2Fingolimod alters inflammatory mediators and vascular permeability in intracerebral hemorrhage显示文摘Intracerebral hemorrhage(ICH) leads to high rates of death and disability. The pronounced infl ammatory reactions that rapidly follow ICH contribute to disease progression. Our recent clinical trial demonstrated that oral administration of an immune modulator fingolimod restrained secondary injury derived from initial hematoma,but the mechanisms remain unknown. In this study,we aim to investigate the effects of fingolimod on inflammatory mediators and vascular permeability in the clinical trial of oral fingolimod for intracerebral hemorrhage(ICH). The results showed that fingolimod decreased the numbers of circulating CD4^+ T,CD8^+ T,CD(19)^+B,NK,and NKT cells and they recovered quickly after the drug was stopped. The plasma ICAM level was decreased and IL-10 was increased by fingolimod. Interestingly,fingolimod protected vascular permeability as indicated by a decreased plasma level of MMP9 and the reduced r T1%. In conclusion,modulation of systemic inflammation by fingolimod demonstrates that it is an effective therapeutic agent for ICH. 3Neural Stem Cells Transfected with Leukemia Inhibitory Factor Promote Neuroprotection in a Rat Model of Cerebral Ischemia显示文摘Leukemia inhibitory factor(LIF) contributes to the neuroprotection by neural stem cells(NSCs) after ischemic stroke. Our aim was to explore whether LIFtransfected NSCs(LIF-NSCs) can ameliorate brain injury and promote neuroprotection in a rat model of cerebral ischemia. To accomplish this goal, we transfected NSCs with a lentivirus carrying the LIF gene to stably overexpress LIF. The LIF-NSCs reduced caspase 3 activation under conditions of oxygen-glucose deprivation in vitro.Transient cerebral ischemia was induced in rats by 2 h of middle cerebral artery occlusion(MCAo), and LIF-NSCs were intravenously injected at 6 h post-ischemia. LIF-NSC treatment reduced the infarction volume and improved neurological recovery. Moreover, LIF-NSCs improved glial cell regeneration and ameliorated white matter injuryin the MCAo rats. 4Synthesis of MoX2 (X =Se or S) monolayers with high-concentration 1T'phase on 4H/fcc-Au nanorods for hydrogen evolution显示文摘Controlled synthesis of transition metal dichalcogenide (TMD) monolayers with unusual crystal phases has attracted increasing attention due to their promising applications in electrocatalysis.However,the facile and large-scale preparation of TMD monolayers with high-concentration unusual crystal phase still remains a challenge.Herein,we report the synthesis of MoX2 (X =Se or S) monolayers with high-concentration semimetallic 1T'phase by using the 4H/face-centered cubic (fcc)-Au nanorod as template to form the 4H/fcc-Au@MoX2 nanocomposite.The concentrations of 1T'phase in the prepared MoSe2 and MoS2 monolayers are up to 86% and 81%,respectively.As a proof-of-concept application,the obtained Au@MoS2 nanocomposite is used for the electrocatalytic hydrogen evolution reaction (HER) in acid medium,exhibiting excellent performance with a low overpotential of 178 mV at the current density of 10 mNcm^2,a small Tafel slope of 43.3 mV/dec,and excellent HER stability.This work paves a way for direct synthesis of TMD monolayers with high-concentration of unusual crystal phase for the electrocatalytic application.Zhengqing Liu Xiao Zhang Yue Gong Qipeng Lu Zhicheng Zhang Hongfei Cheng Qinglang Ma Junze Chen Meiting Zhao Bo Chen Ye Chen Xue-Jun Wu Pengfei Yin Lin Gu Yaping Du Hua Zhang 2019Nano Research2019,12,6:3
5Beclin-1/LC3-II dependent macroautophagy was uninfluenced in ischemia-challenged vascular endothelial cells显示文摘Autophagy has been extensively studied and occurs in many biological settings.However,a question remains as to whether ischemia enhances Beclin-1/LC3-II-dependent macroautophagy in vascular endothelial cells,as has been previously thought.Furthermore,the effect of the level of autophagy on cell or skin flap survival still requires elucidation.We created a lethal ischemia model in human umbilical vascular endothelial cells(HUVECs),performed quantitative proteomics and bioinformatics analyses,and verified the autophagic status and effect both in vitro and in vivo.The significantly upregulated proteins encoded by autophagy-related genes(ATGs)included ATG2A,ATG3,ATG4B,ATG5,ATG7,ATG9A,ATG12,ATG16,and ATG101.The significantly enhanced lysosomal proteins were cathepsin B,cathepsin D,lysosome-associated membrane protein 1(LAMP1),and LAMP2.However,the differentially expressed proteins excluded Beclin-1,microtubule-associated protein light chain 3(LC3)-I,and LC3-II.Western blot analyses verified that the protein expression levels of Beclin-1,LC3-I,and LC3-II were neither upregulated nor downregulated in ischemia-challenged HUVECs.The autophagic status was not enhanced by rapamycin in ischemic HUVECs but appeared to be inhibited by chloroquine.Our in vivo study on rats showed that a downregulation in autophagic status jeopardized skin flap survival.In conclusion,Ischemia neither enhanced nor inhibited Beclin-1/LC3-II-dependent canonical macroautophagy both in vitro and in vivo,in contradiction to previous studies.An appropriate autophagic homeostasis can minimize cell or skin flap damage.Yaping Ma Chaofan Li Yan He Tiwei Fu Li Song Qingsong Ye Fugui Zhang 2022Genes & Diseases2022,9,2:3
6Improved Ant Colony Algorithm for Vehicle Scheduling Problem in Airport Ground Service Support显示文摘Support vehicles are part of the main body of airport ground operations,and their scheduling efficiency directly impacts flight delays.A mathematical model is constructed and the responsiveness of support vehicles for current operational demands is proposed to study optimization algorithms for vehicle scheduling.The model is based on the constraint relationship of the initial operation time,time window,and gate position distribution,which gives an improvement to the ant colony algorithm(ACO).The impacts of the improved ACO as used for support vehicle optimization are compared and analyzed.The results show that the scheduling scheme of refueling trucks based on the improved ACO can reduce flight delays caused by refueling operations by 56.87%,indicating the improved ACO can improve support vehicle scheduling.Besides,the improved ACO can jump out of local optima,which can balance the working time of refueling trucks.This research optimizes the scheduling scheme of support vehicles under the existing conditions of airports,which has practical significance to fully utilize ground service resources,improve the efficiency of airport ground operations,and effectively reduce flight delays caused by ground service support.Yaping Zhang Ye Chen Yu Zhang Jian Mao Qian Luo 2023Journal of Harbin Institute of Technology(New Series)2023,30,1:3

9Amorphization activated FeB_(2) porous nanosheets enable efficient electrocatalytic N_(2) fixation显示文摘Designing active,robust and cost-effective catalysts for the nitrogen reduction reaction(NRR) is of paramount significance for sustainable electrochemical NH3 synthesis.Transition-metal diborides(TMB_2)have been recently theoretically predicted to be a new class of potential NRR catalysts,but direct experimental evidence is still lacking.Herein,we present the first experimental demonstration that amorphous FeB_2 porous nanosheets(a-FeB_2 PNSs) could be a highly efficient NRR catalyst,which exhibited an NH3 yield of 39.8 μg h^(-1) mg^(-1)(-0.3 V) and a Faradaic efficiency of 16.7%(-0.2 V),significantly outperforming their crystalline counterpart and most of existing NRR catalysts.First-principle calculations unveiled that the amorphization could induce the upraised d-band center of a-FeB_2 to boost d-2π~* coupling between the active Fe site and ~*N_2 H intermediate,resulting in enhanced ~*N_2 H stabilization and reduced reaction barrier.Out study may facilitate the development and understanding of earth-abundant TMB_2-based catalysts for electrocatalytic N_2 fixation.Ke Chu Weicong Gu Qingqing Li Yaping Liu Ye Tian Wuming Liu 2021Journal of Energy Chemistry2021,30,2:2

16Mapping of de novo mutations in primary biliary cholangitis to a disease-specific co-expression network underlying homeostasis and metabolism显示文摘Primary biliary cholangitis(PBC) is an autoimmune disease involving dysregulation of a broad array of homeostatic and metabolic processes. Although considerable single-nucleotide polymorphisms have been unveiled, a large fraction of risk factors remains enigmatic. Candidate genes with rare mutations that tend to confer more deleterious effects need to be identified. To help pinpoint cellular and developmental mechanisms beyond common noncoding variants, we integrate whole exome sequencing with integrative network analysis to investigate genes harboring de novo mutations. Prominent convergence has been revealed on a network of disease-specific co-expression comprised of 55 genes associated with homeostasis and metabolism. The transcription factor gene MEF2 D and the DNA repair gene PARP2 are highlighted as hub genes and identified to be up-and down-regulated, respectively, in peripheral blood data set. Enrichment analysis demonstrates that altered expression of MEF2 D and PARP2 may trigger a series of molecular and cellular processes with pivotal roles in PBC pathophysiology. 20SlZF3 regulates tomato plant height by directly repressing SlGA20ox4 in the gibberellic acid biosynthesis pathway显示文摘Plant height is an important target trait for crop genetic improvement.Our previous work has identified a salt-tolerant C2H2 zinc finger,SlZF3,and its overexpression lines also showed a semi-dwarf phenotype,but themolecular mechanism remains to be elucidated.Here,we characterized the dwarf phenotype in detail.The dwarfism is caused by a decrease in stem internode cell elongation and deficiency of bioactive gibberellic acids(GAs),and can be rescued by exogenous GA3 treatment.Gene expression assays detected reduced expression of genes in the GA biosynthesis pathway of the overexpression lines,including SlGA20ox4.Several protein-DNA interaction methods confirmed that SlZF3 can directly bind to the SlGA20ox4 promoter and inhibit its expression,and the interaction can also occur for SlKS and SlKO.Overexpression of SlGA20ox4 in the SlZF3-overexpressing line can recover the dwarf phenotype.Therefore,SlZF3 regulates plant height by directly repressing genes in the tomato GA biosynthesis pathway.Jinying Luo Yunfei Tang Zhuannan Chu Yuxin Peng Jiawei Chen Huiyang Yu Chunmei Shi Jahanzeb Jafar Rong Chen Yaping Tang Yongen Lu Zhibiao Ye Ying Li Bo Ouyang 2023Horticulture Research2023,10,4:0
