| 1 | GDNF基因修饰神经干细胞移植治疗大鼠帕金森病显示文摘目的探讨孕14~15 d胎鼠中脑来源神经干细胞(m NSCs)被胶质源性神经营养因子(GDNF)基因修饰并移植于帕金森病模型大鼠纹状体区的治疗作用。方法取孕14~15 d胎鼠解剖显微镜下分离中脑腹侧组织进行m NSCs培养,培养5 d后行GFP/GDNF基因修饰。PD大鼠模型的建立参照大鼠脑立体定向图谱,立体定向中脑背盖腹侧区、内侧前脑束注射6-羟多巴胺。将PD大鼠随机分组,将基因修饰的干细胞以及无基因修饰干细胞移植到帕金森病大鼠纹状体区,建立空白m NSCs(PBS替代)移植组、GFP基因修饰m NSCs移植组、GDNF基因修饰m NSCs移植组3个实验组。移植细胞前后进行行为学评估,以腹腔注射阿朴吗啡(APO 0.5 mg/kg)诱导PD大鼠旋转圈数作为细胞移植的治疗作用评估手段。免疫荧光组织化学鉴定移植细胞存活、迁移和分化。结果 GDNF基因修饰神经干细胞移植组较对照组和GFP基因修饰神经干细胞移植组有明显的行为改善;移植56 d后,GDNF基因修饰m NSCs移植组较其他两组检测到更多细胞存活,向多巴胺能神经元分化较其他两组都有增加趋势。结论GDNF基因修饰m NSCs移植可显著改善PD大鼠的运动障碍,其分子机制有待进一步研究。 | 段奎甲 王向鹏 杨智勇 王波 王明国 张海龙 邓兴力 | 2016 | 南方医科大学学报2016,36,1: | 14 |
| 2 | 骨髓间充质干细胞联合依达拉奉移植脑梗死大鼠血清炎性因子的表达显示文摘背景:抑制血清炎性因子凝血酶元激活因子的抑制因子1和组织纤维溶酶原激活物表达是治疗脑梗死的潜在靶点之一。目的:观察骨髓间充质干细胞移植联合依达拉奉对脑梗死大鼠血清炎性因子的表达变化。方法:60只SD大鼠采用大脑中动脉线栓法复制局灶性脑梗死模型后,随机等分为模型组、骨髓间充质干细胞移植组和联合组。模型组连续5 d尾静脉注射PBS,骨髓间充质干细胞移植组经尾静脉注射2.0×109 L-1骨髓间充质干细胞悬液1 mL,2次/d,连续5 d。联合组静脉滴注依达拉奉30 mg+尾静脉注射2.0×109 L-1骨髓间充质干细胞悬液1 mL(2次/d),连续5 d。结果与结论:(1)与模型组相比,骨髓间充质干细胞移植组与联合组大鼠m NSS评分较低,血清凝血酶元激活因子的抑制因子1表达水平减少,脑梗死面积减少,脑组织中肿瘤坏死因子αmR NA的表达水平减少;(2)联合组大鼠上述指标变化差异比骨髓间充质干细胞移植组大;(3)结果提示骨髓间充质干细胞移植联合依达拉奉有助于促进脑梗死损伤神经功能的恢复。 | 宋乃光 孙晶晶 张耀龙 孟令海 高书焕 薛建 孙彩月 | 2016 | 中国组织工程研究2016,20,28: | 9 |
| 3 | Substance P combined with epidermal stem cells promotes wound healing and nerve regeneration in diabetes mellitus显示文摘Exogenous substance P accelerates wound healing in diabetes,but the mechanism remains poorly understood.Here,we established a rat model by intraperitoneally injecting streptozotocin.Four wounds(1.8 cm diameter) were drilled using a self-made punch onto the back,bilateral to the vertebral column,and then treated using amniotic membrane with epidermal stem cells and/or substance P around and in the middle of the wounds.With the combined treatment the wound-healing rate was 100% at 14 days.With prolonged time,type I collagen content gradually increased,yet type III collagen content gradually diminished.Abundant protein gene product 9.5-and substance P-immunoreactive nerve fibers regenerated.Partial nerve fiber endings extended to the epidermis.The therapeutic effects of combined substance P and epidermal stem cells were better than with amniotic membrane and either factor alone.Our results suggest that the combination of substance P and epidermal stem cells effectively contributes to nerve regeneration and wound healing in diabetic rats. | Fei-bin Zhu Xiang-jing Fang De-wu Liu Ying Shao Hong-yan Zhang Yan Peng Qing-ling Zhong Yong-tie Li De-ming Liu | 2016 | Neural Regeneration Research2016,11,3: | 3 |

| 5 | 骨髓间充质干细胞移植对脑梗死大鼠神经功能恢复的影响显示文摘目的:观察骨髓间充质干细胞(BMSC)移植对脑梗死大鼠神经功能恢复的影响,并对其相关机制进行探讨。方法:90只大鼠随机分为3组:假手术组、对照组、BMSC移植组,每组30只。对照组和BMSC移植组建立大鼠大脑中动脉阻塞(MCAO)模型,假手术组只需要分离大鼠颈部组织,而不造MCAO模型。BMSC移植组在MCAO模型术后1天经尾静脉注射1 mL/3×10~6 BMSC,对照组注射同剂量的生理盐水,于MCAO术后1 d、3 d、7 d、14 d、21 d、28 d、35 d、42 d、49 d分别对各组大鼠进行神经功能评分(mNSS),术后2个月对BMSC移植组及对照组大鼠脑组织进行免疫组化染色,检测MAP2、TUJ1、Ⅷ因子、GFAP的表达情况。结果:在治疗后的第7天至第35天,BMSC移植组mNSS均显著低于对照组(P<0.05)。术后2个月,BMSC移植组MAP2、TUJ1、Ⅷ因子表达量显著高于对照组,而GFAP表达量显著低于于BMSC对照组(P<0.01)。结论:BMSC移植可以促进脑梗死神经功能的恢复。 | 汪涛 王晓虹 王苏平 杜云霞 赵树芳 王思家 | 2017 | 现代生物医学进展2017,17,22: | 2 |
| 6 | Lentivirus-mediated Persephin overexpression in Parkinson's disease rats显示文摘Persephin, together with glial cell line-derived neurotrophic factor and neurturin, has a neurotrophic effect and promotes the survival of motor neurons cultured in vitro. In this study, dopaminergic neurons in the substantia nigra of rats were transfected with the Persephin gene. One week later 6-hydroxydopamine was injected into the anterior medial bundle to establish a Parkinson's disease model in the rats. Results found that the number of dopaminergic neurons in the substantia nigra increased, tyrosine hydroxylase expression was upregulated and concentrations of dopamine and its metabolites in corpus striatum were increased after pretreatment with Persephin gene. In addition, the rotating effect of the induced Parkinson's disease rats was much less in the group pretreated with the Persephin gene.
