| 2 | Identification of three major Bemisia tabaci biotypes in China based on morphological and DNA polymorphisms显示文摘The sweet potato whitefly, Bemisia tabaci, is a complex species, and many of its biotypes are important agricultural pests. The B and Q biotypes are invasive and coexist with the native Cv biotype on vegetable and ornamental crops in China. In this study, these three bio- types were identified based on their morphological characteristics, RAPD-PCR analysis, and DNA sequences of the COI gene. The ante-rior and posterior wax fringes of the B, Q, and Cv biotypes significantly differed from each other. Based on this morphological characteristic, the three biotypes can easily be distinguished in greenhouses and fields. Genomic DNA RAPD-PCR band patterns also revealed differences between these biotypes using the H16 primer. The B biotype has three bright DNA bands between 250 and 600 bp, the Q biotype only has one bright band at 300 bp, while the Cv biotype has no band between 250 and 500 bp. Both the Cv and Q biotypes have two bright bands at 750 and 1000 bp while the B biotype has only one band at 1000 bp. Based on the COI gene, the genetic identity between B and Cv, B and Q, and Q and CV was 85.8%, 94.7%, and 86.0%, respectively. The MP tree indicated that the phylogenetic relationship between the B and Q biotypes is much closer than that between the B and Cv or the Q and Cv biotypes. | 3 | IL-17A-dependent gut microbiota is essential for regulating diet-induced disorders in mice显示文摘The gut microbiota plays a key role in obesity and related metabolic disorders, and multiple factors including diet, host genotype, and age regulate it. Many studies have examined the contribution of extrinsic factors to the regulation of the gut microbiota, but the importance of the host genetic constitution cannot be ignored. Interleukin 17A(IL-17A), a pro-inflammatory cytokine, is important in the defense against infection and diseases. Here, we investigated the association among IL-17, a high-fat diet(HFD), and the gut microbiota. Mice deficient in IL-17A were resistant to diet-induced obesity and related diseases. Compared with the Ⅱ-17^(-/-)mice, wild-type(WT) mice challenged with HFD showed obvious weight fluctuations, such as those seen in type 2 diabetes, and hematological changes similar to those associated with metabolic syndrome. However, housing WT mice and Ⅱ-17a^(-/-)mice together significantly alleviated these symptoms in the WT mice. A metagenomic analysis of the mouse feces indicated that the microbial community compositions of these two groups differed before HFD feeding. The HFD mediated shifts in the gut microbial compositions, which were associated with the mouse phenotypes.We also identified potentially beneficial and harmful species present during this period, and drew networks of the most abundant species. A functional analysis indicated pathway changes in the WT and Ⅱ-17a^(-/-)mice when fed the HFD. | 5 | Experimental and theoretical studies of the reaction between cationic vanadium oxide clusters and acetylene显示文摘The time of flight mass spectrometer coupled with a laser ablation/supersonic expansion cluster source and a fast flow reactor was adopted to study the reactivity of cationic vanadium oxide clusters(VmOn+) toward acetylene(C2H2) molecules under gas phase(P,~ 1.14 kPa),under near room temperature(T,~ 350 K) conditions.Association products,VmOnC2H2+(m,n = 2,4;2,6;3,7―8;4,9―11;5,12―13;6,13―16,and 7,17),are observed.The oxidation of C2H2 by(V2O5)+n(n = 1―3) is experimentally identified.The reactivity of(V2O5)+ n decreases as n increases.Density functional theory(DFT) calculations were carried out to interpret the reaction mechanisms.The DFT results indicate that a terminal oxygen atom from V2O5+ can transfer overall barrierlessly to C2H2 at room temperature,which is in agreement with the experimental observation.Other experimental results such as the observation of V2O6C2H+2 and non-observation of V2O7,8C2H+2 in the experiments are also well interpreted based on the DFT calculations.The reactivity of vanadium oxide clusters toward acetylene and other hydrocarbons may be considered in identifying molecular level mechanisms for related heterogeneous catalysis. | YIN Shi MA YanPing DU Lin HE ShengGui GE MaoFa | 2008 | Chinese Science Bulletin2008,53,24: | 3 |
| 6 | Development of Cu foam-based Ni catalyst for solar thermal reforming of methane with carbon dioxide显示文摘Using solar energy to produce syngas via the endothermic reforming of methane has been extensively investigated at the laboratory- and pilot plant-scales as a promising method of storing solar energy. One of the challenges to scaling up this process in a tubular reformer is to improve the reactor's performance, which is limited by mass and heat transfer issues. High thermal conductivity Cu foam was therefore used as a substrate to improve the catalyst's thermal conductivity during solar reforming. We also developed a method to coat the foam with the catalytically active component NiMg_3AlO_x. The Cu foam-based NiMg_3AlO_x performs better than catalysts supported on SiSiC foam, which is currently used as a substrate for solar-reforming catalysts, at high gas hourly space velocity(≥400,000 mL/(g·h)) or at low reaction temperatures(≤ 720 °C). | 20 | Fusarivirus accessory helicases present an evolutionary link for viruses infecting plants and fungi显示文摘A significant number of mycoviruses have been identified that are related to plant viruses,but their evolutionary relationships are largely unexplored.A fusarivirus,Rhizoctonia solani fusarivirus 4(RsFV4),was identified in phytopathogenic fungus Rhizoctonia solani(R.solani)strain XY74 co-infected by an alphaendornavirus.RsFV4 had a genome of 10,833 nt(excluding the poly-A tail),and consisted of four non-overlapping open reading frames(ORFs).ORF1 encodes an 825 aa protein containing a conserved helicase domain(Hel1).ORF3 encodes 1550 aa protein with two conserved domains,namely an RNA-dependent RNA polymerase(RdRp)and another helicase(Hel2).The ORF2 and ORF4 likely encode two hypothetical proteins(520 and 542 aa)with unknown functions.The phylogenetic analysis based on Hel2 and RdRp suggest that RsFV4 was positioned within the fusarivirus group,but formed an independent branch with three previously reported fusariviruses of R.solani.Notably,the Hel1 and its relatives were phylogenetically closer to helicases of potyviruses and hypoviruses than fusariviruses,suggesting fusarivirus Hel1 formed an evolutionary link between these three virus groups.This finding provides evidence of the occurrence of a horizontal gene transfer or recombination event between mycoviruses and plant viruses or between mycoviruses.Our findings are likely to enhance the understanding of virus evolution and diversity. | Assane Hamidou Abdoulaye Jichun Jia Aqleem Abbas Du Hai Jiasen Cheng Yanping Fu Yang Lin Daohong Jiang Jiatao Xie | 2022 | Virologica Sinica2022,37,3: | 0 |
