities, currently threatened by quarrying and mining.The grassy matrix of campos rupestres, has long been considered a rich mosaic under the control of local topography and the nature of substrate, but this affirmation has not been well studied.We analyzed whether plant communities varied in relation to edaphic factors within the stony substrate and the sandy substrate of this grassy matrix.We selected 5 sites where occur both grasslands on stony substrate and on sandy substrate, and we carried out vegetation surveys and soil analyses.We counted 222 plant species within our communities, among which38.6% are exclusively found on campos rupestres.Our results show that both soil-types are strongly acidic, nutrient poor and exhibit a seasonal variation.Phosphorus increases and p H and organic carbon decrease during the dry season.Stony soils areslightly richer in nutrients than sandy soils and differences in soil granulometry and composition have led to the formation of distinct plant communities.Some species are confined to either one or the other grassland-type, which makes the plant composition of each community unique.Variations in edaphic factors generate heterogeneous grasslands favorable to a high plant diversity.Conservation programs and restoration actions have to maintain or recreate this heterogeneity.The presence of distinct plant communities implies that different strategies might be adopted to improve the restoration of these ecosystems. | LE STRADIC Soizig BUISSON Elise FERNANDES G.Wilson | 2015 | Journal of Mountain Science2015,12,4: | 3 |
