1A nuclear import inhibitory peptide ameliorates the severity of cholecystokinin-induced acute pancreatitis显示文摘AIM: To assess the effect of our novel cell-permeable nuclear factor-kappaB (NF-κB) inhibitor peptide PN50 in an experimental model of acute pancreatitis. PN50 was produced by conjugating the cell-penetrating penetratin peptide with the nuclear localization signal of the NF-κB p50 subunit.METHODS: Pancreatitis was induced in male Wistar rats by administering 2×100 μg/kg body weight of cholecystokininoctapeptide (CCK) intraperitoneally (IP) at an interval of 1 h. PN50-treated animals received 1 mg/kg of PN50 IP 30 min before or after the CCK injections. The animals were sacrificed 4 h after the first injection of CCK.RESULTS: All the examined laboratory (the pancreatic weight/body weight ratio, serum amylase activity,pancreatic levels of TNF-α and IL-6, degree of lipid peroxidation, reduced glutathione levels, NF-κB binding activity, pancreatic and lung myeloperoxidase activity) and morphological parameters of the disease were improved before and after treatment with the PN50 peptide.According to the histological findings, PN50 protected the animals against acute pancreatitis by favoring the induction of apoptotic, as opposed to necrotic acinar cell death associated with severe acute pancreatitis.CONCLUSION: Our study implies that reversible inhibitors of stress-responsive transcription factors like NF-κB might be clinically useful for the suppression of the severity of acute pancreatitis.Tamás Letoha Csaba Somlai Tamáas Takács Annamária Szabolcs Katalin Jármay Zoltán Rakonczay Jr Péter Hegyi Ilona Varga József Kaszaki István Krizbai Imre Boros Ern(?) 5Role of stromal cell-derived factor 1α pathway in bone metastatic prostate cancer显示文摘Metastatic prostate cancer is one of the leading causes of cancer-related death in men.The primary site of metastasis from prostate cancers is the bone.During the last decade,multiple studies have pointed to the role of the stromal cell-derived factor 1 alpha(SDF1α)/CXCR4 axis in the metastatic spread of the disease,but the mechanisms that underlie this effect are still incompletely understood.In this review,we summarize the current understanding of the role of the SDF1α/CXCR4 pathway in bone metastatic prostate cancer.We also discuss the therapeutic potential of disrupting the interaction between prostate tumor cells and bone environment with focus on the SDF1α pathway.Nisha Gupta Dan G. 6Doege-Potter syndrome by malignant solitary fibrous tumor of the liver: A case report and review of literature显示文摘BACKGROUND Solitary fibrous tumor of the liver (SFTL) is a rare occurrence with a low number of cases reported in literature. SFTL is usually benign but, 10%-20% cases are reported to be malignant with a tendency to metastasize. The majority of malignant SFTL cases are associated with a paraneoplastic hypoglycaemia defined as Doege-Potter syndrome. Surgery is the best therapeutic treatment, however, long- life follow-up is recommended. CASE SUMMARY A 74-year-old man, was admitted to the emergency department after a syncopal episode with detection of hypoglycaemia resistant to medical treatment. The computed tomography revealed a solid mass measuring 15 cm of the left liver. An open left hepatectomy was performed with complete resection of tumor. Histopathological analyses confirmed a malignant SFTL. CONCLUSION Large series with long-term follow-up have not been published neither have clinical trials been undertaken. 7A numerical study on the generation of a distinct type of nonlinear internal wave packet in the South China Sea显示文摘A distinct type of nonlinear internal-wave packet, with the largest internal solitary wave in the middle of the packet, was regularly observed in the South China Sea during the Asian Seas International Acoustics Experiment in 2001. Data analysis shows that the occurrence of the distinct internal wave packet is closely related with the occurrence of lower-high internal tides; the internal tides are mixed in the experimental area and, thus, there is diurnal inequality between the heights of two neighboring internal tides. Modeling of internal tides and internal solitary waves in a shoaling situation suggests that this type of wave packet can be generated in the South China Sea by the large shoaling of internal solitary waves and internal tides. Both the internal solitary waves and the internal tides come from the direction of Luzon Strait. The initial large internal solitary waves contribute to the occurrence of the largest internal solitary wave in the middle of the packet and the waves behind the largest internal solitary wave, while the shoaling internal tides bring about the nonlinear internal waves in front of the largest internal solitary wave via interaction with the local shelf topography.杜涛 严晓海 Timothy DUDA 2010Chinese Journal of Oceanology and Limnology2010,28,3:4
8Nonsense variant of ATP8B1 gene in heterozygosis and benign recurrent intrahepatic cholestasis: A case report and review of literature显示文摘BACKGROUND Benign recurrent intrahepatic cholestasis is a genetic disorder with recurrent cholestatic jaundice due to ATP8B1 and ABCB11 gene mutations encoding for hepato-canalicular transporters.Herein,we firstly provide the evidence that a nonsense variant of ATP8B1 gene(c.1558A>T)in heterozygous form is involved in BRIC pathogenesis.CASE SUMMARY A 29-year-old male showed severe jaundice and laboratory tests consistent with intrahepatic cholestasis despite normal gamma-glutamyltranspeptidase.Acute and chronic liver diseases with viral,metabolic and autoimmune etiology were excluded.Normal intra/extra-hepatic bile ducts were demonstrated by magnetic resonance.Liver biopsy showed:Cholestasis in the centrilobular and intermediate zones with bile plugs and intra-hepatocyte pigment,Kupffer’s cell activation/hyperplasia and preserved biliary ducts.Being satisfied benign recurrent intrahepatic cholestasis diagnostic criteria,ATP8B1 and ABCB11 gene analysis was performed.Surprisingly,we found a novel nonsense variant of ATP8B1 gene(c.1558A>T)in heterozygosis.The variant was confirmed by Sanger sequencing following a standard protocol and tested for familial segregation,showing a maternal inheritance.Immunohistochemistry confirmed a significant reduction of mutated gene related protein(familial intrahepatic cholestasis 1).The patient was treated with ursodeoxycholic acid 15 mg/kg per day and colestyramine 8 g daily with total bilirubin decrease and normalization at the 6th and 12th mo.CONCLUSION A genetic abnormality,different from those already known,could be involved in familial intrahepatic cholestatic disorders and/or pro-cholestatic genetic predisposition,thus encouraging further mutation detection in this field.Mariano Piazzolla Nicola Castellaneta Antonio Novelli Emanuele Agolini Dario Cocciadiferro Leonardo Resta Loren Duda Michele Barone Enzo Ierardi Alfredo Di Leo 2020World Journal of Hepatology2020,12,2:3

10Solution of Inverse Heat Conduction Problem using the Tikhonov Regulariza-tion Method显示文摘It is hard to solve ill-posed problems, as calculated temperatures are very sensitive to errors made while calculating 'measured' temperatures or performing real-time measurements. The errors can create temperature oscillation, which can be the cause of an unstable solution. In order to overcome such difficulties, a variety of techniques have been proposed in literature, including regularization, future time steps and smoothing digital filters. In this paper, the Tikhonov regularization is applied to stabilize the solution of the inverse heat conduction problem.
