1NLRP3-dependent pyroptosis is required for HIV-1 gp120- induced neuropathology显示文摘The human immunodeficiency virus-1(HIV-1)envelope protein gp120 is the major contributor to the pathogenesis of HIVassociated neurocognitive disorder(HAND).Neuroinflammation plays a pivotal role in gp120-induced neuropathology,but how gp120 triggers neuroinflammatory processes and subsequent neuronal death remains unknown.Here,we provide evidence that NLRP3 is required for gp120-induced neuroinflammation and neuropathy.Our results showed that gp120-induced NLRP3-dependent pyroptosis and IL-1βproduction in microglia.Inhibition of microglial NLRP3 inflammasome activation alleviated gp120-mediated neuroinflammatory factor release and neuronal injury.Importantly,we showed that chronic administration of MCC950,a novel selective NLRP3 inhibitor,to gp120 transgenic mice not only attenuated neuroinflammation and neuronal death but also promoted neuronal regeneration and restored the impaired neurocognitive function.In conclusion,our data revealed that the NLRP3 inflammasome is important for gp120-induced neuroinflammation and neuropathology and suggest that NLRP3 is a potential novel target for the treatment of HAND.Xiaolong He Weijun Yang Zhijie Zeng Yi Wei Jie Gao Bao Zhang Li Li Liqun Liu Yu Wan Qing Zeng Zelong Gong Liting Liu Hanyun Zhang Yubin Li Shaojie Yang Tongtong Hu Lixian Wu Eliezer Masliah Shenghe Huang Hong Cao 2020Cellular & Molecular Immunology2020,17,3:11
2Famitinib in combination with concurrent chemoradiotherapy in patients with locoregionally advanced nasopharyngeal carcinoma: a phase 1, open-label, dose-escalation Study显示文摘Background:Famitinib is a tyrosine kinase inhibitor against multiple targets,including vascular endothelial growth factor receptor 2/3,platelet-derived growth factor receptor,and stem cell factor receptor(c-kit).Previous studies have demonstrated anti-tumour activities of famitinib against a wide variety of advanced-stage solid cancers.We aimed to determine the safety and efficacy of famitinib with concurrent chemoradiotherapy(CCRT)in patients with locoregionally advanced nasopharyngeal carcinoma(NPC).We also evaluated the feasibility of contrast-enhanced ultrasound(D-CEUS)as a predictor of early tumour response to famitinib and to correlate functional parameters with clinical efficacy.Methods:The trial was conducted in subjects with stage III or IVa-b NPC using a 3+3 design of escalating fami-tinib doses.Briefly,subjects received 2 weeks of famitinib monotherapy followed by 7 weeks of famitinib plus CCRT.D-CEUS of the neck lymph nodes was performed at day 0,8 and 15 after famitinib was administered before starting concurrent chemoradiotherapy.End points included safety,tolerability and anti-tumour activity.Results:Twenty patients were enrolled(six each for 12.5,16.5 and 20 mg and two for 25 mg).Two patients in the 25 mg cohort developed dose-limiting toxicities,including grade 4 thrombocytopenia and grade 3 hypertension.The most common grade 3/4 adverse events were leukopenia,neutropenia and radiation mucositis.D-CEUS tests showed that more than 60%of patients achieved a perfusion parameter response after 2 weeks taking famitinib alone,and the parameter response was associated with disease improvement.In the famitinib monotherapy stage,three patients(15%)showed partial responses.The complete response rate was 65%at the completion of treatment and 95%3 months after the treatment ended.After a median follow-up of 44 months,the 3-year progression-free survival(PFS)and distant metastasis-free survival were 70%and 75%,respectively.Subjects with a decrease of perfusion parameter response,such as peak intensity decreased at least 30%after 1 week of famitinib treatment,had higher 3-year PFS(90.9%vs.44.4%,95%CI 73.7%-100%vs.11.9%-76.9%,P<0.001)than those with an increase or a reduction of less than 30%.Conclusions:The recommended famitinib dose for phase II trial is 20 mg with CCRT for patients with local advanced NPC.D-CEUS is a reliable and early measure of efficacy for famitinib therapies.Further investigation is required to confirm the effects of famitinib plus chemoradiotherapy.Qiuyan Chen Linquan Tang Na Liu Feng Han Ling Guo Shanshan Guo Jianwei Wang Huai Liu Yanfang Ye Lu Zhang Liting Liu Pan Wang Yingqin Li Qingmei He Xiaoqun Yang Qingnan Tang Yang Li YuJing Liang XueSong Sun Chuanmiao Xie Yunxian Mo Ying Guo Rui Sun Haoyuan Mo Kajia Cao Xiang Guo Musheng Zeng Haiqiang Mai Jun Ma 2018Cancer Communications2018,38,1:6
3AFAP1-AS1: a rising star among oncogenic long non-coding RNAs显示文摘Long non-coding RNAs(lncRNAs)have become a hotspot in biomedical research.This interest reflects their extensive involvement in the regulation of the expression of other genes,and their influence on the occurrence and development of a variety of human diseases.Actin filament associated protein 1-Antisense RNA 1(AFAP1-AS1)is a recently discovered oncogenic lncRNA.It is highly expressed in a variety of solid tumors,and regulates the expression of downstream genes and signaling pathways through adsorption and competing microRNAs,or by the direct binding to other proteins.Ultimately,AFAP1-AS1 promotes proliferation,chemotherapy resistance,and resistance to apoptosis,maintains stemness,and enhances invasion and migration of tumor cells.This paper summarizes the research concerning AFAP1-AS1 in malignant tumors,including the clinical application prospects of AFAP1-AS1 as a potential molecular marker and therapeutic target of malignant tumors.We also discuss the limitations in the knowledge of AFAP1-AS1 and directions of further research.AFAP1-AS1 is expected to provide an example for studies of other lncRNA molecules.Fang Xiong Kunjie Zhu Su Deng Hongbin Huang Liting Yang Zhaojian Gong Lei Shi Yi He Yanyan Tang Qianjin Liao Jianjun Yu Xiaoling Li Yong Li Guiyuan Li Zhaoyang Zeng Wei Xiong Shanshan Zhang Can Guo 2021Science China(Life Sciences)2021,64,10:4
4Building the Chordata Olfactory Receptor Database using more than 400,000 receptors annotated by Genome2OR显示文摘Olfactory receptors are poorly annotated for most genome-sequenced chordates.To address this deficiency,we developed a nhmmer-based olfactory receptor annotation tool Genome2OR(http://gffzz188fe103f8f1460asuo0bvq59vqwk6vv6.ffgz.tsg.suse.edu.cn/To Hanwei/Genome2OR.git),and used it to process 1,695 sequenced chordate genomes in the NCBI Assembly database as of January,2021.In total,765,248 olfactory receptor genes were annotated,with 404,426 functional genes and 360,822 pseudogenes,which represents a four-fold increase in the number of annotated olfactory receptors.Based on the annotation data,we built a database called Chordata Olfactory Receptor Database(CORD,http://gffzz608a37c433994787suo0bvq59vqwk6vv6.ffgz.tsg.suse.edu.cn)for archiving,analysing and disseminating the data.Beyond the primary data,we offer derivative information,including pictures of species,cross references to public databases,structural models,sequence similarity networks and sequence profiles in the CORD.Furthermore,we did brief analyses on these receptors,including building a huge protein sequence similarity network covering all receptors in the database,and clustering them into 20 communities,classifying the 20 communities into three categories based on their presences/absences in ray-finned fish and/or lobe-finned fish.We infer that olfactory receptors should have unique activation and desensitization mechanisms by analysing their sequences and structural models.We believe the CORD can benefit the researchers and the general public who are interested in olfaction.Wei Han Yiran Wu Liting Zeng Suwen Zhao 2022Science China(Life Sciences)2022,65,12:1

6Facile synthesis of CoFe_2O_4 nanoparticles anchored on graphene sheets for enhanced performance of lithium ion battery显示文摘Recently, metal oxides as high capacity anode materials had been investigated for lithium ion batteries.However, the fast capacity fading upon cycling leaded poor durability, which hindered their application as higher energy density of lithium ion battery. In this paper, a nanostructured nanocomposite with graphene supported CoFe_2O_4 nanoparticles(NPs) was prepared via simple hydrothermal reaction. The uniform CoFe_2O_4 NPs were anchored on graphene sheets, which brought a good performance on cyclability. Combined with the optimization of graphene content, the anode delivered a better capacity retention of 944 m A h g^(-1)over 50 cycles at current density of 100 m A g^(-1)and the good reversible capacity as 990 m A h g^(-1)when the rate returned from 5 A g^(-1)to 0.1 A g^(-1)after 60 cycles. 7Burden among caregivers for children with asthma:A mixed-method study in Guangzhou,China显示文摘Objective:Research suggests that caregivers of children with asthma experience a substantial amount of stress,though no comprehensive study examines this problem.Here we both quantitatively and qualitatively examined the burden on caregivers of children with asthma.Methods:Surveys were administered to 138 caregivers of asthmatic children by convenience sampling.The Zarit Caregiver Burden Interview(ZBI)was used to assess the degree of caregiver burden.In addition,13 qualitative semi-structured interviews were carried out via purposive sampling and were used to explore the perception of caregivers.SPSS and content analysis were used to analyze quantitative and qualitative data,respectively.Results:We found that caregiver burden(mean=31.56±14.19)ranged from 24%with no or mild burden,52%with mild to moderate burden,22%with moderate to severe burden and 2%with very severe burden.Caregiver burden was also assessed as 5 different dimensions.Selfcriticism ranked most burdensome(2.09±1.05),followed by sacrifice(1.57±0.94),embarrassment/anger(1.30±0.80),dependency(1.21±0.77),and lastly,loss of control(1.20±0.84).The highest individually scored item on the ZBI was“fear of the future of the relative”(mean=3.04).Interviews of caregiver burden were summarized into three main themes,namely:1)life is too chaotic;2)negative emotions;and 3)gaps in themedical support system.Conclusions:These findings expand our understanding about the burden caregivers of children with asthma face,and may help to propose targeted strategies to help caregivers adapt to their roles,and maintain and promote the health of themselves and their families.Miaolan Guo Guozhen Gao Jinlan Guo Litao Wen Liting Zeng 2015International Journal of Nursing Sciences2015,2,4:1

10Long non-coding RNA AFAP1-AS1 accelerates lung cancer cells migration and invasion by interacting with SNIP1 to upregulate c-Myc显示文摘Actin filament associated protein 1 antisense RNA 1(named AFAP1-AS1)is a long non-coding RNA and overexpressed in many cancers.This study aimed to identify the role and mechanism of AFAP1-AS1 in lung cancer.The AFAP1-AS1 expression was firstly assessed in 187 paraffin-embedded lung cancer and 36 normal lung epithelial tissues by in situ hybridization.The migration and invasion abilities of AFAP1-AS1 were investigated in lung cancer cells.To uncover the molecular mechanism about AFAP1-AS1 function in lung cancer,we screened proteins that interact with AFAP1-AS1 by RNA pull down and the mass spectrometry analyses.AFAP1-AS1 was highly expressed in lung cancer clinical tissues and its expression was positively correlated with lung cancer patients'poor prognosis.In vivo experiments confirmed that AFAP1-AS1 could promote lung cancer metastasis.AFAP1-AS1 promoted lung cancer cells migration and invasion through interacting with Smad nuclear interacting protein 1(named SNIP1),which inhibited ubiquitination and degradation of c-Myc protein.Upregulation of c-Myc molecule in turn promoted the expression of ZEB1,ZEB2,and SNAIL gene,which ultimately enhanced epithelial to mesenchymal transition(EMT)and lung cancer metastasis.Understanding the molecular mechanism by which AFAP1-AS1 promotes lung cancer's migration and invasion may provide novel therapeutic targets for lung cancer patients'early diagnosis and therapy.Yu Zhong Liting Yang Fang Xiong Yi He Yanyan Tang Lei Shi Songqing Fan Zheng Li Shanshan Zhang Zhaojian Gong Can Guo Qianjin Liao Yujuan Zhou Ming Zhou Bo Xiang Xiaoling Li Yong Li Zhaoyang Zeng Guiyuan Li Wei Xiong 2021Signal Transduction and Targeted Therapy2021,6,7:1
11Constructing metal-free heterophotocatalyst using two-dimensional carbon nitride sheets and violet phosphorene for highly efficient visible-light photocatalysis显示文摘Two-dimensional(2D)carbon nitride sheets(CNs)with nanoscale thickness have great potential in solar energy conversion owing to their intrinsic bandgap.However,their photocatalytic efficiency falls far below the practical requirements because the generated electron-hole pairs rapidly recombine.We construct a metal-free 2D p–n junction heterophotocatalyst from CNs and violet phosphorene(VP)using a simple electrostatic self-assembly method.The experimental results and density functional theory calculations show that the built-in electric field of the p–n junction promotes photogenerated carrier transport at the heterojunction interface,thereby promoting the separation of photogenerated electron-hole pairs.Under visible-light illumination,the visible-light photocatalytic hydrogen and CO production rates of the CNs/VP heterojunction were increased by 5.85 and 1.51 times,respectively,when compared to that of CNs.This study provides new insights into the design of metal-free heterogeneous photocatalysts with high catalytic activity。Yong Wang Chengxin Zeng Liting Wu Yalin Dong Yu Zhang Dingyi Yang Wen Hu Jian Hao Hongzhe Pan Rusen Yang 2023Journal of Materials Science & Technology2023,,15:0
12Antioxidant,flavor profile and quality of wheat dough bread incorporated with kiwifruit fermented byβ-glucosidase producing lactic acid bacteria strains显示文摘This study aimed to investigate the effect of incorporation of kiwifruit substrate fermented by 3 highβ-glucosidase producing lactic acid bacteria(LAB)of Lactobacillus harbinensis(M12,M24)and Pediococcus pentosaceus(J28)on physicochemical and rheo-fermentation properties during proofing of wheat dough.Quality,sensory evaluation,antioxidant content and activity,and flavor profiles of bread was evaluated.Results revealed that LAB strains adequately adapted(J28>M12>M24)and produced enzymes during substrate fermentation.In dough,incorporation of fermented substrate increased acidity,soluble dietary fiber,β-glucosidase andα-amylase activity and gas retention during proofing,in a strain dependent manner(J28>M12>M24).The subsequent breads exhibited higher specific volume and had softer crumbs.Furthermore,total flavonoid,total phenolic,antioxidant activity,flavor content and intensity increased in breads incorporated with fermented substrate.Overall sensory acceptance was in order,bread containing substrates fermented by J28(KFB-J28)>M12(KFB-M12)>M24(KFB-M24)>wheat bread(WB)>bread containing unfermented substrate(KFB).Changes observed were attributed to biotransformation byβ-glucosidase which increasingly degraded dietary fibers into soluble dietary fiber,increased acidity,released glycosylated aroma compounds and phenolic compounds in substrate.When incorporated in dough,fermented kiwifruit substrates stabilized gluten network,increased yeast metabolism and gas retention during proofing.Subsequently,KFB-J28,KFB-M12,and KFB-M24 had higher antioxidant content and activity,higher flavor content and intensity,better quality and were more accepted compared to WB and KFB.This suggested the potential role played byβ-glucosidases through LAB fermentation on functionally enriching and adding value to kiwifruit substrate as a novel functional ingredient in bakery industry.Liting Liang Jacob Ojobi Omedi Weining Huang Jianxian Zheng Yongqing Zeng Jing Huang Binle Zhang Liyuan Zhou Ning Li Tiecheng Gao Ruijun Guo 2022Food Bioscience2022,46,2:0
