1Potential antibacterial activity of berberine against multi drug resistant enterovirulent Escherichia coli isolated from yaks(Poephagus grunniens) with haemorrhagic diarrhoea显示文摘Objective:To evaluate the antimicrobial efficacy of berberine,a plant alkaloid.Methods:Five multi-drug resistant(MDR) STEC/EPEC and five MDR ETEC isolates from yaks with haemorrhagic diarrhoea were selected for the study.Antibacterial activity of berberine was evaluated by broth dilution and disc diffusion methods.The binding kinetics of berberine to DNA and protein was also enumerated.Results:For both categories of enterovirulent Escherichia coli(E.roli) isolates, berberine displayed the antibaclerial effect in a dose dependent manner.The MIC50 of berberine chloride for STEC/EPEC isolates varied from 2.07μM to 3.6μM with a mean of(2.95±0.33)μM where as for ETEC strains it varied from 1.75 to 1.96μM with a mean of(1.87±0.03)μM. 2Why do young people with chronic kidney disease die early?显示文摘Cardiovascular disease poses the greatest risk of premature death seen among patients with chronic kidney disease(CKD).Up to 50% of mortality risk in the dialysis population is attributable to cardiovascular disease and the largest relative excess mortality is observed in younger patients.In early CKD,occlusive thrombotic coronary disease is common,but those who survive to reach end-stage renal failure requiring dialysis are more prone to sudden death attributable mostly to sudden arrhythmic events and heart failure related to left ventricular hypertrophy,coronary vascular calcification and electrolyte disturbances.In this review,we discuss the basis of the interaction of traditional risk factors for cardiovascular disease with various pathological processes such as endothelial dysfunction,oxidative stress,low grade chronic inflammation,neurohormonal changes and vascular calcification and stiffness which account for the structural and functional cardiac changes that predispose to excess morbidity and mortality in young people with CKD.Shankar Kumar Richard Bogle Debasish Banerjee 2014World Journal of Nephrology2014,3,4:11
3New insights into estrogenic regulation of O^6-methylguanine DNA-methyltransferase (MGMT) in human breast cancer cells: Co-degradation of ER-α and MGMT proteins by fulvestrant or O^6-benzylguanine indicates fresh avenues for therapy显示文摘Endocrine therapy using estrogen receptor-α(ER-α) antagonists for attenuating horm2one-driven cell proliferation is a major treatment modality for breast cancers.To exploit any DNA repair deficiencies associated with endocrine therapy,we investigated the functional and physical interactions of ER-α with O^6-methylguanine DNA methyltransferase(MGMT),a unique DNA repair protein that confers tumor resistance to various anticancer alkylating agents.The ER-α-positive breast cancer cell lines(MCF-7,T47D) and ER- negative cell lines(MDAMB-468,MDAMB-231),and established inhibitors of ER-α and MGMT,namely,ICI-182,780(Faslodex) and O^6-benzylguanine,respectively,were used to study MGMT- ER interactions.The MGMT gene promoter was found to harbor one full and two half estrogen-responsive elements(EREs) and two antioxidant-responsive elements(AREs).MGMT expression was upregulated by estrogen,downregulated by tamoxifen in Western blot and promoter-linked reporter assays.Similarly,both transient and stable transfections of Nrf-2(nuclear factor-erythroid 2-related factor-2)increased the levels of MGMT protein and activity 3 to 4-fold reflecting novel regulatory nodes for this drugresistance determinant.Of the different ER-α antagonists tested,the pure anti-estrogen fulvestrant was most potent in inhibiting the MGMT activity in a dose,time and ER-α dependent manner,similar to O^6-benzylguanine.Interestingly,fulvestrant exposure led to a degradation of both ER-α and MGMT proteins and O^6-benzylguanine also induced a specific loss of ER-α and MGMT proteins in MCF-7 and T47 D breast cancer cells with similar kinetics.Immunoprecipitation revealed a specific association of ER-α and MGMT proteins in breast cancer cells.Furthermore,silencing of MGMT gene expression triggered a decrease in the levels of both MGMT and ER-α proteins.The involvement of proteasome in the drug-induced degradation of both proteins was also demonstrated.Fulvestrant enhanced the cytotoxicity of MGMT-targeted alkylating agents,namely,temozolomide and BCNU by 3 to 4-fold in ER-α positive cells,but not in ER-negative cells.We conclude that MGMT and ER-α proteins exist as a complex and are co-targeted for ubiquitin-conjugation and subsequent proteasomal degradation.The findings offer a clear rationale for combining alkylating agents with endocrine therapy.Ameya Paranjpe Nathan I. 4Inversion of radiative transfer model for retrieval of wheat biophysical parameters from broadband reflectance measurements显示文摘This study describes the retrieval of wheat biophysical variables of leaf chlorophyll(Cab),leaf area index(LAI),canopy chlorophyll(CCC),and leaf wetness(Cw)from broadband reflectance data corresponding to IRS LISS-3(Linear Imaging Self Scanner)sensor by inversion of PROSAIL5B canopy radiative transfer model.Reflectance data of wheat crop,grown under different treatments,were measured by hand-held spectroradiometer and later integrated to LISS-3 reflectance using its band-wise relative spectral response function.Three inversion techniques were used and their performance was compared using different statistical parameters and target diagram.The inversion techniques tried were:a look up table with best solution(LUT-I),a look up table with mean of best 10%solutions(LUT-II)and an artificial neural network(ANN).All the techniques could estimate the biophysical variables by capturing variability in their observed values,though accuracy of estimation varied among the three techniques.Target diagram clearly depicted the superiority of LUT-II over the other two approaches indicating that a mean of best 10%solutions is a better strategy while ANN was worst performer showing highest bias for all the parameters.In all the three inversion techniques,the general order of retrieval accuracy was LAI>Cab>CCC>Cw.The range of Cw was very narrow and none of the techniques could estimate variations in it.In most of the cases,the parameters were underestimated by model inversion.The best identified LUT-II technique was then applied to retrieve wheat LAI from IRS LISS-3 satellite image of 5-Feb-2012 in Sheopur district.The comparison with ground observations showed that the RMSE of LAI retrieval was about 0.56,similar to that observed in ground experimentation.The findings of this study may help in refining the protocol for generating operational crop biophysical products from IRS LISS-3 or similar sensors.Vinay Kumar Sehgal Debasish Chakraborty Rabi Narayan Sahoo 2016Information Processing in Agriculture2016,3,2:5

19Grain-size distribution in suspension over a sand-gravel bed in open channel flow显示文摘Grain-size distributions of suspended load over a sand-gravel bed at two different flow velocities were studied in a laboratory flume.The experiments had been performed to study the influence of flow velocity and suspension height on grain-size distribution in suspension over a sand-gravel bed.The experimental findings show that with an increase of flow velocity,the grain-size distribution of suspended load changed from a skewed form to a bimodal one at higher suspension heights.This study focuses on the determination of the parameter β_n which is the ratio of the sediment diffusion coefficient to the momentum diffusion coefficient of n th grain-size.A new relationship has been proposed involving β_n,the normalizing settling velocity of sediment particles and suspension height,which is applicable for widest range of normalizing settling velocity available in literature so far.A similar parameter β for calculating total suspension concentration is also developed.The classical Rouse equation is modified with β_n and β and used to compute grain-size distribution and total concentration in suspension,respectively.The computed values have shown good agreement with the measured values of experimental data.Koeli GHOSHAL Debasish PAL 2014International Journal of Sediment Research2014,29,2:1
