2Long-term follow-up after complete ablation of Barrett's esophagus with argon plasma coagulation显示文摘AIM: To report the long-term outcome of patients after complete ablation of non-neoplastic Barrett's esophagus (BE) with respect to BE relapse and development of intraepithelial neoplasia or esophageal adenocarcinoma.METHODS: In 70 patients with histologically proven nonneoplastic BE, complete BE ablation was achieved by argon plasma coagulation (APC) and high-dose proton pump inhibitor therapy (120 mg omeprazole daily). Sixty-six patients (94.4%) underwent further surveillance endoscopy. At each surveillance endoscopy four-quadrant biopsies were taken from the neo-squamous epithelium at 2 cm intervals depending on the pre-treatment length of BE mucosa beginning at the neo-Z-line, and from any endoscopically suspicious lesion.RESULTS: The median follow-up of 66 patients was 51 mo (range 9-85 mo) giving a total of 280.5 patient years.A mean of 6 biopsies were taken during surveillance endoscopies. In 13 patients (19.7%) tongues or islands suspicious for BE were found during endoscopy. In 8 of these patients (12.1%) non-neoplastic BE relapse was confirmed histologically giving a histological relapse rate of 3% per year. 3Retrograde-viewing device improves adenoma detection rate in colonoscopies for surveillance and diagnostic workup显示文摘AIM:To determine which patients might benefit most from retrograde viewing during colonoscopy through subset analysis of randomized,controlled trial data.METHODS:The Third Eye Retroscope Randomized Clinical Evaluation(TERRACE) was a randomized,controlled,multicenter trial designed to evaluate the efficacy of a retrograde-viewing auxiliary imaging device that is used during colonoscopy to provide a second video image which allows viewing of areas on the proximal aspect of haustral folds and flexures that are difficult to see with the colonoscope's forward view.We performed a post-hoc analysis of the TERRACE data to determine whether certain subsets of the patient population would gain more benefit than others from use of the device.Subjects were patients scheduled for colonoscopy for screening,surveillance or diagnostic workup,and each underwent same-day tandem examinations with standard colonoscopy(SC) and Third Eye colonoscopy(TEC),randomized to SC followed by TEC or vice versa.RESULTS:Indication for colonoscopy was screening in 176/345 subjects(51.0%),surveillance after previous polypectomy in 87(25.2%) and diagnostic workup in 82(23.8%).In 4 subjects no indication was specified.Previously reported overall results had shown a net additional adenoma detection rate(ADR) with TEC of 23.2% compared to SC.Relative risk(RR) of missing adenomas with SC vs TEC as the initial procedure was 1.92(P = 0.029).Post-hoc subset analysis shows additional ADRs for TEC compared to SC were 4.4% for screening,35.7% for surveillance,55.4% for diagnostic and 40.7% for surveillance and diagnostic combined.The RR of missing adenomas with SC vs TEC was 1.11(P = 0.815) for screening,3.15(P = 0.014) for surveillance,8.64(P = 0.039) for diagnostic and 3.34(P = 0.003) for surveillance and diagnostic combined.Although a multivariate Poisson regression suggested gender as a possibly significant factor,subset analysis showed that the difference between genders was not statistically significant.Age,bowel prep quality and withdrawal time did not significantly affect the RR of missing adenomas with SC vs TEC.Mean sizes of adenomas detected with TEC and SC were similar at 0.59 cm and 0.56 cm,respectively(P = NS).CONCLUSION:TEC allows detection of significantly more adenomas compared to SC in patients undergoing surveillance or diagnostic workup,but not in screening patients(ClinicalTrials.gov Identifier:NCT01044732).Peter D Siersema Amit Rastogi Anke M Leufkens Paul A Akerman Kassem Azzouzi Richard I Rothstein Frank P Vleggaar Alessandro Repici Giacomo Rando Patrick I Okolo Olivier Dewit Ana Ignjatovic Elizabeth Odstrcil James East Pierre H Deprez Brian P Saunders Anthony N Kalloo Bradley Creel Vikas Singh Anne Marie Lennon Daniel C DeMarco 2012World Journal of Gastroenterology2012,18,26:6
4The fungal UmSrt1 and maize ZmSUT1 sucrose transporters battle for plant sugar resources显示文摘The biotrophic fungus Ustilago maydis causes corn smut disease, inducing tumor formation in its host Zea mays. Upon infection, the fungal hyphae invaginate the plasma membrane of infected maize cells, establishing an interface where pathogen and host are separated only by their plasma membranes. At this interface the fungal and maize sucrose transporters, UmSrt1 and ZmSUT1, compete for extracellular sucrose in the corn smut/maize pathosystem. Here we biophysically characterized ZmSUT1 and UmSrt1 in Xenopus oocytes with respect to their voltage-,pH-and substrate-dependence and determined affinities toward protons and sucrose. In contrast to ZmSUT1,UmSrt1 has a high affinity for sucrose and is relatively pH-and voltage-independent. Using these quantitative parameters, we developed a mathematical model to simulate the competition for extracellular sucrose at the contact zone between the fungus and the host plant. This approach revealed that UmSrt1 exploits the apoplastic sucrose resource, which forces the plant transporter into a sucrose export mode providing the fungus with sugar from the phloem. Importantly, the high sucrose concentration in the phloem appeared disadvantageous for the ZmSUT1, preventing sucrose recovery from the apoplastic space in the fungus/plant interface.Anke Wittek Ingo Dreyer Khaled A.S. 10Modulating effects of acyl-CoA synthetase 5-derived mitochondrial Wnt2B palmitoylation on intestinal Wnt activity显示文摘AIM:To investigate the role of acyl-CoA synthetase 5(ACSL5)activity in Wnt signaling in intestinal surface epithelia.METHODS:Several cell lines were used to investigate the ACSL5-dependent expression and synthesis of Wnt2B,a mitochondrially expressed protein of the Wnt signaling family.Wnt activity was functionally assessed with a luciferase reporter assay.ACSL5-related biochemical Wnt2B modifications were investigatedwith a modified acyl-exchange assay.The findings from the cell culture models were verified using an Apcmin/+mouse model as well as normal and neoplastic diseased human intestinal tissues.RESULTS:In the presence of ACSL5,Wnt2B was unable to translocate into the nucleus and was enriched in mitochondria,which was paralleled by a significant decrease in Wnt activity.ACSL5-dependent S-palmitoylation of Wnt2B was identified as a molecular reason for mitochondrial Wnt2B accumulation.In cell culture systems,a strong relation of ACSL5 expression,Wnt2B palmitoylation,and degree of malignancy were found.Using normal mucosa,the association of ACSL5 and Wnt2B was seen,but in intestinal neoplasias the mechanism was only rudimentarily observed.CONCLUSION:ACSL5 mediates antiproliferative activities via Wnt2B palmitoylation with diminished Wnt activity.The molecular pathway is probably relevant for intestinal homeostasis,overwhelmed by other pathways in carcinogenesis.Christina Klaus Ursula Schneider Christian Hedberg Anke K Schütz Jürgen Bernhagen Herbert Waldmann Nikolaus Gassler Elke Kaemmerer 2014World Journal of Gastroenterology2014,20,40:4
11Transcriptome analysis of Sinorhizobium meliloti nodule bacteria in nifA mutant background显示文摘Gene expression profiles of a Si- norhizobium meliloti 1021 nifA mutant and wild type nodule bacteria were compared using whole genome microarrays. The results revealed a large scale al- teration of gene expression (601 genes) in the nifA minus background. The loss of NifA altered the ex- pression of many functional groups of genes (mac- romolecular metabolism, TCA cycle and respiration, nodulation and nitrogen fixation) and may lead to quite different life stages of the nodule bacteria. Upregulation of fixK and its associated genes was observed in the nifA mutant nodule bacteria. Addi- tional quantitative real-time PCR experiments re- vealed that the transcript levels of fixLJ were signifi- cantly upshifted in the nifA mutant nodule bacteria. 14Feedback structure based entropy approach for multiple-model estimation显示文摘The variable-structure multiple-model(VSMM)approach,one of the multiple-model(MM)methods,is a popular and effective approach in handling problems with mode uncertainties.The model sequence set adaptation(MSA)is the key to design a better VSMM.However,MSA methods in the literature have big room to improve both theoretically and practically.To this end,we propose a feedback structure based entropy approach that could fnd the model sequence sets with the smallest size under certain conditions.The fltered data are fed back in real time and can be used by the minimum entropy(ME)based VSMM algorithms,i.e.,MEVSMM.Firstly,the full Markov chains are used to achieve optimal solutions.Secondly,the myopic method together with particle flter(PF)and the challenge match algorithm are also used to achieve sub-optimal solutions,a trade-off between practicability and optimality.The numerical results show that the proposed algorithm provides not only refned model sets but also a good robustness margin and very high accuracy.Shen-tu Han Xue Anke Guo Yunfei 2013Chinese Journal of Aeronautics2013,26,6:3
15Enhancing point defect scattering in copper antimony selenides via Sm and S Co-doping显示文摘Doping-and alloying-induced point defects lead to mass and strain field fluctuations which can be used as effective strategies to decrease the lattice thermal conductivity and consequently boost the performance of thermoelectric materials. Herein, we report the effects of Sm and S co-doping on thermoelectric transport properties of copper antimony selenides in the temperature range of300 K < T < 650 K. Through the Callaway model, it demonstrates that Sm and S co-doping induces strong mass differences and strain field fluctuations in Cu3 SbSe4. The results prove that doping with suitable elements can increase point defect scattering of heat-carrying phonons,leading to a lower thermal conductivity and a better thermoelectric performance. 16Solids flow diagram of a CFB riser using Geldart B-type powders显示文摘Riser operating modes are vital to designing a circulating fluidized bed (CFB) reactor for a required process of either a gas-solid or a gas-catalytic nature. Different operating modes provide different solids' residence times and mixing behaviors, which define the reactions' efficiency and yield. The literature demonstrates distinct operating modes resulting from observed differences in slip factors and the range of particle velocities and their associated residence time distribution. The present research uses positron emission particle tracking (PEPT) in a riser of B-type bed material to determine the different operating modes by measuring (i) particle velocities and residence time distribution, (ii) population densities of these particles in the cross-sectional area of the riser, and (iii) solids flow pattern at the bottom of the riser. Data treatment defines four distinct solids hold-up regimes in the riser and proposes a 'phase diagram' depicting the existence of the different operating modes (dilute, dense, core-annulus and combined) as a function of the superficial gas velocity and solids circulation flux in the riser. The delineated regimes have good agreement with available literature data and known industrial operations. Comparison with literature data for risers using A-type powders is also fair.
