1Effects of electroacupuncture at Guanyuan(CV 4)or Sanyinjiao(SP 6)on hypothalamus-pituitary-ovary axis and spatial learning and memory in female SAMP8 mice显示文摘OBJECTIVE: To investigate the effects of electroacupuncture(EA) at the Guanyuan(CV 4) or Sanyinjiao(SP 6) acupoints on the hypothalamus-pituitary-ovary(HPO) axis and spatial learning and memory in female mice.METHODS: Nine-month-old female mice with senescence-accelerated mouse prone 8(SAMP8)were divided into three groups: the disease model,EA-Guanyuan and EA-Sanyinjiao groups. Concurrently, 9-month old female mice with senescence-accelerated mouse resistance 1(SAMR1)were set as the control model group. The two treatment groups were given the same pattern of EA stimulation. Gonadotropin-releasing hormone, luteinizing hormone, follicle-stimulating hormone(FSH) and Serum estradiol levels in the Hypothalamus-pituitary-ovary axis were assessed by enzyme-linked immunosorbent assay to determinethe HPO axis function level. Spatial learning and memory were assessed by the Morris Water Maze(MWM) test.RESULTS:(a) HPO axis: compared with the control model group, the disease model group displayed a decrease in E2 levels(P < 0.01), and an increase in Gn RH, LH and FSH levels(P < 0.01). E2 levels were increased in EA treatment groups compared with the disease model group(P < 0.05). In contrast,Gn RH and LH and FSH levels were reduced(P <0.05). EA-Sanyinjiao group was superior than EA-Guanyuan group on increasing E2 and declining Gn RH levels(P < 0.01).(b) The MWM test demonstrated that the response latency in the EA-Sanyinjiao treatment group declined from day 2 to day5 compared with the disease model group(P <0.05), whereas the EA-Guanyuan treatment group showed no significant difference.CONCLUSION: EA can regulate hormone(E2, FSH,LH, Gn RH) levels in the HPO axis and the spatial learning and memory ability in female SAMP8 mice. Moreover, this effect may have been more pronounced in the EA-Sanyinjiao group than the EA-Guanyuan group. 2Regulation of non-classical immune parameters in immune thrombocytopenic purpura mice by a spleen-invigorating,qi-replenishing and blood-containing formula显示文摘Objective:This study investigated the regulatory effect of non-classical immune parameters on immune thrombocytopenic purpura(ITP)mice by a spleen-invigorating,qi-replenishing and blood-containing formula(SQBF).Method:A total of 80 BALB/c mice were randomly divided into four equal groups(20 mice each):control group,model group,prednisone group and spleen-invigorating,qi-replenishing and blood-containing(SQBF)group.Mice in the model group,prednisone group,and SQBF group were administered anti-platelet serum to induce ITP.The dynamic variations of platelet counts in ITP mice were measured with an automatic blood analyzer before modeling and 48 h,and 8,12 and 15 days following APS injection.Levels of b-endorphin(b-EP),vasoactive intestinal peptide(VIP)and salivary IgA(SIgA)were detected by enzyme-linked immunosorbent assay(ELISA)on 15th day of experiment.Results:SQBF enhanced peripheral blood platelet counts in ITP mice similar to that of prednisone,and both groups showed a statistically significant response compared with the model group(P<.01).The SQBF significantly decreased b-EP levels compared with the model and prednisone intervention groups(P<.05),significantly increased the levels of VIP and SIgA in ITP mice compared with the model group(P<.05)and had significant protective effects on the thymus of ITP mice compared with the model group(P<.01).Conclusions:The SQBF had a similar effect to prednisone with regards to enhancing peripheral blood platelet counts in ITP mice.Furthermore,it decreased b-EP levels and increased VIP and SIgA,and protected the thymus.This shows that,on base of the brain-gut axis functions,some non-classical immune vascular active factors or neurotransmitters are also involved in immune responses,and also have relationship with the onset of ITP and bleeding and/or hemostasis.It needs further study to determine whether a change in these active factors is related to immediate hemostasis.Tiantian Li Hao He Li Hou Yue Xu Xiaoyong Wu Yanping Sun Lei Zheng Zhixiong Chen Xinyi Chen Bei Qin 2015Journal of Traditional Chinese Medical Sciences2015,2,2:6
3IL-17A-dependent gut microbiota is essential for regulating diet-induced disorders in mice显示文摘The gut microbiota plays a key role in obesity and related metabolic disorders, and multiple factors including diet, host genotype, and age regulate it. Many studies have examined the contribution of extrinsic factors to the regulation of the gut microbiota, but the importance of the host genetic constitution cannot be ignored. Interleukin 17A(IL-17A), a pro-inflammatory cytokine, is important in the defense against infection and diseases. Here, we investigated the association among IL-17, a high-fat diet(HFD), and the gut microbiota. Mice deficient in IL-17A were resistant to diet-induced obesity and related diseases. Compared with the Ⅱ-17^(-/-)mice, wild-type(WT) mice challenged with HFD showed obvious weight fluctuations, such as those seen in type 2 diabetes, and hematological changes similar to those associated with metabolic syndrome. However, housing WT mice and Ⅱ-17a^(-/-)mice together significantly alleviated these symptoms in the WT mice. A metagenomic analysis of the mouse feces indicated that the microbial community compositions of these two groups differed before HFD feeding. The HFD mediated shifts in the gut microbial compositions, which were associated with the mouse phenotypes.We also identified potentially beneficial and harmful species present during this period, and drew networks of the most abundant species. A functional analysis indicated pathway changes in the WT and Ⅱ-17a^(-/-)mice when fed the HFD. 6A novel method for evaluating the dynamic biocompatibility of degradable biomaterials based on real-time cell analysis显示文摘Degradable biomaterials have emerged as a promising type of medical materials because of their unique advantages of biocompatibility,biodegradability and biosafety.Owing to their bioabsorbable and biocompatible properties,magnesium-based biomaterials are considered as ideal degradable medical implants.However,the rapid corrosion of magnesium-based materials not only limits their clinical application but also necessitates a more specific biological evaluation system and biosafety standard.In this study,extracts of pure Mg and its calcium alloy were prepared using different media based on ISO 10993:12;the Mg^2+ concentration and osmolality of each extract were measured.The biocompatibility was investigated using the MTT assay and xCELLigence real-time cell analysis(RTCA).Cytotoxicity tests were conducted with L929,MG-63 and human umbilical vein endothelial cell lines.The results of the RTCA highly matched with those of the MTT assay and revealed the different dynamic modes of the cytotoxic process,which are related to the differences in the tested cell lines,Mg-based materials and dilution rates of extracts.This study provides an insight on the biocompatibility of biodegradable materials from the perspective of cytotoxic dynamics and suggests the applicability of RTCA for the cytotoxic evaluation of degradable biomaterials.Xiaoxiao Gai Chenghu Liu Guowei Wang Yang Qin Chunguang Fan Jia Liu Yanping Shi 2020Regenerative Biomaterials2020,7,3:3
7CO_2 sensing properties and mechanism of PrFeO_3 and NdFeO_3 thick film sensor显示文摘The CO_2 sensing of PrFeO_3 and NdFeO_3 sensors were investigated. Experimental results show that the resistances for PrFeO_3 and NdFeO_3 in CO_2 gas are larger than those in air and the responses for PrFeO_3and NdFeO_3 sensors increase with an increase in room-temperature relative humidity. When exposed to1000 ppm CO_2, the response of PrFeO_3 thick film based on nano-powders annealed at 700℃can reach8.44 at 160℃for the background of wet air with 58%of room-temperature relative humidity (RH),which is much larger than the corresponding value (3.03) in wet air with 25%RH. The sensing response S of NdFeO_3 thick-film sensor based on nano-powders annealed at 600℃to 3000 ppm CO_2 at the operating temperature 200℃can reach 2.36 for the background of wet air with 72%RH, which is larger than the corresponding value (1.83) in the air with 25%RH. 8Tetrapioid induction of Crassostrea hongkongensis and C.sikamea by inhibiting the polar body 1 release in diploid fertilized eggs显示文摘The production of an all-triploid population by mating tetrapioid males with diploid females is the best and most fundamental method for the large-scale production of triploid oysters.Obtaining a stable tetrapioid population is essential for guaranteed production in industrialized triploid cultivation.C.hongkongensis and C.sikamea are important oyster breeding species in southern China,and have great economic value.However,there are not any published data on inducing tetrapioid C.hongkongensis or C.sikamea.Therefore,we investigated tetrapioid induction in these two oyster species by inhibiting the PB1 release in diploid fertilized eggs using Cytochalasin B(CB)under 31℃,15%o salinity.The results confirmed that the optimal tetrapioid induction conditions for C.hongkongensis were a CB concentration of 0.50 mg/L with induction starting at 9.0 min after fertilization,and stopping at 21.0 min after fertilization;the induction efficiency index reached 0.123 under these conditions.The optimal tetrapioid induction conditions for C.sikamea were a CB concentration of 0.50 mg/L,with induction starting at 7.5 min after fertilization and stopping at 18 min after fertilization;the induction efficiency index could be as high as 0.281 under these conditions.However,we confirmed that the tetrapioid rate decreased with larval growth,and no tetrapioids were detected in the juvenile period of either C.hongkongensis or C.sikamea.This may be attributed to the very low survival of the tetrapioid larvae induced by this method,especially as most tetrapioid larvae died during the first three days.In summary,it is simple to directly induce tetrapioid C.hongkongensis and C.sikamea larvae by inhibiting the PB1 release of diploid zygotes,but the low survival rate makes it challenging to obtain viable juvenile tetrapioids.Yanping Qin Zohaib Noor Xingyou Li Yuehuan Zhang Ziniu Yu Haitao Ma Jun Li Yinyin Zhou Riguan Mo 2021Marine Life Science & Technology2021,3,4:1
9Ca^(2+)-dependent TaCCD1 cooperates with TaSAUR215 to enhance plasma membrane H^(+)-ATPase activity and alkali stress tolerance by inhibiting PP2C-mediated dephosphorylation of TaHA2 in wheat显示文摘Alkali stress is a major constraint for crop production in many regions of saline-alkali land.However,little is known about the mechanisms through which wheat responds to alkali stress.In this study,we identified a calcium ion-binding protein from wheat,TaCCD1,which is critical for regulating the plasma membrane(PM)H^(+)-ATPase-mediated alkali stress response.PM H+-ATPase activity is closely related to alkali tolerance in the wheat variety Shanrong 4(SR4).We found that two D-clade type 2C protein phosphatases,TaPP2C.D1 and TaPP2C.D8(TaPP2C.D1/8),negatively modulate alkali stress tolerance by dephosphorylating the penultimate threonine residue(Thr926)of TaHA2 and thereby inhibiting PM H+-ATPase activity.Alkali stress induces the expression of TaCCD1 in SR4,and TaCCD1 interacts with TaSAUR215,an early auxin-responsive protein.These responses are both dependent on calcium signaling triggered by alkali stress.TaCCD1 enhances the inhibitory effect of TaSAUR215 on TaPP2C.D1/8 activity,thereby promoting the activity of the PM H^(+)-ATPase TaHA2 and alkali stress tolerance in wheat.Functional and genetic analyses verified the effects of these genes in response to alkali stress,indicating that TaPP2C.D1/8 function downstream of TaSAUR215 and TaCCD1.Collectively,this study uncovers a new signaling pathway that regulates wheat responses to alkali stress,in which Ca^(2+)-dependent TaCCD1 cooperates with TaSAUR215 to enhance PM H+-ATPase activity and alkali stress tolerance by inhibiting TaPP2C.D1/8-mediated dephosphorylation of PM H+-ATPase TaHA2 in wheat.Minghan Cui Yanping Li Jianhang Li Fengxiang Yin Xiangyu Chen Lumin Qin Lin Wei Guangmin Xia Shuwei Liu 2023Molecular Plant2023,16,3:1
10Investigation of the structural, electronic and mechanical properties of CaO–SiO_(2) compound particles in steel based on density functional theory显示文摘CaO–SiO_(2)compounds compromise one of the most common series of oxide particles in liquid steels, which could significantly affect the service performance of the steels as crack initiation sites. However, the structural, electronic, and mechanical properties of the compounds in CaO–SiO_(2)system are still not fully clarified due to the difficulties in the experiments. In this study, a thorough investigation of these properties of CaO–SiO_(2)compound particles in steels was conducted based on first-principles density functional theory. Corresponding phases were determined by thermodynamic calculation, including gamma dicalcium silicate(γ-C2S), alpha-prime(L) dicalcium silicate(αL′-C2S), alpha-prime(H) dicalcium silicate(αH′-C2S), alpha dicalcium silicate(α-C2S), rankinite(C3S2), hatrurite(C3S), wollastonite(CS), and pseudowollastonite(Ps-CS). The results showed that the calculated crystal structures of the eight phases agree well with the experimental results. All the eight phases are stable according to the calculated formation energies, and γ-C2S is the most stable. O atom contributes the most to the reactivity of these phases. The Young’s modulus of the eight phases is in the range of 100.63–132.04 GPa. Poisson’s ratio is in the range of0.249–0.281. 12Trends of SARS-CoV-2 Infection in Rural Area in Sentinel Community-Based Surveillance——China,December 2022 to January 2023显示文摘Introduction:In late 2022,a rapid transmission of Omicron variants of severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)occurred throughout China.The purpose of this study was to provide the latest data and evaluate trends of SARS-CoV-2 infection in rural China among the community population.Methods:Data on SARS-CoV-2 infection among approximately 90,000 participants in rural China were collected by the National Sentinel Community-Based Surveillance(NSCS)system.Participants were tested for SARS-CoV-2 infection(defined as positive for SARS-CoV-2 nucleic acid or antigen)twice weekly from December 16,2022 to January 12,2023.The daily average of newly positive rate and its estimated daily percentage change were calculated to describe the national and regional trends of SARS-CoV-2 infection in rural China.Results:In rural China,the daily average new positive rate of SARS-CoV-2 infection peaked at 4.79%between December 20–22,2022 and then decreased to 0.57%between January 10–12,2023,with an average decrease of 29.95%per round.The peak of new SARS-CoV-2 infection was slightly earlier and lower in North China(5.28%between December 20–22,2022)than in South China(5.63%between December 23–26,2022),and then converged from December 30,2022 to January 2,2023.The peak of 6.09%occurred between December 20–22,2022 in eastern China,while the peak of 5.99%occurred later,between December 27–29,2022,in central China.Conclusions:Overall,the epidemic wave in rural China peaked between December 20–22,2022,and passed quickly following the optimization of prevention and control measures.Currently,SARSCoV-2 infection in community populations in rural China is sporadic.Wenzhan Jing Fan Ding Yanping Zhang Guoqing Shi Jue Liu Yu Wu Fangyuan Li Wenxin Yan Chenyuan Qin Min Liu Qun Li Wannian Liang 2023China CDC weekly2023,5,11:1

19Trends of SARS-CoV-2 Infection in Sentinel Community-Based Surveillance After the Optimization of Prevention and Control Measures—China,December 2022-January 2023显示文摘Introduction:On December 7,2022,China implemented the“Ten New Measures”to optimize its prevention and control measures for coronavirus disease 2019(COVID-19).To provide the latest data after the optimization,we evaluated trends of severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)infection among the community population in China.Methods:We utilized data from the National Sentinel Community-Based Surveillance(NSCS)system in China to assess trends of SARS-CoV-2 infection.NSCS is a national community-based surveillance cohort with 0.42 million participants from all 31 provincial-level administrative divisions(PLADs)and Xinjiang Production and Construction Corps(XPCC).Participants were tested for infection twice a week(a total of eight rounds)from December 16,2022 to January 12,2023.SARS-CoV-2 infection was defined as testing positive for SARS-CoV-2 nucleic acid or antigen.We calculated the daily average of newly positive rates of SARS-CoV-2 infection.Results:In this national cohort,the daily average newly positive rate of SARS-CoV-2 infection decreased from 4.13% in Round 1(December 16-19,2022)to 0.69% in Round 8(January 10-12,2023).The epidemic peak occurred in Round 2(December 20-22,2022).Similar trends were observed in urban areas(decreasing from 4.65% to 0.73%),rural areas(decreasing from 2.83% to 0.57%),the eastern region(decreasing from 4.18% to 0.67%),the central region(decreasing from 5.43% to 0.61%),and the western region(decreasing from 3.01%to 0.77%).Conclusions:NSCS data showed that the peak of SARS-CoV-2 infection in China had passed.SARSCoV-2 infection in community populations in China is currently at a low epidemic level.Jue Liu Fan Ding Yu Wu Wenzhan Jing Wenxin Yan Chenyuan Qin Fangyuan Li Yanping Zhang Min Liu Wannian Liang Qun Li 2023China CDC weekly2023,5,7:0
20Morphine Re-arranges Chromatin Spatial Architecture of Primate Cortical Neurons显示文摘The expression of linear DNA sequence is precisely regulated by the three-dimensional(3D)architecture of chromatin.Morphine-induced aberrant gene networks of neurons have been extensively investigated;however,how morphine impacts the 3D genomic architecture of neurons is still unknown.Here,we applied digestion-ligation-only high-throughput chromosome conformation capture(DLO Hi-C)technology to investigate the effects of morphine on the 3D chromatin architecture of primate cortical neurons.After receiving continuous morphine administration for 90 days on rhesus monkeys,we discovered that morphine re-arranged chromosome territories,with a total of 391 segmented compartments being switched.Morphine altered over half of the detected topologically associated domains(TADs),most of which exhibited a variety of shifts,followed by separating and fusing types.Analysis of the looping events at kilobase-scale resolution revealed that morphine increased not only the number but also the length of differential loops.Moreover,all identified differentially expressed genes from the RNA sequencing data were mapped to the specific TAD boundaries or differential loops,and were further validated for changed expression.Collectively,an altered 3D genomic architecture of cortical neurons may regulate the gene networks associated with morphine effects.Our finding provides critical hubs connecting chromosome spatial organization and gene networks associated with the morphine effects in humans.Liang Wang Xiaojie Wang Chunqi Liu Wei Xu Weihong Kuang Qian Bu Hongchun Li Ying Zhao Linhong Jiang Yaxing Chen Feng Qin Shu Li Qinfan Wei Xiaocong Liu Bin Liu Yuanyuan Chen Yanping Dai Hongbo Wang Jingwei Tian Gang Cao Yinglan Zhao Xiaobo Cen 2023Genomics, Proteomics & Bioinformatics2023,21,3:0
