1Inflammasome activation in mouse inner ear in response to MCMV induced hearing loss显示文摘Objective: To identify presence of inflammasome activated in mouse cochlea with sensorineural hearing loss(SNHL) caused by cytomegalovirus(CMV) infection.Method: MCMV was injected into the right cerebral hemisphere in neonatal BALB/c mice at 2000 pfu virus titers. Auditory brainstem responses(ABRs) were tested to evaluate hearing at 21 days. Histopathological studies were conducted to confirm localizations of MCMV infected cells in the inner ear. Expression of inflammasome related factors was assessed by immunofluorescence, Quantitative real-time PCR and Western blotting.Results: In the mouse model of CMV induced SNHL, inflammasome related kinase Caspase-1 and downstream inflammatory factor IL-1b and IL-18 were found increased and activated after CMV infection in the cochlea. These factors could further up-regulate expression of IL-6 and TNFa. These inflammatory factors are neurotoxicity and may contribute to hearing impairment. Furthermore, we also detected significantly increased AIM2 protein that accumulated in the SGN of cochleae with CMV infection.Significance: We have shown that inflammasome as a novel inherent immunity mechanism may contribute to hearing impairment.Conclusion: Our data indicate that imflammasome assemble in mouse inner ear in response to CMV infection. We have revealed a novel pathology event in CMV induced SNHL involving activation of inflammasome in mouse cochlea. 2Preparation of antifouling ultrafiltration membranes from copolymers of polysulfone and zwitterionic poly(arylene ether sulfone)s显示文摘The past few decades have witnessed rapid gains in our demands of antifouling membranes such as water purification membranes and hemodialysis membranes.A variety of methodologies have been proposed for improving the antifouling performance and the hemocompatibility of the membranes.In this study,a series of copolymers(PSF-PESSB)containing polysulfone(PSF)and poly(arylene ether sulfone)bearing pendant zwitterionic sulfobetaine groups(PESSB)were prepared via one-pot polycondensation.Subsequently,the ultrafiltration(UF)membranes were prepared from different zwitterion-containing copolymers.The prepared membranes showed high thermal stability and mechanical properties.Besides,it also displayed attractive antifouling performance and blood compatibility.Compared with the original PSF membrane,the amount of protein absorption on the modified membrane was reduced;the flux recovery ratio and the resistance to blood cells were significantly improved.The results of this work suggest that PSF-PESSB membranes are expected to be applied in blood purification.The introduction of zwitterion-containing polymers to membranes paves ways for developing advanced hemodialysis technologies for crucial process.Sihan Huang Yaohan Chen Xue Wang Jing Guo Yonggang Li Lei Dai Shenghai Li Suobo Zhang 2022Chinese Journal of Chemical Engineering2022,35,9:1
3Vestibular-evoked myogenic potentials in miniature pigs显示文摘Objective: To report detection of vestibular-evoked myogenic potentials(VEMPs) in the miniature pig.Methods: Potentials evoked by 1000 Hz tone bursts were recorded from neck extensor muscles and the masseter muscles in normal adult Bama miniature pigs anesthetized with 3% pentobarbital sodium and Carbachol II.Results: The latency of the first positive wave P from neck extensor muscles was 7.65 ± 0.64 ms, with an amplitude of 1.66 ± 0.34 uv and a rate of successful induction of 75% at 80 d B SPL. The latency of potentials evoked from the masseter muscles was 7.60 ± 0.78 ms, with an amplitude of 1.31 ± 0.28 uv and a rate successful induction of 66% at 80 d B SPL.Conclusion: The latencies and thresholds of VEMPs recorded from the neck extensor muscle and the masseter muscle appear to be comparable in normal adult Bama miniature pigs, although the amplitude recorded from the neck extensor muscle seems to be higher than that from the masseter muscle. 4Fluorescent sensing of anions based on excited state intramolecular proton transfer in N-(3-hydroxy-2- naphthamido)-N′-phenylthiourea显示文摘A neutral N-amidothiourea-based excited state intramolecular proton transfer (ESIPT) anion receptor bearing an o-hydroxynaphthamide fluorophore and a thiourea binding site, N-(3-hydroxy-2-naphthamide)-N’-phenylthiourea (1a), was designed and synthesized. Fluorescence and absorption response of 1a toward anions were assessed in acetonitrile. IR and NMR experiments indicated that the 'OH…O=C' intramolecular hydrogen bond (IHB) in 1a was weak so that it only exhibited the short-wavelength normal emission other than ESIPT fluorescence. Due to the high anion binding affinity of the N-amidothiourea binding site and the formation of a hydrogen binding network in the 1a-anion c omplex, 1a underwent structural change upon anion binding that strengthens the 'OH…O=C' IHB, leading to the ESIPT and the observation of the long-wavelength ESIPT emission whereas the normal fluorescence is quenched. 5The widely reported but poorly studied ciliate family Folliculinidae(Protozoa,Ciliophora,Heterotrichea):a revision with notes on its taxonomy,morphology and phylogenetic relationships显示文摘Ciliates of the heterotrich family Folliculinidae are widespread in various habitats and are distinguished by their transparent loricae of various shapes,conspicuous peristomial lobes,and dimorphic life cycles.They usually attach firmly to the surface of substrates,feed on bacteria and microalgae,and play a significant role in energy flow and material cycling in the microbial food web.However,little is known regarding their biodiversity and systematics.In this work,we establish the terminology of the family Folliculinidae and select six crucial features for genus recognition.Based on previous studies,we revise the classification of Folliculinidae,supply improved diagnoses for each of the 33 folliculinid genera,and provide a key to their identification.Moreover,phylogenetic analyses based on small subunit ribosomal DNA(SSU rDNA)sequences revealed that the family is monophyletic and comprises two subclades(subclades I II)which can be identified by the flexibility of their peristomial lobes and the sculpturing of their necks.Furthermore,we investigate the evolutionary relationships of folliculinids using the six chosen generic features.Tingting Ye Yaohan Jiang Shuchang Chen Yixiang Xu Lifang Li Mann Kyoon Shin Xiangrui Chen 2022Marine Life Science & Technology2022,4,4:0
6Microstructural evolution and oxidation inα/βtitanium alloy under fretting fatigue loading显示文摘Coupling effects of fretting wear and cyclic stress could result in significant fatigue strength degradation,thus potentially causing unanticipated catastrophic fractures.The underlying mechanism of microstructural evolutions caused by fretting wear is ambiguous,which obstructs the understanding of fretting fatigue issues,and is unable to guarantee the reliability of structures for long-term operation.Here,fretting wear studies were performed to understand the microstructural evolution and oxidation behavior of anα/βtitanium alloy up to 108 cycles.Contact surface degradation is mainly caused by surface oxidation and the generation of wear debris during fretting wear within the slip zone.The grain size in the topmost nanostructured layer could be refined to~40 nm.The grain refinement process involves the initial grain rotation,the formation of low angle grain boundary(LAGB;2°–5°),the in-situ increments of the misorientation angle,and the final subdivision,which have been unraveled to feature the evolution in dislocation morphologies from slip lines to tangles and arrays.The formation of hetero microstructures regarding the nonequilibrium high angle grain boundary(HAGB)and dislocation arrays gives rise to more oxygen diffusion pathways in the topmost nanostructured layer,thus resulting in the formation of cracking interface to separate the oxidation zone and the adjoining nanostructured domain driven by tribological fatigue stress.Eventually,it facilitates surface degradation and the formation of catastrophic fractures.Hanqing LIU Xiaohong SHAO Kai TAN Zhenjie TENG Yaohan DU Lang LI Qingyuan WANG Qiang CHEN 2023Friction2023,11,10:0
7Highly stable side-chain-type cardo poly(aryl ether ketone)s membranes for vanadium flow battery显示文摘Vanadium flow batteries(VFBs)have drawn considerable attention as an emerging technology for largescale energy storage systems(ESSs).One of the pivotal challenges is the availability of eligible ion exchange membranes(ICMs)that provide high ion selectivity,proton conductivity,and stability under rigorous condition.Herein,a‘side-chain-type’strategy has been employed to fabricate highly stable phenolphthalein-based cardo poly(arylene ether ketone)s(PAEKs)membrane with low area resistance(0.058Ωcm^(2)),in which flexible alkyl spacers effectively alleviated inductive withdrawing effect from terminal ion exchange groups thus enabling a stable backbone.The assembled VFBs based on PAEKs bearing pendent alkyl chain terminated with quaternary ammonium(Q-PPhEK)demonstrated an energy efficiency above 80%over 700 cycles at 160 mA/cm^(2).Such a remarkable results revealed that the side-chain-type strategy contributed to enhancing the ICMs stability in strong oxidizing environment,meanwhile,more interesting backbones would be woken with this design engaging in stable ICMs for VFBs.Ziming Zhao Qing Dai Sihan Huang Wenjing Lu Yaohan Chen Jifu Zheng Suobo Zhang Shenghai Li Xianfeng Li 2024Chinese Chemical Letters2024,35,3:0
