| 2 | The Late Carboniferous–Early Permian Ocean-Continent Transition in the West Junggar,Central Asian Orogenic Belt: Constraints from Columnar Jointed Rhyolite显示文摘The West Junggar of the western Central Asian Orogenic Belt is one of the typical regions in the term of ocean subduction, contraction and continental growth in the Late Paleozoic. However, it is still controversial on the exact time of ocean-continent transition so far. This study investigates rhyolites with columnar joint in the West Junggar for the first time.Based on zircon U-Pb dating, we determined that the ages of the newly-discovered rhyolites are between 303.6 and 294.5 Ma, belonging to Late Carboniferous–Early Permian, which is the oldest rhyolite with columnar joint preserved in the world at present. Geochemical results show that the characteristics of the major element compositions include a high content of SiO_2(75.78–79.20 wt%) and a moderate content of Al_2O_3(12.21–13.19 wt%). The total alkali content(K_2O +Na_2O) is 6.14–8.05 wt%, among which K_2O is 2.09–4.72 wt% and the rate of K_2O/Na_2O is 0.38–3.05. Over-based minerals such as Ne, Lc, and Ac do not appear. The contents of TiO_2(0.09–0.24 wt%), CaO(0.15–0.99 wt%) and MgO(0.06–0.18 wt%) are low. A/CNK=0.91–1.68, A/NK=1.06–1.76, and as such, these are associated with the quasi-aluminum-weak peraluminous high potassium calc-alkaline and some calc-alkaline magma series. These rhyolites show a significant negative Eu anomaly with relative enrichment of LREE and LILE(Rb, Ba, Th, U, K) and depletion of Sr, HREE and HFSE(Nb, Ta, Ti, P). These rhyolites also have the characteristics of an A2-type granite, similar to the Miaoergou batholith,which indicates they both were affected by post-orogenic extension. | 4 | Ticagrelor inhibits the NLRP3 inflammasome to protect against inflammatory disease independent of the P2Y_(12 ) signaling pathway显示文摘Ticagrelor is the first reversibly binding oralP2Y_(12) receptor antagonist to inhibit platelet activation and has been approved by the Food and Drug Administration for the treatment of coronary artery disease. At present, the other pharmacological functions of ticagrelor remain poorly understood. The NOD-, LRR-, and pyrin domain-containing protein 3 (NLRP3) inflammasome plays a critical role in the innate immune system, but its excessive activation also contributes to the pathogenesis of complex diseases. In this study, we systematically examined the effects of ticagrelor on the NLRP3 inflammasome and found that ticagrelor inhibits NLRP3 inflammasome activation in macrophages independent of its classic inhibitory effect on theP2Y_(12) signaling pathway. Further mechanistic studies demonstrate that ticagrelor attenuates the oligomerization of apoptosis-associated speck-like protein containing a CARD (ASC) by blocking chloride efflux, an effect achieved through the degradation of chloride intracellular channel proteins (CLICs) and blockade of the translocation of CLICs to the plasma membrane. Moreover, experiments on lipopolysaccharide-induced sepsis and alum-induced peritonitis in mice confirmed that ticagrelor mitigates the severity of systemic inflammation independent ofP2Y_(12) receptor antagonism. Importantly, oral administration of ticagrelor rapidly and strongly inhibited NLRP3 inflammasome activation in peripheral blood mononuclear cells from patients with acute coronary syndrome. | 7 | HER4 promotes the growth and metastasis of osteosarcoma via the PI3K/AKT pathway显示文摘Osteosarcoma is the most common primary malignant bone tumor,which occurs in adolescents.As reported by our previous studies,HER4 indicates a poor prognosis of primary osteosarcoma.However,its mechanisms in the pathogenesis of osteosarcoma have not yet been studied.The purpose of this study was to investigate the role of HER4 in osteosarcoma and whether the PI3K/AKT pathway is involved.In this study,western blot analysis was used to investigate the expression of HER4 protein in osteosarcoma tissues and cell lines.CCK8 and transwell assays were used to detect the effects of HER4 on the proliferation,migration,and invasion of osteosarcoma cells in vitro.The effects of HER4 on the growth and metastasis of osteosarcoma in vivo were detected by tumor formation and immunofluorescence in nude mice.The role of the PI3K/AKT pathway in HER4 regulation of the growth and metastasis of osteosarcoma was examined by western blot analysis and immunofluorescence assay.We found that HER4 protein was highly expressed in clinical osteosarcoma specimens and osteosarcoma cells.HER4 markedly promoted the proliferation,migration,and invasion of osteosarcoma cells in vitro as well as the growth and metastasis of osteosarcoma in vivo.HER4 overexpression upregulated the expression of phosphorylated protein kinase B(pAKT),proliferation marker antigen Ki67,and metastasis cell marker matrix metalloproteinase 9(MMP9).Notably,PI3K/AKT inhibitor LY294002 significantly inhibited the effects of HER4 via the downregulation of pAKT,Ki67,and MMP9.Moreover,LY294002 markedly blocked the effects of HER4-induced upregulation of tum or malignancy.The present study suggests that HER4 may promote the growth and metastasis of osteosarcoma via the PI3K/AKT pathway.The HER4/PI3K/AKT pathway could serve as a potential target for the treatment of osteosarcoma. | Xiaodong Li Qingshan Huang Shenglin Wang Zhen Huang Fengqiang Yu Jianhua Lin | 2020 | Acta Biochimica et Biophysica Sinica2020,52,4: | 3 |

| 20 | Epidermal growth factor receptor promotes tumor progression and contributes to gemcitabine resistance in osteosarcoma显示文摘Osteosarcoma(OS)is the most common type of primary malignant tumors that originate in the bone.Resistance to chemotherapy confers a poor prognosis on OS patients.Dysregulation of the epidermal growth factor receptor(EGFR)signaling has been reported in sarcomas.However,the functional contribution of EGFR hyperactivation to the tumor biology and chemoresistance remains largely unexplored in OS.In this study,we aimed to investigate the role of EGFR in OS progression and in the response of OS to gemcitabine treatment.The EGFR expression was found to be upregulated in fibroblastic OS cell lines.EGFR knockdown suppressed OS cell proliferation,migration,and invasion in vitro and tumor formation in vivo.Conversely,EGFR overexpression promoted the growth and motility of OS cells.In terms of mechanism,the levels of phospho-Akt and phospho-ERK were decreased upon EGFR knockdown but increased as a result of EGFR overexpression,implying a possible involvement of PI3K/Akt and ERK pathways in mediating the effects of EGFR on OS cells.Moreover,the level of phospho-EGFR was increased in OS cells when exposed to gemcitabine treatment.A more profound proliferative inhibition and a higher rate of apoptosis were obtained in OS cells via inducing cell cycle arrest at G1 phase upon gemcitabine treatment combined with EGFR knockdown,as compared to gemcitabine alone.On the contrary,EGFR overexpression counteracted the growth-inhibiting and pro-apoptotic effects of gemcitabine in OS cells.The present study suggests that EGFR promotes tumor progression and contributes to gemcitabine resistance in OS. | Shenglin Wang Hongxiang Wei Zhen Huang Xinwen Wang Rongkai Shen Zhaoyang Wu Jianhua Lin | 2021 | Acta Biochimica et Biophysica Sinica2021,53,3: | 0 |
