1Sodium oligomannate therapeutically remodels gut microbiota and suppresses gut bacterial amino acids-shaped neuroinflammation to inhibit Alzheimer's disease progression显示文摘Recently,increasing evidence has suggested the association between gut dysbiosis and Alzheimer's disease(AD)progression,yet the role of gut microbiota in AD pathogenesis remains obscure.Herein,we provide a potential mechanistic link between gut microbiota dysbiosis and neuroinflammation in AD progression.Using AD mouse models,we discovered that,during AD progression,the alteration of gut microbiota composition leads to the peripheral accumulation of phenylalanine and isoleucine,which stimulates the differentiation and proliferation of pro-inflammatory T helper 1(Thl)cells.The brain-infiltrated peripheral Th1 immune cells are associated with the Ml microglia aaivation,contributing to AD-associated neuroinflammation.Importantly,the elevation of phenylalanine and isoleucine concentrations and the increase of Th1 cell frequency in the blood were also observed in two small independent cohorts of patients with mild cognitive impairment(MCI)due to AD.Furthermore,GV-971,a sodium oligomannate that has demonstrated solid and consistent cognition improvement in a phase 3 clinical trial in China,suppresses gut dysbiosis and the associated phenylalanine/isoleucine accumulation,harnesses neuroinflammation and reverses the cognition impairment.Together,our findings highlight the role of gut dysbiosis-promoted neuroinflammation in AD progression and suggest a novel strategy for AD therapy by remodelling the gut microbiota.Xinyi Wang Guangqiang Sun Teng Feng Jing Zhang Xun Huang Tao Wang Zuoquan Xie Xingkun Chu Jun Yang Huan Wang Shuaishuai Chang Yanxue Gong Lingfei Ruan Guanqun Zhang Siyuan Yan Wen Lian Chen Du Dabing Yang Qingli Zhang Feifei Lin Jia Liu Haiyan Zhang Changrong Ge Shifu Xiao Jian Ding Meiyu Geng 2019Cell Research2019,29,10:192

4Enhanced Strength and Ductility Due to Microstructure Refinement and Texture Weakening of the GW102K Alloy by Cyclic Extrusion Compression显示文摘The cyclic extrusion compression(CEC) was applied to severely deform the as-extruded GW102K(Mg–10.0Gd–2.0Y–0.5Zr, wt%) alloy at 350, 400, and 450 °C, respectively. The microstructure, texture, and grain boundary character distribution of the CECed alloy were investigated in the present work. The mechanical properties were measured by uniaxial tension at room temperature. The crack initiation on the longitudinal section near the tensile fracture-surface was investigated by high-resolution scanning electron microscopy(SEM). The result shows that the microstructure was dramatically refined by dynamic recrystallization(DRX). The initial fiber texture was disintegrated and obviously weakened. The 8-passes/350 °C CECed alloy exhibited yield strength of 318 MPa with an elongation-to-fracture of 16.8%, increased by 41.3% and 162.5%, respectively. Moreover, the elongation-to-fracture of the 8-passes/450 °C CECed alloy significantly increased more than 3 times than that of the received alloy. The cracks were mainly initiated at twin boundaries and second phase/matrix interfaces during tensile deformation. The microstructure refinement was considered to result in the dramatically enhanced of the strength and ductility. In addition, the texture randomization during CEC is beneficial for enhancing ductility. 6Influence of physical properties of carrier on the performance of dry powder inhalers显示文摘Dry powder inhalers(DPIs) offer distinct advantages as a means of pulmonary drug delivery and have attracted much attention in the field of pharmaceutical science. DPIs commonly contain micronized drug particles which, because of their cohesiveness and strong propensity to aggregate, have poor aerosolization performance. Thus carriers with a larger particle size are added to address this problem. However, the performance of DPIs is profoundly influenced by the physical properties of the carrier, particularly their particle size, morphology/shape and surface roughness. 7Bi-level Coordinated Planning of Active Distribution Network Considering Demand Response Resources and Severely Restricted Scenarios显示文摘Due to the uncertainty and fluctuation of distributed generation(DG)and load,the operation of active distribution network(ADN)is affected by multi-dimension factors which are described by massive operation scenarios.Efficient and accurate screening of severely restricted scenarios(SRSs)has become a new challenge in ADN planning.In this paper,a novel bi-level coordinated planning model which combines the short-time-scale operation problem with the long-time-scale planning problem is proposed.At the upper level,the demand response(DR)resource,an effective non-component planning resource characterized by low capacity price,high energy price,and short contract term,is co-optimized with the configuration of lines and energy storage systems(ESSs)to achieve the economic trade-off between the investment cost and the operation cost under SRSs.At the lower level,with the planning scheme obtained from the upper level,massive operation problems are optimized to minimize the daily operation cost;and the SRSs are provided to the upper level through a shadow-price-based scenario screening method,which simulates the planning information(i.e.,the restricted degrees of operation scenarios)feedback process from ADN operators to ADN planners.Case studies on a 62-node distribution system in Jianshan New District,Zhejiang Province,China,illustrate the effectiveness of the proposed bi-level coordinated planning model considering DR resources and SRSs.Huang Yixin Lin Zhenzhi Liu Xinyi Yang Li Dan Yangqing Zhu Yanwei Ding Yi Wang Qin 2021Journal of Modern Power Systems and Clean Energy2021,9,5:3
8A comprehensive analysis of the Bencao(herbal)small RNA Atlas reveals novel RNA therapeutics for treating human diseases显示文摘Cross-kingdom herbal mi RNA was first reported in 2012.Using a modified herbal extraction protocol,we obtained 73,677,287sequences by RNA-seq from 245 traditional Chinese Medicine(TCM),of which 20,758,257 were unique sequences.We constructed a Bencao(herbal)small RNA(s RNA)Atlas(http://gffzz4a3adbfa122d4a47h0kxpkvu6co6b65nn.ffgz.tsg.suse.edu.cn),annotated the sequences by sequence-based clustering,and created a nomenclature system for Bencao s RNAs.The profiles of 21,757 mi RNAs in the Atlas were highly consistent with those of plant mi RNAs in mi RBase.Using software tools,our results demonstrated that all human genes might be regulated by s RNAs from the Bencao s RNA Atlas,part of the predicted human target genes were experimentally validated,suggesting that Bencao s RNAs might be one of the main bioactive components of herbal medicines.We established roadmaps for oligonucleotide drugs development and optimization of TCM prescriptions.Moreover,the decoctosome,a lipo-nano particle consisting of 0.5%–2.5%of the decoction,demonstrated potent medical effects.We propose a Bencao(herbal)Index,including small-molecule compounds(SM),protein peptides(P),nucleic acid(N),non-nucleic and non-proteinogenic large-molecule compounds(LM)and elements from Mendeleev's periodic table(E),to quantitatively measure the medical effects of botanic medicine.The Bencao s RNA Atlas is a resource for developing gene-targeting oligonucleotide drugs and optimizing botanical medicine,and may provide potential remedies for the theory and practice of one medicine.Yinghao Cao Yexuan Lin Na Sun Xinyi Du Yixin Dong Song Mei Xingyu Deng Xiaobei Li Shaoting Guo Kegong Tang Jiaqi Liu Xiangyu Qiao Dandan Zhao Yuhao Qin Cong Zhang Tianyi Xin Xiaohu Shi Congzhao Zhou Tao Dong De-an Guo Benedikt MKessler Dong Xu Jingyuan Song Fengming Huang Xiaoyue Wang Chengyu Jiang 2023Science China(Life Sciences)2023,66,10:2
9Hydrodynamic studies on two wiggling hydrofoils in an oblique arrangement显示文摘The propulsive performance of an oblique school of fish is numerically studied using an immersed boundary technique. The effect of the spacing and wiggling phase on the hydrodynamics of the system is investigated. The hydrodynamics of the system is deeply affected by the spacing between each fish in the school. When the horizontal separation is smaller than the length of the fish body,the downstream fish exhibits a larger thrust coefficient and greater propulsive efficiency than the isolated fish. However,the corresponding values for the upstream fish are smaller.The opposite behavior occurs when the horizontal separation increases beyond the length of fish body. 10METTL3-mediated m^(6)A RNA methylation regulates dorsal lingual epithelium homeostasis显示文摘The dorsal lingual epithelium,which is composed of taste buds and keratinocytes differentiated from K14^(+)basal cells,discriminates taste compounds and maintains the epithelial barrier.N6-methyladenosine(m^(6)A)is the most abundant mRNA modification in eukaryotic cells.How METTL3-mediated m^(6)A modification regulates K14^(+)basal cell fate during dorsal lingual epithelium formation and regeneration remains unclear.Here we show knockout of Mettl3 in K14^(+)cells reduced the taste buds and enhanced keratinocytes.Deletion of Mettl3 led to increased basal cell proliferation and decreased cell division in taste buds.Conditional Mettl3 knock-in mice showed little impact on taste buds or keratinization,but displayed increased proliferation of cells around taste buds in a protective manner during post-irradiation recovery.Mechanically,we revealed that the most frequent m^(6)A modifications were enriched in Hippo and Wnt signaling,and specific peaks were observed near the stop codons of Lats1 and FZD7.Our study elucidates that METTL3 is essential for taste bud formation and could promote the quantity recovery of taste bud after radiation.Qiuchan Xiong Caojie Liu Xin Zheng Xinyi Zhou Kexin Lei Xiaohan Zhang Qian Wang Weimin Lin Ruizhan Tong Ruoshi Xu Quan Yuan 2022International Journal of Oral Science2022,14,3:2

12SCM-198 ameliorates endometrial inflammation via suppressing the LPS-JNK-cJUN/cFOS-TLR4-NF-κB pathway显示文摘Endometritis is an inflammatory disease of the endometrium,which is responsible for endometrial dysfunction,decidualization failure,and increased incidence of early pregnancy loss.SCM-198,a synthetic form of leonurine,is well known to possess anti-inflammatory effects.SCM-198 has been reported to display beneficial effects on endometritis.However,the specific mechanisms of SCM-198 in preventing endometritis remain unknown.In this study,we focused on the molecular mechanism of SCM-198 in inhibiting endometritis.The anti-inflammatory effects and the related signaling pathways of SCM-198 were studied in vitro using human endometrial stromal cells(hESCs).Reverse transcriptase-polymerase chain reaction and western blot analysis results demonstrated that SCM-198 markedly inhibited lipopolysaccharide(LPS)-induced endometrial inflammatory response by suppressing the LPS-JNK-cJUN/cFOS-TLR4-NF-κB pathway.The preventive and therapeutic effects of SCM-198 on endometrial inflammation were explored by using a mouse model of LPS-induced endometritis.SCM-198 produced essentially the same effects when administered either post-treatment(after LPS)or pre-treatment(before LPS)via vaginal or intraperitoneal administration.In vivo results indicated that SCM-198 is a potential effective drug for the treatment of endometritis.Yikong Lin Yunyun Li Xinyi Li Xinhua Liu Xiaolin Wang Min Yu Yizhun Zhu Meirong Du 2021Acta Biochimica et Biophysica Sinica2021,53,9:1

15Decentralized MPC-Based Trajectory Generation for Multiple Quadrotors in Cluttered Environments显示文摘Challenges in motion planning for multiple quadrotors in complex environments lie in overall°ight e±ciency and the avoidance of obstacles,deadlock,and collisions among themselves.In this paper,we present a gradient-free trajectory generation method for multiple quadrotors in dynamic obstacle-dense environments with the consideration of time consumption.A model predictive control(MPC)-based approach for each quadrotor is proposed to achieve distributed and asynchronous cooperative motion planning.First,the motion primitives of each quadrotor are formulated as the boundary state constrained primitives(BSCPs)which are constructed with jerk limited trajectory(JLT)generation method,a boundary value problem(BVP)solver,to obtain time-optimal trajectories.They are then approximated with a neural network(NN),pre-trained using this solver to reduce the computational burden.The NN is used for fast evaluation with the guidance of a navigation function during optimization to guarantee°ight safety without deadlock.Finally,the reference trajectories are generated using the same BVP solver.Our simulation and experimental results demonstrate the superior performance of the proposed method.Xinyi Wang Lele Xi Yizhou Chen Shupeng Lai Feng Lin Ben M.Chen 2021Guidance, Navigation and Control2021,1,2:1
16CB1R-stabilized NLRP3 inflammasome drives antipsychotics cardiotoxicity显示文摘Long-term use of antipsychotics is a common cause of myocardial injury and even sudden cardiac deaths that often lead to drug withdrawn or discontinuation.Mechanisms underlying antipsychotics cardiotoxicity remain largely unknown.Herein we performed RNA sequencing and found that NLRP3 inflammasome-mediated pyroptosis contributed predominantly to multiple antipsychotics cardiotoxicity.Pyroptosis-based small-molecule compound screen identified cannabinoid receptor 1(CB1R)as an upstream regulator of the NLRP3 inflammasome.Mechanistically,antipsychotics competitively bond to the CB1R and led to CB1R translocation to the cytoplasm,where CB1R directly interacted with NLRP3 inflammasome via amino acid residues 177–209,rendering stabilization of the inflammasome.Knockout of Cb1r significantly alleviated antipsychotic-induced cardiomyocyte pyroptosis and cardiotoxicity.Multi-organ-based investigation revealed no additional toxicity of newer CB1R antagonists.In authentic human cases,the expression of CB1R and NLRP3 inflammasome positively correlated with antipsychotics-induced cardiotoxicity.These results suggest that CB1R is a potent regulator of the NLRP3 inflammsome-mediated pyroptosis and smallmolecule inhibitors targeting the CB1R/NLRP3 signaling represent attractive approaches to rescue cardiac side effects of antipsychotics.Liliang Li Pan Gao Xinru Tang Zheng Liu Mengying Cao Ruoyu Luo Xiaoqing Li Jing Wang Xinyi Lin Chao Peng Zhihong Li Jianhua Zhang Xian Zhang Zhonglian Cao Yunzeng Zou Li Jin 2022Signal Transduction and Targeted Therapy2022,7,7:1
17High-speed long-distance visible light communication based on multicolor series connection micro-LEDs and wavelength division multiplexing显示文摘Multicolor series connection micro-LED arrays with emission wavelengths of violet, blue, green, and yellow were fabricated, and their optoelectronic properties and communication performances were investigated. The designed series connection micro-LED array exhibited the light output power of multiple milliwatts, whereas mostly keeping a slightly reduced modulation bandwidth, thus, enabling a higher signal-to-noise ratio compared to a single pixel and showing superior performance in the field of long-distance visible light communication(VLC). The achievable data rates of 400-, 451-, 509-, and 556-nm micro-LED arrays using bit/power loading orthogonal frequency division multiplexing were 5.71, 4.86, 4.39, and 0.82 Gbps, respectively. The aggregate data rate of 15.78 Gbps was achieved for the proof-of-concept wavelength division multiplexing system under a transmission distance of 13 m, which was the best data rate-distance product performance for the LED-based VLC to the best of our knowledge. 18Optimized weak measurement of orbital angular momentum-induced beam shifts in optical reflection显示文摘Tiny but universal beam shifts occur when a polarized light beam is reflected upon a planar interface.Although the beam shifts of Gaussian beams have been measured by the weak measurement technique, the weak measurement for orbital angular momentum(OAM)-induced spatial shifts of vortex beams is still missing.Here, by elaborately choosing the preselection and postselection states, the tiny OAM-induced Goos–H?nchen and Imbert–Fedorov shifts are amplified at an air–prism interface. The maximum shifts along directions both parallel and perpendicular to the incident plane are theoretically predicted and experimentally verified with optimal preselection and postselection states. 19Dissecting the epigenomic dynamics of human fetal germ cell development at single-cell resolution显示文摘Proper development of fetal germ cells(FGCs)is vital for the precise transmission of genetic and epigenetic information through generations.The transcriptional landscapes of human FGC development have been revealed;however,the epigenetic reprogramming process of FGCs remains elusive.Here,we profiled the genome-wide DNA methylation and chromatin accessibility of human FGCs at different phases as well as gonadal niche cells at single-cell resolution.First,we found that DNA methylation levels of FGCs changed in a temporal manner,whereas FGCs at different phases in the same embryo exhibited comparable DNA methylation levels and patterns.Second,we revealed the phase-specific chromatin accessibility signatures at the promoter regions of a large set of critical transcription factors and signaling pathway genes.We also identified potential distal regulatory elements including enhancers in FGCs.Third,compared with other hominid-specific retrotransposons,SVA_D might have a broad spectrum of binding capacity for transcription factors,including SOX15 and SOX17.Finally,using an in vitro culture system of human FGCs,we showed that the BMP signaling pathway promoted the cell proliferation of FGCs,and regulated the WNT signaling pathway by orchestrating the chromatin accessibility of its ligand genes.Our single-cell epigenomic atlas and functional assays provide valuable insights for understanding the strongly heterogeneous,unsynchronized,yet highly robust nature of human germ cell development.Li Li Lin Li Qingqing Li Xixi Liu Xinyi Ma Jun Yong Shuai Gao Xinglong Wu Yuan Wei Xiaoye Wang Wei Wang Rong Li Jie Yan Xiaohui Zhu Lu Wen Jie Qiao Liying Yan Fuchou Tang 2021Cell Research2021,31,4:1
20IMU-based motion capture system for rehabilitation applications: A systematic review显示文摘In recent years,the use of inertial measurement unit(IMU)-based motion capture(Mocap)systems in rehabilitation has grown significantly.This paper aimed to provide an overview of current IMUbased Mocap system designs in the field of rehabilitation,explore the specific applications and implementation of these systems,and discuss potential future developments considering sensor limitations.For this review,a systematic literature search was conducted using Scopus,IEEE Xplore,PubMed,and Web of Science from 2013 to 2022.A total of 65 studies were included and analyzed based on their rehabilitation application,target population,and system deployment and measurement.The proportion of rehabilitation assessment,training,and both were 82%,12%,and 6%respectively.The results showed that primary focus of the studies was stroke that was one of the most commonly studied pathological disease.Additionally,general rehabilitation without targeting a specific pathology was also examined widely,with a particular emphasis on gait analysis.The most common sensor configuration for gait analysis was two IMUs measuring spatiotemporal parameters of the lower limb.However,the lack of training applications and upper limb studies could be attributed to the limited battery life and sensor drift.To address this issue,the use of low-power chips and low-consumption transmission pathways was a potential way to extend usage time for long-term training.Furthermore,we suggest the development of a highly integrated multi-modal system with sensor fusion.Chenyu Gu Weicong Lin Xinyi He Lei Zhang Mingming Zhang 2023Biomimetic Intelligence & Robotics2023,3,2:0
