1Metformin induces ferroptosis by targeting miR-324-3p/GPX4 axis in breast cancer显示文摘Metformin is a widely prescribed hypoglycemic drug.Many studies have shown its anti-cancer properties.In the present study,we aimed to explore the effect of metformin on breast cancer and clarify the underlying mechanism.Our results showed that metformin induced ferroptosis in MDA-MB-231 cells through upregulating miR-324-3p expression.Overexpression of miR-324-3p inhibited cancer cell viability.miR-324-3p inhibitor promoted cell viability.Further studies showed that the effect of miR-324-3p was mediated by directly targeting glutathione peroxidase 4(GPX4).miR-324-3p bound to the 3ʹ-UTR of GPX4 and led to the downregulation of GPX4.In vivo studies showed that metformin induced ferroptosis by upregulating miR-324-3p in the xenograft model of breast cancer in mice.Our study suggested that metformin promotes ferroptosis of breast cancer by targeting the miR-324-3p/GPX4 axis.Metformin could act as a potential anti-cancer agent through the induction of ferroptosis.Yifeng Hou Shuang Cai Shouyang Yu Hui Lin 2021Acta Biochimica et Biophysica Sinica2021,53,3:11

4Association study of the single nucleotide polymorphisms in adiponectin-associated genes with type 2 diabetes in Han Chinese显示文摘Single-nucleotide polymorphisms (SNPs) of ADIPOQ, ADIPOR1, and ADIPOR2 have been associated with type 2 diabetes mellitus (T2DM), but there are many conflicting results especially in Chinese populations. To investigate the contribution of the adiponectin genes and their receptors to T2DM, a case-control study was performed and 11 SNPs of ADIPOQ, ADIPOR1, and ADIPOR2 were genotyped in 985 T2DM and 1,050 control subjects. rs16861194 (-11426 A>G) in the putative promoter of ADIPOQ was associated with T2DM (P = 0.007; OR = 1.29, 95% CI 1.08-1.55). None of the other 10 SNPs were associated with T2DM in this study, although rs2241766 and rs1501299 were reported to be associated with T2DM in previous Chinese studies. There was also no significant difference found from the ADIPOQ haplotype analysis, which contains rs16861194. In addition, we also assessed potential gene-gene interactions in three genes and no interactions were found. 6Investigation on surface roughness,residual stress and fatigue property of milling in-situ TiB_2/7050Al metal matrix composites显示文摘For higher efficiency and precision manufacturing,more and more attentions are focused on the surface roughness and residual stress of machined parts to obtain a good fatigue life.At present,the in-situ TiB_2/7050 Al metal matrix composites are widely researched due to its attractive properties such as low density,good wear resistance and improved strength.It is of great significance to investigate the machined surface roughness,residual stress and fatigue life for higher efficiency and precision manufacturing of this new kind material.In this study,the surface roughness including two-dimensional and three-dimensional roughness,residual stress and fatigue life of milling in-situ TiB_2/7050 Al metal matrix composites were analyzed.It was found from comparative investigation that the three-dimensional surface roughness would be more appropriate to represent the machined surface profile of milling particle reinforced metal matrix composites.The cutting temperature played a great role on the residual stress.However,the effect of increasing cutting force could slow down the transformation from compressive stress to tensile stress under 270°C.An exponential relationship between three-dimensional roughness and fatigue life was established and the main fracture mechanism was brittle fracture with observation of obvious shellfish veins,river pattern veins and wave shaped veins in fracture surface.Yifeng XIONG Wenhu WANG Yaoyao SHI Ruisong JIANG Chenwei SHAN Xiaofen LIU Kunyang LIN 2021Chinese Journal of Aeronautics2021,34,4:3

14Protective effects of combined Astragalus membranaceus and magnesium ions on global brain ischemia显示文摘The traditional Chinese herb Astragalus membranaceus is a well-known treatment for neurological diseases and is considered to exhibit anti-dementia properties.This study investigated the synergistic effects of magnesium ions and Astragalus membranaceus on global brain ischemia in rats.4'-6-diamidino-2-phenylindole staining demonstrated that the number of living neurons was significantly greater in the rat hippocampus after administration of a combination of Astragalus membranaceus and magnesium,compared with a vehicle group,an Astragalus membranaceus alone group,and a magnesium alone group.Western blot assay revealed that cleaved Caspase-3 protein expression was significantly reduced in the rat hippocampus in the combined Astragalus membranaceus and magnesium group compared with the Astragalus membranaceus alone group and the magnesium alone group.The results suggested that the combination of Astragalus membranaceus and magnesium exhibits a stronger neuroprotective effect on global brain ischemia in rats compared with Astragalus membranaceus or magnesium alone.This effect was associated with decreased Caspase-3 expression.Yifeng Miao Bing Li Yuchang Lin Xiaojie Lu Bin Wu Yongming Qiu 2011Neural Regeneration Research2011,6,15:1

17How psychiatrists coordinate treatment for COVID-19: a retrospective study and experience from China显示文摘Background Patients with C0VID-19 are at high risk of developing mental health problems;however,the prevalence and management of mental disorders and how psychiatrists coordinate the treatment are unclear.Aims We aimed to investigate the mental health problems of patients infected with C0VID-19 and to identify the role of psychiatrists in the clinical treatment team during the pandemic.We also share the experience of psychiatric consultations of patients with COVID-19 in Shanghai,China.Methods We analysed data from the psychiatric medical records of 329 patients with COVID-19 in the Shanghai Public Health Clinical Center from 20 January to 8 March 2020.We collected information including sociodemographic characteristics,whether patients received psychiatric consultation,mental health symptoms,psychiatric diagnoses,psychiatric treatments and severity level of COVID-19.Results Psychiatric consultations were received by 84(25.5%)patients with COVID-19.The most common symptoms of mental health problems were sleep disorders(75%),anxiety(58.3%)and depressive symptoms(11.9%).The psychiatric consultation rate was highest in critically ill patients(69.2%),with affective symptoms or disturbed behaviour as their main mental health problems.Psychiatric diagnoses for patients who received consultation included acute stress reaction(39.3%),sleep disorders(33.3%),anxiety(15.5%),depression(7.1%)and delirium(4.8%).In terms of psychiatric treatments,86.9%of patients who received psychiatric consultation were treated with psychotropic medications,including non-benzodiazepine sedative-hypnotic agents(54.8%),antidepressants(26.2%),benzodiazepines(22.6%)and antipsychotics(10.7%).Among the 76 patients who were discharged from the hospital,79%had recovered from mental health problems and were not prescribed any psychotropic medications.The symptoms of the remaining 21%of patients had improved and they were prescribed medications to continue the treatment.Conclusions This is the first study to report psychiatric consultations for patients with COVID-19.Our study indicated that a considerable proportion of patients with COVID-19,especially critically ill cases,experienced mental health problems.Given the remarkable effect of psychiatric treatments,we recommend that psychiatrists be timely and actively involved in the treatment of COVID-19.Ling Yue Jingyi Wang Mingliang Ju Yi Zhu Lianghu Chen Lin Shi Bohai Shi Jun Chen Yifeng Shen 2020General Psychiatry2020,33,4:1

19Deletion of CHD8 in cerebellar granule neuron progenitors leads to severe cerebellar hypoplasia,ataxia,and psychiatric behavior in mice显示文摘CHD8 is a candidate gene for autism spectrum disorders and neurological development delay.It has been reported to be essential for neurogenesis in the cerebral cortex,but the function of CHD8 in cerebellum has not been comprehensively investigated.The potential relationship of cerebellum dysplasia with psychiatric disorders in patients with CHD8 mutations is still not clear.In this study,we establish different conditional knockout mouse models to investigate the roles of CHD8 in cerebellar development.Mice with neural stem cell-specific Chd8 deletion exhibit significant reduction of cerebellum volume and no layering structure is detected.Genetic deletion of Chd8 in cerebellar granule neuron progenitors(GNPs)leads to cerebellar hypoplasia,absent of proliferation layer and ectopic of Purkinje neuron.However,no substantial cerebellar dysplasia is detected in mice with Purkinje neuron-or oligodendrocyte-specific Chd8 ablation.Single-cell RNA sequencing indicates that ribosome-related genes and pathways are most significantly disrupted in GNPs,indicating the potential mechanism.Importantly,in addition to the ataxia phenotype,mice with GNPspecific Chd8 ablation present a neuropsychiatric phenotype in three-chamber and light/dark tests.Taken together,our results provide insights not only into the function of CHD8 in cerebellar development,but also the pathogenesis of neuropsychiatric disorders in patients with CHD8 mutations.Xiang Chen Tong Chen Chen Dong Huiyao Chen Xinran Dong Lin Yang Liyuan Hu Huijun Wang Bingbing Wu Ye Yao Yu Xiong Man Xiong Yifeng Lin Wenhao Zhou 2022Journal of Genetics and Genomics2022,49,9:1
