| 1 | NLRP3-dependent pyroptosis is required for HIV-1 gp120- induced neuropathology显示文摘The human immunodeficiency virus-1(HIV-1)envelope protein gp120 is the major contributor to the pathogenesis of HIVassociated neurocognitive disorder(HAND).Neuroinflammation plays a pivotal role in gp120-induced neuropathology,but how gp120 triggers neuroinflammatory processes and subsequent neuronal death remains unknown.Here,we provide evidence that NLRP3 is required for gp120-induced neuroinflammation and neuropathy.Our results showed that gp120-induced NLRP3-dependent pyroptosis and IL-1βproduction in microglia.Inhibition of microglial NLRP3 inflammasome activation alleviated gp120-mediated neuroinflammatory factor release and neuronal injury.Importantly,we showed that chronic administration of MCC950,a novel selective NLRP3 inhibitor,to gp120 transgenic mice not only attenuated neuroinflammation and neuronal death but also promoted neuronal regeneration and restored the impaired neurocognitive function.In conclusion,our data revealed that the NLRP3 inflammasome is important for gp120-induced neuroinflammation and neuropathology and suggest that NLRP3 is a potential novel target for the treatment of HAND. | Xiaolong He Weijun Yang Zhijie Zeng Yi Wei Jie Gao Bao Zhang Li Li Liqun Liu Yu Wan Qing Zeng Zelong Gong Liting Liu Hanyun Zhang Yubin Li Shaojie Yang Tongtong Hu Lixian Wu Eliezer Masliah Shenghe Huang Hong Cao | 2020 | Cellular & Molecular Immunology2020,17,3: | 11 |

| 3 | Experimental Study on Thermal Management of Cylindrical Li-ion Battery with Flexible Microchannel Plates显示文摘A novel thermal management system of cylindrical Li-ion battery with the liquid cooling in flexible microchannel plate was established in the study. The experiments were conducted with R141 b in flexible microchannel plates. The cooling system with the flexible aluminum microchannels can effectively transfer heat from battery to the cooling refrigerant R141 b based on flow boiling. A battery module with five cells along flow channel was chosen to study the effects of contact surface area and mass flux on the thermal performance and electrochemical characteristics in the experiments. Three types of structure with different contact areas were studied and their performances were compared with the experiments without cooling structures. The experiments were carried out at the same discharge rate with the inlet mass flow rates of 0–10 kg/h. For the inlet mass flow rate of 5.98 kg/h, the surface temperature and temperature uniformity of battery were the best, and the output voltage and capacity of batteries were higher than those under other mass flow rates. With given inlet mass flow rates, the five series cells exhibited different electrochemical performances, including output voltage and discharge capacity, due to the different refrigerant flow states in the microchannels. | 5 | The effect of pore size and porosity of Ti6Al4V scaffolds on MC3T3-E1 cells and tissue in rabbits显示文摘Electron beam melting(EBM) allows the fabrication of specific porous titanium implants, whereas their in vitro and in vivo biological performance should be further investigated. In this study, we examined the porous Ti6 Al4 V scaffolds(low, 334.1 μm pore size with 55.4% porosity;middle, 383.2 μm pore size with 65.2% porosity;and high, 401.6 μm pore size with 78.1%porosity) fabricated through EBM. The structural characterization and mechanical properties of porous Ti6 Al4 V scaffolds were measured through micro-computed tomography(micro-CT), scanning electron microscopy, and a material testing system.MC3 T3-E1 cells were used to assess the proliferation and differentiation of the cells on different scaffolds at day 7 and day 14 based on the expression levels of genes, including alkaline phosphatase, bone morphogenetic protein-2, osteopontin and runtrelated transcription factor-2. Rabbits with distal femoral defects were utilized to evaluate bone ingrowth in the porous titanium.All of the samples were subjected to micro-CT and histological analysis after 12 weeks. Results showed that compressive Young’s modulus of 0.3–1.1 GPa was similar to the trabecular bone. The three types of porous Ti6 Al4 V scaffolds were inclined to promote cell proliferation, whereas cell differentiation and bone ingrowth into the porous scaffolds were biased to the porous titanium with relatively large pores and porosity(middle and high). | 6 | Cell Membrane-camouflaged Multi-functional Dendritic Large Pore Mesoporous Silica Nanoparticles for Combined Photothermal Therapy and Radiotherapy of Cancer显示文摘Combining photothermal therapy and radiotherapy(PTT-RT) with reducing tumor hypoxia acts as an important antitumor modality. However, it is a great challenge to realize photothermal therapy, radiotherapy and exogenous oxygen supply in one nanosystem. To realize a combination of the three functions, we fabricated a red blood cell membrane(RBCm)-camouflaged, red blood cell content(RBCc) and the copper sulfide(CuS) co-loaded dendritic large pore mesoporous silica nanoparticle(DLMSN/CuS/RBCc/ RBCm). The cell membrane coating endowed the nanoparticles with good stability in the physiological environment, and CuS allowed the nanoparticle exhibiting good photothermal and radiosensitization properties. RBCc loaded nanoparticle DLMSN/CuS/RBCc enhanced superior anti-tumor effect than DLMSN/CuS during combined PTT-RT therapy because the introduction of RBCc increased the exogenous oxygen supply. The in vitro study further demonstrated that the combination of photothermal therapy and radiotherapy induced superior antitumor efficacy than single therapy. | 7 | Building the Chordata Olfactory Receptor Database using more than 400,000 receptors annotated by Genome2OR显示文摘Olfactory receptors are poorly annotated for most genome-sequenced chordates.To address this deficiency,we developed a nhmmer-based olfactory receptor annotation tool Genome2OR(http://gffzz188fe103f8f1460asp0xq5bncf6u96b5x.ffgz.tsg.suse.edu.cn/To Hanwei/Genome2OR.git),and used it to process 1,695 sequenced chordate genomes in the NCBI Assembly database as of January,2021.In total,765,248 olfactory receptor genes were annotated,with 404,426 functional genes and 360,822 pseudogenes,which represents a four-fold increase in the number of annotated olfactory receptors.Based on the annotation data,we built a database called Chordata Olfactory Receptor Database(CORD,http://gffzz608a37c433994787sp0xq5bncf6u96b5x.ffgz.tsg.suse.edu.cn)for archiving,analysing and disseminating the data.Beyond the primary data,we offer derivative information,including pictures of species,cross references to public databases,structural models,sequence similarity networks and sequence profiles in the CORD.Furthermore,we did brief analyses on these receptors,including building a huge protein sequence similarity network covering all receptors in the database,and clustering them into 20 communities,classifying the 20 communities into three categories based on their presences/absences in ray-finned fish and/or lobe-finned fish.We infer that olfactory receptors should have unique activation and desensitization mechanisms by analysing their sequences and structural models.We believe the CORD can benefit the researchers and the general public who are interested in olfaction. | Wei Han Yiran Wu Liting Zeng Suwen Zhao | 2022 | Science China(Life Sciences)2022,65,12: | 1 |
| 8 | Phase-field simulation tending to depict practical electrodeposition process in lithium-based batteries显示文摘Lithium dendrite growth due to uneven electrodeposition usually leads to the potential hazard of internal short circuit and shorter lifetime of lithium-based batteries. Extensive efforts have been devoted to explore the effects of single or two factors on dendrite growth, involving the diffusion coefficient, exchange current density, electrolyte concentration, temperature, and applied voltage. However, these factors interrelate during battery operation, signifying that a understanding of how they jointly influence the electrodeposition is of paramount importance for the effective suppression of dendrites. Here, we incorporate the dependent relationships among key factors into the phase-field model to capture their synergistic effects on electrodeposition. All the simulations are implemented in our self-written MATLAB code under a unified modeling framework. Following this, five groups of experimentally common dendrite patterns are reproduced and the corresponding electrodeposition driving forces are identified. Unexpectedly, we find that with the decrease of the ratio of exchange current density(or applied voltage) to diffusion coefficient, the electrodeposition morphology changes from needle-like dendrites to columnar dendrites and to uniform deposition. | 10 | Shen Qi Wan attenuates renal interstitial fibrosis through upregulating AQP1显示文摘Renal interstitial fibrosis(RIF)is the crucial pathway in chronic kidney disease(CKD)leading to the end-stage renal failure.However,the underlying mechanism of Shen Qi Wan(SQW)on RIF is not fully understood.In the current study,we investigated the role of Aquaporin 1(AQP1)in SQW on tubular epithelial-to-mesenchymal transition(EMT).A RIF mouse model induced by adenine and a TGF-β1-stimulated HK-2 cell model were etablished to explore the involvement of AQP 1 in the protective effect of SQW on EMT in vitro and in vivo.Subsequently,the molecular mechanism of SQW on EMT was explored in HK-2 cells with AQP1 knockdown.The results indicated that SQW alleviated kidney injury and renal collagen deposition in the kidneys of mice induced by adenine,increased the protein expression of E-cadherin and AQP1 expression,and decreased the expression of vimentin andα-smooth muscle actin(α-SMA).Similarly,treatmement with SQW-containing serum significantly halted EMT process in TGF-β1 stimulated HK-2 cells.The expression of snail and slug was significantly upregulated in HK-2 cells after knockdown of AQP1.AQP1 knockdown also increased the mRNA expression of vimentin andα-SMA,and decreased the expression of E-cadherin.The protein expression of vimentin increased,while the expression of E-cadherin and CK-18 significantly decreased after AQP1 knockdown in HK-2 cells.These results revealed that AQP1 knockdown promoted EMT.Furthermore,AQP1 knockdown abolished the protective effect of SQW-containing serum on EMT in HK-2 cells.In sum,SQW attentuates EMT process in RIF through upregulation of the expression of AQP1. | LIN Yiyou WEI Jiale ZHANG Yehui HUANG Junhao WANG Sichen LUO Qihan YU Hongxia JI Liting ZHOU Xiaojie LI Changyu | 2023 | Chinese Journal of Natural Medicines2023,21,5: | 1 |

| 13 | Effect of Roucongrong(Herba Cistanches Deserticolae) decoction on the substantia nigra through Wnt/β-catenin signaling pathway in rats with Parkinson's disease induced by 6-hydroxydopamine hydrochloride显示文摘OBJECTIVE:To investigate the effect of Roucongrong(Herba Cistanches Deserticolae)decoction on the substantia nigra in rats with Parkinson’s disease(PD)induced by 6-hydroxydopamine hydrochloride(6-OHDA).To further determine whether the Wnt/β-catenin signaling pathway is involved in the action.METHODS:A rat model of PD was established by intracranial injection of 6-OHDA.Subsequently,three concentrations of Roucongrong(Herba Cistanches Deserticolae)decoction were prepared and administered to rats by gavage therapy for 14 d.Behavioral changes were measured in PD rats.In vivo tyrosine hydroxylase(TH)levels in the substantia nigra were examined by immunohistochemistry.Additionally,gene and protein expression levels of members of the Wnt/β-catenin signaling pathway were examined by Western blotting and polymerase chain reaction.Lastly,a Wnt/β-catenin inhibitor was used to investigate the mechanism of action in 1-methyl-4-phenylpyridinium(MPP+)-treated MES23.5 cells in vitro.RESULTS:Roucongrong(Herba Cistanches Deserticolae)decoction improved performance in the stride and gait adjustment tests in PD rats.It also increased TH in the substantia nigra and altered the expression of genes and proteins in the Wnt/β-catenin signaling pathway.Wnt/β-catenin inhibitor reduced the effect of Roucongrong(Herba Cistanches Deserticolae)decoction in MPP+-treated MES23.5 cells.CONCLUSION:Roucongrong(Herba Cistanches Deserticolae)decoction may promote neuronal survival in PD in vivo and in vitro by increasing TH content in the substantia nigra and by activating the Wnt/β-catenin signaling pathway. | XU Qian QIN Wei WU Fangzhen LIN Yao HONG Liting CHEN Dan HU Xuefeng CAI Jing | 2021 | Journal of Traditional Chinese Medicine2021,41,5: | 1 |
