1Single-cell transcriptomic atlas of primate cardiopulmonary aging显示文摘Aging is a major risk factor for many diseases,especially in highly prevalent cardiopulmonary comorbidities and infectious diseases including Coronavirus Disease 2019(COVID-19).Resolving cellular and molecular mechanisms associated with aging in higher mammals is therefore urgently needed.Here,we created young and old non-human primate single-nucleus/cell transcriptomic atlases of lung,heart and artery,the top tissues targeted by SARS-CoV-2.Analysis of cell type-specific aging-associated transcriptional changes revealed increased systemic inflammation and compromised virus defense as a hallmark of cardiopulmonary aging.With age,expression of the SARS-CoV-2 receptor angiotensin-converting enzyme 2(ACE2)was increased in the pulmonary alveolar epithelial barrier,cardiomyocytes,and vascular endothelial cells.We found that interleukin 7(IL7)accumulated in aged cardiopulmonary tissues and induced ACE2 expression in human vascular endothelial cells in an NF-κB-dependent manner.Furthermore,treatment with vitamin C blocked IL7-induced ACE2 expression.Altogether,our findings depict the first transcriptomic atlas of the aged primate cardiopulmonary system and provide vital insights into age-linked susceptibility to SARS-CoV-2,suggesting that geroprotective strategies may reduce COVID-19 severity in the elderly.Shuai Ma Shuhui Sun Jiaming Li Yanling Fan Jing Qu Liang Sun Si Wang Yiyuan Zhang Shanshan Yang Zunpeng Liu Zeming Wu Sheng Zhang Qiaoran Wang Aihua Zheng Shuguang Duo Yang Yu Juan Carlos Izpisua Belmonte Piu Chan Qi Zhou Moshi Song Weiqi Zhang Guang-Hui Liu 2021Cell Research2021,31,4:10
2Stabilization of heterochromatin by CLOCK promotes stem cell rejuvenation and cartilage regeneration显示文摘Accumulating evidence indicates an association between the circadian clock and the aging process.However,it remains elusive whether the deregulation of circadian clock proteins underlies stem cell aging and whether they are targetable for the alleviation of aging-associated syndromes.Here,we identified a transcription factor-independent role of CLOCK,a core component of the molecular circadian clock machinery,in counteracting human mesenchymal stem cell(hMSC)decay.CLOCK expression was decreased during hMSC aging.In addition,CLOCK deficiency accelerated hMSC senescence,whereas the overexpression of CLOCK,even as a transcriptionally inactive form,rejuvenated physiologically and pathologically aged hMSCs.Mechanistic studies revealed that CLOCK formed complexes with nuclear lamina proteins and KAP1,thus maintaining heterochromatin architecture and stabilizing repetitive genomic sequences.Finally,gene therapy with lentiviral vectors encoding CLOCK promoted cartilage regeneration and attenuated age-related articular degeneration in mice.These findings demonstrate a noncanonical role of CLOCK in stabilizing heterochromatin,promoting tissue regeneration,and mitigating aging-associated chronic diseases.Chuqian Liang Zunpeng Liu Moshi Song Wei Li Zeming Wu Zehua Wang Qiaoran Wang Si Wang Kaowen Yan Liang Sun Tomoaki Hishida Yanning Cai Juan Carlos lzpisua Belmonte Pedro Guillen Piu Chan Qi Zhou Weiqi Zhang Jing Qu Guang-Hui Liu 2021Cell Research2021,31,2:8
3Single small molecule-assembled nanoparticles mediate efficient oral drug delivery显示文摘Oral drug delivery,which requires surviving the harsh environment in the gastrointestinal(Gl)tract and penetrating the intestinal epithelium,has not bee n achieved using simple formulatio n nan oparticles(NPs).Medici nal natural products(MNPs)have bee n widely used in traditi onal medicine for disease management through oral consumption.However,most pharmacologically active compounds within MNPs do not have the properties suitable for oral applicatio ns.We hypothesize that some MNPs contain n atural nano materials that can convert those compounds into oral formulations by forming NPs.After screening 66 MNPs,we identified five classes of small molecules that form NPs,many of which are capable of efficient drug encapsulation and Gl penetration.We show that one of them,dehydrotrametenolic acid(DTA),is capable of mediating oral delivery for effective disease treatment.We determined that DTA NPs assemble through hydrogen bonding and penetra怕the Gl tract via apical sodium-depe ndent bile acid tran sporter.Our study reveals a no vel class of single comp orient,small molecule-assembled NPs for oral drug delivery,and suggests a n ovel approach to modernizi ng MNPs through nano material discovery.Xin Yang Chao Ma Zeming Chen Jun Liu Fuyao Liu Rongbin Xie Haitian Zhao Gang Deng Ann TChen Ningbo Gong Lei Yao Pengjian Zuo Kangkang Zhi Jiacheng Wang Xiaobin Gao Jing Wang Louzhen Fan Jiangbing Zhou 2019Nano Research2019,12,10:4
4Synthesis,Characterization of Rare-Earth Metal Chlorides Bearing Indolyl-Based NCN Pincer Ligand and Their Catalytic Activity toward 1,4-cis Polymerization of Isoprene显示文摘A novel indolyl-based NCN pincer ligand precursor HL(HL=(1-(2-(CH_(2))_(4)NCH_(2)CH_(2))-3-(2,6-ipr_(2)C_(6)H_(3)N=CH)C_(8)H_(5)N))was rationally and precisely designed,and successfully prepared in a high yield.Lithiation of HL with ^(n)BuLi in n-hexane,followed by treatment with RECl_(3) in THF led to the isolation of rare-earth metal chlorides in monomeric form(κ^(3)-L)RECI_(2)(THF)(RE=Sc(2),Yb(3)),and dimeric form[(κ^(3)-L)RECI(THF)(μ-Cl)]_(2)(RE=Er(4),Y(5))bearing the novel indolyl-based NCN pincer ligand in designed pincer ligation depending on different metal ionic radii;and the chloride bridged rare-earth metal and lithium mixed complexes in form(κ^(2)-L)_(2)RE(μ-Cl)_(2)Li(THF)_(2)(RE=Y(6),Dy(7)).All complexes were fully characterized and their structures were determined.The diamagnetic complexes 2,5 and 6 were furtherly characterized by the NMR spectroscopic method.The catalytic activity studies revealed that the central metal ions,borates,aluminum alkyls,and ligation of the central metal ions have influences on catalytic activity,and regio-and stereoselectivity for the polymerization of isoprene.With cooperation of the cocatalysts,the pincer-type yttrium chloride 5 initiated the isoprene polymerization with a high activity producing polymers with high regio-and stereoselectivity(1,4-cis polymers up to 99.6%),and the Mn as high as 8.76×10^(5).Zeming Huang Shaowu Wang Xiancui Zhu Yun Wei Qingbing Yuan Shuangliu Zhou Xiaolong Mu Hua Wang 2021Chinese Journal of Chemistry2021,39,12:2
5Pan-sharpening:a fast variational fusion approach显示文摘A fast variational fusion model based on partial differential equations (PDEs) is presented for pansharpening. The functional framework consists of several energy terms. The gradient energy term is created by calculating the gradient vector field of the panchromatic image and the geometry of the pan is injected into the multi-spectral bands. The radiometric reduction energy term and the channel correlation energy term are defined to decrease the radiometric distortion and preserve the correlation of multi-spectral channels while enforcing the inter-bands coherence. Inspired by the shock-filtering model, an inverse diffusion term for image enhancement is put to PDEs which are deduced by minimizing the energy functional. In comparison with the state-of-the-art fusion approaches based on `a trous wavelet and non-sampled contourlet, our model can obtain the fused image with a high spatial or spectral quality by adjusting the weight coefficients of the energy terms. It can also achieve a rather good trade-off between the spatial resolution improvement and the spectral quality preserving. 9P-Wave Velocity in Rocks of Dabieshan, China at High Pressure and High Temperature: Constraints for Composition of Lower Crust and Crust-Mantle Recycling显示文摘P-wave velocities in the rocks of Dabieshan, central China were measured at pressures up to 5.0 GPa and temperatures up to 1 300℃. The ultrahigh pressure eclogites have the highest density and P-wave velocity (Vp) and lower anisotropy. Pressure derivatives of the eclogites range from 0. 22 to 0. 33 km. s-1 GPa-1. Average temperature derivative of the eclogites is - 3. 41×10-4 km. s-1. °C -1. The density and VP of the eclogites imply that there will be two united possibilities related to crust-mantle recycling after the eclogite formed in the deep lithosphere. One is that some eclogites in the deep lithosphere were detached and sunk into deeper mantle due to their denser density. 10Brain-targeting,acid-responsive antioxidant nanoparticles for stroke treatment and drug delivery显示文摘Stroke is the leading cause of death and disability.Currently,there is no effective pharmacological treatment for this disease,which can be partially attributed to the inability to efficiently deliver therapeutics to the brain.Here we report the development of natural compound-derived nanoparticles(NPs),which function both as a potent therapeutic agent for stroke treatment and as an efficient carrier for drug delivery to the ischemic brain.First,we screened a collection of natural nanomaterials and identified betulinic acid(BA)as one of the most potent antioxidants for stroke treatment.Next,we engineered BA NPs for preferential drug release in acidic ischemic tissue through chemically converting BA to betulinic amine(BAM)and for targeted drug delivery through surface conjugation of AMD3100,a CXCR4 antagonist.The resulting AMD3100-conjugated BAM NPs,or A-BAM NPs,were then assessed as a therapeutic agent for stroke treatment and as a carrier for delivery of NA1,a neuroprotective peptide.We show that intravenous administration of A-BAM NPs effectively improved recovery from stroke and its efficacy was further enhanced when NA1 was encapsulated.Due to their multifunctionality and significant efficacy,we anticipate that A-BAM NPs have the potential to be translated both as a therapeutic agent and as a drug carrier to improve the treatment of stroke.Shenqi Zhang Bin Peng Zeming Chen Jiang Yu Gang Deng Youmei Bao Chao Ma Fengyi Du Wendy C.Sheu W.Taylor Kimberly J.Marc Simard Daniel Coman Qianxue Chen Fahmeed Hyder Jiangbing Zhou Kevin N.Sheth 2022Bioactive Materials2022,7,10:1

12Metamorphic P-T-t Path of UHT Granulites from the North Tongbai Orogen, Central China显示文摘Although zircon U-Pb geochronometer has been widely used for dating metamorphism in moderate-to high-grade metamorphic rocks, it is still difficult to link the zircon U-Pb age to pressure and temperature(P-T) conditions. In this study, zircon trace elements and Hf isotopes and REE partitioning between zircon and garnet are adopted to track the formation condition of zircon in the granulites from North Tongbai Orogen, Central China. Combined with previous metamorphic P-T path results, a quantitative integrated anticlockwise P-T-t path was established for Tongbai granulites. These granulites recorded an early low-P heating followed by a dramatic pressure increase. Evidence for the prograde history(M1) is provided by cordierite, orthopyroxene and biotite inclusions in garnet. The prograde metamorphism occurred at around 443±3 Ma, with P-T conditions of ca. 730–820 ℃ and <6 kbar. The peak metamorphic(M2) condition is >920 ℃ and 8.5–10 kbar and the peak metamorphism age is ca. 432±4 Ma. At around 419 Ma, the granulites suffered an amphibolite-facies retrograde metamorphism(M3), represented by the replacement of garnet by biotite and plagioclase, and clinopyroxene by amphibole, with metamorphic condition of ca. 700 ℃ and ca. 7 kbar. The last retrograde metamorphism(M4) is a greenschist-facies overprint with an age of ca. 404 Ma. It is concluded that the metamorphism of Tongbai granulite lasted for more than 40 Ma, including a stage of more than 20 Ma granulite-facies metamorphism. 14Variational PCA fusion for Pan-sharpening very high resolution imagery显示文摘The Pan-sharpening approach based on principle component analysis(PCA) is affected by severe spectral distortion. To address this problem, a new pan-sharpening model based on PCA and variational technique is proposed to construct the substitute image of the first principal component(PC1). The energy functional consists of three terms. The first term injects PC1 with the geometric structure of the panchromatic(Pan) image. The second term preserves the spectral pattern of the multi-spectral image in the merged result.And the third term guarantees the smoothness of the functional optimization solution. The fusion result is given by the minimum of the energy functional, which is computed with the gradient descend flow. The experiments on QuickBird and IKONOS datasets validate the effectiveness of the proposed model. 20Unusual selective reactivity of the rare-earth metal complexes bearing a ligand with multiple functionalities显示文摘Ligands play a key role in controlling activity of organometallic complexes so that development of new ligands to overcome the challenge is the main topic of modern chemistry.The first example of 1,1-hydride migratory insertion and intramolecular redox reaction has been realized in this work by applying a new ligand in rare-earth metal chemistry.The novel rare-earth metal complexes L^(Mes)RECH2TMS(THF)(RE=Y(1a),Dy(1b),Er(1c),Yb(1d),L^(Mes)=1-(3-(2,6-iPr_(2)C_(6)H_(3)N=CH)C8H4N)-CH_(2)CH_(2)-3-(2-CH2–4,6-Me_(2)C_(6)H_(2))-(N(CH)_(2)NC),THF=tetrahydrofuran)bearing a ligand with imino,indolyl,NHC(N-heterocyclic carbene)multiple functionalities were synthesized and characterized.Treatment of complexes 1 with silanes(PhSiH3or PhSiH2Me or PhSiD3)selectively produced the unprecedented 1,1-hydride(or deuterated H)migratory insertion of the indolyl moiety of the novel unsymmetrical dinuclear rare-earth metal complexes 2.The complex 2a reacts with Ph_(2)C=O to give the selective C=O double bond insertion to the RE–Co-methylene-Mesbond product 3a which further reacts with another Ph_(2)C=O(or DMAP,4-N,N-dimethylaminopyridine)affording the novelμ-η^(2):η^(3)-dianionic 3-iminoindolyl dinuclear rare-earth metal complex 4a.The latter is formed through an unusual intramolecular redox reaction(through electron migration from the 2-carbanion of the indolyl ring to the imino motif)resulting in the re-aromatization of the indolyl ring.Dongjing Hong Thayalan Rajeshkumar Shan Zhu Zeming Huang Shuangliu Zhou Xiancui Zhu Laurent Maron Shaowu Wang 2023Science China Chemistry2023,66,1:0
