| 1 | Multiple checkpoints keep follicular helper T cells under control to prevent autoimmunity显示文摘Follicular helper T(Tfh)cells select mutated B cells in germinal centres,which can then differentiate into long-lived high affinity memory B cells and plasma cells.Tfh cells are regulated by a unique molecular programme orchestrated by the transcriptional repressor Bcl6.This transcription factor turns down expression of multiple genes,including transcriptional regulators of other T helper lineages and a vast amount of microRNAs.This enables Tfh cells to express a suite of chemokine receptors,stimulatory ligands and cytokines that enable migration into B-cell follicles,and provision of effective help to B cells.Not surprisingly,dysregulation of this powerful helper subset can lead to a range of autoantibody-mediated diseases;indeed,aberrant accumulation of Tfh cells has been linked with systemic lupus erythematosus,Sjogren’s disease and autoimmune arthritis.Here we dissect multiple checkpoints that operate throughout Tfh cell development and maturation to maintain immunological tolerance while mounting robust and long-lasting antibody responses. | Di Yu Carola G Vinuesa | 2010 | Cellular & Molecular Immunology2010,7,3: | 16 |
| 2 | Covalently closed-circular hepatitis B virus DNA reduction with entecavir or lamivudine显示文摘AIM: To investigate the reduction in hepatitis B virus(HBV) covalently closed-circular DNA(ccc DNA) with entecavir(ETV) or lamivudine(LAM). METHODS: This analysis included patients who had participated in the randomized Phase Ⅲ study ETV-022 comparing ETV vs LAM in nucleos(t)ide-naive, HBe Agpositive patients. Patients received ETV(0.5 mg daily) or LAM(100 mg daily) for a minimum of 52 wk. Patients were eligible to participate in this sub-study if they had paired biopsies at baseline and week 48 with evaluable measurements for hepatic HBV ccc DNA and total hepatic HBV DNA. The main objective was to compare changes in hepatic HBV ccc DNA and total hepatic HBV DNA at week 48 of ETV or LAM treatment, which was a secondary endpoint of study ETV-022. Additional post hoc analyses included linear regression analyses to assess associations of baseline levels and on-treatment changes of ccc DNA with other baseline factors [sex,age, serum HBV DNA, alanine aminotransferase(ALT), Knodell necroinflammatory score, Ishak fibrosis score, total hepatic HBV DNA, and HBV genotype], or ontreatment factors(changes from baseline at week 48 in serum HBV DNA, ALT, Knodell necroinflammatory score, Ishak fibrosis score, total hepatic HBV DNA, and HBe Ag loss at week 48).RESULTS: Overall, 305 patients(ETV = 159; LAM = 146) of ETV-022 had paired baseline and week 48 liver biopsies with evaluable measurements for hepatic HBV ccc DNA and total hepatic HBV DNA, and were included in this analysis. Baseline demographics and disease characteristics were comparable between the two arms. After 48 wk, ETV resulted in significantly greater reductions in hepatic HBV ccc DNA [-0.9 log10 copies/human genome equivalent(HGEq) vs-0.7 log10 copies/HGEq; P = 0.0033] and total hepatic DNA levels(-2.1 log10 copies/HGEq vs-1.6 log10 copies/HGEq; P < 0.0001) than LAM. Virologic, biochemical, and histologic response rates at week 48 were also greater with ETV than with LAM. Baseline HBV ccc DNA levels were positively associated with baseline levels of serum HBV DNA and total hepatic HBV DNA, and negatively associated with HBV genotype F. | 4 | Co-effect of Demand-control-support Model and Effort-reward Imbalance Model on Depression Risk Estimation in Humans: Findings from Henan Province of China显示文摘Objective To investigate the co-effect of Demand-control-support(DCS) model and Effort-reward Imbalance(ERI) model on the risk estimation of depression in humans in comparison with the effects when they are used respectively. Methods A total of 3 632 males and 1 706 females from 13 factories and companies in Henan province were recruited in this cross-sectional study. Perceived job stress was evaluated with the Job Content Questionnaire and Effort-Reward Imbalance Questionnaire(Chinese version). Depressive symptoms were assessed by using the Center for Epidemiological Studies Depression Scale(CES-D). Results DC(demands/job control ratio) and ERI were shown to be independently associated with depressive symptoms. The outcome of low social support and overcommitment were similar. High DC and low social support(SS), high ERI and high overcommitment, and high DC and high ERI posed greater risks of depressive symptoms than each of them did alone.ERI model and SS model seem to be effective in estimating the risk of depressive symptoms if they are used respectively. Conclusion The DC had better performance when it was used in combination with low SS. The effect on physical demands was better than on psychological demands. | 5 | Genome resequencing and transcriptome profiling reveal structural diversity and expression patterns of constitutive disease resistance genes in Huanglongbing-tolerant Poncirus trifoliata and its hybrids显示文摘Huanglongbing(HLB)is the most destructive bacterial disease of citrus worldwide.While most citrus varieties are susceptible to HLB,Poncirus trifoliata,a close relative of Citrus,and some of its hybrids with Citrus are tolerant to HLB.No specific HLB tolerance genes have been identified in P.trifoliata but recent studies have shown that constitutive disease resistance(CDR)genes were expressed at much higher levels in HLB-tolerant Poncirus hybrids and the expression of CDR genes was modulated by Candidatus Liberibacter asiaticus(CLas),the pathogen of HLB.The current study was undertaken to mine and characterize the CDR gene family in Citrus and Poncirus and to understand its association with HLB tolerance in Poncirus.We identified 17 CDR genes in two citrus genomes,deduced their structures,and investigated their phylogenetic relationships.We revealed that the expansion of the CDR family in Citrus seems to be due to segmental and tandem duplication events.Through genome resequencing and transcriptome sequencing,we identified eight CDR genes in the Poncirus genome(PtCDR1-PtCDR8).The number of SNPs was the highest in PtCDR2 and the lowest in PtCDR7.Most of the deletion and insertion events were observed in the UTR regions of Citrus and Poncirus CDR genes.PtCDR2 and PtCDR8 were in abundance in the leaf transcriptomes of two HLB-tolerant Poncirus genotypes and were also upregulated in HLB-tolerant,Poncirus hybrids as revealed by real-time PCR analysis.These two CDR genes seem to be good candidate genes for future studies of their role in citrus-CLas interactions. | Nidhi Rawat Brajendra Kumar Ute Albrecht Dongliang Du Ming Huang Qibin Yu Yi Zhang Yong-Ping Duan Kim D Bowman Fred G Gmitter Jr Zhanao Deng | 2017 | Horticulture Research2017,4,1: | 5 |
| 6 | 帕劳国预应力连续箱梁长期下挠分析显示文摘帕劳共和国科罗尔岛-巴伦尔图阿普岛桥(简称KB桥)为主跨241 m的预应力混凝土箱梁,通车后KB桥发生长期下挠,严重影响其正常运营,最后垮塌,其教训值得反思。该桥于1977竣工,运营18年后挠度达到1.61 m,1996年对该桥进行了体外预应力加固,却在加固后3个月发生垮塌。挠度分析结果表明:传统的梁理论与三维空间分析相比,误差高达20%,且宽跨比越大,误差越大。三维有限元空间分析采用多种徐变收缩计算模型:美国ACI模型、日本JSCE模型、CEB-FIP模型和GL模型,其挠度计算值要比(18年后)实测值低50%~70%,预测的变形也与实测不符;预应力损失计算值为22%~24%,远低于实测值50%。唯一基于理论的B3模型,采用经验值参数计算得到预应力损失40%,18年后挠度计算值比实测值低42%。如果根据长期实测值调整输入参数,结果可与实际更符合。对于早期挠度及其预测,B3模型能够合理地考虑因截面厚度差异产生的收缩和干燥徐变的速率差异。此外,B3模型还可考虑温度差异和顶板可能发生的开裂。 | 叶华文 王力武 曲浩博 Bazant Z P Yu Q Li G H | 2017 | 中外公路2017,37,5: | 5 |
