| 1 | Diversity characterization and association analysis of agronomic traits in a Chinese peanut (Arachis hypogaea L.) mini-core collection显示文摘Association mapping is a powerful approach for exploring the molecular basis of phenotypic variations in plants.A peanut(Arachis hypogaea L.)mini-core collection in China comprising 298 accessions was genotyped using 109 simple sequence repeat(SSR)markers,which identified 554 SSR alleles and phenotyped for 15 agronomic traits in three different environments,exhibiting abundant genetic and phenotypic diversity within the panel.A model-based structure analysis assigned all accessions to three groups.Most of the accessions had the relative kinship of less than 0.05,indicating that there were no or weak relationships between accessions of the mini-core collection.For 15 agronomic traits in the peanut panel,generally the Q t K model exhibited the best performance to eliminate the false associated positives compared to the Q model and the general linear model-simple model.In total,89SSR alleles were identified to be associated with 15 agronomic traits of three environments by the Q t K model-based association analysis.Of these,eight alleles were repeatedly detected in two or three environments,and 15 alleles were commonly detected to be associated with multiple agronomic traits.Simple sequence repeat allelic effects confirmed significant differences between different genotypes of these repeatedly detected markers.Our results demonstrate the great potential of integrating the association analysis and marker-assisted breeding by utilizing the peanut mini-core collection. | Huifang Jiang Li Huang Xiaoping Ren Yuning Chen Xiaojing Zhou Youlin Xia Jiaquan Huang Yong Lei Liying Yan Liyun Wan Boshou Liao | 2014 | Journal of Integrative Plant Biology2014,56,2: | 14 |
| 2 | Image formation of brain function in patients suffering from knee osteoarthritis treated with moxibustion显示文摘OBJECTIVES:Functional magnetic resonance imaging(fMRI) technology was used to study changes to the resting state blood flow in the brains of patients with knee osteoarthritis(KOA) before and after treatment with moxibustion at the acupoint of the left Dubi(ST 35) and to probe the cerebral mechanism underlying the effect of moxibustion.METHODS:The resting state brain function of 30 patients with left KOA was scanned with fMRI before and after treatment with moxibustion.The analytic methods of fractional amplitude of low frequency fluctuation(fALFF) and regional homogeneity(ReHo) were used to observe changes in resting state brain function.RESULTS:The fALFF values of the right cerebrum,extra-nucleus,left cerebellum,left cerebrum and white matter of patients after moxibustion treatment were higher than before treatment,and the fALFF values of the precentral gyrus,frontal lobe and occipital lobe were lower than before treatment(P<0.05,K≥85).The ReHo values of the thalamus,extra-nucleus and parietal lobe of patients were much higher than those before moxibustion treatment,and the ReHo values of the right cerebrum,left cerebrum and frontal lobe were lower than before treatment(P<0.05,K≥85).CONCLUSION:The influence of moxibustion on obvious changes in brain regions basically conforms to the way that pain and warmth is transmitted in the body,and the activation of sensitive systems in the body may be objective evidence of channel transmission.The regulation of brain function by moxibustion is not in a single brain region but rather in a network of many brain regions. | Hongwu Xie Fangming Xu Rixin Chen Tianyou Luo Mingren Chen Weidong Fang Fajin Lü Fei Wu Yune Song Jun Xiong | 2013 | Journal of Traditional Chinese Medicine2013,33,2: | 7 |
| 3 | Correlation between muscular strength and basal nuclei ischemic/hemorrhagic stroke-induced corticospinal tract injury,as detected by diffusion tensor imaging and tractography显示文摘BACKGROUND:Conventional neuroimaging diagnosis does not assist with the monitoring or evaluation of basal nuclei ischemic and hemorrhagic stroke,or motor functional recovery.Magnetic resonance,diffusion tensor imaging,and diffusion tensor tractography have all been used to observe features of cerebral white matter fibrous structures.In addition,diffusion tensor tractography is the only non-invasive imaging method to display the corticospinal tract in vivo.OBJECTIVE:To evaluate the impairment degree of corticospinal tract induced by basal nuclei ischemic and hemorrhagic stroke through the use of magnetic resonance,diffusion tensor imaging,and diffusion tensor tractography,and to analyze the correlation to muscular strength.DESIGN,TIME AND SETTING:A retrospective case analysis was performed at the Department of Medical Imaging,Neurology and Neurosurgery,Fuzhou General Hospital of Nanjing Military Area Command of Chinese PLA between November 2002 and June 2008.PARTICIPANTS:A total of 15 patients with acute or subacute cerebral ischemic stroke and nine with hemorrhagic stroke in the basal nuclei were selected.METHODS:Magnetic resonance,diffusion tensor imaging,and diffusion tensor tractography results and data were analyzed.Fractional anisotropy and directionally encoded color maps were obtained.Three-dimensional tractography of bilateral corticospinal tract was created,and corticospinal tract integrity was graded.Fractional anisotropy of infarct region and corresponding contralateral normal regions were measured,and hematoma volume in hemorrhagic stroke patients was determined.Hand motor function ability was evaluated using Brunstorm criteria.MAIN OUTCOME MEASURES:Fractional anisotropy of infarct region and corresponding contralateral normal regions;hematoma volume in hemorrhagic stroke patients;correlation between muscular strength and corticospinal tract impairment degree in ischemic stroke and hemorrhagic stroke patients before and after treatment.RESULTS:In ischemic stroke patients,the fractional anisotropy value was significantly lower in the infarct area of white matter than in the normal hemisphere(P < 0.01).The impairment degree of corticospinal tract negatively correlated with muscular strength of the corresponding hand(r =-0.97 P < 0.01).The hematoma volume of hemorrhagic stroke patients significantly negatively correlated with Spearman test results for muscular strength of the corresponding hand(r =-0.88,P < 0.01).CONCLUSION:Corticospinal tract impairment severity negatively correlated with muscular strength and motor functional recovery,which suggested that diffusion tensor imaging and diffusion tensor tractography could be used to evaluate corticospinal tract motor function. | Shangwen Xu Ziqian Chen Jinhua Chen Youqiang Ye Yuning Lin Ping Ni Hui Xiao Gennian Qian | 2010 | Neural Regeneration Research2010,5,13: | 7 |
| 4 | AFF4 regulates osteogenic differentiation of human dental follicle cells显示文摘As a member of the AFF(AF4/FMR2)family,AFF4 is a transcription elongation factor that is a component of the super elongation complex.AFF4 serves as a scaffolding protein that connects transcription factors and promotes gene transcription through elongation and chromatin remodelling.Here,we investigated the effect of AFF4 on human dental follicle cells(DFCs)in osteogenic differentiation.In this study,we found that small interfering RNA-mediated depletion of AFF4 resulted in decreased alkaline phosphatase(ALP)activity and impaired mineralization.In addition,the expression of osteogenic-related genes(DLX5,SP7,RUNX2 and BGLAP)was significantly downregulated.In contrast,lentivirus-mediated overexpression of AFF4 significantly enhanced the osteogenic potential of human DFCs.Mechanistically,we found that both the mRNA and protein levels of ALKBH1,a critical regulator of epigenetics,changed in accordance with AFF4 expression levels.Overexpression of ALKBH1 in AFF4-depleted DFCs partially rescued the impairment of osteogenic differentiation.Our data indicated that AFF4 promoted the osteogenic differentiation of DFCs by upregulating the transcription of ALKBH1. | Qingyue Xiao Yuning Zhang Xingying Qi Yaqian Chen Rui Sheng Ruoshi Xu Quan Yuan Chenchen Zhou | 2020 | International Journal of Oral Science2020,12,4: | 4 |

| 7 | Single-/fused-band dual-mode mid-infrared imaging with colloidal quantum-dot triple-junctions显示文摘Image data acquired with fused multispectral information can be used for effective identification and navigation owing to additional information beyond human vision,including thermal distribution,night vision,and molecular composition.However,the construction of photodetectors with such capabilities is hindered by the structural complexity arising from the integration of multiple semiconductor junctions with distinct energy gaps and lattice constants.In this work,we develop a colloidal quantum-dot dual-mode detector capable of detecting,separating,and fusing photons from various wavelength ranges.Using three vertically stacked colloidal quantum-dot homojunctions with alternating polarity,single-band short-wave infrared imaging and fused-band imaging(short-wave and mid-wave infrared)can be achieved with the same detector by controlling bias polarity and magnitude.The dual-mode detectors show detectivity up to 8×10^(10)Jones at the fused-band mode and 3.1×10^(11)Jones at the single-band mode,respectively.Without image post-processing algorithms,the dual-mode detectors could provide both night vision and thermal information-enhanced night vision imaging capability.To the best of our knowledge,this is the first colloidal quantum-dot detector that can achieve such functionality.The operation mode can be changed at a high frequency up to 1.7 MHz,making it possible to achieve simultaneously dual-mode imaging and remote temperature sensing. | SHUO ZHANG GE MU JIE CAO YUNING LUO QUN HAO MENGLU CHEN YIMEI TAN PENGFEI ZHAO XIN TANG | 2022 | Photonics Research2022,10,8: | 3 |

| 10 | Efficient and high-fidelity base editor with expanded PAM compatibility for cytidine dinucleotide显示文摘Cytidine base editor(CBE),which is composed of a cytidine deaminase fused to Cas9 nickase,has been widely used to induce C-to-T conversions in a wide range of organisms.However,the targeting scope of current CBEs is largely restricted to protospacer adjacent motif(PAM)sequences containing G,T,or A bases.In this study,we developed a new base editor termed“nNme2-CBE”with excellent PAM compatibility for cytidine dinucleotide,significantly expanding the genome-targeting scope of CBEs.Using nNme2-CBE,targeted editing efficiencies of 29.0%-55.0%and 17.3%—52.5%were generated in human cells and rabbit embryos,respectively.In contrast to conventional nSp-CBE,the nNme2-CBE is a natural high-fidelity base editing platform with minimal DNA off-targeting detected in vivo.Significantly increased efficiency in GC context and precision were determined by combining nNme2Cas9 with rationally engineered cytidine deaminases.In addition,the Founder rabbits with accurate single-base substitutions at Fgf5 gene loci were successfully generated by using the nNme2-CBE system.These novel nNme2-CBEs with expanded PAM compatibility and high fidelity will expand the base editing toolset for efficient gene modification and therapeutic applications. | Zhiquan Liu Siyu Chen Yingqi Jia Huanhuan Shan Mao Chen Yuning Song Liangxue Lai Zhanjun Li | 2021 | Science China(Life Sciences)2021,64,8: | 2 |

| 15 | Identification of major QTL for seed number per pod on chromosome A05 of tetraploid peanut(Arachis hypogaea L.)显示文摘The inheritance of pod-and seed-number traits(PSNT) in peanut(Arachis hypogaea L.) is poorly understood. In the present study, a recombinant inbred line(RIL) population of 188 lines was used to map quantitative trait loci(QTL) for number of seeds per pod(NSP),number of pods per plant(NPP), and numbers of one-, two-, and three-seeded pods per plant(N1 PP, N2 PP, and N3 PP) in four environments. A total of 28 consensus QTL and 14 single QTL were identified, including 11 major and stable QTL. Four major and stable QTL including qN3 PPA5.2, q N3 PPA5.4, qN3 PPA5.5, and qN3 PPA5.7 each explained 12.3%–33.0% of phenotype variation. By use of another integrated linkage map for the A5 group(hereafter referred to as INT A5 group), QTL for PSNT were located in seven intervals of 0.73–9.68 Mb in length on chromosome A05, and candidate genes underlying N3 PP were suggested. These findings shed light on the genetic basis of PSNT.
