3Efficacy and safety of a novel anti-HER2 therapeutic antibody RC48 in patients with HER2-overexpressing,locally advanced or metastatic gastric or gastroesophageal junction cancer:a single-arm phase II study显示文摘Background:Current treatment options for human epidermal growth factor receptor 2(HER2)-overexpressing gastric cancer at third-line have shown limited clinical benefit.Further,there is no specific treatment for HER2 immunohistochemistry(IHC)2+and fluorescence in-situ hybridization-negative patients.Here,we report the efficacy and safety of a novel anti-HER2 antibody RC48 for patients with HER2-overexpressing,advanced gastric or gastroesophageal junction cancer.Methods:Patients with HER2-overexpressing(IHC 2+or 3+),locally advanced or metastatic gastric or gastroesophageal junction cancer who were under at least second-line therapy were eligible and received RC482.5 mg/kg alone every 2 weeks.The primary endpoint was the objective response rate(ORR)assessed by an independent review committee.Secondary endpoints included progressionfree survival(PFS),overall survival(OS),duration of response,time to progression,disease control rate,and safety.Results:Of 179 patients screened,125 were eligible and received RC48 treatment.The ORR was 24.8%(95%confidence interval[CI]:17.5%-33.3%).The median PFS and OS were 4.1 months(95%CI:3.7-4.9 months)and 7.9 months(95%CI:6.7-9.9 months),respectively.The most frequently reported adverse events were decreased white blood cell count(53.6%),asthenia(53.6%),hair loss(53.6%),decreased neutrophil count(52.0%),anemia(49.6%),and increased aspartate aminotransferase level(43.2%).Serious adverse events(SAEs)occurred in 45(36.0%)patients,and RC48-related SAEs were mainly decreased neutrophil count(3.2%).Seven patients had adverse events that led to death were not RC48-related.Conclusions:RC48 showed promising activity with manageable safety,suggesting potential application in patients with HER2-overexpressing,advanced gastric or gastroesophageal junction cancer who have previously received at least two lines of chemotherapy.Zhi Peng Tianshu Liu Jia Wei Airong Wang Yifu He Liuzhong Yang Xizhi Zhang Nanfeng Fan Suxia Luo Zhen Li Kangsheng Gu Jianwei Lu Jianming Xu Qingxia Fan Ruihua Xu Liangming Zhang Enxiao Li Yuping Sun Guohua Yu Chunmei Bai Yong Liu Jiangzheng Zeng Jieer Ying Xinjun Liang Nong Xu Chao Gao Yongqian Shu Dong Ma Guanghai Dai Shengmian Li Ting Deng Yuehong Cui Jianmin Fang Yi Ba Lin Shen 2021Cancer Communications2021,41,11:29
4BMAL1 knockout macaque monkeys display reduced sleep and psychiatric disorders显示文摘Circadian disruption is a risk factor for metabolic, psychiatric and age-related disorders, and non-human primate models could help to develop therapeutic treatments. Here, we report the generation of BMAL1 knockout cynomolgus monkeys for circadian-related disorders by CRISPR/Cas9 editing of monkey embryos. These monkeys showed higher nocturnal locomotion and reduced sleep, which was further exacerbated by a constant light regimen. Physiological circadian disruption was reflected by the markedly dampened and arrhythmic blood hormonal levels. Furthermore, BMAL1-deficient monkeys exhibited anxiety and depression, consistent with their stably elevated blood cortisol, and defective sensory processing in auditory oddball tests found in schizophrenia patients. Ablation of BMAL1 up-regulated transcriptional programs toward inflammatory and stress responses, with transcription networks associated with human sleep deprivation, major depressive disorders, and aging. 5Comparison of Undernutrition Prevalence of Children under 5 Years in China between 2002 and 2013显示文摘Objective To describe the undernutrition status of children under 5-year in China, and study the trend between 2002 and 2013. Methods The study was based on two national surveys. Undernutrition was determined against WHO's 2006 growth standards. The prevalence in 2013 and 2002 was weighted by China sixth National Population Census(2010). The relationship between undernutrition and gender/age groups/different areas use weighted logistic regression. Results The results indicated the overall prevalence of stunting, underweight, and wasting of Chinese children under 5-year was 8.1%, 2.4%, and 1.9% in 2013, respectively. The prevalence of stunting was higher for children aged 12-47 month, while underweight was higher for children aged 48-59 month. The prevalence of undernutrition was higher in rural areas than in urban areas, especially in poor rural areas. There was a decline of stunting, underweight, and wasting between 2002 and 2013 among the children, with greater reduction in rural areas than in urban areas. Conclusion The prevalence of undernutrition of children under 5-year remains high in rural areas especially in poor rural areas in China. 6Polysaccharides from Angelica sinensis alleviate neuronal cell injury caused by oxidative stress显示文摘Angelica sinensis has antioxidative and neuroprotective effects. In the present study, we aimed to determine the neuroprotective effect of polysaccharides isolated from Angelica sinensis. In a preliminary experiment, Angelica sinensis polysaccharides not only protected PC12 neuronal cells from H 2 O 2-induced cytotoxicity, but also reduced apoptosis and intracellular reactive oxygen species levels, and increased the mitochondrial membrane potential induced by H 2 O 2 treatment. In a rat model of local cerebral ischemia, we further demonstrated that Angelica sinensis polysaccharides enhanced the antioxidant activity in cerebral cortical neurons, increased the number of microvessels, and improved blood flow after ischemia. 8Liver Enzymes Concentrations Are Closely Related to Pre-diabetes:Findings of the Shanghai Diabetes Study Ⅱ (SHDS Ⅱ)显示文摘Objective To investigate the relationship of liver enzymes with hyperglycemia in a large population in Shanghai and identify the association between liver enzymes and insulin resistance.Methods A total of 3 756 participants were enrolled.Each participant underwent an oral glucose tolerance test and completed a questionnaire.Anthropometric indices were recorded and serum samples were collected for measurement.Results Liver enzymes concentrations were independently associated with i-IGT,IFG+IGT,and diabetes.With the increase of ALT and GGT concentrations,ORs for i-IGT,IFG+IGT,and diabetes increased gradually.By comparing patients in the highest quartile of GGT concentrations or ALT concentrations with those in the lowest quartile (Q1),ORs for i-IGT,IFG+IGT,or diabetes was significant after adjustment.Both ALT and GGT concentrations were linearly correlated with HOMA-IR and independently associated with HOMA-IR [ALT OR (95% CI):2.56 (1.51-4.34) P=0.00;GGT OR (95% CI):2.66 (1.53-4.65) P=0.00].Conclusion Serum ALT and GGT concentrations were closely related to pre-diabetes and diabetes in the Shanghai population and positively associated with insulin resistance.GAO Fei PAN Jie Min HOU Xu Hong FANG Qi Chen LU Hui Juan TANG Jun Ling GU Hui Lin PAN Zhi Jian YAO You Hua SHEN Wei Zhen JIA Wei Ping 2012Biomedical and Environmental Sciences2012,25,1:15
9Myelin Oligodendrocyte Glycoprotein-IgG Contributes to Oligodendrocytopathy in the Presence of Complement, Distinct from Astrocytopathy Induced by AQP4-IgG显示文摘Immunoglobulin G against myelin oligodendrocyte glycoprotein(MOG-Ig G) is detectable in neuromyelitis optica spectrum disorder(NMOSD) without aquaporin-4 Ig G(AQP4-Ig G), but its pathogenicity remains unclear.In this study, we explored the pathogenic mechanisms of MOG-Ig G in vitro and in vivo and compared them with those of AQP4-Ig G. MOG-Ig G-positive serum induced complement activation and cell death in human embryonic kidney(HEK)-293 T cells transfected with human MOG. In C57 BL/6 mice and Sprague-Dawley rats, MOG-Ig G only caused lesions in the presence of complement. Interestingly, AQP4-Ig G induced astroglial damage, while MOGIg G mainly caused myelin loss. MOG-Ig G also induced astrocyte damage in mouse brains in the presence ofcomplement. Importantly, we also observed ultrastructural changes induced by MOG-Ig G and AQP4-Ig G. These findings suggest that MOG-Ig G directly mediates cell death by activating complement in vitro and producing NMOSDlike lesions in vivo. 10Ultrafast and broadband photodetectors based on a perovskite/organic bulk heterojunction for large-dynamic-range imaging显示文摘Organic-inorganic hybrid perovskite(OIHP)photodetectors that simultaneously achieve an ultrafast response and high sensitivity in the near-infrared(NIR)region are prerequisites for expanding current monitoring,imaging,and optical communication capbilities.Herein,we demonstrate photodetectors constructed by OIHP and an organic bulk heterojunction(BHJ)consisting of a low-bandgap nonfullerene and polymer,which achieve broadband response spectra up to 1μm with a highest external quantum efficiency of approximately 54%at 850 nm,an ultrafast response speed of 5.6 ns and a linear dynamic range(LDR)of 191 dB.High sensitivity,ultrafast speed and a large LDR are preeminent prerequisites for the practical application of photodetectors.Encouragingly,due to the high-dynamicrange imaging capacity,high-quality visible-NIR actual imaging is achieved by employing the OIHP photodetectors.We believe that state-of-the-art OIHP photodetectors can accelerate the translation of solution-processed photodetector applications from the laboratory to the imaging market.Chenglong Li Hailu Wang Fang Wang Tengfei Li Mengjian Xu Hao Wang Zhen Wang Xiaowei Zhan Weida Hu Liang Shen 2020Light(Science & Applications)2020,9,1:13
11Expressions of chromogranin A and cathepsin D in human primary hepatocellular carcinoma显示文摘AIM To determine the expression and clinicalsignificance of chromogranin A and cathepsin Din hepatocellular carcinoma(HCC).METHODS Double immunofluorescence stain-ing techniques combined with laser confocalscanning microscopy(LSCM)was used toinvestigate chromogranin A and cathepsin Dexpressions in 85 HCC patients.RESULTS Cathepsin D was expressed in :3normal liver tissues,while in HCC the stainingshowed regional variation and the fraction ofstrongly stained cells increased as the tumorsbecame less differentiated and usually clinicallymore malignant.Cells which showed strongpositivity for cathepsin D were present in 71/85(83.5%)cases.Strong expression of cathepsinD in cancer cells was related tohistopathological features.They were morecommon in grade 3-4(26/28,92.9%)and grade2(46/53,86.8%)tumors than in grade 1 tumors(1/4,25.0%)(P<0.01).No significantcorrelation was found between age andcathepsin D expression.In patients with positivecathepsin D reaction,the mean age was 52.1±2.8 years(range 32-68 years)and in the groupwith negative reaction,the mean age was 51.3±4.5 years(range 28-71 years).No obvious relationship was observed between CgAexpression in cancer cells and thehistopathological features.The CgA positiverate was 75.0%(3/4)in grade 1,71.7%(38/53)in grade 2,and 71.4%(20/28)in grade 3-4(P>0.05)tumors.The coexpression of CgA andcathepsin D was found by double labeledimmunofluorescence staining techniques.Theprocessing of cathepsin D was disturbed in HCCcells and accumulated in the cells.Cathepsin Dhad proteolytic activity and autocrine mitogeniceffect,suggesting their functions in invasion.These findings demonstrated that the expressionof cathepsin D in HCC had prognostic value.CONCLUSION Chromogranin A and cathepsin Dare expressed in a high proportion of HCC andthe existence of cathepsin D in HCC might berelated to processing of CgA.This is clearly asubject for further studies because of itspotential clinical applications.Xiao Feng Huang Chun Mei Wang Xiao Wen Dai Zhen Jiang Li Bo Rong Pan Li Bin Yu Bin Qian Li Fang 2000World Journal of Gastroenterology2000,6,5:12
12Timing and splitting of nitrogen fertilizer supply to increase crop yield and efficiency of nitrogen utilization in a wheat–peanut relay intercropping system in China显示文摘Agronomically optimizing the timing and rates of nitrogen(N) fertilizer application can increase crop yield and decrease N loss to the environment. Wheat(Triticum aestivum L.)–peanut(Arachis hypogaea L.) relay intercropping systems are a mainstay of economic and food security in China. We performed a field experiment to investigate the effects of N fertilizer on N recovery efficiency, crop yield, and N loss rate in wheat–peanut relay intercropping systems in the Huang-Huai-Hai Plain, China during 2015–2017. The N was applied on the day before sowing, the jointing stage(G30) or the booting stage(G40) of winter wheat, and the anthesis stage(R1) of peanut in the following percentage splits: 50-50-0-0(N1), 35-35-0-30(N2), and 35-0-35-30(N3), using 300 kg N ha-1, with 0 kg N ha-1(N0) as control. ^(15)N-labeled(20.14 atom %) urea was used to trace the fate of N in microplots. The yields of wheat and peanut increased by 12.4% and 15.4% under the N2 and N3 treatments, relative to those under the N1 treatment. The ^(15)N recovery efficiencies( ^(15)NRE) were 64.9% and 58.1% for treatments N2 and N3, significantly greater than that for the N1 treatment(45.3%). The potential N loss rates for the treatments N2 and N3 were23.7% and 7.0%, significantly lower than that for treatment N1(30.1%). Withholding N supply until the booting stage(N3) did not reduce the wheat grain yield; however, it increased the N content derived from ^(15)N-labeled urea in peanuts, promoted the distribution of ^(15)N to pods, and ultimately increased pod yields in comparison with those obtained by topdressing N at jointing stage(N2). In comparison with N2, the N uptake and N recovery efficiency(NRE) of N3 was increased by 12.0% and 24.1%,respectively, while the apparent N loss decreased by 16.7%. In conclusion, applying N fertilizer with three splits and delaying topdressing fertilization until G40 of winter wheat increased total grain yields and NRE and reduced N loss. 14Developing gene-tagged molecular markers for evaluation of genetic association of apple SWEET genes with fruit sugar accumulation显示文摘Sugar content is an important component of fruit quality.Although sugar transporters are known to be crucial for sugar accumulation,the role of genes encoding SWEET sugar transporters in fruit sugar accumulation remains elusive.Here we report the effect of the SWEET genes on fruit sugar accumulation in apple.A total of 25 MdSWEET genes were identified in the apple genome,and 9 were highly expressed throughout fruit development.Molecular markers of these 9 MdSWEET genes were developed and used for genotyping of 188 apple cultivars.The association of polymorphic MdSWEET genes with soluble sugar content in mature fruit was analyzed.Three genes,MdSWEET2e,MdSWEET9b,and MdSWEET15a,were significantly associated with fruit sugar content,with MdSWEET15a and MdSWEET9b accounting for a relatively large proportion of phenotypic variation in sugar content.Moreover,both MdSWEET9b and MdSWEET15a are located on chromosomal regions harboring QTLs for sugar content.Hence,MdSWEET9b and MdSWEET15a are likely candidates regulating fruit sugar accumulation in apple.Our study not only presents an efficient way of implementing gene functional study but also provides molecular tools for genetic improvement of fruit quality in apple-breeding programs.Qiaoling Zhen Ting Fang Qian Peng Liao Liao Li Zhao Albert Owiti Yuepeng Han 2018Horticulture Research2018,5,1:12

16Improving winter wheat grain yield and water-/nitrogen-use efficiency by optimizing the micro-sprinkling irrigation amount and nitrogen application rate显示文摘Available irrigation resources are becoming increasingly scarce in the North China Plain (NCP),and nitrogen-use efficiency of crop production is also relatively low.Thus,it is imperative to improve the water-use efficiency (WUE) and nitrogen fertilizer productivity on the NCP.Here,we conducted a two-year field experiment to explore the effects of different irrigation amounts (S60,60 mm;S90,90 mm;S120,120 mm;S150,150 mm) and nitrogen application rates (150,195 and 240 kg ha^(–1);denoted as N1,N2 and N3,respectively) under micro-sprinkling with water and nitrogen combined on the grain yield(GY),yield components,leaf area index (LAI),flag leaf chlorophyll content,dry matter accumulation (DM),WUE,and nitrogen partial factor productivity (NPFP).The results indicated that the GY and NPFP increased significantly with increasing irrigation amount,but there was no significant difference between S120 and S150;WUE significantly increased first but then decreased with increasing irrigation and S120 achieved the highest WUE.The increase in nitrogen was beneficial to improving the GY and WUE in S60 and S90,while the excessive nitrogen application (N3) significantly reduced the GY and WUE in S120 and S150 compared with those in the N2 treatment.The NPFP significantly decreased with increasing nitrogen rate under the same irrigation treatments.The synchronous increase in spike number (SN) and 1 000-grain weight (TWG)was the main reason for the large increase in GY by micro-sprinkling with increasing irrigation,and the differences in SN and TGW between S120 and S150 were small.Under S60 and S90,the TGW increased with increasing nitrogen application,which enhanced the GY,while N2 achieved the highest TWG in S120 and S150.At the filling stage,the LAI increased with increasing irrigation,and greater amounts of irrigation significantly increased the chlorophyll content in the flag leaf,which was instrumental in increasing DM after anthesis and increasing the TGW.Micro-sprinkling with increased amounts of irrigation or excessive nitrogen application decreased the WUE mainly due to the increase in total water consumption (ET)and the small increase or decrease in GY.Moreover,the increase in irrigation increased the total nitrogen accumulation or contents (TNC) of plants at maturity and reduced the residual nitrate-nitrogen in the soil (SNC),which was conducive to the increase in NPFP,but there was no significant difference in TNC between S120 and S150.Under the same irrigation treatments,an increase in nitrogen application significantly increased the residual SNC and decreased the NPFP.Overall,micro-sprinkling with 120 mm of irrigation and a total nitrogen application of 195 kg ha^(–1) can lead to increases in GY,WUE and NPFP on the NCP.LI Jin-peng ZHANG Zhen YAO Chun-sheng LIU Yang WANG Zhi-min FANG Bao-ting ZHANG Ying-hua 2021Journal of Integrative Agriculture2021,20,2:12

19Recent Advances in Purely Organic Room Temperature Phosphorescence Polymer显示文摘Room temperature phosphorescence(RTP) has drawn increasing attention for its great potential in practical applications.Polymers with large molecular weights and long chains tend to form coil, which can endow them with a high degree of possible rigidity and result in the much restricted non-radiative transition. Also, the intertwined structure of polymers could isolate the oxygen and humidity effectively, thus reducing the consumption of triplet excitons. In consideration of these points, organic polymers would be another kind of ideal platform to realize RTP effect.
