| 1 | IntegrinαEβ7^(+)T cells direct intestinal stem cell fate decisions via adhesion signaling显示文摘Intestinal stem cell(ISC)differentiation is regulated precisely by a niche in the crypt,where lymphocytes may interact with stem and transient amplifying(TA)cells.However,whether and how lymphocyte–stem/TA cell contact affects ISC differentiation is largely unknown.Here,we uncover a novel role of T cell–stem/TA cell contact in ISC fate decisions.We show that intestinal lymphocyte depletion results in skewed ISC differentiation in mice,which can be rescued by T cell transfer.Mechanistically,integrinαEβ7 expressed on T cells binds to E-cadherin on ISCs and TA cells,triggering E-cadherin endocytosis and the consequent Wnt and Notch signaling alterations.BlockingαEβ7−E-cadherin adhesion suppresses Wnt signaling and promotes Notch signaling in ISCs and TA cells,leading to defective ISC differentiation.Thus,αEβ7^(+)T cells regulate ISC differentiation at single-cell level through cell–cell contact-mediatedαEβ7−E-cadherin adhesion signaling,highlighting a critical role of the T cell–stem/TA cell contact in maintaining intestinal homeostasis. | Shiyang Chen Yajuan Zheng Xiaojuan Ran Hui Du Hua Feng Lei Yang Yating Wen Changdong Lin Shihui Wang Mengwen Huang Zhanjun Yan Dianqing Wu Hongyan Wang Gaoxiang Ge An Zeng Yi Arial Zeng Jianfeng Chen | 2021 | Cell Research2021,31,12: | 3 |

| 5 | DDG1 and G Protein α Subunit RGA1 Interaction Regulates Plant Height and Senescence in Rice(Oryza sativa)显示文摘Many studies have already shown that dwarfism and moderate delayed leaf senescence positively impact rice yield,but the underlying molecular mechanism of dwarfism and leaf senescence remains largely unknown.Here,using map-based cloning,we identified an allele of DEP2,DDG1,which controls plant height and leaf senescence in rice.The ddg1 mutant displayed dwarfism,short panicles,and delayed leaf senescence.Compared with the wild-type,ddg1 was insensitive to exogenous gibberellins(GA)and brassinolide(BR).DDG1 is expressed in various organs,especially in stems and panicles.Yeast two-hybrid assay,bimolecular fluorescent complementation and luciferase complementation image assay showed that DDG1 interacts with theα-subunit of the heterotrimeric G protein.Disruption of RGA1 resulted in dwarfism,short panicles,and darker-green leaves.Furthermore,we found that ddg1 and the RGA1 mutant was more sensitive to salt treatment,suggesting that DDG1 and RGA1 are involved in regulating salt stress response in rice.Our results show that DDG1/DEP2 regulates plant height and leaf senescence through interacting with RGA1. | Xi Liu Chuxuan Zhao Di Wang Gen Pan Xiaonan Ji Su Gao Tanxiao Du Yating Feng Wenjing Chen | 2023 | Phyton-International Journal of Experimental Botany2023,92,7: | 1 |
| 6 | A Genome‑Wide Association Study for Susceptibility to Axial Length in Highly Myopic Eyes显示文摘High myopia has long been highly prevalent worldwide with a largely yet unexplained genetic contribution.To identify novel susceptibility genes for axial length(AL)in highly myopic eyes,a genome-wide association study(GWAS)was performed using the genomic dataset of 350 deep whole-genome sequencing data from highly myopic patients.Top single nucleotide polymorphisms(SNPs)were functionally annotated.Immunofluorescence staining,quantitative polymerase chain reaction,and western blot were performed using neural retina of form-deprived myopic mice.Enrichment analyses were further performed.We identified the four top SNPs and found that ADAM Metallopeptidase With Thrombospondin Type 1 Motif 16(ADAMTS16)and Phosphatidylinositol Glycan Anchor Biosynthesis Class Z(PIGZ)had the potential of clinical signifi-cance.Animal experiments confirmed that PIGZ expression could be observed and showed higher expression level in form-deprived mice,especially in the ganglion cell layer.The messenger RNA(mRNA)levels of both ADAMTS16 and PIGZ were significantly higher in the neural retina of form-deprived eyes(p=0.005 and 0.007 respectively),and both proteins showed significantly upregulated expression in the neural retina of deprived eyes(p=0.004 and 0.042,respectively).Enrichment analysis revealed a significant role of cellular adhesion and signal transduction in AL,and also several AL-related pathways including circadian entrainment and inflammatory mediator regulation of transient receptor potential channels were proposed.In conclusion,the current study identified four novel SNPs associated with AL in highly myopic eyes and confirmed that the expression of ADAMTS16 and PIGZ was significantly upregulated in neural retina of deprived eyes.Enrichment analyses provided novel insight into the etiology of high myopia and opened avenues for future research interest. | Qiang Lu Yu Du Ye Zhang Yuxi Chen Hao Li Wenwen He Yating Tang Zhennan Zhao Yinglei Zhang Jihong Wu Xiangjia Zhu Yi Lu | 2023 | Phenomics2023,3,3: | 0 |

| 9 | Effects of soybean oil exposure on the survival,reproduction,biochemical responses,and gut microbiome of the earthworm Eisenia fetida显示文摘With increasing production of kitchen waste,cooking oil gradually enters the soil,where it can negatively affect soil fauna.In this study,we explored the effects of soybean oil on the survival,growth,reproduction,tissue structure,biochemical responses,mRNA expression,and gut microbiome of earthworms(Eisenia fetida).The median lethal concentration of soybean oil was found to be 15.59%.Earthworm growth and reproduction were significantly inhibited following exposure to a sublethal concentration of soybean oil(1/3 LC50,5.2%).The activity of the antioxidant enzymes total superoxide dismutase(T-SOD),peroxidase(POD),and catalase(CAT)were affected under soybean oil exposure.The glutathione(GSH)content decreased significantly,whereas that of the lipid peroxide malondialdehyde(MDA)increased significantly after soybean oil exposure.mRNA expression levels of the SOD,metallothionein(MT),lysenin and lysozyme were significantly upregulated.The abundance of Bacteroides species,which are related to mineral oil repair,and Muribaculaceae species,which are related to immune regulation,increased within the earthworm intestine.These results indicate that soybean oil waste is toxic to earthworms.Thus,earthworms deployed defense mechanisms involving antioxidant system and gut microbiota for protection against soybean oil exposure-induced stress. | Yating Du Guangshen Shang Junjie Zhai Xing Wang | 2023 | Journal of Environmental Sciences2023,,11: | 0 |
