| 1 | Whole genome sequencing of 10K patients with acute ischaemic stroke or transient ischaemic attack: design, methods and baseline patient characteristics显示文摘Background and purpose Stroke is the second leading cause of death worldwide and the leading cause of mortality and long-term disability in China,but its underlying risk genes and pathways are far from being comprehensively understood.We here describe the design and methods of whole genome sequencing(WGS)for 10914 patients with acute ischaemic stroke or transient ischaemic attack from the Third China National Stroke Registry(CNSR-III).Methods Baseline clinical characteristics of the included patients in this study were reported.DNA was extracted from white blood cells of participants.Libraries are constructed using qualified DNA,and WGS is conducted on BGISEQ-500 platform.The average depth is intended to be greater than 30×for each subject.Afterwards,Sentieon software is applied to process the sequencing data under the Genome Analysis Toolkit best practice guidance to call genotypes of single nucleotide variants(SNVs)and insertion-deletions.For each included subject,21 fingerprint SNVs are genotyped by MassARRAY assays to verify that DNA sample and sequencing data originate from the same individual.The copy number variations and structural variations are also called for each patient.All of the genetic variants are annotated and predicted by bioinformatics software or by reviewing public databases.Results The average age of the included 10914 patients was 62.2±11.3 years,and 31.4%patients were women.Most of the baseline clinical characteristics of the 10914 and the excluded patients were balanced.Conclusions The WGS data together with abundant clinical and imaging data of CNSR-III could provide opportunity to elucidate the molecular mechanisms and discover novel therapeutic targets for stroke. | Si Cheng Zhe Xu Yang Liu Jinxi Lin Yong Jiang Yilong Wang Xia Meng Anxin Wang Xinying Huang Zhimin Wang Guohua Chen Songdi Wu Zhengchang Jia Yongming Chen Xuerong Qiu Jun Wu Binbin Song Weizhong Ji Zhongping An Wenjun Xue Lili Zhao Yu Geng Hongyan Li Hao Li Yongjun Wang | 2021 | Stroke & Vascular Neurology2021,6,2: | 8 |
| 2 | A Scenario-Based Evaluation of COVID-19-Related Essential Clinical Resource Demands in China显示文摘The coronavirus disease 2019(COVID-19)pandemic is a global crisis,and medical systems in many countries are overwhelmed with supply shortages and increasing demands to treat patients due to the surge in cases and severe illnesses.This study aimed to assess COVID-19-related essential clinical resource demands in China,based on different scenarios involving COVID-19 spreads and interventions.We used a susceptible–exposed–infectious–hospitalized/isolated–removed(SEIHR)transmission dynamics model to estimate the number of COVID-19 infections and hospitalizations with corresponding essential healthcare resources needed.We found that,under strict non-pharmaceutical interventions(NPIs)or mass vaccination of the population,China would be able to contain community transmission and local outbreaks rapidly.However,under scenarios involving a low intensity of implemented NPIs and a small proportion of the population vaccinated,the use of a peacetime–wartime transition model would be needed for medical source stockpiles and preparations to ensure a normal functioning healthcare system.The implementation of COVID-19 vaccines and NPIs in different periods can influence the transmission of COVID-19 and subsequently affect the demand for clinical diagnosis and treatment.An increased proportion of asymptomatic infections in simulations will not reduce the demand for medical resources;however,attention must be paid to the increasing difficulty in containing COVID-19 transmission due to asymptomatic cases.This study provides evidence for emergency preparations and the adjustment of prevention and control strategies during the COVID-19 pandemic.It also provides guidance for essential healthcare investment and resource allocation. | Ting Zhang Qing Wang Zhiwei Leng Yuan Yang Jin Yang Fangyuan Chen Mengmeng Jia Xingxing Zhang Weiran Qi Yunshao Xu Siya Chen Peixi Dai Libing Ma Luzhao Feng Weizhong Yang | 2021 | Engineering2021,7,7: | 7 |

| 4 | Rarefaction and Temperature Gradient Effect on the Performance of the Knudsen Pump显示文摘The prediction of the multiscale flow in the Knudsen pump is important for understanding its pumping mechanism.However,there is little research on such interesting multiscale phenomenon in the Knudsen pumps.In this paper,a novel numerical analysis method combining the direct simulation Monte Carlo(DSMC) method with the smoothed particle hydrodynamics(SPH) method is presented for simulating the multiscale flow,which is often encountered in the application of the Knudsen pumps.Validity and accuracy of the new method are given by comparing its results with that of the previous research.Using the coupled multiscale approach,the rarefaction and the temperature drive are studied,which are two main factors on the performance of the Knudsen pumps.To investigate the effect of rarefaction on the performance of the Knudsen pump,various pump operation pressures are compared.The flow characteristics and pumping ability at different rarefaction are analyzed,and the phenomenon of the multiscale flow is also discussed.Several cases with different linear or nonlinear temperature gradients are set to investigate the effect of temperature gradient on the performance of the Knudsen pump.The flow characteristics of the Knudsen pump such as the velocity,pressure increase,and the mass flowrate are presented.A unique phenomenon,the reverse transpiration effect caused by the nonlinear temperature gradient is studied,and the reason of the significant pressure increase in the pump channel is also analyzed.Since the multiscale gas flow is widely encountered in the microflow systems,the above method and its results can also be greatly beneficial and provide significant insights for the design of the MEMS devices. | YE Jianjun YANG Jian ZHENG Jinyang DING Xianting WONG Ieong LI Weizhong CHEN Cong | 2012 | Chinese Journal of Mechanical Engineering2012,25,4: | 6 |
| 5 | Hydrogen production from coke oven gas over LiNi/γ-Al_2O_3 catalyst modified by rare earth metal oxide in a membrane reactor显示文摘The performance of LiNi/γ-Al2O3 catalysts modified by rare earth metal oxide(La2O3 or CeO2 ) packed on BCFNO membrane reactor was discussed for the partial oxidation of methane(POM) in coke oven gas(COG) at 875 C. The NiO/γ-Al2O3 catalysts with different amounts of La2O3 and CeO2 were prepared with the same preparation method and under the same condition in order to compare the reaction performance(oxygen permeation,CH 4 conversion,H 2 and CO selectivity) on the membrane reactor. The results show that the oxygen permeation flux increased significantly with LiNiREO x /γ-Al2O3(RE = La or Ce) catalysts by adding the element of rare earth especially the Ce during the POM in COG. Such as,the Li15wt%CeO2 9wt%NiO/γ-Al2O3 catalyst with an oxygen permeation flux of 24.71 ml cm 2 min 1 and a high CH 4 conversion was obtained in 875 C. The resulted high oxygen permeation flux may be due to the added Ce that inhibited the strong interaction between Ni and Al2O3 to form the NiAl2O4 phase. | 8 | Revisiting the methods for gas permeability measurement in tight porous medium显示文摘Permeability is a key parameter to describe fluid transport properties of porous medium; however, the permeability measurement is extremely difficult for tight porous medium, e.g. fine-grained rock or dense soil. In this paper, three methods for gas permeability measurement, i.e. steady state method, pulse decay method(PDM) and pressure oscillation method(POM), are first reviewed and then their advantages and drawbacks are discussed. Both analytical and numerical solutions of gas permeability are presented for the tight porous medium. The results show that the analytical method is relatively simple but only valid under certain conditions, whilst the numerical method is more robust and generic, which can take into account several factors such as porosity, saturation, gas leakage, and unconventional boundary conditions. The influence of the effective porosity on the permeability determination is further analyzed using the proposed numerical method. | 9 | On the critical particle size of soil with clogging potential in shield tunneling显示文摘Shield tunneling is easily obstructed by clogging in clayey strata with small soil particles.However,soil clogging rarely occurs in strata with coarse-grained soils.Theoretically,a critical particle size of soils should exist,below which there is a high risk of soil clogging in shield tunneling.To determine the critical particle size,a series of laboratory tests was carried out with a large-scale rotary shear apparatus to measure the tangential adhesion strength of soils with different particle sizes and water contents.It was found that the tangential adhesion strength at the soilesteel interface gradually increased linearly with applied normal pressure.When the particle size of the soil specimen was less than 0.15 mm,the interfacial adhesion force first increased and then decreased as the water content gradually increased;otherwise,the soil specimens did not manifest any interfacial adhesion force.The amount of soil mass adhering to the steel disc was positively correlated with the interfacial adhesion force,thus the interfacial adhesion force was adopted to characterize the soil clogging risk in shield tunneling.The critical particle size of soils causing clogging was determined to be 0.15 mm.Finally,the generation mechanism of interfacial adhesion force was explored for soils with different particle sizes to explain the critical particle size of soil with clogging risk in shield tunneling. | Shuying Wang Zihao Zhou Pengfei Liu Zhao Yang Qiujing Pan Weizhong Chen | 2023 | Journal of Rock Mechanics and Geotechnical Engineering2023,15,2: | 3 |
| 10 | The Incoming Influenza Season—China,the United Kingdom,and the United States,2021–2022显示文摘Introduction:Seasonal influenza activity has declined globally since the widespread of severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)transmission.There has been scarce information to understand the future dynamics of influenza—and under different hypothesis on relaxation of nonpharmaceutical interventions(NPIs)in particular—after the disruptions to seasonal patterns.Methods:We collected data from public sources in China,the United Kingdom,and the United States,and forecasted the influenza dynamics in the incoming 2021–2022 season under different NPIs.We considered Northern China and Southern China separately,due to the sharp difference in the patterns of seasonal influenza.For the United Kingdom,data were collected for England only.Results:Compared to the epidemics in 2017–2019,longer and blunter influenza outbreaks could occur should NPIs be fully lifted,with percent positivity varying from 10.5 to 18.6 in the studying regions.The rebounds would be smaller if the maskwearing intervention continued or the international mobility stayed low,but sharper if the mask-wearing intervention was lifted in the middle of influenza season.Further,influenza activity could stay low under a much less stringent mask-wearing intervention coordinated with influenza vaccination.Conclusions:The results added to our understandings of future influenza dynamics after the global decline during the coronavirus disease 2019(COVID-19)pandemic.In light of the uncertainty on the incoming circulation strains and the relatively low negative impacts of mask wearing on society,our findings suggested that wearing mask could be considered as an accompanying mitigation measure in influenza prevention and control,especially for seasons after long periods of low-exposure to influenza viruses.Seasonal influenza activity declines globally during the coronavirus disease 2019(COVID-19)pandemic(1–4).For instance,in China,influenza activity,as measured by percentage of submitted specimens testing positive,dropped from 11.8%to 2.0%in 2020–2021 influenza season,compared to the past 5 years(5).The long-period of low-exposure to influenza viruses adds great uncertainty on preparedness for the incoming 2021–2022 influenza season.Influenza vaccination is one of the most effective measures in seasonal influenza prevention and control,but with only a few influenza viruses circulating,it could be difficult to determine the targeted strains for vaccination.In this context,it is of primary importance to identify alternative mitigation measures for the incoming 2021–2022 influenza season,the first season after long periods of virtually no influenza outbreaks worldwide.Using data from China,the United Kingdom,and the United States,we forecasted the influenza activity in the incoming 2021–2022 influenza season under hypothetical scenarios without non-pharmaceutical interventions(NPIs)and with different assumptions on mask-wearing and mobility levels. | Shasha Han Ting Zhang Yan Lyu Shengjie Lai Peixi Dai Jiandong Zheng Weizhong Yang Xiaohua Zhou Luzhao Feng | 2021 | China CDC weekly2021,3,49: | 3 |

| 12 | Diagnosis and clinical manifestations of subacute combined degeneration of the spinal cord: Analysis of 21 cases显示文摘BACKGROUND: Subacute combined degeneration of the spinal cord is caused by vitamin B12 deficiency and is a kind of degenerative disease owing the characteristics of nervous system diseases. In addition, different patients have variously clinical manifestations and various prognoses after vitamin B12 therapy. OBJECTIVE: To investigate and analyze diagnosis, clinical manifestations and prognosis of subacute combined degeneration of the spinal cord. DESIGN: Case analysis. SETTING: Department of Neurology, the Third Hospital of Peking University. PARTICIPANTS: A total of 21 subacute combined degeneration of the spinal cord patients including 14 males and 7 females aged from 33 to 82 years were selected from Department of Neurology, the Third Hospital of Peking University from January 1999 to December 2005. Duration from onset to final diagnosis lasted for 1.5–108 months. All patients had typically clinical manifestations; meanwhile, level of serum vitamin B12 was decreased and/or vitamin B12 therapy was effective. All patients provided the confirmed consent. METHODS: Clinical data of 21 subacute combined degeneration of the spinal cord patients were retrospectively analyzed, while general data and clinical characteristics were recorded at the same time. Levels of blood routine, serum vitamin B12 and homocysteine were measured at the phase of hospitalization. Normal value of serum vitamin B12 was 187–1 059 ng/L and normal value of serum homocysteine was 5– 15 μmol/L. All patients received neuroelectrophysiological examination and 15 patients received MRI examinations of spinal cord. After final diagnosis, patients were given vitamin B12 therapy. And follow-up was performed to investigate the prognosis. MAIN OUTCOME MEASURES: ① Levels of blood routine, serum vitamin B12 and homocysteine; ② results of neuroelectrophysiological examination; ③ results of MRI examination of spinal cord; ④ prognosis. RESULTS: Clinical data of 21 patients and follow-up data of 20 patients were involved in the final analysis and 1 patient was lost because of living in the other province. ① Clinical manifestations: All 21 patients had typically clinical manifestations. The original symptoms included numbness of lower and/or upper limbs (5 cases), unstable gait (3 cases), limb asthenia (4 cases), limb numbness combined with light asthenia (5 cases), limb numbness combined with unskillful activity (3 cases), and limb numbness combined with unstable gait (1 case). ② Experimental results: Eight subacute combined degeneration of the spinal cord patients accompanied with mild-severe anemia and mean corpuscular volume of 13 patients were increased. Among 13 subacute combined degeneration of the spinal cord patients not administrating vitamin B12 before hospitalization, the levels of serum vitamin B12 of 2 patients were not measured but those of other patients were decreased. After vitamin B12 therapy,the levels of serum vitamin B12 of 8 patients were normal or increased. In addition, the levels of serum homocysteine of 6 patients were not measured but those of 7 patients were increased. While, the levels of homocysteine of 5 following-up patients were normal. The levels of serum vitamin B12 of 8 patients who received with vitamin B12 therapy before hospitalization were normal or increased. Among them,the levels of homocysteine were not measured in 4 patients, those of 3 patients were increased, and that of 1 patient was normal. ③ Results of neuroelectrophysiological examination: Among all patients, 95% (20/21) patients had abnormal sensory-evoked potential, 89% (8/9) patients had abnormal motor evoked potential, 67% (10/15) patients had abnormal nerve conduction, 13% (2/15) patients had neurogenic muscle injury showed by electromyography (EMG), 70% (7/10) patients had abnormal brain-stem auditory evoked potential, and 40% (4/10) patients had abnormal visual evokedpotential. ④ Results of MRI examination of spinal cord: MRI examination demonstrated that 40% (6/15) patients had spinal cord lesion, but spinal cord lesion disappeared in 2 patients during follow up. In addition, clinical manifestations of patients were improved after standard vitamin B12 therapy. CONCLUSION: Nervous system lesion caused by vitamin B12 deficiency is not only involved in spinal cord, also in peripheral nerve, optic nerve, auditory pathway, etc. Diagnosis of the lesion depends on clinical characteristics and level of serum vitamin B12. | 13 | Neuronal tolerance to hypoxia-ischemia through recombinant adeno-associated viral vectors expressing neuronal and inducible nitric oxide synthase An in vivo study显示文摘BACKGROUND: Studies have confirmed that neuronal nitric oxide synthase (nNOS) mediates neurotoxic effects during the early stages of hypoxia-ischemia,while inducible nitric oxide synthase (iNOS) mediates delayed neurotoxicity during advanced stages of hypoxia-ischemia. OBJECTIVE: This study was designed to observe neuronal apoptosis and the expressions of nNOS,iNOS,p38 mitogen-activated protein kinase (MAPK),and caspase-3 mRNA following transfection of recombinant adeno-associated viral vectors separately expressing nNOS and iNOS antisense (rAAV-AsnNOS and rAVV-AsiNOS,respectively) into rat brains subjected to cerebral ischemia; to analyze mechanisms underlying elevated neuronal tolerance to hypoxia-ischemia. DESIGN: A randomized controlled in vivo experiment. SETTING: Fujian Institute of Neurosurgery & Department of Neurosurgery,Union Hospital,Fujian Medical University. MATERIALS: Eighty healthy adult male Sprague Dawley rats of clean grade were provided by the Zhejiang Laboratory Animal Center,China. The protocol was performed in accordance with ethical guidelines for the use and care of animals. The following vectors,rAAV-AsnNOS,rAAV-AsiNOS,and rAAV expressing the β-galactosidase gene (rAAV-LacZ),were successfully constructed by Fujian Institute of Neurosurgery. Rabbit anti-mouse nitrotyrosine (NT) monoclonal antibody (Zhongshan Jinqiao Biotechnology Co.,Ltd.,Beijing,China) and reverse transcription-polymerase chain reaction (RT-PCR) kit (two-step method) (Promega Company,USA) were used in this study. METHODS: This study was performed at the Fujian Institute of Neurosurgery in December 2003. Sixty rats were randomly divided into 3 groups,with 20 rats in each group: rAAV-AsnNOS group,rAAV-AsiNOS group,and rAAV-LacZ group. The remaining 20 rats served as controls. Pre-treated viral vectors (rAAV-AsnNOS,rAAV-AsiNOS,and rAAV-LacZ,respectively; each 50 μL,virus titer of 2×109 viral particles/mL) were transfected into the cerebral cortex of the targeted. Phosphate buffer saline (50 μL) was perfused identically into the rat cerebral cortex of the control group. All rats were subjected to cerebral ischemia by occluding the right middle cerebral artery with suture. Five time points (0,1,6,24,72 hours of ischemia,4 rats for each time point) were allotted to each group. Ischemic brain tissue specimens were prepared for index measurement. MAIN OUTCOME MEASURES: The percentage of NT-positive cells and apoptotic cells in the rat ischemic brain tissue specimens were measured by flow cytometry. The expressions of nNOS,iNOS,p38MAPK,and caspase-3 mRNA were measured by RT-PCR. RESULTS: During the early stages of ischemia (at 1 and 6 hours) with rAAV-AsnNOS transfection,the percentage of NT-positive cells,apoptotic cells,and the expressions of nNOS,p38MAPK and caspase-3 mRNA in the brain nerve cells were remarkably reduced compared to the control,rAAV-LacZ,and rAAV-AsiNOS groups. During the advanced stages of ischemia (at 24 and 72 hours) with rAAV-AsiNOS transfection,the above-mentioned indices were significantly reduced compared to control,rAAV-LacZ,and rAAV-AsnNOS groups. CONCLUSION: Following rAAV transfection,neuronal cells resisted ischemic injury in the MCAO ischemic rat model. Neurons transfected with rAAV-AsnNOS inhibited expression of nNOS,p38MAPK,and caspase-3 during the early stages of ischemia,and those transfected by rAAV-AsiNOS inhibited above-mentioned index expression during the advanced stages of ischemia. | 14 | Herbal formula of Bushen Jianpi(补肾健脾方)combined with sorafenib inhibits hepatocellular carcinoma growth by promoting cell apoptosis and blocking the cell cycle显示文摘OBJECTIVE:To investigate the efficacy of an herbal formula of Bushen Jianpi(补肾健脾方,BSJP)combined with sorafenib on hepatocellular carcinoma(HCC)in vitro and in vivo,and to study the underlying mechanisms of action.METHODS:BSJP,a mixture of 12 raw herbs,was extracted in 70%alcohol/30%water and freeze-dried into a powder.The in vitro effects of BSJP alone,sorafenib alone,and their combination on cell survival,apoptosis,and cell cycle distribution were evaluated in HCC cell lines HCCLM3,HepG2,and SMMC-7721.The expression of B-cell lymphoma-2(Bcl-2),caspase-3,and caspase-9 in HCCLM3 cells was measured using Western blots after drug administration.The in vivo effects of BSJP and sorafenib were evaluated in a tumor surgical resection model using 4-week old male athymic BALB/c nude mice injected with HCCLM3 cells.Immunohistochemical analysis of tumor tissues was performed to evaluate the effects of BSJP alone,sorafenib alone,and their combination on the expression of caspase-3,caspase-9,and Bcl-2.RESULTS:BSJP decreased the survival rate of HCC cell lines,and the combination of BSJP and sorafenib further decreased the survival rate.BSJP significantly promoted cell apoptosis and blocked cell-cycle progression in HCCLM3,HepG2,and SMMC-7721 cells in a dose-dependent manner.Furthermore,the administration of BSJP and sorafenib inhibited the growth of HCCLM3 cell xenografts in nude mice,with no reduction in body weight.In vivo and in vitro experiments showed that BSJP combined with sorafenib could significantly decrease the expression of Bcl-2.CONCLUSION:Our findings suggest that the herbal formula of BSJP is a potential HCC antitumor agent. | ZHOU Zhangjie LIU Xinhua WU Tingting QUE Zujue WU Zhonghua WU Weizhong FU Shujuan ZHANG Shiqiang YANG Yun JIANG Haiyan XIA Xiaoting LV Junqiang DU Boqian LI Yun LU Tao ZHANG Zhihui ZHONG Yi | 2021 | Journal of Traditional Chinese Medicine2021,41,2: | 3 |
| 15 | Development of a fine-resolution atmosphere-wave-ocean coupled forecasting model for the South China Sea and its adjacent seas显示文摘A 72-h fine-resolution atmosphere-wave-ocean coupled forecasting system was developed for the South China Sea and its adjacent seas. The forecasting model domain covers from from 15°S to 45°N in latitude and 99°E to135°E in longitude including the Bohai Sea, the Yellow Sea, the East China Sea, the South China Sea and the Indonesian seas. To get precise initial conditions for the coupled forecasting model, the forecasting system conducts a 24-h hindcast simulation with data assimilation before forecasting. The Ensemble Adjustment Kalman Filter(EAKF) data assimilation method was adopted for the wave model MASNUM with assimilating Jason-2 significant wave height(SWH) data. The EAKF data assimilation method was also introduced to the ROMS model with assimilating sea surface temperature(SST), mean absolute dynamic topography(MADT) and Argo profiles data. To improve simulation of the structure of temperature and salinity, the vertical mixing scheme of the ocean model was improved by considering the surface wave induced vertical mixing and internal wave induced vertical mixing. The wave and current models were integrated from January 2014 to October 2015 driven by the ECMWF reanalysis 6 hourly mean dataset with data assimilation. Then the coupled atmosphere-wave-ocean forecasting system was carried out 14 months operational running since November 2015. The forecasting outputs include atmospheric forecast products, wave forecast products and ocean forecast products. A series of observation data are used to evaluate the coupled forecasting results, including the wind, SHW, ocean temperature and velocity.The forecasting results are in good agreement with observation data. | 16 | Preparation of Ceramic Foams Suitable for Aircraft Arresting by the Airport Runway Based on a Protein Foaming Agent显示文摘Crushable ceramic foams are more suitable to be used as an arrestor material applied in engineered materials arresting system(EMAS) for airport runway for their properties of widely controllable strength, negligible crushing-rebounding behavior, durability, and chemically-inert composition, comparing with traditional concrete foams. The synthesis of ceramic foams adopted direct-foaming method and used an animal protein as foaming agent. Kaolin, talc powder and alumina were the main raw materials. Effects of the ratios of raw materials, calcination temperatures, heating rates, holding time, viscosities of polyvinyl alcohol(PVA) solution as well as the amounts of protein foaming agent and water on microscopic structure, densities, compressive strength and open porosities of ceramic foams were investigated systematically. The results indicate that ceramic foams with typical pore sizes 100-300 μm, open porosities from 73.1% to 91.5%, densities from 0.25 to 0.62 g·cm-3, compressive strength from 0.19 to 4.89 MPa, are obtained by properly adjusting the parameters mentioned above. And the mechanical strength meets the requirement for the EMAS for airport runway. In addition, good correlations are observed among compressive strength, open porosity, microscopic structure, and crystal phase. Its total conductivity was measured to be 3.6 S·cm-1 at 600°C. Though the total conductivity of the BCFNO membrane is much smaller than that of the Ba0.5Sr0.5Co0.8Fe0.2O3-δ (BSCFO) membrane,the oxygen permeability of the BCFNO membrane is similar to that of the BSCFO membrane. SEM observation and EDX analysis of the BCFNO and BSCFO membranes indicated that no segregation of metal ions was found for the used BCFNO membrane while the original perovskite phase of BSCFO decomposed under the experimental condition.
