1Alterations of glutathione S-transferase and matrix metalloproteinase-9 expressions are early events in esophageal carcinogenesis显示文摘AIM: To investigate the role of glutathione S-transferase (GST) and matrix metalloproteinase-9 (MMP-9) expres-sions in the development and progression of reflux es-ophagitis-Barrett’s metaplasia-dysplasia-adenocarcinoma sequence in the esophagus.METHODS: GST and MMP-9 expressions were analyzed in 51 paraffin-embedded tissue samples by immunohisto-chemistry including patients with reflux esophagitis (n = 7), Barrett’s metaplasia (n = 14), Barrett and esophagi-tis (n = 8), Barrett and dysplasia (n = 7), esophageal adenocarcinoma (n = 8) and a control group without any histological changes (n = 7). Immunostaining was determined semiquantitatively. Statistical analysis with one-way ANOVA, LSD test and correlation analysis were performed. P value of < 0.05 was considered significant.RESULTS: GST expression was significantly higher while MMP-9 expression was significantly lower in control group compared to Barrett’s metaplasia and the other groups. No major changes were observed between Bar-rett, esophagitis, and Barrett and concomitant esophagi-tis. Barrett and concomitant dysplasia, and adenocarci-noma revealed a significant lower expression of GST and higher levels of MMP-9 compared to all other groups. Adenocarcinoma showed almost no expression of GST and significantly higher levels of MMP-9 than Barrett and concomitant dysplasia. Alterations of GST and MMP-9 were inversely correlated (r = - 0.82).CONCLUSION: Decreased GST and increased ex-pression of MMP-9 in Barrett’s metaplasia-dysplasia-adenocarcinoma sequence as compared to normal tissue suggest their association with esophageal tumorigenesis. Loss of GST and gain of MMP-9 in Barrett with dyspla-sia compared to non-dysplastic metaplasia indicate that these alterations may be early events in carcinogenesis. 2Interleukins and interleukin receptors in rheumatoid arthritis: Research, diagnostics and clinical implications显示文摘Rheumatoid arthritis(RA) is an autoimmune disease, resulting in a chronic, systemic inflammatory disorder. It may affect many tissues and organs, but it primarily affects the flexible joints. In clinical practice patient care generates many questions about diagnosis, prognosis, and treatment. It is challenging for health care specialists to keep up to date with the medical literature. This review summarizes the pathogenesis, the polymorphisms of interleukin and interleukin genes and the standard available and possible future immunologictargets for RA treatment. The identification of diseaseassociated interleukin and interleukin receptor genes can provide precious insight into the genetic variations prior to disease onset in order to identify the pathways important for RA pathogenesis. 3Genetic update on inflammatory factors in ulcerative colitis: Review of the current literature显示文摘Ulcerative colitis(UC) is one of the main types of inflammatory bowel disease, which is caused by dysregulated immune responses in genetically predisposed individuals. Several genetic factors, including interleukin and interleukin receptor gene polymorphisms and other inflammation-related genes play central role in mediating and modulating the inflammation in the human body, thereby these can be the main cause of development of the disease. It is clear these data are very important for understanding the base of the disease, especially in terms of clinical utility and validity, but summarized literature is exiguous for challenge health specialist that can used in the clinical practice nowadays. This review summarizes the current literature on inflammationrelated genetic polymorphisms which are associated with UC. 4Primary gastric mucosa associated lymphoid tissue lymphoma: Clinical data predicted treatment outcome显示文摘AIM: To determine clinical characteristics and treatment outcome of gastric lymphoma after chemotherapy and immuno-chemotherapy. METHODS: Thirty four patients with primary gastric mucosa associated lymphoid tissue (MALT) lymphoma (Ann Arbor stages Ⅰ to Ⅳ) were enrolled. All had upper gastric endoscopy, abdominal ultrasonography, CT and H pylori status assessment (histology and serology). After anti-H pylori treatment and initial chemotherapy, patients were re-examined every 4 mo. RESULTS: Histological regression of the lymphoma was complete in 22/34 (64.7%) and partial in 9 (26.5%) patients. Median follow up time for these 31 responders was 60 mo (range 48-120). No regression was noted in 3 patients. Among the 25 (73.5%) H pylori positive patients, the eradication rate was 100%.CONCLUSION: Using univariate analysis, predictive factors for overall survival were international prognostic index (IPI) score, hemoglobin level, erythrocyte sedimentation rate (ESR), and platelet numbers (P < 0.005). 6Long-term omeprazole and esomeprazole treatment does not significantly increase gastric epithelial cell proliferation and epithelial growth factor receptor expression and has no effect on apoptosis and p53 expression显示文摘AIM: To study the effect of proton pump inhibitor (PPI)treatment on patients with reflux esophagitis and its in vivo effect on apoptosis, p53- and epidermal growth factor receptor (EGFR) expression.METHODS: After informed consent was obtained, gastric biopsies of the antrum were taken from patients with reflux oesophagitis prior to and after 6 mo of 20 mg omeprazole (n = 14) or 40 mg esomeprazole (n = 12) therapy.Patients did not take any other medications known to affect the gastric mucosa. All patients were Helicobacter pylori negative as confirmed by rapid urease test and histology,respectively. Cell proliferation, apoptosis, EGFR, and p53expression were measured by immunohistochemical techniques. At least 600 glandular epithelial cells were encountered and results were expressed as percentage of total cells counted. Was considered statistically significant.RESULTS: Although there was a trend towards increase of cell proliferation and EGFR expression both in omeprazole and esomeprazole treated group, the difference was not statistically significant. 7心电图有心肌缺血改变而冠状动脉造影正常者的冠状动脉血流的意义显示文摘目的 以计帧法评价心电图有心肌缺血改变而冠状动脉造影正常者的相关冠状动脉血流的意义。方法 静息心电图或踏板运动试验 (treadm ill test,TMT)有缺血改变 (连续 2个以上相关导联 ST段水平型或下斜型压低≥0 .0 5 m V或 T波深倒置 )但冠状动脉造影正常的 19例患者为缺血组。有心绞痛样症状而静息心电图、踏板运动试验以及冠状动脉造影正常的 5 2例为对照组。测定缺血相关性冠状动脉血流心肌梗死溶栓治疗试验 (thrombolysisin myocardial infarcton,TIMI)计帧值 (frame count,TIMI- FC)。结果 缺血组心电图示前壁心肌缺血者的冠状动脉前降支血流和下壁缺血者的右冠状动脉血流以及高侧壁缺血者的左回旋支血流的 TIMI- FC值显著大于对照组(分别为 35 .3± 16 .1vs19.9± 13.7,P<0 .0 5 ;38.9± 11.7vs2 1.7± 14 .1,P<0 .0 5及 35 .9± 16 .7vs.14 .1± 9.5 ,P<0 .0 1) ;TIMI- FC测定结果以 <2 7为血流正常 ,≥ 2 7为血流缓慢。血流缓慢 (slow coronary flow,SCF)者 2 2例 ,血流正常 (norm al coronary flow,NCF)者 4 9例。SCF者中心电图有缺血改变的患者 (15例 ,6 8.2 % )显著多于 NCF者中心电图有缺血的患者 (4例 ,8.2 % ) ;NCF者中心电图正常者 (4 5例 ,91.8% )显著多于 SCF者中心电图正常者(7例 ,金振一 Bela Merkely 丁大植 崔兰 沈铁宽 2004中国心血管杂志2004,9,3:4

9Prognostic value of pre-treatment F-18-FDG PET-CT in patients with hepatocellular carcinoma undergoing radioembolization显示文摘AIM To evaluate the value of pre-treatment 18F-FDG PET/CT in patients with HCC following liver radioembolization.METHODS We identified 34 patients with HCC who underwent an FDG PET/CT scan prior to hepatic radioembolization at our institution between 2009 and 2013. Patients wereseen in clinic one month after radioembolization and then at 2-3 mo intervals. We assessed the influence of FDG tumor uptake on outcomes including local liver control(LLC), distant liver control(DLC), time to distant metastases(DM), progression free survival(PFS) and overall survival(OS).RESULTS The majority of patients were males(n = 25, 74%), and had Child Pugh Class A(n = 31, 91%), with a median age of 68 years(46-84 years). FDG-avid disease was found in 19(56%) patients with SUVmax ranging from 3 to 20. Female patients were more likely to have an FDG-avid HCC(P = 0.02). Median follow up of patients following radioembolization was 12 months(1.2-62.8 mo). FDG-avid disease was associated with a decreased 1 year LLC, DLC, DM and PFS(P < 0.05). Using multivariate analysis, FDG avidity predicted for LLC, DLC, and PFS(all P < 0.05).CONCLUSION In this retrospective study, pre-treatment HCC FDGavidity was found to be associated with worse LLC, DLC, and PFS following radioembolization. 10Preparation and Structural Characterization of Rapidly Solidified Al-Cu Alloys显示文摘Rapidly solidified Al100-x—Cux alloys(x = 5,10,1 5,25,35 wt%) were prepared and analyzed.High cooling rate increased the Cu solubility in 9.-AI matrix.The influence of the cooling rate on Cu solubility extension in Al was experimentally simulated.Thus the pouring was performed in metallic die and by melt spinning-low pressure(MSLP) technique.Melt processing by liquid quenching was performed using a self-designed melt spinning set-up which combined the cooling technology of a melt jet on the spinning disc with the principle of the mold feeding from low pressure casting technology.The thickness of the melt-spun ribbons was in the range of30—70 μm.The cooling rate provided by MS-LP was within 105—106 K/s after the device calibration.The obtained alloys were characterized from structural,thermal and mechanical point of view.Optical microscopy and scanning electron microscopy were employed for the microstructural characterization which was followed by X-ray analysis.The thermal properties were evaluated by dilatometric and differential scanning calorimetric measurements.Vickers microhardness measurements were performed in the study.In the case of the hypereutectic alloy with 35 wt%Cu obtained by MS-LP method,the microhardness value increased by 45%compared to the same alloy obtained by gravity casting method.This was due to the extended solubility of the alloying element in the α-AI solid solution.Iuliana Lichioiu Ildiko Peter Bela Varga Mario Rosso 2014Journal of Materials Science & Technology2014,30,4:3

12Blood flow restriction in human skeletal muscle during rest periods after high-load resistance training down-regulates miR-206 and induces Pax7显示文摘Backgroud:Blood flow re striction(BFR) with low-intensity re sistance training has been shown to result in hypertrophy of skeletal muscle.In this study,we tested the hypothesis that BFR during the rest periods between acute,high-intensity resistance exercise sessions(70% of 1 repetition maximum,7 sets with 10 repetitions) enhances the effects of the resistance training.Methods:A total of 7 healthy young men performed squats,and between sets BFR was carried out on one leg while the other leg served as a control.Because BFR was applied during rest periods,even severe occlusion pressure(approximately 230 mmHg),which almost completely blocked blood flow,was well-tolerated by the participants.Five muscle-specific microRNAs were measured from the biopsy samples,which were taken 2 h after the acute training.Results:Doppler data showed that the pattern of blood flow recovery changed significantly between the first and last BFR.microRNA-206 levels significantly decreased in the BFR leg compared to the control.The mRNA levels of RAC-β serine/threonine-protein kinase v22,nuclear re spiratory factor 1,vascular endothelial growth factor,lupus Ku autoantigen protein p70 genes(p <0.05),and paired box 7(p <0.01) increased in the BFR leg.The protein levels of paired box 7,nuclear respiratory factor 1,and peroxisome proliferator-activated receptor y coactivator 1α did not differ between the BFR leg and the control leg.Conclusion:BFR,during the rest periods of high-load resistance training,could lead to mRNA elevation of those proteins that regulate angiogenesis,mitochondrial biogenesis,and muscle hypertrophy and repair.However,BFR also can cause DNA damage,judging from the increase in mRNA levels of lupus Ku autoantigen protein p70.Ferenc Torma Zoltan Gombos Marcell Fridvalszki Gergely Langmar Zsofia Tarcza Bela Merkely Hisashi Naito Noriko Ichinoseki-Sekine Masaki Takeda Zsolt Murlasits Peter Osvath Zsolt Radak 2021Journal of Sport and Health Science2021,10,4:3
13Minimally invasive image-guided therapy of primary and metastatic pancreatic cancer显示文摘Pancreatic cancer is a challenging malignancy with limited treatment options and poor life expectancy.The only curative option is surgical resection,but only 15%-20%of patients are resectable at presentation because more than 50%of patients has distant metastasis at diagnosis and the rest of them has locally advanced pancreatic cancer(LAPC).The standard of care first line treatment for LAPC patients is chemotherapy with or without radiation therapy.Recent developments in minimally invasive ablative techniques may add to the treatment armamentarium of LAPC.There are increasing number of studies evaluating these novel ablative techniques,including radiofrequency ablation,microwave ablation,cryoablation and irreversible electroporation.Most studies which included pancreatic tumor ablation,demonstrated improved overall survival in LAPC patients.However,the exact protocols are yet to set up to which stage of the treatment algorithm ablative techniques can be added and in what kind of treatment combinations.Patients with metastatic pancreatic cancer has dismal prognosis with 5-year survival is only 3%.The most common metastatic site is the liver as 90%of pancreatic cancer patients develop liver metastasis.Chemotherapy is the primary treatment option for patients with metastatic pancreatic cancer.However,when the tumor is not responding to chemotherapy or severe drug toxicity develops,locoregional liver-directed therapies can provide an opportunity to control intrahepatic disease progression and improve survival in selected patients.During the last decade new therapeutic options arose with the advancement of minimally invasive technologies to treat pancreatic cancer patients.These new therapies have been a topic of increasing interest due to the severe prognostic implications of locally advanced and metastatic pancreatic cancer and the low comorbid risk of these procedures.This review summarizes new ablative options for patients with LAPC and percutaneous liver-directed therapies for patients with liver-dominant metastatic disease.Andras Bibok Dae Won Kim Mokenge Malafa Bela Kis 2021World Journal of Gastroenterology2021,27,27:3
14Glycidyl methacrylate-compatibilized poly(lactic acid)/hemp hurd biocomposites: Processing, crystallization, and thermo-mechanical response显示文摘Poly(lactic acid)-based biocomposites were developed with hemp hurd(Cannabis sativa L.) with grafting-based interfacial compatibilization. Poly(lactic acid) was extruded with hemp hurd and glycidyl methacrylate as the polymer/hurd interfacial compatibilizer, and injection molded. 17mRNA expression, functional profiling and multivariate classification of colon biopsy specimen by cDNA overall glass microarray显示文摘AIM: To understand the local pathophysiological altera- tions and gene ontology-based functional classification of colonic biopsies into inflammatory and neoplastic dis- eases. METHODS: Total RNA was extracted from frozen biop- sies and amplified by T7-method. Expression profile was evaluated by Atlas Glass 1K microarrays. After microar- ray quality control, applicable data were available from 10 adenomas, 6 colorectal adenocarcinomas (CRCs), and 6 inflammatory bowel diseases (IBDs). Multivariate statistical and cell functional analyses were performed. Real-time RT-PCR and immunohistochemistry were used for validation. RESULTS: Discriminant analysis of selected genes, could correctly reclassify all 22 samples using 4 parameters (heat shock transcription factor-1, bystin-like, calgranu- lin-A, TRAIL receptor 3). IBD samples were characterized by overregulated chemokine (C-X-C motif) ligand 13, replication protein A1, E74-like factor 2 and downregu- lated TNF receptor-associated factor 6, BCL2-interacting killer genes. In adenomas upregulation of TNF receptor- associated factor 6, replication protein A1, E74-like factor 2 and underexpression of BCL2-associated X protein, cal- granulin-A genes were found. CRC cases had significantly increased epidermal growth factor receptor, topoisomer- ase-1, v-jun, TNF receptor-associated factor 6 and TRAIL receptor 3, and decreased RAD51 and RAD52 DNA repair gene, protein phosphatase-2A and BCL2-interacting killer mRNA levels. Epidermal growth factor receptor RT-PCR and immunohistochemistry, topoisomerase-1 RT-PCRconfirmed the chip results. CONCLUSION: Different histological alterations can be reclassified by functional, multivariate analysis using cDNA microarrays. 19Influence of Genista Tinctoria L or Methylparaben on Subchronic Toxicity of Bisphenol A in Rats显示文摘Objective To evaluate the influence of an extract of Genista tinctoria L. herba(GT) or methylparaben(MP) on histopathological changes and 2 biomarkers of oxidative stress in rats subchronicly exposed to bisphenol A(BPA). Methods Adult female Wistar rats were orally exposed for 90 d to BPA(50 mg/kg), BPA+GT(35 mg isoflavones/kg) or BPA+MP(250 mg/kg). Plasma and tissue samples were taken from liver, kidney, thyroid, uterus, ovary, and mammary gland after 30, 60, and 90 d of exposure respectively. Lipid peroxidation and in vivo hydroxyl radical production were evaluated by histological analysis along with malondialdehyde and 2,3-dihydroxybenzoic acid detection. Results The severity of histopathological changes in liver and kidneys was lower after GT treatment than after BPA or BPA+MP treatment. A minimal thyroid receptor antagonist effect was only observed after BPA+MP treatment. The abnormal folliculogenesis increased in a time-dependent manner, and the number of corpus luteum decreased. No significant histological alterations were found in the uterus. The mammary gland displayed specific estrogen stimulation changes at all periods. Both MP and GT revealed antioxidant properties reducing lipid peroxidation and BPA-induced hydroxyl radical generation. Conclusion GT L. extract ameliorates the toxic effects of BPA and is proved to have antioxidant potential and antitoxic effect. 20Splenectomy with chemotherapy vs surgery alone as initial treatment for splenic marginal zone lymphoma显示文摘AIM:To evaluate the clinical characteristics of splenic marginal-zone lymphoma (SMZL) following antigen expression and the influence of therapeutic approaches on clinical outcome and overall survival (OS).METHODS: A total of 30 patients with typical histological and immunohistochemical SMZL patterns were examined. Splenectomy plus chemotherapy was applied in 20 patients, while splenectomy as a single treatment-option was performed in 10 patients. Prognostic factor and overall survival rate were analyzed.RESULTS: Complete remission (CR) was achieved in 20 (66.7%), partial remission (PR) in seven (23.3%), and lethal outcome due to disease progression occurred in three (10.0%) patients. Median survival of patients with a splenectomy was 93.0 mo and for patients with splenectomy plus chemotherapy it was 107.5 mo (Log rank=0.056, P>0.05). Time from onset of first symptoms to the beginning of the treatment (mean 9.4 mo) was influenced by spleen dimensions, as measured by computerized tomography and ultra-sound (t=2.558, P=0.018). Strong positivity (+++) of CD20 antigen expression in splenic tissue had a positive influence on OS (Log rank=5.244, P<0.05). The analysis of factors interfering with survival (by the Kaplan-Meier method) revealed that gender, general symptoms, clinical stage, and spleen infiltration type (nodular vs diffuse) had no significant (P>0.05) effects on the OS.
