| 2 | Three-dimensional genome architectural CCCTC-binding factor makes choice in duplicated enhancers at Pcdhα locus显示文摘During development, gene expression is spatiotemporally regulated by long-distance chromatin interactions between distal enhancers and target promoters. However, how specificity of the interactions between enhancers and promoters is achieved remains largely unknown. As there are far more enhancers than promoters in mammalian genomes, the complexities of enhancer choice during gene regulation remain obscure. CTCF, the CCCTC-binding factor that directionally binds to a vast range of genomic sites known as CBSs(CTCF-binding sites), mediates oriented chromatin looping between a substantial set of distal enhancers and target promoters. To investigate mechanisms by which CTCF engages in enhancer choice, we used CRISPR/Cas9-based DNA-fragment editing to duplicate CBS-containing enhancers and promoters in the native genomic locus of the clustered Pcdhα genes. We found that the promoter is regulated by the proximal one among duplicated enhancers and that this choice is dependent on CTCF-mediated directional enhancer-promoter looping. In addition, gene expression is unaltered upon the switch of enhancers. Moreover, after promoter duplication, only the proximal promoter is chosen by CTCF-mediated directional chromatin looping to contact with the distal enhancer. Finally, we demonstrated that both enhancer activation and chromatin looping with the promoter are essential for gene expression. | 3 | Luminescence of halophosphate solid-solution Ca5(PO4)3(F0.8Cl0.2):Eu^2+,Mn^2+ for WLED显示文摘Solid-solution based luminescent materials have been widely explored due to their tunable optical properties in recent years. In this work, instead of more common cation-substitution approach, we investigated the luminescence properties of Eu^(2+) and Mn^(2+) co-doped halophosphate solid solution Ca_5(PO_4)_3(F_(0.8)Cl_(0.2)) via anion substitution synthesized by high temperature solid state reaction method.The formation of the solid solution was confirmed by X-ray diffraction(XRD) characterization, which indicates that the introduction of certain proportion of Cl^-will not make significant change on the Ca_5(PO_4)_3F lattice. We also studied the energy transfer from Eu^(2+) to Mn^(2+) in the host so as to obtain white light emission via adjusting the doping concentrations of Eu^(2+) and Mn^(2+). The white light emission was achieved through combination of Eu^(2+) blue emission and Mn^(2+) red-orange emission with appropriate proportions. | 4 | A novel oil spill detection method from synthetic aperture radar imageries via a bidimensional empirical mode decomposition显示文摘Oil spills pose a major threat to ocean ecosystems and their health. Synthetic aperture radar(SAR) sensors can detect oil spills on the sea surface. These oil spills appear as dark spots in SAR images. However, dark formations can be caused by a number of phenomena. It is aimed to distinguishing oil spills or look-alike objects. A novel method based on a bidimensional empirical mode decomposition is proposed. The selected dark formations are first decomposed into several bidimensional intrinsic mode functions and the residue. Subsequently, 64 dimension feature sets are calculated using the Hilbert spectral analysis and five new features are extracted with a relief algorithm. Mahalanobis distances are then used for classification. Three data sets containing oil spills or look-alikes are used to test the accuracy rate of the method. The accuracy rate is more than 90%. | 8 | VUV Spectroscopy of GdPO_4∶Eu^(3+) and GdBO_3∶Eu^(3+)显示文摘The emission and the excitation spectra of GdPO4∶Eu3+ and GdBO3∶Eu3+ prepared by solid state reaction method were investigated using the synchrotron radiation source of SUPERLUMI station of HASYLAB. The energy transfer between Gd3+ and Eu3+ was discussed with the probability of quantum cutting process. In the excitation spectra monitoring the red emission from Eu3+, the distinct lines corresponding to the intraconfigurational 4f-4f transitions from Gd3+ were observed for both samples, indicating an efficient energy transfer from host Gd3+ ions to the doped Eu3+ ions. The efficient energy transfer is necessary for the quantum cutting process based on the two-step energy transfer from Gd3+ to Eu3+. However, the overlapping of the lines corresponding to Gd3+∶8S7/2→6GJ and the broad excitation band (180~270 nm) due to Eu3+-O2- charge transfer states (CTS) around 200 nm cause excitation energy on ()6GJ levels to dissipate into CTS by direct energy transfer, unfavorable to the cross relaxation energy transfer between Gd3+ and Eu3+, thus unfavorable to the quantum cutting process.
