1Defined tumor antigen-specific T cells potentiate personalized TCR-T cell therapy and prediction of immunotherapy response显示文摘Personalized immunotherapy targeting tumor-specific antigens(TSAs)could generate efficient and safe antitumor immune response without damaging normal tissues.Although neoantigen vaccines have shown therapeutic effect in clinic trials,precise prediction of neoantigens from tumor mutations is still challenging.The host antitumor immune response selects and activates T cells recognizing tumor antigens.Hence,T cells engineered with T-cell receptors(TCRs)from these naturally occurring tumor antigen-specific T(Tas)cells in a patient will target personal TSAs in his/her tumor.To establish such a personalized TCR-T cell therapy,we comprehensively characterized T cells in tumor and its adjacent tissues by single-cell mRNA sequencing(scRNA-seq),TCR sequencing(TCR-seq)and in vitro neoantigen stimulation.Compared to bystander T cells circulating among tissues,Tas cells were characterized by tumor enrichment,tumor-specific clonal expansion and neoantigen specificity.We found that CXCL13 is a unique marker for both CD4^(+)and CD8^(+)Tas cells.Importantly,TCR-T cells expressing TCRs from Tas cells showed significant therapeutic effects on autologous patient-derived xenograft(PDX)tumors.Intratumoral Tas cell levels measured by CXCL13 expression precisely predicted the response to immune checkpoint blockade,indicating a critical role of Tas cells in the antitumor immunity.We further identified CD200 and ENTPD1 as surface markers for CD4^(^(+))and CD8^(^(+))Tas cells respectively,which enabled the isolation of Tas cells from tumor by Fluorescence Activating Cell Sorter(FACS)sorting.Overall,our results suggest that TCR-T cells engineered with Tas TCRs are a promising agent for personalized immunotherapy,and intratumoral Tas cell levels determine the response to immunotherapy.Jingjing He Xinxin Xiong Han Yang Dandan Li Xuefei Liu Shuo Li Shuangye Liao Siyu Chen Xizhi Wen Kuai Yu Lingyi Fu Xingjun Dong Kaiyu Zhu Xiaojun Xia Tiebang Kang Chaochao Bian Xiang Li Haiping Liu Peirong Ding Xiaoshi Zhang Zhenjiang Liu Wende Li Zhixiang Zuo Penghui Zhou 2022Cell Research2022,32,6:10

5Particle swarm optimization and its application to seismic inversion of igneous rocks显示文摘In order to improve the fine structure inversion ability of igneous rocks for the exploration of underlying strata, based on particle swarm optimization(PSO), we have developed a method for seismic wave impedance inversion. Through numerical simulation, we tested the effects of different algorithm parameters and different model parameterization methods on PSO wave impedance inversion, and analyzed the characteristics of PSO method. Under the conclusions drawn from numerical simulation, we propose the scheme of combining a cross-moving strategy based on a divided block model and high-frequency filtering technology for PSO inversion. By analyzing the inversion results of a wedge model of a pitchout coal seam and a coal coking model with igneous rock intrusion, we discuss the vertical and horizontal resolution, stability and reliability of PSO inversion. 6Allotransplantation of adult spinal cord tissues after complete transected spinal cord injury: long-term survival and functional recovery in canines显示文摘Spinal cord injury(SCI), especially complete transected SCI, leads to loss of cells and extracellular matrix and functional impairments. In a previous study, we transplanted adult spinal cord tissues(aSCTs) to replace lost tissues and facilitate recovery in a rat SCI model. However, rodents display considerable differences from human patients in the scale, anatomy and functions of spinal cord systems, and responses after injury. Thus, use of a large animal SCI model is required to examine the repair efficiency of potential therapeutic approaches. In this study, we transplanted allogenic aSCTs from adult dogs to the lesion area of canines after complete transection of the thoracic spinal cord, and investigated the long-term cell survival and functional recovery. To enhance repair efficiency, a growth factor cocktail was added during aSCT transplantation, providing a favorable microenvironment. The results showed that transplantation of a SCTs, in particular with the addition of growth factors, significantly improves locomotor function restoration and increases the number of neurofilament-, microtubule-associated protein2-, 5-hydroxytryptamine-, choline acetyltransferase-and tyrosine hydroxylase-positive neurons in the lesion area at 6 months post-surgery. In addition, we demonstrated that donor neurons in a SCTs can survive for a long period after transplantation. 7Fully integrated hybrid microwave photonic receiver显示文摘Microwave photonic receivers are a promising candidate in breaking the bandwidth limitation of traditional radio-frequency(RF)receivers.To further balance the performance superiority with the requirements regarding size,weight,and power consumption(SWa P),the implementation of integrated microwave photonic microsystems has been considered an upgrade path.However,up to now,to the best of our knowledge,chip-scale fully integrated microwave photonic receivers have not been reported due to the limitation of material platforms.In this paper,we report a fully integrated hybrid microwave photonic receiver(FIH-MWPR)obtained by comprising the indium phosphide(In P)laser chip and the monolithic silicon-on-insulator(SOI)photonic circuit into the same substrate based on the low-coupling-loss micro-optics method.Benefiting from the integration of all optoelectronic components,the packaged FIH-MWPR exhibits a compact volume of 6 cmand low power consumption of 1.2 W.The FIH-MWPR supports a wide operation bandwidth from 2 to 18 GHz.Furthermore,its RF-link performance to down-convert the RF signals to the intermediate frequency is experimentally characterized by measuring the link gain,the noise figure,and the spurious-free dynamic range metrics across the whole operation frequency band.Moreover,we have utilized it as a de-chirp receiver to process the broadband linear frequencymodulated(LFM)radar echo signals at different frequency bands(S-,C-,X-,and Ku-bands)and successfully demonstrated its high-resolution-ranging capability.To the best of our knowledge,this is the first realization of a chip-scale broadband fully integrated microwave photonic receiver,which is expected to be an important step in demonstrating the feasibility of all-integrated microwave photonic microsystems oriented to miniaturized application scenarios.JIACHEN LI SIGANG YANG HONGWEI CHEN XINGJUN WANG MINGHUA CHEN WEIWEN ZOU 2022Photonics Research2022,10,6:2
8Multiplexed intracellular detection based on dual-excitation/dualemission upconversion nanoprobes显示文摘Multiplexed intracellular detection is desirable in biomedical sciences for its higher eficiency and accuracy compared to the single-analyte detection.However,it is very challenging to construct nanoprobes that possess multiple fluorescent signals to recognize the different intracellular species synchronously.Herein,we proposed a novel dual-excitation/dual-emission upconversion strategy for multiplexed detection through the design of upconversion nanoparticles(UCNP)loaded with two dyes for sensitization and quenching of the upconversion luminescence(UCL),respectively.Based on the two independent energy transfer processes of near-infrared(NIR)dye IR845 to UCNP and UCNP to visible dye PAPS-Zn,CIO-and Zn2+were simultaneously detected with a limit of detection(LOD)of41.4 and 10.5 nM,respectively.By tilizing a purpose built 830/980 nm dual-laser confocal microscope,both intrinsic and exogenous CIO and Zn2+in live MCF-7 cells have been accurately quantified.Such dual-excitation/dual-emission ratiometric UCL detection mode enables not only monitoring multiple intracellular analytes but also eliminating the detection deviation caused by inhomogeneous probe distribution in cells.Through modulation of NIR dye and visible dye with other reactive groups,the nanoprobes can be extended to analyze various intraellular species,which provides a promising tool to study the biological activities in live cells and diagnose diseases.Jianxi Ke Shan Lu Zhuo Li Xiaoying Shang Xingjun Li Renfu Li Datao Tu Zhuo Chen Xueyuan Chen 2020Nano Research2020,13,7:2
9Phase Boundary Mapping in ZrNiSn Half-Heusler for Enhanced Thermoelectric Performance显示文摘The solubility range of interstitial Ni in the ZrNi1+xSn half-Heusler phase is a controversial issue,but it has an impact on the thermoelectric properties.In this study,two isothermal section phase diagrams of the Zr-Ni-Sn ternary system at 973K and 1173 K were experimentally constructed based on the binary phase diagrams of Zr-Ni,Zr-Sn,and Ni-Sn.The thermodynamic equilibrium phases were obtained after a long time of heating treatment on the raw alloys prepared by levitation melting.Solubilities of x<0:07 at 973 K and x<0:13 at 1173 K were clearly indicated.An intermediate-Heusler phase with a partly filled Ni void was observed,which is believed to be beneficial to the lowered lattice thermal conductivity.The highest ZT value~0:71 at 973 K was obtained for ZrNi_(1.11)Sn_(1.04).The phase boundary mapping provides an important instruction for the further optimization of ZrNiSn-based materials and other systems.Xiaofang Li Pengbo Yang Yumei Wang Zongwei Zhang Dandan Qin Wenhua Xue Chen Chen Yifang Huang Xiaodong Xie Xinyu Wang Mujin Yang Cuiping Wang Feng Cao Jiehe Sui Xingjun Liu Qian Zhang 2020Research2020,,1:2
10Thermodynamic database of the phase diagrams in the Mg-Al-Zn-Y-Ce system显示文摘The Mg-Al-Zn-Y-Ce system is one of the key systems for designing high-strength Mg alloys. The purpose of the present article is to develop a thermodynamic database for the Mg-Al-Zn-Y-Ce multicomponent system to design Mg alloys using the calculation of phase diagrams (CALPHAD) method, where the Gibbs energies of solution phases such as liquid, fcc, bcc, and hcp phases were described by the subregular solution model, whereas those of all the compounds were de- scribed by the sublattice model. The thermodynamic parameters describing Gibbs energies of the different phases in this database were evaluated by fitting the experimental data for phase equilibria and thermodynamic properties. On the basis of this database, a lot of information concerning stable and metastable phase equilibria of isothermal and vertical sections, mo- lar fractions of constituent phases, the liquidus projection, etc., can be predicted. 11Design of large-displacement asymmetric piezoelectric microgripper based on flexible mechanisms显示文摘The output displacement of the traditional symmetrical microgripper is large,but its micro-components or parts are easily damaged due to the uneven force exerted on the left and right jaws of the gripper.The output force of the traditional asymmetric microgripper is stable.However,its output displacement is small,typically half the output displacement of the symmetric microgripper.To solve these problems,in this study,we designed a large-displacement asymmetric microgripper.First,we calculated the relationship between the theoretical input and output variables based on their geometric relationship.Then,we analyzed the performance of the microgripper using finite element software.Lastly,we used a piezoelectric actuator as the input driver of the microgripper.The errors associated with the theoretical and simulated output displacements were 7.05%and 9.24%,respectively.At 150 V of driving voltage,the maximum output displacement was 224μm,and the actual magnificationwas 11.2 times.Microparts can be gripped in parallel and stably,which confirms the validity of the design.Xiaodong Chen Siya Hu Zilong Deng Jinhai Gao Xingjun Gao 2019Nanotechnology and Precision Engineering2019,2,4:2

19Microwave frequency upconversion employing a coupling-modulated ring resonator显示文摘We present a method to generate a frequency-doubled microwave signal by employing a coupling-modulated ring resonator. Critical coupling is achieved when the resonator intrinsic loss is perfectly balanced by the external coupling enabled by a Mach–Zehnder interferometer coupler. 20Enhanced thermoelectric performance in Ti(Fe, Co, Ni)Sb pseudoternary Half-Heusler alloys显示文摘TiFe0.5Ni0.5Sb-based half-Heusler compounds have the intrinsic low lattice thermal conductivity and the adjustable band structure.Inspired by the previously reports to achieve both p-and n-type components by tuning the ratio of Fe and Ni based on the same parent TiFe0.5Ni0.5Sb,we selected Co as the amphoteric dopants to prepare both n-type and p-type pseudo-ternary Ti(Fe,Co,Ni)Sb-based halfHeusler alloys.The carrier concentration,as well as the density of states effective mass was significantly increased by Co doping,contributing to the enhanced power factor of 1.80 mW m^(-1) K^(-2) for n-type TiFe0.3Co_(0.2)Ni_(0.5)Sb and 2.21 mW m^(-1) K^(-2) for p-type TiFe_(0.5)Co_(0.15)Ni_(0.35)Sb at 973 K.Combined with the further decreased lattice thermal conductivity due to the strain field and mass fluctuation scattering induced by alloying Hf on the Ti site,peak ZTs of 0.65 in n-type Ti0.8Hf_(0.2)Fe_(0.3)Co_(0.2)Ni_(0.5)Sb and 0.85 in ptype Ti0.8Hf_(0.2)Fe_(0.5)Co_(0.15)Ni_(0.35)Sb were achieved at 973 K,which is of great significance for the thermoelectric power generation applications.Qingmei Wang Xiaodong Xie Shan Li Zongwei Zhang Xiaofang Li Honghao Yao Chen Chen Feng Cao Jiehe Sui Xingjun Liu Qian Zhang 2021Journal of Materiomics2021,7,4:0
