| 1 | Herbal decoctosome is a novel form of medicine显示文摘Traditionally, herbal medicine is consumed by drinking decoctions produced by boiling herbs with water. The functional components of the decoction are heat stable. Small RNAs(sRNAs) were reported as a new class of functional components in decoctions. However, the mechanisms by which sRNAs survive heat treatment of the decoction and enter cells are unclear.Previous studies showed that plant-derived exosome-like nanoparticles(ELNs), which we call botanosomes, could deliver therapeutic reagents in vivo. Here, we report that heat-stable decoctosomes(ELNs) from decoctions have more therapeutic effects than the decoctions in vitro and demonstrate therapeutic efficacy in vivo. Furthermore, sRNAs, such as HJT-sRNA-m7 and PGY-sRNA-6, in the decoctosome exhibit potent anti-fibrosis and anti-inflammatory effects, respectively. Decoctosome is comprised of lipids, chemical compounds, proteins, and s RNAs. A medical decoctosome mimic is called bencaosome. A single lipid sphinganine(d22:0) identified in the decoctosome was mixed and heated with the synthesized sRNAs to form the simplest bencaosome. This simple bencaosome structure was identified by critical micelle concentration(cmc) assay that sRNAs coassembled with sphinganine(d22:0) to form the lipid layers of vesicles. The heating process facilitates co-assembly of sRNAs and sphinganine(d22:0) until a steady state is reached. The artificially produced sphinganine-HJT-sRNA-m7 and sphinganinePGY-sRNA-6 bencaosomes could ameliorate bleomycin-induced lung fibrosis and poly(I:C)-induced lung inflammation, respectively, following oral administration in mice. | 2 | Plant-derived phosphocholine facilitates cellular uptake of anti-pulmonary fibrotic HJT-sRNA-m7显示文摘Pulmonary fibrosis, a progressive chronic disease with a high mortality rate, has limited treatment options. Currently, lung transplantation remains the only effective treatment. Here we report that a small RNA, HJT-sRNA-m7, from a Chinese herbal medicine Hong Jing Tian(HJT, RHODIOHAE CRENULATAE RADIX ET RHIZOMA, Rhodiola crenulata) can effectively reduce the expressions of fibrotic hallmark genes and proteins both in alveolar in vitro and in mouse lung tissues in vivo. We also discovered over one hundred oil-soluble chemicals from HJT decoctions, most of which are found in lipid extracts from other Chinese herbals decoctions, including Pu Gong Ying(PGY, TARAXACI HERBA, Taraxacum mongolicum), Chuan Xin Lian(CXL, changed to 'ANDROGRAPHIS HERBA, Andrographis paniculata'), and Jin Yin Hua(JYH, lonicera japonica or Honeysuckle). We identified the active component in these decoctions as two forms of phosphocholines, PC(18:0/18:2) and PC(16:0/18:2). These PCs potentially could form liposomes with small RNAs to enter human alveolar and gastric cells. | 3 | Large-scale analysis of small RNAs derived from traditional Chinese herbs in human tissues显示文摘Plant-derived microRNAs have recently been reported to function in human blood and tissues. Controversy was immediately raised due to possible contamination and the lack of large sample sizes. Here, we report thousands of unique small RNAs derived from traditional Chinese medicine(TCM) herbs found in human blood cells and mouse lung tissues using a large-scale analysis.We extracted small RNAs from decoctions of 10 TCM plants(Ban Zhi Lian, Chai Hu, Chuan Xin Lian, Di Ding Zi Jin, Huang Qin, Jin Yin Hua, Lian Qiao, Pu Gong Ying, Xia Ku Cao, and Yu Xing Cao) and obtained millions of RNA sequences from each herb. We also obtained RNA-Seq data from the blood cells of humans who consumed herbal decoctions and from the lung tissues of mice administered RNAs from herbal decoctions via oral gavage. We identified thousands of unique small RNA sequences in human blood cells and mouse lung tissues. Some of these identified small RNAs from Chuan Xin Lian and Hong Jing Tian could be mapped to the genomes of the herbs, confirming their TCM plant origin. The laser system, operating with typical pulse durations of 3–5 ns and beam diameter 60 mm, employs a sequence of successive rod amplifiers to achieve 100-J-level energy at 1053 nm at3 ns. The frequency conversion can provide energy of 50-J level at 351 nm. | 9 | Nonlinear short-term heart rate variability prediction of spontaneous ventricular tachyarrhythmia显示文摘As malign ventricular tachyarrhythmias triggering sudden cardiac death (SCD), both ventricular tachycardia (VT) and ventricular fibrillation (VF) are major causes of mortality. The most efficient ther- apy for SCD prevention is implantable cardioverter defibrillators (ICD). The ICD can accurately and ef- fectively identify the forthcoming of fatal ventricular tachyarrhythmias and deliver a shock in order to restore patients’ normal sinus rhythm. In this study, two nonlinear complexity measures based on en- tropy: approximate entropy (ApEn) and sample entropy (SampEn) as well as two time linear indices: the mean RR interval (the average of time intervals between consecutive R-waves) and the standard devia- tion of RR intervals were used for short-term forecasting of VT-VF occurrence. The last small sections of interbeat intervals preceding 135 VT-VF episodes from 78 patients stored by the ICD were analyzed and compared with individually acquired control time series (CON series) from the same patients, which are normally intrinsic sinus rhythms. The results demonstrate that in addition to an obvious in- crease in heart rates of the patients, the values of two entropy measures are significantly smaller for VT-VF episodes than those for CON series. Conclusions can be drawn that when a ventricular tach- yarrhythmia approaches, the sympathetic tone of the patients is increased, and the complexity of their RR intervals immediately before the onset of VT-VF events is obviously lower than that of RR intervals recorded during sinus rhythms. For a better separation, the optimal range of threshold r is determined for two algorithms. | 10 | The Uniform Distribution of SiC Particles in an A356-SiC_p Composite Produced by the Tilt-blade Mechanical Stirring显示文摘The tilt-blade mechanical stirring of A356-2.5vol.%SiCp liquid was conducted in a cylindrical crucible to solve the problem of nonhomogeneous radial distribution of SiC particles in conventional straight-blade mechanical stirring. The radial distribution of SiC particles in A356 liquid was studied under the conditions of 25° for horizontal tilt angle α of the blade, 200 r/min for rotating speed of stirrer and 10 mm/s for speed of moving up and down of stirrer. The results show that there exists a nonlinear relationship between circumferential tilt angle β of the blade and radial relative deviation d of SiCp content in A356 liquid. When β is smaller than 26°, the d of SiCp content in A356 liquid between the center and the periphery of crucible decreases with increasing β. Conversely, when β is bigger than 26 , d increases with increasing β. Only when β is about 26°, d can be equal to nought i.e. uniform radial distribution of SiC particles can be realized.
