| 1 | A complete sequence and comparative analysis of a SARS-associated virus(Isolate BJ01)显示文摘The genome sequence of the Severe Acute Respiratory Syndrome (SARS)-associated virus provides essential information for the identification of pathogen(s), exploration of etiology and evolution, interpretation of transmission and pathogenesis, development of diagnostics, prevention by future vaccination, and treatment by developing new drugs. We report the complete genome sequence and comparative analysis of an isolate (BJ01) of the coronavirus that has been recognized as a pathogen for SARS. The genome is 29725 nt in size and has 11 ORFs (Open Reading Frames). It is composed of a stable region encoding an RNA-dependent RNA polymerase (composed of 2 ORFs) and a variable region representing 4 CDSs (coding sequences) for viral structural genes (the S, E, M, N proteins) and 5 PUPs (putative uncharacterized proteins). Its gene order is identical to that of other known coronaviruses. The sequence alignment with all known RNA viruses places this virus as a member in the family of Coronaviridae. Thirty putative substitutions have been identified by comparative analysis of the 5 SARS- associated virus genome sequences in GenBank. Fifteen of them lead to possible amino acid changes (non-synonymous mutations) in the proteins. Three amino acid changes, with predicted alteration of physical and chemical features, have been detected in the S protein that is postulated to beinvolved in the immunoreactions between the virus and its host. Two amino acid changes have been detected in the Mprotein, which could be related to viral envelope formation. Phylogenetic analysis suggests the possibility of non-human origin of the SARS-associated viruses but provides noevidence that they are man-made. | 3 | Baicalin prevents LPS-induced activation of TLR4/NF-κB p65 pathway and inflammation in mice via inhibiting the expression of CD14显示文摘Previous studies have shown that baicalin,an active ingredient of the Chinese traditional medicine Huangqin,attenuates LPS-induced inflammation by inhibiting the activation of TLR4/NF-κBp65 pathway,but how it affects this pathway is unknown.It has been shown that CD14 binds directly to LPS and plays an important role in sensitizing the cells to minute quantities of LPS via chaperoning LPS molecules to the TLR4/MD-2 signaling complex.In the present study we investigated the role of CD14 in the anti-inflammatory effects of baicalin in vitro and in vivo.Exposure to LPS(1μg/mL)induced inflammatory responses in RAW264.7 cells,evidenced by marked increases in the expression of MHC II molecules and the secretion of NO and IL-6,and by activation of MyD88/NF-κB p65 signaling pathway,as well as the expression of CD14 and TLR4.These changes were dose-dependently attenuated by pretreatment baicalin(12.5–50μM),but not by baicalin post-treatment.In RAW264.7 cells without LPS stimulation,baicalin dose-dependently inhibit the protein and mRNA expression of CD14,but not TLR4.In RAW264.7 cells with CD14 knockdown,baicalin pretreatment did not prevent inflammatory responses and activation of MyD88/NF-κB p65 pathway induced by high concentrations(1000μg/mL)of LPS.Furthermore,baicalin pretreatment also inhibited the expression of CD14 and activation of MyD88/NF-κB p65 pathway in LPS-induced hepatocyte-derived HepG2 cells and intestinal epithelial-derived HT-29 cells.In mice with intraperitoneal injection of LPS and in DSS-induced UC mice,oral administration of baicalin exerted protective effects by inhibition of CD14 expression and inflammation.Taken together,we demonstrate that baicalin pretreatment prevents LPS-induced inflammation in RAW264.7 cells in CD14-dependent manner.This study supports the therapeutic use of baicalin in preventing the progression of LPS-induced inflammatory diseases. | Ya-jun Fu Bo Xu Shao-wei Huang Xia Luo Xiang-liang Deng Shuang Luo Chang Liu Qing Wang Jin-yan Chen Lian Zhou | 2021 | Acta Pharmacologica Sinica2021,42,1: | 31 |

| 6 | Insight-HXMT observations of the first binary neutron star merger GW170817显示文摘Finding the electromagnetic(EM) counterpart of binary compact star merger, especially the binary neutron star(BNS) merger,is critically important for gravitational wave(GW) astronomy, cosmology and fundamental physics. On Aug. 17, 2017,Advanced LIGO and Fermi/GBM independently triggered the first BNS merger, GW170817, and its high energy EM counterpart,GRB 170817 A, respectively, resulting in a global observation campaign covering gamma-ray, X-ray, UV, optical, IR, radio as well as neutrinos. The High Energy X-ray telescope(HE) onboard Insight-HXMT(Hard X-ray Modulation Telescope) is the unique high-energy gamma-ray telescope that monitored the entire GW localization area and especially the optical counterpart(SSS17 a/AT2017 gfo) with very large collection area(~1000 cm^2) and microsecond time resolution in 0.2-5 MeV. In addition,Insight-HXMT quickly implemented a Target of Opportunity(ToO) observation to scan the GW localization area for potential X-ray emission from the GW source. Although Insight-HXMT did not detect any significant high energy(0.2-5 MeV) radiation from GW170817, its observation helped to confirm the unexpected weak and soft nature of GRB 170817 A. Meanwhile,Insight-HXMT/HE provides one of the most stringent constraints(~10^(-7) to 10^(-6) erg/cm^2/s) for both GRB170817 A and any other possible precursor or extended emissions in 0.2-5 MeV, which help us to better understand the properties of EM radiation from this BNS merger. | 8 | Molybdenum recovery from oxygen pressure water leaching residue of Ni-Mo ore显示文摘This article investigated molybdenum recovery from oxygen pressure water leaching residue of Ni-Mo ore using alkaline leaching, followed by chemical treatment of leach liquor. Parameters affecting Mo leaching rate, such as sodium hydroxide concentration, reaction time, a liquid-to-solid ratio, and temperature for the preliminary alkaline leaching were experimentally determined. The results showed that more than 88% of molybdenum was leached under the optimum conditions (2.5 ml·g-1 NaOH, 80 °C, a liquid to solid ratio 3 ml·g-1 , and reaction time 3 h). After the purification of leach liquor, a CaMoO 4 product of 99.2% purity could be obtained by CaCl 2 precipitation method. | 9 | Selective laser melting(SLM)of CX stainless steel:Theoretical calculation,process optimization and strengthening mechanism显示文摘In the present work,selective laser melting(SLM)technology was utilized for manufacturing CX stainless steel samples under a series of laser parameters.The effect of laser linear energy density on the microstructure characteristics,phase distribution,crystallographic orientation and mechanical properties of these CX stainless steel samples were investigated theoretically and experimentally via scanning electron microscope(SEM),X-ray diffraction(XRD),electron backscatter diffraction(EBSD)and transmission electron microscope(TEM).Based on the systematic study,the SLM CX stainless steel sample with best surface roughness(Ra=4.05±1.8μm)and relative density(Rd=99.72%±0.22%)under the optimal linear density(η=245 J/m)can be obtained.SLM CX stainless steel was primarily constituted by a large number of fine martensite(α’phase)structures(i.e.,cell structures,cellular dendrites and blocky grains)and a small quantity of austenite(γphase)structures.The pre ferred crystallographic orientation(i.e.,<111>direction)can be determined in the XZ plane of the SLM CX sample.Furthermore,under the optimal linear energy density,the good combinations with the highest ultimate tensile strength(UTS=1068.0%±5.9%)and the best total elongation(TE=15.70%±0.26%)of the SLM CX sample can be attained.Dislocation strengthening dominates the strengthening mechanism of the SLM CX sample in as-built state. | Dongdong Dong Cheng Chang Hao Wang Xingchen Yan Wenyou Ma Min Liu Sihao Deng Julien Gardan Rodolphe Bolot Hanlin Liao | 2021 | Journal of Materials Science & Technology2021,,14: | 11 |

| 11 | The High Energy X-ray telescope (HE) onboard the Insight-HXMT astronomy satellite显示文摘The Insight-Hard X-ray Modulation Telescope(Insight-HXMT) is a broadband X-ray and γ-ray(1-3000 ke V) astronomy satellite. One of its three main telescopes is the High Energy X-ray telescope(HE). The main detector plane of HE comprises 18 Na I(Tl)/Cs I(Na) phoswich detectors, where Na I(Tl) is used as the primary detector to measure ~ 20-250 ke V photons incident from the field of view(FOV) defined by collimators, and Cs I(Na) is used as the active shielding detector to Na I(Tl) by pulse shape discrimination. Additionally, Cs I(Na) is used as an omnidirectional γ-ray monitor. The HE collimators have a diverse FOV,i.e. 1.1°×5.7°(15 units), 5.7°×5.7°(2 units), and blocked(1 unit). Therefore, the combined FOV of HE is approximately5.7°×5.7°. Each HE detector has a diameter of 190 mm resulting in a total geometrical area of approximately 5100 cm2, and the energy resolution is ~15% at 60 ke V. For each recorded X-ray event by HE, the timing accuracy is less than 10 μs and the deadtime is less than 10 μs. HE is used for observing spectra and temporal variability of X-ray sources in the 20-250 ke V band either by pointing observations for known sources or scanning observations to unveil new sources. Additionally, HE is used for monitoring the γ-ray burst in 0.2-3 Me V band. | 12 | Origin and Evolution of Core Components Responsible for Monitoring Light Environment Changes during Plant Terrestrialization显示文摘Light serves as the source of energy as well as an information signal for photosynthetic plants. During evolution, plants have acquired the ability to monitor environmental light radiation and adjust their developmental patterns to optimally utilize light energy for photosynthesis. The mechanisms of light perception and signal transduction have been comprehensively studied in past decades, mostly in a few model plants, including Arabidopsis thaliana. However, systematic analyses of the origin and evolution of core components involved in light perception and signaling are still lacking. In this study, we took advantage of the recently sequenced genomes and transcriptomes covering all the main Archaeplastida clades in the public domain to identify orthologous genes of core components involved in light perception and signaling and to reconstruct their evolutionary history. Our analyses suggested that acclimation to different distribution of light quality in new environments led to the origination of specific light signaling pathways in plants. The UVR8 (UV Resistance Locus 8) signaling pathway originated during the movement of plants from the deeper sea to shallow water and enabled plants to deal with ultraviolet B light (UV-B). After acquisition of UV-B adaptation, origination of the phytochrome signaling pathway helped plants to colonize water surface where red light became the prominent light energy source. The seedling emergence pathway, which is mediated by a combination of light and phytohormone signals that orchestrate plant growth pattern transitions, originated before the emergence of seed plants. | 13 | Organic amendments increase corn yield by enhancing soil resilience to climate change显示文摘A 22-year field experiment was conducted in Gongzhuling, Jilin province, China to investigate corn yield response to fertilization practice. Compared to an unfertilized control(CK), all fertilization treatments, including inorganic nitrogen fertilizer only(N), balanced inorganic fertilizers(NPK), NPK plus corn straw(SNPK), and NPK plus farmyard manure(MNPK), resulted in significant increases in corn yield. However, only organic matter amendments sustained increasing yield trends, with annual rates of 0.137 and 0.194 t ha-1for the SPNK and MNPK treatments, respectively(P < 0.05). During the 22 years, the daily mean, maximum and minimum temperatures increased by 0.50, 0.53, and 0.46 °C per decade, whereas precipitation displayed no significant change but showed large seasonal variation. According to a regression analysis, increased air temperature exerted positive effects on corn yields under the SNPK and the MNPK treatments. Under both treatments,soil organic carbon contents and soil nutrient availabilities increased significantly compared to their initial levels in 1990, whereas soil bulk density and total porosity changed slightly under the two treatments, which showed higher soil water storage than other treatments. In contrast, significant increases in soil bulk density and decreases in soil total porosity and soil nutrient availability were observed under the CK, N and NPK treatments. The contributions of soil fertility to corn yield were 28.4%, 37.9%, 38.4%, 39.0%,and 42.9% under CK, N, NPK, SNPK, and MNPK treatments, respectively, whereas climate changes accounted for 27.0%, 14.6%, 12.4%, 11.8%, and 10.8%. Abnormal expression of CMTM is associated with the development of various diseases. This article summarizes the relevant research on the role of the CMTM family in immune disorders. This information will increase our understanding of pathogenesis and identify promising targets for the diagnosis and treatment of autoimmune diseases. The CMTM family is highly expressed in peripheral blood mononuclear cells. CKLF1 may be involved in the development of arthritis through its interaction with C-C chemokine receptor 4. CKLF1 is associated with the pathogenesis of lupus nephritis and psoriasis. Both CMTM4 and CMTM5 are associated with the pathogenesis of systemic lupus erythematosus. CMTM1, CMTM2, CMTM3, and CMTM6 play a role in rheumatoid arthritis, systemic sclerosis, Sj?gren syndrome, and anti-phospholipid syndrome, respectively. The CMTM family has been implicated in various autoimmune diseases.
