4Efficiency and Inheritance of Targeted Mutagenesis in Maize Using CRISPR-Cas9显示文摘CRISPR/Cas(clustered regularly interspaced short palindromic repeats/CRISPR-associated proteins) is an adaptive immune system in bacteria and archaea to defend against invasion from foreign DNA fragments. Recently, it has been developed as a powerful targeted genome editing tool for a wide variety of species. However, its application in maize has only been tested with transiently expressed somatic cells or with a limited number of stable transgenic T0 plants. The exact efficiency and specificity of the CRISPR/Cas system in the highly complex maize genome has not been documented yet. Here we report an extensive study of the well-studied type II CRISPR-Cas9 system for targeted genome editing in maize, with the codon-optimized Cas9 protein and the short non-coding guide RNA generated through a functional maize U6 sn RNA promoter. Targeted gene mutagenesis was detected for 90 loci by maize protoplast assay, with an average cleavage efficiency of 10.67%. Stable knockout transformants for maize phytoene synthase gene(PSY1) were obtained. Mutations occurred in germ cells can be stably inherited to the next generation. Moreover, no off-target effect was detected at the computationally predicted putative off-target loci. No significant difference between the transcriptomes of the Cas9 expressed and non-expressed lines was detected. 6A stable artificial protective layer for high capacity dendrite-free lithium metal anode显示文摘The metallic lithium(Li)is considered as the most promising anode material for high-e nergy batteries.Nevertheless,the uncon trollable growth of Li den drite and un stable electrolyte/electrode in terface still hin der the developme nt of Li-based battery.In this work,a no vel strategy has been proposed to stabilize Li anode by in-situ polymerizing polypyrrole(PPy)layer on Ni foam(PPy@Ni foam)as an artificial protective layer.The PPy protective layer can effectively decrease the contact between Li metal and electrolyte during cycling.In addition,the morphology characterization shows that the PPy layer can help the even Li deposition undemeath the layer,leading to a dendrite-free Li anode.As a result,when deposited 2 mAh-cm-2 Li metal,the PPy@Ni foam can keep stable Coulombic efficiency(99%)during nearly 250 cycles,much better than the pure Ni foam(100 cycles).Even in the case of the Li capacity of 10 mAh-cm-2,the stable cycling performance for 60 cycles can still be achieved.Furthermore,when assembled with LiFePO4 material as the cathode for a full cell,the PPy@Ni foam can keep high capacity retention of 85.5%at 500 cycles.In our work,we provide a simple and effective method to enhanee the electrochemical performances of Li metal-based batteries,and reveal a new avenue to design three-dimensional(3D)metallic curre nt collector for protecting the Li metal ano de.Zhipeng Wen Yueying Peng Jianlong Cong Haiming Hua Yingxin Lin Jian Xiong Jing Zeng Jinbao Zhao 2019Nano Research2019,12,10:6
7Defective Kernel 39 encodes a PPR protein required for seed development in maize显示文摘RNA editing is a posttranscriptional process that is important in mitochondria and plastids of higher plants. All RNA editing-specific trans-factors reported so far belong to PLS-class of pentatricopeptide repeat(PPR)proteins. Here, we report the map-based cloning and molecular characterization of a defective kernel mutant dek39 in maize. Loss of Dek39 function leads to delayed embryogenesis and endosperm development, reduced kernel size, and seedling lethality. Dek39 encodes an E subclass PPR protein that targets to both mitochondria and chloroplasts, and is involved in RNA editing in mitochondrial NADH dehydrogenase3(nad3) at nad3-247 and nad3-275. C-to-U editing of nad3-275 is not conserved and even lost in Arabidopsis, consistent with the idea that no close DEK39 homologs are present in Arabidopsis. However, the amino acids generated by editing nad3-247 and nad3-275 are highly conserved in many other plant species, and the reductions of editing at these two sites decrease the activity of mitochondria NADH dehydrogenase complex I,indicating that the alteration of amino acid sequence is necessary for Nad3 function. 8Polybrominated diphenyl ether (PBDE) in blood from children (age 9–12) in Taizhou, China显示文摘Polybrominated diphenyl ethers (PBDEs) as ubiquitous persistent organic pollutants have attracted much attention in recent years. Exposure to PBDEs could induce a high health risk for children. The aim of this study was to investigate the PBDEs exposure of children (9–12 years) from Taizhou, China. Fifty-eight blood samples were collected in one school in a mountainous area in Taizhou. The concentrations of ∑9PBDEs (sum of BDE-28, -47, -99, -100, -153, -154, -183, -197 and -209) ranged from 2.66 to 33.9 ng/g lipid wet (lw) with a median of 7.22 ng/g lw. These concentrations were lower than those of children in USA, but close to European and Asian general population levels. The results showed that children in Taizhou countryside were at a low level of PBDEs exposure. The predominant congener was BDE-209, followed by BDE-28, -47, -197 and -153. 9(Cr_(0.2)Mn_(0.2)Fe_(0.2)Co_(0.2)Mo_(0.2))B:A novel high-entropy monoboride with good electromagnetic interference shielding performance in K-band显示文摘A novel equimolar high-entropy(HE)transition metal monoboride,(Cr_(0.2)Mn_(0.2)Fe_(0.2)Co_(0.2)Mo_(0.2))B,was designed and prepared in powder and bulk form by high temperature elemental reaction method and spark plasma sintering(SPS)method,respectively.XRD analysis shows that HE(Cr_(0.2)Mn_(0.2)Fe_(0.2)Co_(0.2)Mo_(0.2))B possesses orthorhombic structure with Pnma space group.Through Rietveld refinement,the lattice parameters of HE(Cr_(0.2)Mn_(0.2)Fe_(0.2)Co_(0.2)Mo_(0.2))B are a=5.6675,b=2.9714,c=4.2209 and the theoretical density is 6.95 g/cm~3.The Vickers hardness and electrical conductivity of HE(Cr_(0.2)Mn_(0.2)Fe_(0.2)Co_(0.2)Mo_(0.2))B bulk with relative density of 90%is 12.3±0.5 GPa and 0.49±0.04×10~6 S/m,respectively.Due to high electrical conductivity,HE(Cr_(0.2)Mn_(0.2)Fe_(0.2)Co_(0.2)Mo_(0.2))B bulk with 3.0 mm thickness displays superior EMI shielding performance in 18.0–26.5 GHz(K-band),and the average values of SET,SER,and SEAare 23.3 dB,13.9 dB,and 9.4 dB,respectively.The EMI shielding mechanism of HE(Cr_(0.2)Mn_(0.2)Fe_(0.2)Co_(0.2)Mo_(0.2))B mainly results from reflection.Haiming Zhang Biao Zhao Fu-Zhi Dai Huimin Xiang Zhili Zhang Yanchun Zhou 2021Journal of Materials Science & Technology2021,,18:5

12Maize WI5 encodes an endo-1,4-β-xylanase required for secondary cell wall synthesis and water transport in xylem显示文摘Water transport from roots to leaves through xylem is important for plant growth and development.Defects in water transport can cause drought stress,even when there is adequate water in the soil.Here,we identi-fied the maize(Zea mays)wilty5(wi5)mutant,which ex-hibits marked dwarfing and leaf wilting throughout most of its life cycle under normal growth conditions.wilty5 seed-lings exhibited lower xylem conductivity and wilted more rapidly under drought,NaCl,and high temperature treat-ments than wild-type plants.Map-based cloning revealed that WI5 encodes an active endo-1,4-β-xylanase from gly-cosyl dehydration family 10,which mainly functions in degrading and reorganizing cell wall xylan.Reverse-transcription polymerase chain reaction andβ-glucuronidase assays revealed that WI5 is highly expressed in stems,especially in internodes undergoing secondary wall as-sembly.RNA sequencing suggested that WI5 plays a unique role in internode growth.Immunohistochemistry and electron microscopy confirmed that wi5 is defective in xylan deposition and secondary cell wall thickening.Lignin deposition and xylan content were markedly re-duced in wi5 compared to the wild-type plants.Our results suggest that WI5 functions in xylem cell wall thickening through its xylanase activity and thereby regulates xylem water transport,the drought stress response,and plant growth in maize.Xiaojiao Hu Yang Cui Xiaomin Lu Weibin Song Lei Lei Jinjie Zhu Jinsheng Lai Lizhu E Haiming Zhao 2020Journal of Integrative Plant Biology2020,62,10:3
13Parameter studies and evaluation principles of delamination damage in laminated composites显示文摘Delamination represents one of the most severe failure modes in composite laminates,especially when they are subjected to uniaxial compression loads.The evaluation of the delamination damage has always been an essential issue of composite laminates for durability and damage tolerance in engineering practice.Focusing on the most typical and representative elliptical delamination issue,an analytical model simultaneously considering the conservative buckling process and non-conservative delamination propagation process is implemented.Various computational cases considering different delamination depths,directions,aspect ratios,and areas are established,and the predicted results based on the analytical model are carefully compared.Effects of these geometrical delamination parameters on the buckling,delamination propagation,and failure behaviors of composite laminates are thoroughly analyzed,and innovative evaluation principles of the delamination damage have been concluded.It is found that the delamination area is the key factor that truly affecting the failure behaviors of delaminated composites,and the local/global buckling and failure loads show clear linearity with the delamination area,whilst the delamination depth and direction only have slight effects.Kangkang WANG Libin ZHAO Haiming HONG Jianyu ZHANG Yu GONG 2021Chinese Journal of Aeronautics2021,34,7:3
14OS1 functions in the allocation of nutrients between the endosperm and embryo in maize seeds显示文摘Uncovering the genetic basis of seed develop-ment will provide useful tools for improving both crop yield and nutritional value, However, the genetic regulatory networks of maize (Zea mays) seed development remain largely unknown. The maize opaque endosperm and small germ 1 (osl) mutant has opaque endosperm and a small embryo,Here, we cloned OSi1 and show that it encodes a putative transcription factor containing an RWP-RK domain-Transcriptional analysis indicated that OSi1 expression is elevated in early endosperm development, especially in the basal endosperm transfer layer (BETL), conducting zone (CZ), and central starch endospcrm (CSE) cells. RNA sequencing (RNA-Seq) analysis of the osi mutant revealed sharp downregulation of certain genes in specific cell types, including ZmMRP-1 and Megl in BETL cells and a majority of zein-and starch-related genes in CSE cells. Using a haploid induction system, we show that wild-type endosperm could rescue the smaller size of osi embryo, which suggests that nutrients are allocated by tho wild type endosperm. Therefore, our dsta imply that the network regulated by 051 accomplishes a key step in nutrient allocation between endosperm and embryo within maize seeds. 15Molecular characterization and developmental expression patterns of thyroid hormone receptors(TRs) and their responsiveness to TR agonist and antagonist in Rana nigromaculata显示文摘Considering some advantages of Rana nigromaculata as an experimental species, we propose that this species, like Xenopus laevis, could be used to assay thyroid hormone(TH) signaling disrupting actions. To validate the utilizability of R. nigromaculata, we investigated the responsiveness of R. nigromaculata to a TH receptor(TR) agonist(T3) and antagonist(amiodarone) by analyzing expression, based on characterizing TR cDNA and developmental expression patterns. With high levels of identity with the corresponding genes in X. laevis, both TRα and TRβ in R. nigromaculata exhibited roughly similar developmental expression patterns to those of X. laevis, in spite of some species-specific differences. Both TRα and TRβ expression had greater changes in the liver and intestine than in the tail and brain during metamorphosis. T3 exposure for 2 days induced more dramatic increases of TRβ expression in stage 27 than in stage34 tadpoles but not in stage 42 tadpoles, showing that the responsiveness of R. nigromaculata to TH decreased with development and disappeared at the onset of metamorphic climax.Corresponding to greater changes of TRβ expression in the liver and intestine than in the tail and brain during metamorphosis, the liver and intestine had higher responsiveness to exogenous T3 than the tail and brain. Amiodarone inhibited T3-induced TRβ expression. 17NaV6O15:A promising cathode material for insertion/extraction of Mg^2+with excellent cycling performance显示文摘The rechargeable magnesium batteries(RMBs)are getting more and more attention because of their high-energy density,high-security and low-cost.Nevertheless,the high charge density of Mg^2+makes the diffusion of Mg2+in the conventional cathodes very slow,resulting in a lack of appropriate electrode materials for RMBs.In this work,we enlarge the layer spacing of V2Os by introducing Na^2+in the crystal structure to promote the diffusion kinetics of Mg^2+.The NaVeO15(NVO)synthesized by a facile method is studied as a cathode material for RMBs with the anhydrous pure Mg^2+electrolyte.As a result,the NVO not only exhibits high discharge capacity(119.2 mAh:g^-1 after 100 cycles at the current density of 20 mA:g^-1)and working voltage(above 1.6 V vs.Mg^2+/Mg),but also expresses good rate capability.Besides,the eX-situ characterizations results reveal that the Mg^2+storage mechanism in NVO is based on the intercalation and deintercalation.The density functional theory(DFT)calculation results further indicate that Mg^2+tends to occupy the semi-occupied sites of Na^+in the NVO.Moreover,the galvanostatic intermittent titration technique(GITT)demonstrates that NVO electrode has the fast diffusion kinetics of Mg^2+during discharge process ranging from 7.55×10^-13 to2.41×10^-11 cm^2·s^-1.Our work proves that the NVO is a potential cathode material for RMBs.Dongzheng Wu Jing Zeng Haiming Hua Junnan Wu Yang Yang Jinbao Zhao 2020Nano Research2020,13,2:2
18Hierarchical N-doped carbon nanocages/carbon textiles as a flexible O2 electrode for Li–O2 batteries显示文摘The conventional Li–O2 battery(LOB)has hardly been considered as a next-generation flexible electronics thus far,since it is bulk,inflexible and limited by the absence of an adjustable cell configuration.Here,we present a flexible Li–O2 cell using N-doped carbon nanocages grown onto the carbon textiles(NCNs/CTs)as a self-standing and binder-free O2 electrode.The highly flexible NCNs/CTs exhibits an excellent mechanic durability,a promising catalytic activity towards the ORR and OER,a considerable cyclability of more than 70 cycles with an overpotential of 0.36 V on the 1 stcycle at a constant current density of 0.2 m A/cm2,a good rate capability,a superior reversibility with formation and decomposition of desired Li2 O2,and a highly electrochemical stability even under stringent bending and twisting conditions.Our work represents a promising progress in the material development and architecture design of O2 electrode for flexible LOBs.Jia Liu Dan Li Siqi Zhang Ying Wang Guiru Sun Zhao Wang Haiming Xie Liqun Sun 2020Journal of Energy Chemistry2020,29,7:2
19Clinical significance of interventional therapeutic bronchoscopy combined with bronchial arterial embolization in the treatment of hypervascular primary airway tumors in children显示文摘Importance Pediatric hypervascular primary airway tumors are progressive,fatal lesions with a low incidence,and the disease is often more serious than that in adults.Objective To evaluate the clinical efficacy and safety of interventional therapeutic bronchoscopy combined with conservative treatment and bronchial arterial embolization in children with primary airway tumors.Methods We retrospectively analyzed the clinical data of four pediatric patients with hypervascular primary airway tumor between 2017 and 2019 at Beijing Children’s Hospital.Results Two patients were low-grade bronchial mucoepidermoid carcinoma,one patient was pleomorphic adenoma,and one was bronchial leiomyoma.Interventional therapeutic bronchoscopy combined with bronchial arterial embolization was used for treatment(all four patients received general anesthesia).The tumors were safely resected in all patients via interventional bronchoscopy.There were no severe complications related to the procedures.All patients were followed up for 5–12 months,and one low-grade bronchial mucoepidermoid carcinoma recurred.Interpretation Interventional therapeutic bronchoscopy combined with bronchial arterial embolization appears to be a safe and efficient therapeutic method associated with less trauma and fewer complications,including no serious adverse events,in children with hypervascular primary airway tumors without bronchus wall infiltration.Haiming Yang Jie Yin Xiaolei Tang Gan Li Huimin Li Shunying Zhao 2021Pediatric Investigation2021,5,2:2
20Synthesis of an ordered porous carbon with the dual nitrogen-doped interfaces and its ORR catalysis performance显示文摘Both nitrogen-doping feature and pore structure are critical factors for developing nitrogen-doped carbons based catalysts with a high performance toward oxygen reduction reaction(ORR).Herein,a simple one-step CVD of acetylene and acetonitrile vapor method using silanized SBA-15 as a template has been developed to synthesize an ordered porous carbon(OPC) with dual nitrogen-doped interfaces.The optimized sample as prepared with the CVD of 4 h at 750℃ contains two types of ordered mesopores that one type is the ordered cylindrical pores inheriting from the pores of SBA-15 and has a pore width of4.0~5.0 nm,the other type is the ordered quasi-hexagonal pores with a width of 3.0~4.0 nm produced by etching the pore walls of SBA-15.These two types of pores whose pore walls are built by the nitrogen doped carbon layers resulted by the CVD and thus it actually makes the dual nitrogen-doped interfaced OPC(DN-OPC).Meanwhile,DN-OPC contains a few of micropores and a large SSA of 1430 m~2/g.This dualordered pores and dual nitrogen-doped interfaces cannot only facilitate mass transport but also utilize the active sites of DN-OPC for ORR.Therefore,as metal-free ORR catalyst,DN-OPC exhibits a good activity close to commercial Pt/C catalyst,and an excellent durability and methanol tolerance.Hongwei Zhao Yanqiu Zhang Li Lixiang Xin Geng Haiming Yang Weimin Zhou Chengguo Sun Baigang An 2021Chinese Chemical Letters2021,32,1:2
