| 1 | MicroRNA-21 as a potential colon and rectal cancer biomarker显示文摘Colorectal cancer(CRC)is one of the most common malignant diseases worldwide and the prognosis is still poor although much progress has been achieved in recent years.In order to reduce CRC-related deaths,many studies are aimed at identifying novel screeningand prognosis-related biomarkers.MicroRNAs(miRNAs)are a class of 18-27-nucleotide single-stranded RNA molecules that regulate gene expression at the posttranscriptional level.It has been demonstrated that miRNAs regulate a variety of physiological functions,including development,cell differentiation,proliferation,and apoptosis.They play important roles in various physiologic and developmental processes and in the initiation and progression of various human cancers.It has been shown that miRNAs can critically regulate tumor cell gene expression,and evidence suggests that they may function as both oncogenes and tumor suppressor genes.In CRC,miRNAs-21 is one of the most important miRNAs and is rapidly emerging as a novel biomarker in CRC,with good potential as a diagnostic and therapeutic target.In this review,we summarize the latest research findings of the clinicopathological relevance of miRNAs-21 in CRC initiation,development,and progress,highlighting its potential diagnostic,prognostic,and therapeutic application,as well as discuss-ing future prospects. | Tao Li Mei Ha Leong Bruce Harms Gregory Kennedy Lin Chen | 2013 | World Journal of Gastroenterology2013,19,34: | 16 |

| 3 | A STUDY OF STRUCTURE AND PROPERTY CHANGES OF BIODEGRADABLE POLYGLYCOLIDE AND POLY(GLYCOLIDE-co-LACTIDE)FIBERS DURING PROCESSING AND IN VITRO DEGRADATION显示文摘Structure and properties of bioabsorbable polyglycolide (PGA) and poly(glycolide-co-lactide) (PGA-co-PLA)fibers were investigated during several industrial processing stages and in vitro degradation by means of wide-angle X-ray diffraction (WAXD), dynamic mechanical analysis (DMA) and mechanical property tests. In the orientation stage, the PGA fibers were found to have higher degrees of crystallinity than corresponding PGA-co-PLA samples produced under similar conditions. In the hot-stretching and post-annealing stages, after fibers were braided, PGA samples were found to gain more crystallinity and higher Tg than PGA-co-PLA samples. The higher crystallinity in PGA fibers resulted in a slower rate of degradation. DMA results showed that a great deal of internal stress that was built during orientation and hot-stretching stages was released in the post-annealing stage for all PGA and PGA-co-PLA samples. During earlier stages ofin vitro degradation, both PGA and PGA-co-PLA samples exhibited the typical cleavage-induced crystallization mechanism. | 4 | ALDH2 contributes to melatonin-induced protection against APP/PS1 mutation-prompted cardiac anomalies through cGAS-STING-TBK1-mediated regulation of mitophagy显示文摘Ample clinical evidence suggests a high incidence of cardiovascular events in Alzheimer’s disease(AD),although neither precise etiology nor effective treatment is available.This study was designed to evaluate cardiac function in AD patients and APP/PS1 mutant mice,along with circulating levels of melatonin,mitochondrial aldehyde dehydrogenase(ALDH2)and autophagy.AD patients and APP/PS1 mice displayed cognitive and myocardial deficits,low levels of circulating melatonin,ALDH2 activity,and autophagy,ultrastructural,geometric(cardiac atrophy and interstitial fibrosis)and functional(reduced fractional shortening and cardiomyocyte contraction)anomalies,mitochondrial injury,cytosolic mtDNA buildup,apoptosis,and suppressed autophagy and mitophagy.APP/PS1 mutation downregulated cyclic GMP-AMP synthase(cGAS)and stimulator of interferon genes(STING)levels and TBK1 phosphorylation,while promoting Aβaccumulation.Treatment with melatonin overtly ameliorated unfavorable APP/PS1-induced changes in cardiac geometry and function,apoptosis,mitochondrial integrity,cytosolic mtDNA accumulation(using both immunocytochemistry and qPCR),mitophagy,and cGAS-STING-TBK1 signaling,although these benefits were absent in APP/PS1/ALDH2−/−mice.In vitro evidence indicated that melatonin attenuated APP/PS1-induced suppression of mitophagy and cardiomyocyte function,and the effect was negated by the nonselective melatonin receptor blocker luzindole,inhibitors or RNA interference of cGAS,STING,TBK1,and autophagy.Our data collectively established a correlation among cardiac dysfunction,low levels of melatonin,ALDH2 activity,and autophagy in AD patients,with compelling support in APP/PS1 mice,in which melatonin rescued myopathic changes by promoting cGAS-STING-TBK1 signaling and mitophagy via an ALDH2-dependent mechanism. | Shuyi Wang Lin Wang Xing Qin Subat Turdi Dongdong Sun Bruce Culver Russel JReiter Xiaoming Wang Hao Zhou Jun Ren | 2020 | Signal Transduction and Targeted Therapy2020,5,1: | 9 |
