| 1 | Nitrogen-doped fluorescence carbon dots as multi-mechanism detection for iodide and curcumin in biological and food samples显示文摘Iodine ion is one of the most indispensable anions in living organisms,particularly being an important substance for the synthesis of thyroid hormones.Curcumin is a yellow-orange polyphenol compound derived from the rhizome of Curcuma longa L.,which has been commonly used as a spice and natural coloring agent,food additives,cosmetics as well as Chinese medicine.However,excess curcumin may cause DNA inactivation,lead to a decrease in intracellular ATP levels,and trigger the tissue necrosis.Therefore,quantitative detection of iodine and curcumin is of great significance in the fields of food and life sciences.Herein,we develop nitrogen-doped fluorescent carbon dots(NCDs)as a multi-mechanism detection for iodide and curcumin in actual complex biological and food samples,which was prepared by a one-step solid-phase synthesis using tartaric acid and urea as precursors without adding any other reagents.An assembled NCDs-Hg^(2+) fluorescence-enhanced sensor for the quantitative detection of I^(-) was established based on a fluorescence“turn-off-on”mechanism in a linear range of 0.3-15μM with a detection limit of 69.4 nM and successfully quantified trace amounts of I^(-) in water samples and urine sample.Meanwhile,the as-synthesized NCDs also can be used as a fluorescent quenched sensor for curcumin detection based on the synergistic internal filtration effect(IFE)and static quenching,achieving a good linear range of 0.1-20μM with a satisfactory detection limit of 29.8 nM.These results indicate that carbon dots are potential sensing materials for iodine and curcumin detection for the good of our health. | Xiaodan Tang Hongmei Yu Brian Bui Lingyun Wang Christina Xing Shaoyan Wang Mingli Chen Zhizhi Hu Wei Chen | 2021 | Bioactive Materials2021,6,6: | 11 |
| 2 | LAMOST Experiment for Galactic Understanding and Exploration (LEGUE)——The survey's science plan显示文摘We describe the current plans for a spectroscopic survey of millions of stars in the Milky Way galaxy using the Guo Shou Jing Telescope (GSJT, formerly called the Large sky Area Multi-Object fiber Spectroscopic Telescope - LAMOST). The survey will obtain spectra for 2.5 million stars brighter than r < 19 during dark/grey time, and 5 million stars brighter than r < 17 or J < 16 on nights that are moonlit or have low transparency. The survey will begin in the fall of 2012, and will run for at least four years. The telescope's design constrains the optimal declination range for observations to 10 < δ < 50 , and site conditions lead to an emphasis on stars in the direction of the Galactic anticenter. The survey is divided into three parts with different target selection strategies: disk, anticenter, and spheroid. The resulting dataset will be used to study the merger history of the Milky Way, the substructure and evolution of the disks, the nature of the first generation of stars through identification of the lowest metallicity stars, and star formation through study of open clusters and OB associations. Novel therapeutic targets are desperately needed because the meager improvement in the cure rate of about 10% realized by adjunctive treatments to surgery is unacceptable as > 50% patients with localized gastric cancer succumb to their disease. Either postoperative chemoradiotherapy(United States),pre-and post-operative chemotherapy(Europe),and adjuvant chemotherapy after a D2 resection(Asia) can all be regarded as standards of care in the localized gastric cancer management. In metastatic disease the addition of trastuzumab to chemotherapy is standard of care in Her2 positive disease. | 4 | Drought Stress and Preharvest Aflatoxin Contamination in Agricultural Commodity: Genetics, Genomics and Proteomics显示文摘Throughout the world, aflatoxin contamination is considered one of the most serious food safety issues concerning health. Chronic problems with preharvest aflatoxin contamination occur in the southern US, and are particularly troublesome in corn, peanut, cottonseed, and tree nuts. Drought stress is a major factor to contribute to preharvest aflatoxin contamination. Recent studies have demonstrated higher concentration of defense or stress-related proteins in corn kernels of resistant genotypes compared with susceptible genotypes, suggesting that preharvest field condition (drought or not drought) influences gene expression differently in different genotypes resulting in different levels of 'end products': PR(pathogenesis-related) proteins in the mature kernels. Because of the complexity of Aspergillus-plant interactions, better understanding of the mechanisms of genetic resistance will be needed using genomics and proteomics for crop improvement. Genetic improvement of crop resistance to drought stress is one component and will provide a good perspective on the efficacy of control strategy. Proteomic comparisons of corn kernel proteins between resistant or susceptible genotypes to Aspergillus flavus infection have identified stress-related proteins along with antifungal proteins as associated with kernel resistance. | 6 | Central Asian aridification during the late Eocene to early Miocene inferred from preliminary study of shallow marine-eolian sedimentary rocks from northeastern Tajik Basin显示文摘The tempo-spatial development of the Cenozoic Asian aridification across the Eocene-Oligocene and its controlling factors are important scientific topics in Earth Sciences, which are pertinent to regional and global tectonic and climatic events. However, sedimentary rocks preserving the record of aridification during this time from central Asia(ACA) are rare. Here we present a preliminary analysis of sedimentary facies of the lower Paleogene in the northeastern Tajik Basin, which reveals that: the lower part of the studied section is dominated by shallow marine deposits of the Paratethys, the middle part is typical of alternations of eolian dune and fluvial deposits, the upper part is represented by eolian loess-sandy loess(L&SL) like facies, and the top exhibits alternations of fluvial-lacustrine and loess like facies. Based on a chronological framework derived from preliminary magnetostratigraphy, published U-Pb dating of a volcanic ash, and regional litho-stratigraphic correlations, we determine that eolian and L&SL facies accumulated in the northeastern Tajik Basin during the Late Eocene and through most of the Oligocene. These sedimentary units indicate that semi-arid to arid environments of ACA had developed at least since the late Eocene. | 8 | Smartphone-Imaged HIV-1 Reverse-Transcription Loop-Mediated Isothermal Ampliflcation(RT-LAMP) on a Chip from Whole Blood显示文摘Viral load measurements are an essential tool for the long-term clinical care of hum an immunodeficiency virus(HIV)-positive individuals. The gol d standards in viral load instrumentation, however, are still too limited by their size, cost, and sophisticated operation for these measurements to be ubiquitous in remote settings with poor healthcare infrastructure, including parts of the world that are disproportionately affected by HIV infection. The challenge of developing a point-of-care platform capable of making viral load more accessible has been frequently approached but no solution has yet emerged that meets the practical requirements of low cost, portability, and ease-of-use. In this paper, we perform reverse-transcription loop-mediated isothermal amplification(RT-LAMP) on minimally processed HIV-spiked whole blood samples with a microfluidic and silicon microchip platform, and perform fluorescence measurements with a consumer smartphone. Our integrated assay shows amplification from as few as three viruses in a ~ 60 nL RTLAMP droplet, corresponding to a whole blood concentration of 670 viruses per μL of whole blood. The technology contains greater power in a digital RT-LAMP approach that could be scaled up for the determination of viral load from a flnger prick of blood in the clinical care of HIV-positive individuals. Methodology Human tongue carcinoma Tca8113 cells were cultured with DAPT.Cell growth was determined using Indigotic Reduction method.The cell cycle and apoptosis were analyzed by flow cytometry.Real-time PCR and Immuno-Fluorescence(IF) were employed to determine the intracellular expression levels. Results DAPT inhibited the growth of human tongue carcinoma Tca8113 cells by inducing G_0-G_1 cell cycle arrest and apoptosis.The mRNA levels of Hairy/Enhancer of Split-1(Hes-1),a target of Notch activation,were reduced by DAPT in a dose-dependent manner.Coincident with this observation,DAPT induced a dose-dependent promotion of constitutive Caspase-3 in Tca8113 cells. | 10 | Biomarkers in schizophrenia:A focus on blood based diagnostics and theranostics显示文摘Identifying biomarkers that can be used as diagnostics or predictors of treatment response(theranostics) in people with schizophrenia(Sz) will be an important step towards being able to provide personalized treatment. Findings from the studies in brain tissue have not yet been translated into biomarkers that are practical in clinical use because brain biopsies are not acceptable and neuroimaging techniques are expensive and the results are inconclusive. Thus, in recent years, there has been search for blood-based biomarkers for Sz as a valid alternative. Although there are some encouraging preliminary data to support the notion of peripheral biomarkers for Sz, it must be acknowledged that Sz is a complex and heterogeneous disorder which needs to be further dissected into subtype using biological based and clinical markers. The scope of this review is to critically examine published blood-based biomarker of Sz, focusing on possible uses for diagnosis, treatment response, or their relationship with schizophreniaassociated phenotype. We sorted the studies into six categories which include:(1) brain-derived neurotrophic factor;(2) inflammation and immune function;(3) neurochemistry;(4) oxidative stress response and metabolism;(5) epigenetics and micro RNA; and(6) transcriptome and proteome studies. This review also summarized the molecules which have been conclusively reported as potential blood-based biomarkers for Sz in different blood cell types. | 12 | Viable but nonculturable bacteria and their resuscitation:implications for cultivating uncultured marine microorganisms显示文摘Culturing has been the cornerstone of microbiology since Robert Koch first successfully cultured bacteria in the late nineteenth century. However, even today, the majority of microorganisms in the marine environment remain uncultivated. There are various explanations for the inability to culture bacteria in the laboratory, including lack of essential nutrients, osmotic support or incubation conditions, low growth rate, development of micro-colonies, and the presence of senescent or viable but nonculturable (VBNC) cells. In the marine environment, many bacteria have been associated with dormancy, as typified by the VBNC state. VBNC refers to a state where bacteria are metabolically active, but are no longer culturable on routine growth media. It is apparently a unique survival strategy that has been adopted by many microorganisms in response to harsh environmental conditions and the bacterial cells in the VBNC state may regain culturability under favorable conditions. The resuscitation of VBNC cells may well be an important way to cultivate the otherwise uncultured microorganisms in marine environments. Many resuscitation stimuli that promote the restoration of culturability have so far been identified;these include sodium pyruvate, quorum sensing autoinducers, resuscitation-promoting factors Rpfs and YeaZ, and catalase. In this review, we focus on the issues associated with bacterial culturability, the diversity of bacteria entering the VBNC state, mechanisms of induction into the VBNC state, resuscitation factors of VBNC cells and implications of VBNC resuscitation stimuli for cultivating these otherwise uncultured microorganisms. | 14 | Resistance to Aspergillus flavus in maize and peanut:Molecular biology, breeding, environmental stress,and future perspectives显示文摘The colonization of maize(Zea mays L.) and peanut(Arachis hypogaea L.) by the fungal pathogen Aspergillus flavus results in the contamination of kernels with carcinogenic mycotoxins known as aflatoxins leading to economic losses and potential health threats to humans. The regulation of aflatoxin biosynthesis in various Aspergillus spp. has been extensively studied, and has been shown to be related to oxidative stress responses. Given that environmental stresses such as drought and heat stress result in the accumulation of reactive oxygen species(ROS) within host plant tissues, host-derived ROS may play an important role in cross-kingdom communication between host plants and A. flavus. Recent technological advances in plant breeding have provided the tools necessary to study and apply knowledge derived from metabolomic, proteomic, and transcriptomic studies in the context of productive breeding populations. Here, we review the current understanding of the potential roles of environmental stress, ROS, and aflatoxin in the interaction between A.flavus and its host plants, and the current status in molecular breeding and marker discovery for resistance to A. flavus colonization and aflatoxin contamination in maize and peanut.
