| 1 | Development of composite PLGA microspheres containing exenatide-encapsulated lecithin nanoparticles for sustained drug release显示文摘This study aimed to prepare poly(D, L-lactic-co-glycolic acid) microspheres(PLGA-Ms)by a modified solid-in-oil-in-water(S/O/W) multi-emulsion technique in order to achieve sustained release with reduced initial burst and maintain efficient drug concentration for a prolonged period of time. Composite PLGA microspheres containing exenatideencapsulated lecithin nanoparticles(Ex-NPs-PLGA-Ms) were obtained by initial fabrication of exenatide-loaded lecithin nanoparticles(Ex-NPs) via the alcohol injection method,followed by encapsulation of Ex-NPs into PLGA microspheres. Compared to Ms prepared by the conventional water-in-oil-in-water(W/O/W) technique(Ex-PLGA-Ms), Ex-NPs-PLGAMs showed a more uniform particle size distribution, reduced initial burst release, and sustained release for over 60 d in vitro. Cytotoxicity studies showed that Ms prepared by both techniques had superior biocompatibility without causing any detectable cytotoxicity.In pharmacokinetic studies, the effective drug concentration was maintained for over 30 d following a single subcutaneous injection of two types of Ms formulation in rats, potentially prolonging the therapeutic action of Ex. | 2 | Comparison of Clinical and Epidemiological Characteristics of Asymptomatic and Symptomatic SARS-CoV-2 Infection in Children显示文摘To understand the epidemiological and clinical features of the symptomatic and asymptomatic pediatric cases of COVID-19,we carried out a prospective study in Shanghai during the period of January 19 to April 30,2020.A total of 49 children(mean age 11.5±5.12 years)confirmed with SARS-CoV-2 infection were enrolled in the study,including 11(22.4%)domestic cases and 38(77.6%)imported cases.Nine(81.8%)local cases and 12(31.6%)imported cases had a definitive epidemiological exposure.Twenty-eight(57.1%)were symptomatic and 21(42.9%)were asymptomatic.Neither asymptomatic nor symptomatic cases progressed to severe diseases.The mean duration of viral shedding for SARS-CoV-2 in upper respiratory tract was 14.1±6.4 days in asymptomatic cases and 14.8±8.4 days in symptomatic cases(P>0.05).Forty-five(91.8%)cases had viral RNA detected in stool.The mean duration of viral shedding in stool was 28.1±13.3 days in asymptomatic cases and 30.8±18.6 days in symptomatic participants(P>0.05).Children<7 years shed viral RNA in stool for a longer duration than school-aged children(P<0.05).Forty-three(87.8%)cases had seropositivity for antibodies against SARS-CoV-2 within 1–3 weeks after confirmation with infection.In conclusion,asymptomatic SARS-CoV-2 infection may be common in children in the community during the COVID-19 pandemic wave.Asymptomatic cases shed viral RNA in a similar pattern as symptomatic cases do.It is of particular concern that asymptomatic individuals are potentially seed transmission of SARS-CoV-2 and pose a challenge to disease control. | Jiehao Cai Xiangshi Wang Jun Zhao Yanling Ge Jin Xu He Tian Hailing Chang Aimei Xia Jiali Wang Jinqiang Zhang Zhongqiu Wei Jingjing Li Chuning Wang Jianshe Wang Qirong Zhu Xiaowen Zhai Mei Zeng | 2020 | Virologica Sinica2020,35,6: | 4 |

The spectra of BSA solution and BSA dry powder were mainly reflected the formation of hydrogen bonds between water and BSA. Further investigation indicated that the concentrations of calcium cations in BSA aqueous solution also affected the secondary structural change of the protein. Amide I band was red shifted and amide II band was blue shifted in aqueous environment compared with the dry BSA powder, and the addition of calcium cations further changed the amide bands position, which led to the change of the secondary structure. | 7 | FTIR-ATR study for adsorption of trypsin in aqueous environment on bare and TiO2 coated ZnSe surfaces显示文摘Undesired adsorption of proteins brings big troubles to marine structures.The settled proteins change the physical and chemical properties of the surfaces,which allow marine fouling organisms to settle down on the structures.Therefore,to understand the adsorption mechanism of proteins is very helpful to find an environment-friendly solution against biofouling.Many approaches have been developed to study protein adsorption,but most of them are insufficient to give the chemical interaction information between proteins and surfaces.Fourier transform infrared spectroscopy with attenuated total reflection(FTIR-ATR)is an efficient,fast and non-destructive method for in situ surface measurement,which greatly minimizes the interference of water to infra red spectra,because of the very small depth of penetration of the evanescent wave.In this paper,an in situ FTIR-ATR technology was used to investigate the adsorption process of trypsin on a bare ZnSe surface and on a TiO2 coated ZnSe surface,and the effect of calcium cation strength and ultraviolet light irradiation on the secondary structure of trypsin were also evaluated.FTIR spectra of trypsin showed that Amide I band red shift and AmideⅡband blue shift in aqueous environment on both surfaces compared with the dry trypsin powder,and the addition of calcium cations further changed the Amide bands position,which indicated that the change of the secondary structure could be interfered by the environment.The hydrogen bond formation between water and trypsin,the interaction between surface and trypsin,the interaction between hydrated calcium cations and trypsin,are major facto rs to change the secondary structure of trypsin,and UV light irradiation also showed its influence for the secondary structure. | Chune Guo Xiaomi Guo Wubo Chu Nan Jiang He Li | 2020 | Chinese Chemical Letters2020,31,1: | 1 |

| 11 | Combined superposition effect of hypertension and dyslipidemia on left ventricular hypertrophy显示文摘Background : Hypertension and dyslipidemia are considered reversible risk factors for cardiovascular disease. The purpose of this study was to explore the impact of traditional and nontraditional blood lipid profiles on the risk of left ventricular hypertrophy(LVH) and to explore the superposition effect of dyslipidemia combined with hypertension.Methods : Data on 9134 participants(53.5 ±10.3 years old) from the Northeast China Rural Cardiovascular Health Study(NCRCHS) were statistically analyzed. The blood lipid profile was measured by total cholesterol(TC), low-density lipoprotein cholesterol(LDL-C), high-density lipoprotein cholesterol(HDL-C), total glyceride(TG), and calculated nontraditional blood lipid indices including non-HDL-C, atherosclerosis index(AI), TC/HDL-C, and residual cholesterol(RC).Results : After the adjustment of age and gender, the odds ratios(ORs) of LVH in patients with hypertension, high LDL-C, high non-HDL-C, high AI, and high TC/HDL-C were 3.97(3.31– 4.76), 1.27(1.02– 1.59), 1.21(1.04– 1.39), 1.33(1.15– 1.53), and 1.42(1.22– 1.65), respectively. After full adjustment of potential confounding factors, high AI and TC/HDL-C were associated with LVH rather than traditional blood lipid indices.The combination of hypertension and nontraditional dyslipidemia(defined by high AI and TC/HDL-C) was associated with the highest risk of LVH, especially in participants under 45 years of age. It was significant to understand the bio-interaction between the microorganisms and the surface. Attenuated total reflection(ATR) Fourier transform infrared(FTIR) spectroscopy was used to study the initial stages of marine microorganism adhering to surfaces, because it could probe the microorganism interaction to the surface regardless of the water interference. Bacillus sp. and Escherichia coli were selected to study the initial attachment on different surfaces, because they were typical fouling microorganisms and showed opposite Gram stain results. The assays were conducted respectively in dried and settled bacteria on two different surfaces(ZnSe, Al2 O3 coated on ZnSe). IR spectra of settled bacteria showed amide I band red shift and amide II band blue shift in aqueous environment on both surfaces compared with the dry bacteria. The reasons of amide bands shift were investigated and it was discovered that the hydrogen bond between the water and the protein of the bacteria led to the protein secondary structure change.
