1Efficacy and safety of a novel anti-HER2 therapeutic antibody RC48 in patients with HER2-overexpressing,locally advanced or metastatic gastric or gastroesophageal junction cancer:a single-arm phase II study显示文摘Background:Current treatment options for human epidermal growth factor receptor 2(HER2)-overexpressing gastric cancer at third-line have shown limited clinical benefit.Further,there is no specific treatment for HER2 immunohistochemistry(IHC)2+and fluorescence in-situ hybridization-negative patients.Here,we report the efficacy and safety of a novel anti-HER2 antibody RC48 for patients with HER2-overexpressing,advanced gastric or gastroesophageal junction cancer.Methods:Patients with HER2-overexpressing(IHC 2+or 3+),locally advanced or metastatic gastric or gastroesophageal junction cancer who were under at least second-line therapy were eligible and received RC482.5 mg/kg alone every 2 weeks.The primary endpoint was the objective response rate(ORR)assessed by an independent review committee.Secondary endpoints included progressionfree survival(PFS),overall survival(OS),duration of response,time to progression,disease control rate,and safety.Results:Of 179 patients screened,125 were eligible and received RC48 treatment.The ORR was 24.8%(95%confidence interval[CI]:17.5%-33.3%).The median PFS and OS were 4.1 months(95%CI:3.7-4.9 months)and 7.9 months(95%CI:6.7-9.9 months),respectively.The most frequently reported adverse events were decreased white blood cell count(53.6%),asthenia(53.6%),hair loss(53.6%),decreased neutrophil count(52.0%),anemia(49.6%),and increased aspartate aminotransferase level(43.2%).Serious adverse events(SAEs)occurred in 45(36.0%)patients,and RC48-related SAEs were mainly decreased neutrophil count(3.2%).Seven patients had adverse events that led to death were not RC48-related.Conclusions:RC48 showed promising activity with manageable safety,suggesting potential application in patients with HER2-overexpressing,advanced gastric or gastroesophageal junction cancer who have previously received at least two lines of chemotherapy.Zhi Peng Tianshu Liu Jia Wei Airong Wang Yifu He Liuzhong Yang Xizhi Zhang Nanfeng Fan Suxia Luo Zhen Li Kangsheng Gu Jianwei Lu Jianming Xu Qingxia Fan Ruihua Xu Liangming Zhang Enxiao Li Yuping Sun Guohua Yu Chunmei Bai Yong Liu Jiangzheng Zeng Jieer Ying Xinjun Liang Nong Xu Chao Gao Yongqian Shu Dong Ma Guanghai Dai Shengmian Li Ting Deng Yuehong Cui Jianmin Fang Yi Ba Lin Shen 2021Cancer Communications2021,41,11:29
2African Swine Fever Virus MGF360-12L Inhibits Type Ⅰ Interferon Production by Blocking the Interaction of Importin α and NF-κB Signaling Pathway显示文摘African swine fever(ASF)is an infectious transboundary disease of domestic pigs and wild boar and spreading throughout Eurasia.There is no vaccine and treatment available.Complex immune escape strategies of African swine fever virus(ASFV)are crucial factors affecting immune prevention and vaccine development.MGF360 genes have been implicated in the modulation of the IFN-Ⅰresponse.The molecular mechanisms contributing to innate immunity are poorly understood.In this study,we demonstrated that ASFV MGF360-12 L(MGF360 families 12 L protein)significantly inhibited the mRNA transcription and promoter activity of IFN-βand NF-κB,accompanied by decreases of IRF3,STING,TBK1,ISG54,ISG56 and AP-1 m RNA transcription.Also,MGF360-12 L might suppress the nuclear localization of p50 and p65 mediated by classical nuclear localization signal(NLS).Additionally,MGF360-12 L could interact with KPNA2,KPNA3,and KPNA4,which interrupted the interaction between p65 and KPNA2,KPNA3,KPNA4.We further found that MGF360-12 L could interfere with the NF-κB nuclear translocation by competitively inhibiting the interaction between NF-κB and nuclear transport proteins.These findings suggested that MGF360-12 L could inhibit the IFN-Ⅰproduction by blocking the interaction of importinαand NF-κB signaling pathway,which might reveal a novel strategy for ASFV to escape the host innate immune response.Yisha Zhuo Zeheng Guo Tongtong Ba Cheng Zhang Lihua He Cuiping Zeng Hanchuan Dai 2021Virologica Sinica2021,36,2:21
3Corrosion behavior of Mg-3Gd-1Zn-0.4Zr alloy with and without stacking faults显示文摘To develop biodegradable magnesium alloy with desirable corrosion properties,a low Gd-containing Mg-3Gd-1Zn-0.4Zr(wt%,GZ31K)alloy was prepared.The as-cast ingot was solution treated and then hot extruded.Microstructures were characterized by scanning electron microscopy(SEM).Corrosion behavior of the alloy under each condition was studied by hydrogen evolution and quasi in-situ corrosion methods.It has been found that the as-cast alloy is composed ofα-Mg,stacking faults(SFs)at the outer edge of the matrix grains,and eutectic phase along the grain boundaries.After solution treatment,the SFs disappear and precipitates rich in Zn and Zr elements form in the grain interior and boundaries.The microstructure is significantly refined after extrusion.Hydrogen evolution tests show that the as-cast alloy exhibits the best corrosion resistance,and the solution-treated alloy has the worst corrosion resistance.Corrosion rate of the alloy under each condition decreases first and then increases with prolonging immersion time.Corrosion experiments demonstrate thatα-Mg was corroded preferentially,the eutectic phase and precipitates exhibit better corrosion resistance.The as-extruded alloy demonstrates uniform corrosion due to fine and homogeneous microstructure.Xiaobo Zhang Jianwei Dai Ruifeng Zhang Zhixin Ba Nick Birbilis 2019Journal of Magnesium and Alloys2019,7,2:4
4Influence of chemical liquids on the fatigue crack growth of the AZ31 magnesium alloy显示文摘The fatigue crack growth behavior of an AZ31 magnesium alloy was investigated by comparing the effect of zirconate and phos-phate chemical liquids.The morphology,components,and phase compositions of the chemical depositions at the fatigue crack tip were analyzed by employing scanning electron microscopy(SEM),energy dispersive spectroscopy(EDS),and X-ray diffraction(XRD),respectively.For samples with and without the chemical liquids,their stress-intensity factor values at the fatigue crack tip were compared by using a stress-strain gauge.The results demonstrated that a zirconate film(ZrxOy·ZnxOy) and a phosphate film(Zn3(PO4)2·4H2O and MgZnP2O7) could be formed on the fatigue crack-surface at the fatigue crack tip.The stress distribution was changed because of the chemical depositions and the causticity of the chemical liquids.This could decrease the stress-intensity factor value and thus effectively cause fatigue crack closure,which reduces the fatigue crack growth rate.Moreover,it was found that the fatigue crack closure effect of zirconates was more positive than that of phosphates.Zhang-zhong Wang Xian-cong He Yun-qiang Bai Zhi-xin Ba Yu-ming Dai Heng-zhi Zhou 2012International Journal of Minerals,Metallurgy and Materials2012,19,3:2
5Friction and wear behaviors of biodegradable Mg-6Gd-0.5Zn-0.4Zr alloy under simulated body fluid condition显示文摘The friction and wear behaviors of biodegradable Mg-6Gd-0.5Zn-0.4Zr(wt%,GZ60K)alloy were evaluated under simulated body fluid(SBF)condition using ball-on-disk configuration and compared with those under dry sliding condition.The results show that under dry sliding and SBF conditions,the friction coefficient declines with increasing applied load and keeps stable with prolonging sliding time.The friction coefficient of the alloy effectively decreases in SBF as compared to dry sliding due to lubrication caused by SBF.The real wear rates under SBF condition are lower than those under dry sliding condition for each parameter.Nevertheless,the nominal wear rates are higher in SBF which are attributed to the more mass loss caused by corrosion but not wear.Both the nominal wear rate in SBF and the dry sliding wear rate increase with increasing applied load,and they decline firstly and then keep stable with prolonging sliding time.It is concluded that the wear of the alloy is restricted by the SBF,but the corrosion of the alloy is aggravated by the wear.Jianwei Dai Xiaobo Zhang Qiao Yin Shengnan Ni Zhixin Ba Zhangzhong Wang 2017Journal of Magnesium and Alloys2017,5,4:2
6Protective effect and mechanisms of action of Mongolian medicine Sulongga-4 on pyloric ligation-induced gastroduodenal ulcer in rats显示文摘BACKGROUND Sulongga-4(SL-4)is a herbal formula used in traditional Mongolian medical clinics for the treatment of peptic ulcers and gastroenteritis,even though its pharmacological mechanism has not been well characterized.AIM To evaluate the protective effect and identify the mechanisms of action of SL-4 on gastroduodenal ulcer induced by pyloric ligation(PL)in rats.METHODS PL was performed to induce gastric and duodenal ulcers in rats,which were then treated with oral SL-4(1.3,2.6,or 3.9 g/kg per day)for 15 d.PL-induced gastroduodenal ulceration.Therapeutic effects were characterized by pathological and histological evaluations and inflammatory indicators were analyzed by enzyme-linked immunosorbent assay.Microarray analyses were conducted to identify gene expression profiles of gastroduodenal tissue in PL rats with or without SL-4 treatment.The candidate target genes were selected and verified by quantitative reverse transcription polymerase chain reaction(qRT-PCR).RESULTS SL-4 decreased histopathological features in the PL-induced ulcerated rats.SL-4 significantly (P < 0.05) decreased expression of tumor necrosis factor-α,interleukin (IL)-1β, IL-6, endotoxin, platelet-activating factor, and increasedprostaglandin E2 and epidermal growth factor in ulcer tissue. Microarray analysiswas used to identify a panel of candidate target genes for SL-4 acting on PLinducedulceration. Genes included some complement and coagulation cascadeand retinol metabolism pathways that are closely associated with inflammatoryresponses and gastric mucosal protective mechanisms. qRT-PCR showed thataltered expression of the selected genes, such as CYP2b2, UGT2b1, A2m, andMASP1 was consistent with the microarray results.CONCLUSIONSL-4 exerts protective effects against PL-induced gastroduodenal ulcers viareducing inflammatory cytokines and elevating expression of gastric acidinhibitory factors. 8Composite silicone rubber of high piezoresistance repeatability filled with nanoparticles显示文摘The ruthenium oxide nanoparticles with size less than 20 nm were fabricated by annealing the metallic ruthenium nanoparticles in air,which were synthesized by using the thermal reduction in the polyol solution.The rutile structure of the ruthenium oxide was proved by using transmission electron microscopy(TEM)and X-ray photoelectron spectroscopy(XPS).The oxide has good electron conductivity. The surface of the ruthenium oxide was modified by a vinyl silane coupling agent.The assembling of the silane to the oxide surface was proved by Infrared(IR)absorption spectroscopy.By mixing the nanoparticles with poly(methylvinylsiloxane)(PMVS)silicone rubber,a composite filled with dispersive conducting phase was fabricated.The temperature dependent conductivity shows that the electron transportation through composite is mainly dominated by tunneling.The measurement of piezoresistance shows that the composite at low strain has high piezoresistance repeatability.The 3D reconstruction images of the composite filled with carbon black or ruthenium oxide show that the aggregation of the nanoparticles differs much for two composites.The narrow distribution range of the particle size was thought to be the main factor for the high piezoresistance recurrence.WU JuYing ZHOU ChengXi ZHU QingWen LI EnRong DAI Ge BA Long HUANG YuHong Mei Jun 2009Science China(Technological Sciences)2009,52,12:1

16Effect of γ phase on mechanical behavior and detwinning evolution of directionally solidified Ni-Mn-Ga alloys under uniaxial compression显示文摘Specimens with single-phase martensite, net-like γ/martensite mixed structure and lamella-likeγ/martensite mixed structure were designed to investigate the effect of the γ phase on the mechanical behavior and the detwinning of non-modulated(NM) martensitic variant in Ni-rich Ni-Mn-Ga alloys under uniaxial compression. It can be found the existence of the γ phase significantly enhances the compressive stresses, and the net-like γ phase specimen presents a higher value of compressive strain than that of the lamella-like γ phase specimen. Especially, the detwinning plateau of the lamella-like γ phase specimen is almost invisible due to the martensite colonies with low Schmid factors. Finally, according to the calculation of deformation gradient tensor, we found that the tensors along compression direction(εzz) of net-like γ phase/martensite mixed structure specimen and single-phase martensite specimen are lower than that of the specimen with lamella-like γ phase/martensite mixed structure, which well explained the detwinning strain for these specimens. 17The Role of Renewable Protocatechol Acid in Epoxy Coating Modification: Significantly Improved Antibacterial and Adhesive Properties显示文摘It is of great significance to design epoxy coatings with superior antibacterial properties and high adhesive properties, as well as excellent processing, superior durability, and high transparency. However, it is still a challenge because of the common complex design and synthesis. Herein, the bio-based monomer protocatechuic acid(PCA) was used as raw material, the catechol structure with high bonding and antibacterial properties was introduced into the flexible alkane segment of ethylene glycol diglycidyl ether(EGDE) through an efficient, and green method, and it was cured with isophorone diamine(IPDA) to prepare corresponding thermosets. The cured resins exhibited excellent allaround qualities, particularly in bonding and antibacterial. When 30% PCA was added to pure epoxy resin, the adhesion between substrate and coating increased from 4.40 MPa to 13.60 MPa and the antibacterial rate of coating against E. coli and S. aureus could approach 100%. All of this is due to the fact that the catechol structure present in PCA has the ability to interact with various substrates and alter the permeability of bacterial cell membranes. 18Direction finding for two-dimensional incoherently distributed sources with Hadamard shift invariance in non-uniform orthogonal arrays显示文摘This paper proposes a novel algorithm for Two-Dimensional(2D) central Directionof-Arrival(DOA) estimation of incoherently distributed sources. In particular, an orthogonal array structure consisting of two Non-uniform Linear Arrays(NLAs) is considered. Based on first-order Taylor series approximation, the Generalized Array Manifold(GAM) model can first be established to separate the central DOAs from the original array manifold. Then, the Hadamard rotational invariance relationships inside the GAMs of two NLAs are identified. With the aid of such relationships, the central elevation and azimuth DOAs can be estimated through a search-free polynomial rooting method. Additionally, a simple parameter pairing of the estimated 2D angular parameters is also accomplished via the Hadamard rotational invariance relationship inside the GAM of the whole array. A secondary but important result is a derivation of closed-form expressions of the Cramer-Rao lower bound. The simulation results show that the proposed algorithm can achieve a remarkably higher precision at less complexity increment compared with the existing low-complexity methods, which benefits from the larger array aperture of the NLAs. 20Lomatogonium rotatum extract alleviates diabetes mellitus induced by a high-fat,high-sugar diet and streptozotocin in rats显示文摘BACKGROUND Lomatogonium rotatum(LR)is traditionally used in Mongolian folk medicine as a hypoglycemic agent,but its evidence-based pharmacological effects and mechanisms of action have not been fully elucidated.AIM To emphasize the hypoglycemic action mechanism of LR in a type 2 diabetic rat model and examine potential biomarkers to obtain mechanistic understanding regarding serum metabolite modifications.METHODS A high-fat,high-sugar diet and streptozotocin injection-induced type 2 diabetic rat model was established.The chemical composition of the LR was identified by high performance liquid chromatography.LR extract administrated as oral gavage at 0.5 g/kg,2.5 g/kg,and 5 g/kg for 4 wk.Anti-diabetic effects of LR extract were evaluated based on histopathological examination as well as the measurement of blood glucose,insulin,glucagon-like peptide 1(GLP-1),and lipid levels.Serum metabolites were analyzed using an untargeted metabolomics approach.RESULTS According to a chemical analysis,swertiamarin,sweroside,hesperetin,coumarin,1.7-dihydroxy-3,8-dimethoxyl xanthone,and 1-hydroxy-2,3,5 trimethoxanone are the principal active ingredients in LR.An anti-diabetic experiment revealed that the LR treatment significantly increased plasma insulin and GLP-1 levels while effectively lowering blood glucose,total cholesterol,triglycerides,low-density lipoprotein cholesterol,and oral glucose tolerance test compared to the model group.Furthermore,untargeted metabolomic analysis of serum samples detected 236 metabolites,among which 86 were differentially expressed between the model and the LR group.It was also found that LR considerably altered the levels of metabolites such as vitamin B6,mevalonate-5P,Dproline,L-lysine,and taurine,which are involved in the regulation of the vitamin B6 metabolic pathway,selenium amino acid metabolic pathway,pyrimidine metabolic pathway,and arginine and proline metabolic pathways.CONCLUSION These findings indicated that LR may have a hypoglycemic impact and that its role may be related to changes in the serum metabolites and to facilitate the release of insulin and GLP-1,which lower blood glucose and lipid profiles.Li-Li Dai Sung-Bo Cho Hui-Fang Li Li-Sha A Xiao-Ping Ji Sirigunqiqige Pan Ming-Lan Bao Laxinamujila Bai Gen-Na Ba Ming-Hai Fu 2023World Journal of Diabetes2023,14,6:0
