| 2 | Preoperative diagnosis of malignant pulmonary nodules in lung cancer screening with a radiomics nomogram显示文摘Background:Lung cancer is the most commonly diagnosed cancer worldwide.Its survival rate can be significantly improved by early screening.Biomarkers based on radiomics features have been found to provide important physiological information on tumors and considered as having the potential to be used in the early screening of lung cancer.In this study,we aim to establish a radiomics model and develop a tool to improve the discrimination between benign and malignant pulmonary nodules.Methods:A retrospective study was conducted on 875 patients with benign or malignant pulmonary nodules who underwent computed tomography(CT)examinations between June 2013 and June 2018.We assigned 612 patients to a training cohort and 263 patients to a validation cohort.Radiomics features were extracted from the CT images of each patient.Least absolute shrinkage and selection operator(LASSO)was used for radiomics feature selection and radiomics score calculation.Multivariate logistic regression analysis was used to develop a classification model and radiomics nomogram.Radiomics score and clinical variables were used to distinguish benign and malignant pulmonary nodules in logistic model.The performance of the radiomics nomogram was evaluated by the area under the curve(AUC),calibration curve and Hosmer-Lemeshow test in both the training and validation cohorts.Results:A radiomics score was built and consisted of 20 features selected by LASSO from 1288 radiomics features in the training cohort.The multivariate logistic model and radiomics nomogram were constructed using the radiomics score and patients’age.Good discrimination of benign and malignant pulmonary nodules was obtained from the training cohort(AUC,0.836;95%confidence interval[CI]:0.793-0.879)and validation cohort(AUC,0.809;95%CI:0.745-0.872).The Hosmer-Lemeshow test also showed good performance for the logistic regression model in the training cohort(P=0.765)and validation cohort(P=0.064).Good alignment with the calibration curve indicated the good performance of the nomogram.Conclusions:The established radiomics nomogram is a noninvasive preoperative prediction tool for malignant pulmonary nodule diagnosis.Validation revealed that this nomogram exhibited excellent discrimination and calibration capacities,suggesting its clinical utility in the early screening of lung cancer. | Ailing Liu Zhiheng Wang Yachao Yang Jingtao Wang Xiaoyu Dai Lijie Wang Yuan Lu Fuzhong Xue | 2020 | Cancer Communications2020,40,1: | 22 |

| 4 | Oxymatrine reduces neuroinflammation in rat brain A signaling pathway显示文摘Cerebral neuroinflammation models were established by injecting 10 μg lipopolysaccharide into the hippocampus of male Sprague-Dawley rats. The rats were treated with an intraperitoneal injection of 120, 90, or 60 mg/kg oxymatrine daily for three days prior to the lipopolysaccharide injection. Twenty-four hours after model induction, the hippocampus was analyzed by real-time quantitative PCR, and the cerebral cortex was analyzed by enzyme-linked immunosorbent assay and western blot assay. The results of the enzyme-linked immunosorbent assay and the real-time quantitative PCR showed that the secretion and mRNA expression of the pro-inflammatory cytokines interleukin-1β and tumor necrosis factor-α were significantly decreased in the hippocampus and cerebral cortex of model rats treated with oxymatrine. Western blot assay and real-time quantitative PCR analysis indicated that toll-like receptor 4 mRNA and protein expression were significantly decreased in the groups receiving different doses of oxymatrine. Additionally, 120 and 90 mg/kg oxymatrine were shown to reduce protein levels of nuclear factor-κB p65 in the nucleus and of phosphorylated IκBα in the cytoplasm of brain cells, as detected by western blot assay. | 8 | Two-Phase Rate Adaptation Strategy for Improving Real-Time Video QoE in Mobile Networks显示文摘With the popularity of smart handheld devices, mobile streaming video has multiplied the global network traffic in recent years. A huge concern of users' quality of experience(Qo E) has made rate adaptation methods very attractive. In this paper, we propose a two-phase rate adaptation strategy to improve users' real-time video Qo E. First, to measure and assess video Qo E, we provide a continuous Qo E prediction engine modeled by RNN recurrent neural network. Different from traditional Qo E models which consider the Qo E-aware factors separately or incompletely, our RNN-Qo E model accounts for three descriptive factors(video quality, rebuffering, and rate change) and reflects the impact of cognitive memory and recency. Besides, the video playing is separated into the initial startup phase and the steady playback phase, and we takes different optimization goals for each phase: the former aims at shortening the startup delay while the latter ameliorates the video quality and the rebufferings. | 9 | Tectonic Implications and Petrogenesis of the Various Types of Magmatic Rocks from the Zedang Area in Southern Tibet显示文摘In this study,we report systematically field observations,geochronology,whole-rock geochemistry and Sr-Nd-Hf isotopic dataset on the various types of magmatic rocks collected from the Zedang area.Chemically,the diabase and gabbro have a low-K calc-alkaline affinity,whereas the basalt and plagiogranite have medium to high-K calc-alkaline characteristics.In addition,the basalts are highly enriched in light rare earth elements(LREE)and large ion lithophile elements(LILE),but strongly depleted in high strength field elements(HFSE),indicating that their magma source probably was derived from a subduction-or arc-related setting.In contrast,both the gabbro and diabase mainly demonstrate an N-MORB-like affinity consistent with normal mid-oceanic ridge basalt(N-MORB)origin.The zircon U-Pb dating results suggest that the basalts were crystallized earlier at ca.158–161 Ma(Oxfordian stage),but the gabbro was crystallized at ca.131 Ma(Hauterivian stage of Early Cretaceous).The zircon U-Pb dating results correspond with the field observations that the veins of gabbro intruded basalt.Furthermore,the plagiogranite has a weighted mean age of ca.160 Ma(MSWD=2.1)consistent with the basalt within the uncertainty.The basalt and the plagiogranite have significantly positiveεHf(t)values(+5.8 to+15.6 and+8.6 to+16.1,respectively),suggesting that they were originated from partial melting of a depleted source.However,basalt and plagiogranite are characterized by the wide variations ofεHf(t)values indicating minor amounts of exotic crustal material input during the later magma evolution.Additionally,the basalt shows duplex geochemical features of island-arc and mid-oceanic ridge basalt,corresponding to the supra-subduction zone-(SSZ)type affinity.To sum up,two distinct magmatic events identified in this study probably suggest an intra-oceanic arc system existing in the Zedang area during the Late Jurassic,but the intra-oceanic arc subduction extinguished in the Early Cretaceous as suggested by the N-MORB-like gabbro and diabase.Integrated with regional background and different rock types,as well as geochemical features,we conclude that intra-oceanic arc subduction setting developed during the Late Jurassic in the Zedang area,southern Tibet. | Yuanku Mengc Fahui Xiong Jingsui Yang Zhao Liu Kieran AIles Paul TRobinson Xiangzhen Xu | 2019 | Journal of Earth Science2019,30,6: | 2 |

| 18 | SHANK2 is a frequently amplified oncogene with evolutionarily conserved roles in regulating Hippo signaling显示文摘Dysfunction of the Hippo pathway enables cells to evade contact inhibition and provides advantages for cancerous overgrowth.However,for a significant portion of human cancer,how Hippo signaling is perturbed remains unknown.To answer this question,we performed a genome-wide screening for genes that affect the Hippo pathway in Drosophila and cross-referenced the hit genes with human cancer genome.In our screen,Prosap was identified as a novel regulator of the Hippo pathway that potently affects tissue growth.Interestingly,a mammalian homolog of Prosap,SHANK2,is the most frequently amplified gene on 11 q13,a major tumor amplicon in human cancer.Gene amplification profile in this 11q13 amplicon clearly indicates selective pressure for SHANK2 amplification.More importantly,across the human cancer genome,SHANK2 is the most frequently amplified gene that is not located within the Myc amplicon.Further studies in multiple human cell lines confirmed that SHANK2 overexpression causes deregulation of Hippo signaling through competitive binding for a LATS1 activator,and as a potential oncogene,SHANK2 promotes cellular transformation and tumor formation in vivo.In cancer cell lines with deregulated Hippo pathway,depletion of SHANK2 restores Hippo signaling and ceases cellular proliferation.Taken together,these results suggest that SHANK2 is an evolutionarily conserved Hippo pathway regulator,commonly amplified in human cancer and potently promotes cancer.Our study for the first time illustrated oncogenic function of SHANK2,one of the most frequently amplified gene in human cancer.Furthermore,given that in normal adult tissues,SHANK2 s expression is largely restricted to the nervous system,SHANK2 may represent an interesting target for anticancer therapy. | Liang Xu Peixue Li Xue Hao Yi Lu Mingxian Liu Wenqian Song Lin Shan Jiao Yu Hongyu Ding Shishuang Chen Ailing Yang Yi Arial Zeng Lei Zhang Hai Jiang | 2021 | Protein & Cell2021,12,3: | 1 |
