1Identification of a novel mutation in POU3F4 for prenatal diagnosis in a Chinese family with X-linked nonsyndromic hearing loss显示文摘We present the clinical and genetic findings for a Chinese family with X-linked non-syndromic hearing loss in which the affected males showed congenital profound sensorineural hearing impairment. In two affected brothers, the computer tomography of temporal bone showed bilateral dilation of the internal auditory canal with fistulous communication between the lateral canal and the basal cochlear turn, which is consistent with the typical DFNX2 phenotype. A missense mutation (c.647G→A) in the POU3F4 gene caused a substitu- tion from glycine to glutamic acid at position 216 (p.G216E), and this mutation was found to consistently cosegregate with the deafness phenotype in the family. The mutation resulted in the loss of function of the POU3F4 by decreasing the affinity between the protein and DNA, as shown in silico by the structural analysis. Prenatal diagnosis of pregnant proband of this family revealed the c.647G→A muta- tion in DNA extracted from the amniotic fluid surrounding the fetus. 2Effects of Mahuang(Herba Ephedra Sinica) and Wuweizi(Fructus Schisandrae Chinensis) medicated serum on chemotactic migration of alveolar macrophages and inters regions macrophages in rats显示文摘OBJECTIVE: To observe the effects of serum containing Mahuang(Herba Ephedra Sinica) or Wuweizi(Fructus Schisandrae Chinensis) on the migration of alveolar macrophages(AM) and interstitial macrophages(IM) from normal rats, and to analyze and compare the mechanisms leading to cell migration differences.METHODS: Rats were randomly divided into three groups: Mahuang(Herba Ephedra Sinica), Wuweizi(Fructus Schisandrae Chinensis), and blank serum. After treatment with the herbs, serum was extracted from the rats. AM and IM were isolated from normal rats and cultured. The effects of Mahuang(Herba Ephedra Sinica) and Wuweizi(Fructus Schisandrae Chinensis) medicated serum on normal rat AM and IM chemotactic migration were determined by transwell assays. The CC chemokine receptor(CCR) 2, CCR5, voltage-gated Kvl. 3 K^+channel(Kv1. 3), and voltage-gated Kvl. 5 K^+channel(Kv1. 5) protein levels were analyzed by western blotting.RESULTS: The migration quantities of AM and IM in the Mahuang(Herba Ephedra Sinica) and Wuweizi(Fructus Schisandrae Chinensis) medicated serum groups were significantly higher than those in the blank serum group(P < 0.01). Compared with the Wuweizi(Fructus Schisandrae Chinensis) medicated serum group, the migration quantity of cultured rat AM in the Mahuang(Herba Ephedra Sinica) medicated serum group was significantly increased(P <0.01). Meanwhile, compared with the Mahuang(Herba Ephedra Sinica) medicated serum group, the migration quantity of cultured rat IM in the Wuweizi(Fructus Schisandrae Chinensis) medicated serum group was significantly increased(P < 0.01).CCR2, CCR5, Kv1. 3, and Kv1. 5 proteins were expressed on the AM cell surface, and showed significantly higher expression in the Mahuang(Herba Ephedra Sinica) medicated serum group compared with the Wuweizi(Fructus Schisandrae Chinensis)medicated serum group. In contrast, CCR5, Kv1.3,and Kv1.5 proteins were expressed on the IM cell surface, and showed significantly higher expression in the Wuweizi(Fructus Schisandrae Chinensis)medicated serum group compared with the Mahuang(Herba Ephedra Sinica) medicated serum group.CONCLUSION: Mahuang(Herba Ephedra Sinica)and Wuweizi(Fructus Schisandrae Chinensis) can promote AM and IM migration ability, with Mahuang(Herba Ephedra Sinica) targeting AM more apparently and Wuweizi(Fructus Schisandrae Chinensis) targeting IM more apparently. 4Efficacy of Rebixiao Chinese herbal tablets and Chinese formula granules in acute gout arthritis patients:a randomized,multicenter,double-blind,controlled trial显示文摘OBJECTIVE:To evaluate the clinical efficacy and safety of Rebixiao(RBX)Chinese herbal tablets(CHT)and Chinese formula granules(CFG)in the treatment of acute gout arthritis(AGA).METHODS:This randomized,multicenter,double-blind,controlled trial included 165 AGA patients with the damp-heat symptom pattern who were randomly divided into an RBX CHT group and an RBX CFG group and treated for 7 d at three centers.The total effective rates of the joint symptom score,Traditional Chinese Medicine(TCM)symptoms score,erythrocyte sedimentation rate(ESR),and C-reactive protein(CRP)were used to evaluate the clinical efficacy.Safety assessments were also performed.RESULTS:Of the 165 enrolled patients,147 completed the clinical observation.There was no difference in baseline between the two groups.The total effective rates of the joint symptom score were 94.36%and 97.36%,and the total effective rates of the TCM symptoms score were 95.77%and 97.36%in the CFG group and CHT group,respectively.No statistical difference was found between the two groups(P>0.05).Additionally,ESR and CRP were similar in both groups(P>0.05).Furthermore,treatment efficacy regarding TCM and joint symptoms,the ESR,and CRP were consistent within each center and among the different centers(P>0.05).In addition,the incidence of adverse events was 4.22%and 2.63%in the CFG group and CHT group,respectively,and no difference was observed between the two groups(P>0.05).CONCLUSION:RBX CFG and CHT have significant and similar efficacy in the treatment of AGA,and CFG did not increase adverse side effects.Wang Shasha Zhang Ying Dai Min Yang Min Leng Wenfei Peng Xiufeng Li Guojun Yang Jinbing Gan Jianping Zhu Fenglin Li Yanping Wu Bin 2020Journal of Traditional Chinese Medicine2020,40,4:3

7Variation of high-molecular-weight glutenin subunits and glutenin macropolymer particle distribution in wheat grains produced under different water regimes显示文摘The components and contents of high-molecular-weight glutenin subunits(HMW-GS) in wheat grains affect glutenin macropolymer(GMP) size, which is considered an important flour quality trait in wheat. Four wheat cultivars(Shiluan 02-1, Yannong 24, Jinan 17 and Lumai 21) with different end-use qualities were used to investigate the HMW-GS and GMP contents, and the GMP particle distributions in grain produced under irrigated and rainfed conditions. The percent volume of GMP particles and the contents of HMW-GS and GMP were affected by genotype and soil water. Genotype × soil water interaction was significant only for GMP particles <12 μm and >100 μm in the growing season of 2010–2011. Irrigated and rainfed conditions had different influences on the GMP particle distribution in the four cultivars. Compared to irrigated treatment, the rainfed treatment had higher accumulations of HMW-GS and GMP, especially in cultivars Yannong 24, Jinan 17 and Lumai 21. Rainfed conditions also increased the proportion of large size particles of GMP, indicating that different water regimes had an effect on grain quality. 18Enhanced degradation of organic contaminants by Fe(Ⅲ)/peroxymonosulfate process with L-cysteine显示文摘The difficulty in Fe(Ⅲ)/Fe(Ⅱ) conversion in the Fe(Ⅲ)/peroxymonosulfate(PMS) process limits its efficiency and application.Herein,L-cysteine(Cys),a green natural organic ligand with reducing capability,was innovatively introduced into Fe(Ⅲ)/PMS to construct an excellent Cys/Fe(Ⅲ)/PMS process.The Cys/Fe(Ⅲ)/PMS process,at room temperature,can degrade a variety of organic contaminants,including dyes,phenolic compounds,and pharmaceuticals.In subsequent experiments with acid orange 7(A07),the AO7 degradation efficiency followed pseudo-first-order kinetic which exhibited an initial 'fast stage' and a second 'slow stage'.The rate constant values ranged depending on the initial Cys,Fe(Ⅲ),PMS,and AO7 concentrations,reaction temperature,and pH values.In addition,the presence of Cl^(-),NO_(3)^(-),and SO_(4)^(2-) had negligible impact while HCO_(3)^(-) and humic acid inhibited the degradation of AO7.Furthermore,radical scavenger experiments and methyl phenyl sulfoxide(PMSO) transformation assay indicated that sulfate radical,hydroxyl radical,and ferryl ion(Fe(Ⅳ)) were the dominant reactive species involved in the Cys/Fe(Ⅲ)/PMS process.Finally,based on the results of gas chromatography-mass spectrometry,several AO7 degradation pathways,including N=N cleavage,hydroxylation,and ring opening were proposed.This study provided a new insight to improve the efficiency of Fe(Ⅲ)/PMS process by accelerating Fe(Ⅲ)/Fe(Ⅱ)cycle with Cys.Chengdu Qi Yanni Wen Yijie Zhao Yinhao Dai Yanping Li Chenmin Xu Shaogui Yang Huan He 2022Chinese Chemical Letters2022,33,4:1
19Morphine Re-arranges Chromatin Spatial Architecture of Primate Cortical Neurons显示文摘The expression of linear DNA sequence is precisely regulated by the three-dimensional(3D)architecture of chromatin.Morphine-induced aberrant gene networks of neurons have been extensively investigated;however,how morphine impacts the 3D genomic architecture of neurons is still unknown.Here,we applied digestion-ligation-only high-throughput chromosome conformation capture(DLO Hi-C)technology to investigate the effects of morphine on the 3D chromatin architecture of primate cortical neurons.After receiving continuous morphine administration for 90 days on rhesus monkeys,we discovered that morphine re-arranged chromosome territories,with a total of 391 segmented compartments being switched.Morphine altered over half of the detected topologically associated domains(TADs),most of which exhibited a variety of shifts,followed by separating and fusing types.Analysis of the looping events at kilobase-scale resolution revealed that morphine increased not only the number but also the length of differential loops.Moreover,all identified differentially expressed genes from the RNA sequencing data were mapped to the specific TAD boundaries or differential loops,and were further validated for changed expression.Collectively,an altered 3D genomic architecture of cortical neurons may regulate the gene networks associated with morphine effects.Our finding provides critical hubs connecting chromosome spatial organization and gene networks associated with the morphine effects in humans.Liang Wang Xiaojie Wang Chunqi Liu Wei Xu Weihong Kuang Qian Bu Hongchun Li Ying Zhao Linhong Jiang Yaxing Chen Feng Qin Shu Li Qinfan Wei Xiaocong Liu Bin Liu Yuanyuan Chen Yanping Dai Hongbo Wang Jingwei Tian Gang Cao Yinglan Zhao Xiaobo Cen 2023Genomics, Proteomics & Bioinformatics2023,21,3:0
20Short-term dynamics of nutrients influenced by upwelling in a small oligotrophic coastal ecosystem,Gan Bay,in the northwest Philippines显示文摘We present a time-series analysis of nutrient and pCO2 (partial pressure of CO2) levels in an oligotrophic coastal ecosystem (Gan Bay), which was likely to be influenced by upwelled subsurface water. Gan Bay is of Currimao Harbor, in the northwest Philippines and is located at the boundary of the South China Sea (SCS). This 42-h time-series observation was conducted in December 2006. In addition to continuous observations of dissolved oxygen (DO) and pCO2, discrete samples were collected at a depth of 5 m every 3 h for measurements of nutrients, including soluble reactive phosphorus (SRP) and inorganic nitrogen (NO3 + NO2 ) in order to examine their dynamics and possible physical and biological controls. We observed remarkably large short-term variations in the surface water, spanning a 10-fold change for SRP (32-330 nM) and from <0.3 lM to 4.3 lM for NO3 + NO2 . DO also varied substantially from a lower end of 171 to 205 lM O2. Surface water pCO2 changed from an equilibrium stage with the atmosphere ( 386 latm) to a stage where it was a significant source for the atmospheric CO2 (seawater pCO2 469 latm). We found that the variation of nutrients was driven neither by tidal mixing nor by biological activities, as was suggested by the variations in the total bacterial abundance and chlorophyll a. Instead, our inverse T-S relationship suggested a two end-member mixing process during the observation period. The N:P ratio throughout the observation period was 13.2, which is characteristic of SCS subsurface and deep waters. Moreover, pCO2 was correlated inversely with the sea surface temperature. It is likely, therefore, that an upwelled subsurface cold water with high nutrients, low-temperature and high-pCO2 existed.
