| 1 | Evolution of strontium isotopic composition of sea-water from Late Permian to Early Triassic based on study of marine carbonates,Zhongliang Mountain,Chongqing,China显示文摘Collected from a Late Permian to Early Triassic sedimentary section in the Zhongliang Mountain of Chongqing, Southwest China, sixty marine carbonate samples were measured for the 87Sr/86Sr ratios, and corresponding evolution curve was constructed. The concentrations of SiO2, CaO, MgO, Mn and Sr are used to evaluate reservation of strontium isotopic composition for original seawater and the credi-bility of the dissolution method for sample preparation. The results show that most of the samples (except seven samples with the Mn/Sr ratios higher than 2) contain the original geochemistry signa-tures of ancient seawater. Compared to the published 87Sr/86Sr ratios from the Late Permian to Early Triassic, our database reported here is the largest and the curve constructed is the most complete. The strontium isotopic curve from Late Permian to Early Triassic is consistent globally and exhibits a gen-eral trend of steady increase during this period. The minimum of 87Sr/86Sr ratios (0.707011) occurs in the Late Permian (30 m in thickness below the Permian-Triassic boundary), and the maximum (0.708281), near the Early-Middle Triassic boundary. The lack of land plants and the rapid continental weathering result in the increase of 87Sr/86Sr ratios during the interval. The Permian-Triassic boundary in Zhongli-ang Mountain Section has been accepted internationally. The 87Sr/86Sr ratios of six samples near the boundary vary from 0.70714 to 0.70715 with an average of 0.70714, which is consistent with the value of 0.70715 (samples are from articulate brachiopod shells) from Korte et al. published in 2006 (within the error range in experiment). Accordingly, the strontium isotope composition in the Permian-Triassic boundary in this paper is of global significance. | 2 | A strategy with label-free quantification of the targeted peptides for quantitative peptidome analysis of human serum显示文摘Peptidomics draws more and more attention in discovering useful biomarkers for early diagnosis of disease. However, there is lack of efficient quantification strategy in peptidome analysis. In this study, a strategy with label-free quantification of the targeted endogenous peptides based on peak intensity using μUPLC-Q-TOF-MS/MS was developed for quantitative peptidome analysis of human serum. Different amounts of standard BSA tryptic digesting peptides were added into the same serum extracts for evaluation of the developed strategy, and it was observed that the average relative error of the targeted peptides was 6.42%, which was superior to the result obtained directly by commercially available software PLGS. It was also demonstrated that this quantification strategy could obviously increase the detection sensitivity of the peptide by DDA analysis. Then, this strategy was applied to comparatively analyze the peptides extracted from the serum of HCC or breast cancer patients and healthy individuals, respectively. Peptides with charge states up to 5 and molecular weight over 4000 can be reliably identified and quantified. This quantitative analysis method based on μUPLC-Q-TOF-MS/MS exhibited superior sensitivity than that by MALDI-TOF-MS commonly used in peptidome analysis. | 3 | Corrosion Behavior of Anodic Oxidized TiO_2 Film in Seawater显示文摘TiO2 films were formed on metallic titanium substrates by the anodic oxidation method in H2SO4 solution under the 80V D.C..Phase component and microstructure were characterized by X-ray diffraction (XRD) and scanning electron microscopy (SEM).Water contact angles on titanium oxide film surface were measured under both dark and sunlight illumination conditions.Corrosion tests were carried out in seawater under different illumination conditions by electrochemistry impedance spectrum (EIS) and polarization curves.The result showed that the TiO2 film prepared by the anodic oxidation method was anatase with a uniform structure and without obvious pores or cracks on its surface.The average water contact angle of the film was 116.4? in dark, in contrast to an angle of 42.7? under the UV illumination for 2 hours, which demonstrates good hydrophobic property.The anti-corrosion behavior of the TiO2 film was declining with the extended immersion time.Under dark conditions, however, the hydrophobic TiO2 film retarded the water infiltrating into the substrate.The impedance changed slowly and the corrosion current density was 2 orders of magnitude lower than that with the film illuminated by sunlight.All of those mentioned above indicate that the TiO2 film possesses much better performance under dark condition, and it can be applied as an engineering material under dark seawater environment. | WANG Min WANG Wei HE Benlin SUN Mingliang YIN Yansheng LIU Lan ZOU Wuyuan XU Xuefei | 2010 | Journal of Ocean University of China2010,9,4: | 2 |

| 20 | Targeting TRPV1-mediated autophagy attenuates nitrogen mustard-induced dermal toxicity显示文摘Nitrogen mustard(NM)causes severe vesicating skin injury,which lacks effective targeted therapies.The major limitation is that the specific mechanism of NM-induced skin injury is not well understood.Recently,autophagy has been found to play important roles in physical and chemical exposure-caused cutaneous injuries.However,whether autophagy contributes to NM-induced dermal toxicity is unclear.Herein,we initially confrmed that NM dose-dependently caused cell death and induced autophagy in keratinocytes.Suppression of autophagy by 3-methyladenine,chloroquine,and baflomycin A1 or ATG5 siRNA attenuated NM-induced keratinocyte cell death.Furthermore,NM increased transient receptor potential vanilloid 1(TRPV1)expression,intracellular Ca2+content,and the activities of Car+/calmodulin-dependent kinase kinaseβ(CaMKKB),AMP-activated protein kinase(AMPK),unc-51-like kinase 1(ULK1),and mammalian target of rapamycin(mTOR).NM-induced autophagy in keratinocytes was abolished by treatment with inhibitors of TRPV1(capsazepine),CaMKKβ(STO-609),AMPK(compound C),and ULK1(SBI-0206965)as well as TRPV1,CaMKKB,and AMPK siRNA transfection.In addition,an mTOR inhibitor(rapamycin)had no signifcant effect on NM-stimulated autophagy or cell death of keratinocytes.Finally,the results of the in vivo experiment in NM-treated skin tissues were consistent with the findings of the in vitro experiment.In conclusion,NM-caused dermal toxicity by overactivating autophagy partially through the activation of TRPV1-Ca2+-CaMKKB-AMPK-ULK1 signaling pathway.These results suggest that blocking TRPV1-dependent autophagy could be a potential treatment strategy for NM-caused cutaneous injury. | Mingliang Chen Xunhu Dong Haoyue Deng Feng Ye Yuanpeng Zhao Jin Cheng Guorong Dan Jiqing Zhao Yan Sai Xiuwu Bian Zhongmin Zou | 2021 | Signal Transduction and Targeted Therapy2021,6,2: | 0 |
