| 3 | Bacterial flora concurrent with Helicobacter pylori in the stomach of patients with upper gastrointestinal diseases显示文摘AIM:To investigate the non-Helicobacter pylori(H.pylori) bacterial flora concurrent with H.pylori infection.METHODS:A total of 103 gastric biopsy specimens from H.pylori positive patients were selected for bacterial culture.All the non-H.pylori bacterial isolates were identified by matrix-assisted laser desorption ionization time-of-flight mass spectrometry(MALDI-TOF MS).RESULTS:A total of 201 non-H.pylori bacterial isolates were cultivated from 67(65.0%) of the 103 gastric samples,including 153 isolates identified successfully at species level and 48 at genus level by MALDI-TOF MS.The dominant species were Streptococcus,Neisseria,Rothia and Staphylococcus,which differed from the predominantly acid resistant species reported previously in healthy volunteers.The prevalence of non-H.pylori bacteria was higher in non-ulcer dyspepsia group than in gastric ulcer group(100%vs 42.9%,P < 0.001).Six bacterial species with urease activity(Staphylococcus epidermidis,Staphylococcus warneri,Staphylococcus capitis,Staphylococcus aureus,Brevibacterium spp.and Klebsiella pneumoniae) were also isolated.CONCLUSION:There is a high prevalence of the non-H.pylori bacteria concurrent with H.pylori infection,and the non-H.pylori bacteria may also play important as-yet-undiscovered roles in the pathogenesis of stomach disorders. | Yuan Hu Li-Hua He Di Xiao Guo-Dong Liu Yi-Xin Gu Xiao-Xia Tao Jian-Zhong Zhang | 2012 | World Journal of Gastroenterology2012,18,11: | 20 |
| 4 | Combination of small interfering RNA and lamivudine on inhibition of human B virus replication in HepG2.2.15 cells显示文摘AIM: To observe the inhibition of hepatitis B virus (HBV) replication and expression by combination of siRNA and lamivudine in HepG2.2.15 cells. METHODS: Recombinant plasmid psil-HBV was constructed and transfected into HepG2.2.15 cells. The transfected cells were cultured in lamivudine-containing medium (0.05 μmol/L) and harvested at 48, 72 and 96 h. The concentration of HBeAg and HBsAg was determined using ELISA. HBV DNA replication was examined by real- time PCR and the level of HBV mRNA was measured by RT-PCR. RESULTS: In HepG2.2.15 cells treated with combination of siRNA and lamivudine, the secretion of HBeAg and HBsAg into the supernatant was found to be inhibited by 91.80% and 82.40% (2.89 ± 0.48 vs 11.73 ± 0.38, P < 0.05; 4.59 ± 0.57 vs 16.25 ± 0.48, P < 0.05) at 96 h, respectively; the number of HBV DNA copies within culture medium was also significantly decreased at 96 h (1.04 ± 0.26 vs 8.35 ± 0.33, P < 0.05). Moreover, mRNA concentration in HepG2.2.15 cells treated with combination of siRNA and lamivudine was obviously lower compared to those treated either with siRNA or lamivudine (19.44 ± 0.17 vs 33.27 ± 0.21 or 79.9 ± 0.13, P < 0.05). | 5 | SLIP VELOCITY MODEL OF POROUS WALLS ABSORBED BY HYDROPHOBIC NANOPARTICLES SIO_2显示文摘According to new slip effects on nanopatterned interfaces,the mechanism of enhancing water injection into hydrophobic nanomaterial SiO2 was proposed. When Hydrophobic Nanoparticles(HNPs)are adsorbed on surfaces of porous walls,hydrophobic nanoparticles layers are formed instead of hydrated layer, and slip effects appear on the pore wall when a driving pressure is applied to the rock cores sample. It makes fluid to move more quickly and the flow capacity increases greatly. Experiments on changing wettability of porous walls were conducted, and the phenomenon that porous walls surfaces were adsorbed by nanoparticles was validated with the Environment Scan Electron Microscopy(ESEM). The results of displacement experiments show that flowing resistance is greatly reduced , and water-phase effective permeability is increased by 47 % averagely after being treated by nanofluid. These results indicate that the slip effect may occur on nanoparticle film of porous walls. Based on this new mechanism of enhancing water injection about hydrophobic nanomaterial SiO2,a slip velocity model in uniform porous media was introduced, and some formulas for the ratio of slip length to radius, slip length ,stream slip velocity and flux increment were deduced. and calculated results indicate that the ratio of slip length to radius is about 3.54%-6.97%, and the slip length is about 0.024μm -0.063μm. The mission aims at determining the equation of state of matter at supra-nuclear density, measuring effects of QED, and understanding the dynamics of matter in strong-field gravity. In addition to investigating fundamental physics, eXTP will be a very powerful observatory for astrophysics that will provide observations of unprecedented quality on a variety of galactic and extragalactic objects. | 9 | Effects of anesthetic methods on preserving anti-tumor T-helper polarization following hepatectomy显示文摘AIM:To investigate the impact of different anesthetic techniques on T-helper(Th) cell subsets in hepatocellular carcinoma(HCC) patients undergoing hepatectomy.METHODS:Sixty-one HCC patients who received hepatectomies were randomized into an epidural combined general anesthesia(G + E;n = 31) or a general anesthesia(G;n = 30) group.Blood samples were obtained the morning before the operation(d0),and on the second(d2) and seventh(d7) day after the operation.Th cell contents were evaluated using flow cytometry,realtime reverse transcription-polymerase chain reaction and enzyme-linked immunosorbent assay.RESULTS:In all 61 patients,Th1 and Th2 cell frequencies,and interferon-(IFN-) mRNA expression markedly increased on d2,compared to d0.They recovered slightly on d7,and the Th1/Th2 ratio increased markedly on d7,compared with d2.In contrast,Th17,regulatory T cell(Treg),and interleukin-17(IL-17) levels and FOXP3 mRNA expression showed no significant change on d2,and then markedly decreased on d7.Similarly,plasma IFN-concentration on d2 was much higher than that on d0,and then partly recovered on d7.As compared with the G group,in the G + E group,Th1 cell frequencies and the Th1/Th2 ratio were slightly higher on d2 and significantly higher on d7,while Th2,Th17,and Treg cell frequencies were slightly lower on d2,and significantly lower on d7.Consistently,on d7,IFN-mRNA and protein levels and the IFN-/IL-4 ratio in the G + E group were higher than those in the G group.In contrast,the IL-17 mRNA level,and IL-17 and transforming growth factor-1 concentrations in the G + E group were lower than those in the G group.CONCLUSION:G + E is superior to G in shifting the Th1/Th2 balance towards Th1,while decreasing Th17 and Treg,potentially benefiting HCC patients by promoting anti-tumor Th polarization. | Di Zhou Fang-Ming Gu Qiang Gao Quan-Lin Li Jian Zhou Chang-Hong Miao, | 2012 | World Journal of Gastroenterology2012,18,24: | 11 |
| 10 | Rebalancing of the gut flora and microbial metabolism is responsible for the anti-arthritis effect of kaempferol显示文摘Kaempferol is a natural flavonol that possesses various pharmacological activities,including anti-arthritis effects,yet the underlying mechanisms remain controversial.To evaluate the anti-arthritis efficacy and the underlying mechanisms of kaempferol,collagen-induced arthritis(CIA)mice were treated with kaempferol intragastrically(200 mg·kg^−1·d^−1)and intraperitoneally(20 mg·kg^−1·d^−1).Pharmacodynamic and pharmacokinetic studies showed that the oral administration of kaempferol produced distinct anti-arthritis effects in model mice with arthritis in terms of the spleen index,arthritis index,paw thickness,and inflammatory factors;the bioavailability(1.5%,relative to that of the intraperitoneal injection)and circulatory exposure of kaempferol(Cmax=0.23±0.06 ng/mL)and its primary metabolite kaempferol-3-O-glucuronide(Cmax=233.29±89.64 ng/mL)were rather low.In contrast,the intraperitoneal injection of kaempferol caused marginal anti-arthritis effects,although it achieved a much higher in vivo exposure.The much higher kaempferol content in the gut implicated a potential mechanism involved in the gut.Analysis of 16S ribosomal RNA revealed that CIA caused imbalance of 14 types of bacteria at the family level,whereas kaempferol largely rebalanced the intestinal microbiota in CIA mice.A metabolomics study showed that kaempferol treatment significantly reversed the perturbation of metabolites involved in energy production and the tryptophan,fatty acid and secondary bile acid metabolisms in the gut contents of the CIA mice.In conclusion,we demonstrate for the first time that the high level of kaempferol in the gut regulates the intestinal flora and microbiotic metabolism,which are potentially responsible for the anti-arthritis activities of kaempferol. | Li-xiang Aa Fei Fei Qi Qi Run-bin Sun Sheng-hua Gu Zi-zhen Di Ji-ye Aa Guang-ji Wang Chang-xiao Liu | 2020 | Acta Pharmacologica Sinica2020,41,1: | 9 |

| 13 | Shuanghuanglian injection downregulates nuclear factor-kappa B expression in mice with viral encephalitis显示文摘A mouse model of viral encephalitis was induced by intracranial injection of a Coxsackie virus B3 suspension. Quantitative real-time reverse transcription-PCR and western blot assay were applied to detect mRNA and protein expression of intelectin-2 and nuclear factor-kappa B in the viral encephalitis and control groups. Nuclear factor-kappa B and intelectin-2 mRNA and protein expression were significantly increased in mice with viral encephalitis. After intraperitoneal injection of Shuanghuanglian at a dose of 1.5 mg/kg for 5 successive days, intelectin-2 and nuclear factor-kappa B protein and mRNA expression were significantly decreased. To elucidate the relationship between intelectin-2 and nuclear factor-kappa B, mice with viral encephalitis were administered an intracerebral injection of 107 pfu recombinant lentivirus expressing intelectin shRNA. Both protein and mRNA levels of intelectin and nuclear factor-kappa B in brain tissue of mice were significantly decreased. | 15 | One-pot synthesis of nitrogen and sulfur co-doped activated carbon supported AuCl_3 as efficient catalysts for acetylene hydrochlorination显示文摘Commercialization of acetylene hydrochlorination using AuCl3 catalysts has been impeded by its poor stability.We have been studying that nitrogen-modified Au/NAC catalyst delivered a stable performance which can improve acetylene hydrochlorination activity and has resistance to catalytic deactivation.Here we show that nitrogen and sulfur co-doped activated carbon supported AuCl_3 catalyst worked as efficient catalysts for the hydrochlorination of acetylene to vinyl chloride. Au/NSAC catalyst demonstrated high activity comparative to Au/AC catalyst. Furthermore, it also delivered stable performance within the selectivity of acetylene, reaching more than 99.5%, and there was only a 3.3%C2H2 conversion loss after running for 12 h under the reaction conditions of a temperature of 180 8C and a C_2H_2 hourly space velocity of 1480 h^(-1). | 17 | Ultrasound-triggered noninvasive regulation of blood glucose levels using microgels integrated with insulin nanocapsules显示文摘Diabetes is a serious public health problem affecting 422 million people worldwide. Traditional diabetes management often requires multiple daily insulin injections, associated with pain and inadequate glycemia control. Herein, we have developed an ultrasound-triggered insulin delivery system capable of pulsatile insulin release that can provide both long-term sustained and fast on-demand responses. In this system, insulin-loaded poly(lactic-co-glycolic acid) (PLGA) nanocapsules are encapsulated within chitosan microgels. The encapsulated insulin in nanocapsules can passively diffuse from the nanoparticle but remain restricted within the microgel. Upon ultrasound treatment, the stored insulin in microgels can be rapidly released to regulate blood glucose levels. | 18 | Optical observations of LIGO source GW 170817 by the Antarctic Survey Telescopes at Dome A,Antarctica显示文摘The LIGO detection of gravitational waves(GW) from merging black holes in 2015 marked the beginning of a new era in observational astronomy. The detection of an electromagnetic signal from a GW source is the critical next step to explore in detail the physics involved. The Antarctic Survey Telescopes(AST3),located at Dome A, Antarctica, is uniquely situated for rapid response time-domain astronomy with its continuous night-time coverage during the austral winter. We report optical observations of the GW source(GW 170817) in the nearby galaxy NGC 4993 using AST3. The data show a rapidly fading transient at around 1 day after the GW trigger, with the i-band magnitude declining from 17:23 ± 0:13 magnitude to 17:72 ± 0:09 magnitude in ~1:8 h. The brightness and time evolution of the optical transient associated with GW 170817 are broadly consistent with the predictions of models involving merging binary neutron stars. | 19 | Dark matter direct search sensitivity of the PandaX-4T experiment显示文摘The Panda X-4T experiment, a 4-ton scale dark matter direct detection experiment, is being planned at the China Jinping Underground Laboratory. In this paper we present a simulation study of the expected background in this experiment. In a 2.8-ton fiducial mass and the signal region between 1-10 keV electron equivalent energy, the total electron recoil background is found to be 4.9 × 10^(-5) kg^(-1) d^(-1) keV^(-1). The nuclear recoil background in the same region is 2.8 × 10^(-7) kg^(-1) d^(-1) keV^(-1). | 20 | Activation of β-catenin signaling in aggrecan-expressing cells in temporomandibular joint causes osteoarthritis-like defects显示文摘β-Catenin plays a critical role in cartilage formation and development. To further understand the role of β-catenin in osteoarthritis(OA) development in temporomandibular joint(TMJ), we have generated β-catenin conditional activation mice(β-cat(ex3)^(Agc1CreER))by breeding Agc1-CreER mice with β-catenin^(flox(ex3)/+)mice. Results of histologic analysis showed the progressive TMJ defects in 3-and 6-month-old β-cat(ex3)^(Agc1CreER)mice(tamoxifen induction was performed at 2 weeks of age), including decreased chondrocyte numbers in the superficial layer associated with less Alcian blue staining, increased numbers of hypertrophic chondrocytes in deep layers, and rough articular surface. Compared to the TMJ phenotype of β-cat(ex3)^(Col2CreER)mice, β-cat(ex3)^(Agc1CreER)mice showed much severe morphological defects in the superficial layer of TMJ. This may reflect that Agc1-CreER mice could efficiently target cells in the superficial layer of TMJ. Results of immunostaining showed significantly increased expression of MMP13, Col-X, Adamts4,and Adamts5 in TMJ of β-cat(ex3)^(Agc1CreER)mice. Results of proliferating cell nuclear antigen(PCNA), Ki67, and terminal deoxinucleotidyl transferase-mediated d UTP-fluorescein nick end labeling(TUNEL) staining further demonstrated that cell proliferation was decreased and cell apoptosis was increased in condylar cartilage of β-cat(ex3)^(Agc1CreER)mice.
