1Study on gastric cancer blood plasma based on surface-enhanced Raman spectroscopy combined with multivariate analysis显示文摘A surface-enhanced Raman spectroscopy (SERS) method combined with multivariate analysis was developed for non-invasive gastric cancer detection. SERS measurements were performed on two groups of blood plasma samples: one group from 32 gastric patients and the other group from 33 healthy volunteers. Tentative assignments of the Raman bands in the measured SERS spectra suggest interesting cancer-specific biomolecular changes, including an increase in the relative amounts of nucleic acid, collagen, phospholipids and phenylalanine and a decrease in the percentage of amino acids and saccharide in the blood plasma of gastric cancer patients as compared with those of healthy subjects. Principal components analysis (PCA) and linear discriminant analysis (LDA) were employed to develop effective diagnostic algorithms for classification of SERS spectra between normal and cancer plasma with high sensitivity (79.5%) and specificity (91%). A receiver operating characteristic (ROC) curve was employed to assess the accuracy of diagnostic algorithms based on PCA-LDA. 4Three-dimensional reconstruction of Picea wilsonii Mast. pollen grains using automated electron microscopy显示文摘Three-dimensional electron microscopy(3 D-EM) has attracted considerable attention because of its ability to provide detailed information with respect to developmental analysis. However, large-scale high-resolution 3 D reconstruction of biological samples remains challenging. Herein, we present a 3 D view of a Picea wilsonii Mast. pollen grain with 100 nm axial and38.57 nm lateral resolution using AutoCUTS-SEM(automatic collector of ultrathin sections-scanning electron microscopy). We established a library of 3,127 100 nm thick serial sections of pollen grains for preservation and observation, demonstrating that the protocol can be used to analyze large-volume samples. After obtaining the SEM images, we reconstructed an entire pollen grain comprising 734 serial sections. The images produced by 3D reconstruction clearly revealed the main components of the P.wilsonii pollen grain, i.e., two sacci and pollen corpus, tube cell, generative cell, and two degenerated prothallial cells, and their internal organization. In addition, we performed a quantitative analysis of the different pollen grain cells, including sacci, and found that there were 202 connections within a saccus SEM image. 7Criteria of limiting temperature and parametric analysis of the large deflection behavior for fully restrained steel beams in fire显示文摘This paper mainly involve 3 parts:1) To apply the minimum principle of acceleration in dynamics of elastic-plastic continua at finite deformation to the statics problems,a computing model is presented for the restrained steel beams exposed to the fire.In this model,both effects of large deflection and thermal expansion deformation are taken into account,and the constitutive equations with the temperature effects are used.Then a dynamic finite difference(DFD) method is presented by using the dis-crete technique,which can be used in simulating the response of the steel beams at elevated temperature,and the large deflec-tion behavior and catenary action effects of the beams can be adequately expressed.The primary numerical results show that the method is valid and credible.Compared with other methods,this technique is very simple,and it can also be further devel-oped to simulate the behavior of steel beams subjected to the coupling loading of explosion and fire when both effects of strain rate and inertia are considered.2) By using this DFD method,detailed parametric analysis are presented so as to check the consistency of response results for several different formulas of thermal expansion deformation and retention factors of steel at elevated temperature,the influence of these parameters on the critical temperature is examined.3) Based on the analysis for the curves of temperature-generalized yield function comprised by the axial force and bending moment,both criteria to determine the limiting temperature(or failure temperature) of large deflection steel beams are presented more explicitly,that is,both lim-iting temperatures can be determined by if the catenary force begins to appear or arrives at the maximum value,respectively.It is shown by numerical results that both limiting temperatures are close to the both critical temperatures which are correspond-ing to the maximum deflections equal to span/20 and span/10,respectively.This conclusion may be helpful to make rational fire resisting design for the steel beams.XI Feng LUAN YanPing 2012Science China(Technological Sciences)2012,55,1:2
8A single-center retrospective study on epidemiological and Traditional Chinese Medicine syndrome characteristics of 21010 patients with reflux/heartburn symptoms显示文摘OBJECTIVE:To provide a basis for the clinical identification of true and false reflux,integrated traditional Chinese and Western medicine,and psychosomatic treatment,we conducted a retrospective study of the etiology and epidemiological and Traditional Chinese Medicine(TCM) syndrome characteristics of patients with reflux/heartburn symptoms.METHODS:The 210 10 patients with reflux/heartburn treated at Tianjin Nankai Hospital from January 1,2016,to December 31,2019,were divided into four groups according to their pathogenesis.Sex,age,course of disease,incidence rate,gastroscopy,24-h p Himpedance,esophageal manometry,Hamilton Anxiety Scale(HAMA)/Hamilton Depression Scale(HAMD) score,8-week proton pump inhibitor(PPI) treatment effect,and TCM syndrome characteristics were statistically analyzed.RESULTS:A total of 21010 patients(8864 men and 12146 women),with reflux/heartburn symptoms were screened,including 6284(29.9%) patients with reflux esophagitis(RE),10427(49.6%) patients with nonerosive reflux esophagitis(NERD),2430(11.6%) patients with reflux hypersensitivity(RH),and 1870(8.9%) patients with functional heartburn(FH).The incidence of the disease was higher in women than in men(P <0.0001).The ranking of the incidence of anxiety and depression in these four groups was FH > RH > NERD > RE(P < 0.0001).There were more women than men in the groups with anxiety and more men than women in the groups with depression(P < 0.0001),and there was no significant difference in the distribution of anxiety and depression between men and women(P = 0.5689).There were significant differences in TCM syndrome characteristics between NERD,RE,and functional esophageal diseases(P < 0.01).The highest proportion of functional esophageal disease TCM symptoms was Qi stagnation and phlegm obstruction syndrome(36.16%),and there was no significant difference between RH and FH.The effective rates of PPI treatment at 8 weeks in patients in the RE,NERD,RH,and FH groups were 89%,72%,54%,and 0%,respectively.RE was classified into grades A,B,C,and D according to the Los Angeles grading system.The ranking of the incidence of these four grades was A > B > C > D(P < 0.0001).The effective rates of PPI treatment at 8 weeks were 91%,81%,69%,and 63% in patients with grade A,B,C,and D RE,respectively(P < 0.0001).The highest proportion of TCM syndrome types of NERD and RE was the stagnated heat syndrome in the liver and stomach syndrome,38.99% and 33.90%,respectively.CONCLUSION:Reflux/heartburn symptoms are relatively common in middle-aged women,and NERD is the most common etiology,followed by RE,RH,and FH.The most common TCM syndrome characteristics in NERD and RE were stagnated heat syndrome in the liver and stomach syndrome,and Qi stagnation and phlegm obstruction syndrome in functional esophageal diseases.Most patients with reflux/heartburn symptoms also experienced anxiety and depression.TANG Yanping LI Peicai LIU Xi LIU Lei GONG Yanxia WEI Xiaodong LIU Lina YANG Li 2023Journal of Traditional Chinese Medicine2023,43,3:2

16Jumonji domain-containing 6 (JMJD6) identified as a potential therapeutic target in ovarian cancer显示文摘Jumonji domain-containing 6(JMJD6)is a candidate gene associated with tumorigenesis,and JMJD6 overexpression predicts poor differentiation and unfavorable survival in some cancers.However,there are no studies reporting the expression of JMJD6 in ovarian cancer,and no JMJD6 inhibitors have been developed and applied to targeted cancer therapy research.In the present study,we found that the high expression of JMJD6 in ovarian cancer was correlated with poor prognosis in ovarian cancer.A potential inhibitor(SKLB325)was designed based on the crystal structure of the jmjC domain of JMJD6.This molecule significantly suppressed proliferation and induced apoptosis in a dose-dependent manner in SKOV3 cell lines as detected by CCK-8 cell proliferation assays and flow cytometry.A Matrigel endothelial tube formation assay showed that SKLB325 inhibited capillary tube organization and migration in HUVECs in vitro.We also observed that JMJD6 colocalized with p53 protein in the nucleus,with mRNA and protein expression of p53 as well as its downstream effectors significantly increasing both in vitro and in intraperitoneal tumor tissues treated with SKLB325.In addition,SKLB325 significantly reduced the intraperitoneal tumor weight and markedly prolonged the survival of tumor-bearing mice.Taken together,our findings suggest that JMJD6 may be a marker of poor prognosis in ovarian cancer and that SKLB325 may be a potential candidate drug for the treatment of ovarian cancer.Heng Zheng Yan Tie Zhen Fang Xiaoai Wu Tao Yi Shuang Huang Xiao Liang Yanping Qian Xi Wang Ruyu Pi Siyuan Chen Yong Peng Shengyong Yang Xia Zhao Xiawei Wei 2019Signal Transduction and Targeted Therapy2019,4,1:1

20Numerical Simulation of the Influence of Pore Structure on Resistivity, Formation Factor and Cementation Index in Tight Sandstone显示文摘Tight sandstone,with severe diagenesis and complex pore structure,differs greatly from conventional sandstone in terms of rock electrical parameters.In subsurface rock electrical experiments,various electrical parameters are confounded and can only be analyzed qualitatively.The lack of quantitative analysis for each individual electrical parameter presents a challenge for the evaluation of oil and gas saturation in tight sandstone.Based on the 2D pore-throat model and the features of pore structure in the tight sandstone of the Penglaizhen and Shaximiao Formations in the upper and middle Jurassic of the Western Sichuan Depression,this paper presents 3D micro pore-throat models for three types of tight sandstone.It proposes a finite element-based rock electrical simulation method to analyze the influence of pore structure parameters,such as throat radius and throat tortuosity,on electrical parameters such as resistivity,formation factor,and cementation index quantitatively.The research revealed the following results:(1)Throats of tight sandstone usually have lamellar or curved lamellar shapes that are slender and narrow.The lamellar throat used in the proposed porethroat model is more consistent with the features of tight sandstone than the tubular throat used in the original model.(2)The throat determines the conductivity of tight sandstone.The throat parallel to the electric potential has the greatest influence on conductivity,and the throat perpendicular to the potential has the least influence.(3)In tight sandstone grades I to III,as the porosity decreases,the formation factor increases and the cementation index decreases.(4)The results of the rock electrical simulation are consistent with the results of the rock electrical experiment,which indicates that the proposed rock electrical simulation method of tight sandstone is effective and accurate.WU Feng WEN Zhu YAO Cong WANG Xianhu XI Yanping CONG Linlin 2020Acta Geologica Sinica(English Edition)2020,94,2:0
