1Advantages of white LED lamps and new detector technology in photometry显示文摘Light emitting diode(LED)lighting is becoming more and more popular,as incandescent lamps are being phased out globally.LEDs have several advantages over incandescent lamps,including energy efficiency,robustness,long lifetime,and good temporal stability.The three latter features make LEDs attractive candidates as new photometric standards.Because the spectra of white LEDs are limited to the visible wavelength range,a novel method for the realization of photometric units based on the predictable quantum efficient detector(PQED)can be utilized.The method eliminates the need of photometric filters that are traditionally used in photometry,and instead relies on carrying out the photometric weighting numerically based on the measured relative spectrum of the source.The PQED-based realization simplifies the traceability chain of photometric measurements significantly as compared with the traditional filter-based method.The measured illuminance values of a white LED deviate by only 0.03%when determined by the new and the traditional methods.The new PQED method has significantly lower expanded uncertainty of 0.26%(k=52)as compared with that of the traditional filter-based method of 0.42%(k=52).Furthermore,when filtered photometers that measure LED lighting are calibrated using LED lamps as calibration sources instead of incandescent lamps,a significant decrease in the uncertainty related to the spectral mismatch correction can be obtained.The maximum spectral mismatch errors of LED measurements decreased on average by a factor of 3 when switching from an incandescent lamp to an LED calibration source.Tomi Pulli Timo Donsberg Tuomas Poikonen Farshid Manoocheri Petri Karha Erkki Ikonen 2015Light(Science & Applications)2015,4,1:8
2基于磁珠和时间分辨荧光免疫分析的微囊藻毒素LR单链抗体筛选与鉴定显示文摘【目的】从鼠源合成噬菌体抗体库中,快速分离获得微囊藻毒素LR单链抗体。【方法】采用磁珠筛选和负筛选方法,设计两轮微囊藻毒素LR单链抗体的筛选方案。用噬菌体产出投入比和多克隆噬菌体免疫分析,对每轮筛选后微囊藻毒素LR特异性噬菌体的富集效果进行鉴定。再将第2轮筛选获得的次级库提取单链抗体基因,转入可溶性表达载体;诱导表达后,采用时间分辨荧光免疫法,对单个噬菌体克隆鉴定并测序。【结果】两轮筛选的噬菌体产出投入比分别为4.8×10-8和2.88×10-6。第2轮筛选后的次级库,经多克隆噬菌体免疫分析得到的荧光信噪比,比原始抗体库提高了22.8倍。最终鉴定得到5株不同的阳性噬菌体克隆,其中最佳的克隆对微囊藻毒素LR的检测灵敏度(IC10)为13 ng.mL-1,抑制中浓度(IC50)为435 ng.mL-1,线性范围在31—5 952 ng.mL-1。【结论】本研究筛选到的单链抗体,有潜力应用于微囊藻毒素LR的免疫学检测或免疫亲和柱的制备,同时也探索了一种高效的微囊藻毒素LR单链抗体的库筛选、鉴定方法。刘媛 Huovinen Tuomas 刘贤金 梁颖 张存政 谢雅晶 贺江 王耘 张霄 2012中国农业科学2012,45,2:8
3时间分辨荧光免疫分析方法检测烟草中菌核净残留显示文摘建立了简便、灵敏地检测烟草中菌核净残留的时间分辨荧光免疫分析方法(TRFIA),及配套的无净化的快速前处理技术。采用镧系元素螯合物Eu-N1标记亲和纯化后的羊抗兔IgG示踪抗体。采用间接竞争TRFIA法建立了菌核净标准抑制曲线,方法的检出限I10为2.0μg/L;抑制终浓度I50为32.00μg/L;检测线性范围为4~128μg/L。考察了丙酮提取后33%,10%和1%的烟草基质对菌核净-TRFIA分析方法的影响,表明在1%烟叶基质条件下,建立的菌核净的抑制曲线的线性范围与标准抑制曲线趋于平行,确定烟草样品的前处理稀释倍数为100倍,计算得到基质影响因子(Im)为17.1。对烟叶样品中添加1,7和24mg/L的菌核净标样,连续3d的添加回收实验表明,方法的回收率为73%~128%,相对标准标准偏差(RSD)在4.3%~13.2%之间;烟草中菌核净的实际最低检出限为1mg/L。本方法可望用于烟草中菌核净残留的快速筛选检测。刘媛 刘蓉蓉 刘贤金 方敦煌 宋春满 Huovinen Tuomas Vehniinen Markus Lvgren Timo 2012分析化学2012,40,7:6
4Cellulose structure and lignin distribution in normal and compression wood of the Maidenhair tree(Ginkgo biloba L.)显示文摘We studied in detail the mean micro fibril angle and the width of cellulose crystals from the pith to the bark of a 15-year-old Maidenhair tree(Ginkgo biloba L.). The orientation of cellulose micro fibrils with respect to the cell axis and the width and length of cellulose crystallites were determined using Xray diffraction. Raman microscopy was used to compare the lignin distribution in the cell wall of normal/opposite and compression wood, which was found near the pith. Ginkgo biloba showed a relatively large mean micro fibril angle,varying between 19° and 39° in the S2 layer, and the average width of cellulose crystallites was 3.1–3.2 nm. Mild compression wood without any intercellular spaces or helical cavities was observed near the pith. Slit-like bordered pit openings and a heavily lignified S2 L layer con firmed the presence of compression wood. Ginkgo biloba showed typical features present in the juvenile wood of conifers. The micro fibril angle remained large over the 14 annual rings. The entire stem disc,with a diameter of 18 cm, was considered to consist of juvenile wood. 5Synchrotron radiation investigations of the Sr_2MgSi_2O_7:Eu^(2+),R^(3+) persistent luminescence materials显示文摘The electronic and defect energy level structure of polycrystalline Sr2MgSi2O7:Eu2+,R3+ persistent luminescence materials were studied with thermoluminescence and different synchrotron radiation spectroscopies (UV-VUV emission and excitation, X-ray absorption near-edge spectroscopy (XANES) and extended X-ray absorption fine structure (EXAFS)). Special attention was paid on the effect of the R3+ co-dopants on the persistent luminescence properties of the materials. Theoretical calculations using the density functional theory (DFT) were carried out simultaneously with the experimental work. The experimental band gap energy (Eg) value of ca. 7.1 eV agreed very well with the DFT value of 6.7 eV. The variation of the Eg value was attempted to relate with the trap structure as well as with the different properties of the R3+ co-dopants. The trap level energy distribution depended strongly on the R3+ co-dopant except for the shallowest trap energy above the room temperature remaining practically the same, however. 7Converting lateral scanning into axial focusing to speed up three-dimensional microscopy显示文摘In optical microscopy,the slow axial scanning rate of the objective or the sample has traditionally limited the speed of volumetric imaging.Recently,by conjugating either a movable mirror to the image plane in a remote-focusing geometry or an electrically tuneable lens(ETL)to the back focal plane,rapid axial scanning has been achieved.However,mechanical actuation of a mirror limits the axial scanning rate(usually only 10–100 Hz for piezoelectric or voice coil-based actuators),while ETLs introduce spherical and higher-order aberrations that prevent high-resolution imaging.In an effort to overcome these limitations,we introduce a novel optical design that transforms a lateral-scan motion into a spherical aberration-free axial scan that can be used for high-resolution imaging.Using a galvanometric mirror,we scan a laser beam laterally in a remote-focusing arm,which is then back-reflected from different heights of a mirror in the image space.We characterize the optical performance of this remote-focusing technique and use it to accelerate axially swept light-sheet microscopy by an order of magnitude,allowing the quantification of rapid vesicular dynamics in three dimensions.We also demonstrate resonant remote focusing at 12 kHz with a two-photon raster-scanning microscope,which allows rapid imaging of brain tissues and zebrafish cardiac dynamics with diffraction-limited resolution.Tonmoy Chakraborty Bingying Chen Stephan Daetwyler Bo-Jui Chang Oliver Vanderpoorten Etai Sapoznik Clemens F.Kaminski Tuomas P.J.Knowles Kevin M.Dean Reto Fiolka 2020Light(Science & Applications)2020,9,1:4
8Additive QTLs on three chromosomes control flowering time in woodland strawberry(Fragaria vesca L.)显示文摘Flowering time is an important trait that affects survival,reproduction and yield in both wild and cultivated plants.Therefore,many studies have focused on the identification of flowering time quantitative trait locus(QTLs)in different crops,and molecular control of this trait has been extensively investigated in model species.Here we report the mapping of QTLs for flowering time and vegetative traits in a large woodland strawberry mapping population that was phenotyped both under field conditions and in a greenhouse after flower induction in the field.The greenhouse experiment revealed additive QTLs in three linkage groups(LG),two on both LG4 and LG7,and one on LG6 that explain about half of the flowering time variance in the population.Three of the QTLs were newly identified in this study,and one co-localized with the previously characterized FvTFL1 gene.An additional strong QTL corresponding to previously mapped PFRU was detected in both field and greenhouse experiments indicating that gene(s)in this locus can control the timing of flowering in different environments in addition to the duration of flowering and axillary bud differentiation to runners and branch crowns.Several putative flowering time genes were identified in these QTL regions that await functional validation.Our results indicate that a few major QTLs may control flowering time and axillary bud differentiation in strawberries.We suggest that the identification of causal genes in the diploid strawberry may enable fine tuning of flowering time and vegetative growth in the closely related octoploid cultivated strawberry.Samia Samad Takeshi Kurokura Elli Koskela Tuomas Toivainen Vipul Patel Katriina Mouhu Daniel James Sargent Timo Hytönen 2017Horticulture Research2017,4,1:3

13Dead standing pine trees in a boreal forest landscape in the Kalevala National Park,northern Fennoscandia: amount,population characteristics and spatial pattern显示文摘Background: After their death, Scots pine trees can remain standing for decades and sometimes up to 200 years,forming long-lasting and ecologically important structures in boreal forest landscapes. Standing dead pines decay very slowly and with time develop into ‘kelo' trees, which are characterized by hard wood with silvery-colored appearance. These kelo trees represent an ecologically important, long lasting and visually striking element of the structure of natural pine-dominated forests in boreal Fennoscandia that is nowadays virtually absent from managed forest landscapes.Methods: We examined and mapped the amount, structural features, site characteristics and spatial distribution of dead standing pine trees over a ten hectare area in an unmanaged boreal forest landscape in the Kalevala National Park in Russian Viena Karelia.Results: The mean basal area of dead standing pine trees in the forested part of the landscape was 1.7 m^2?ha^(-1) and the estimated volume 12.7 m^3?ha^(-1). From the total number of standing dead pine trees 65% were kelo trees, with a basal area of 1.1 m^2?ha^(-1) and volume of 8.0 m^3?ha^(-1), the remainder consisting of standing dead pines along the continuum between a recently dead tree and a kelo tree. Overall, standing dead pines were distributed throughout the study area, but there was a tendency towards spatial clustering up to <100 m distances. Standing dead pines were most commonly situated on flat ground or in the mid slope in the local topography.In addition, standing dead pines contributed to substrate diversity also by commonly having charred wood and broken tops. Based on the presence of dead pine snags in different stage of transition from a recently dead pine to a kelo with silvery surface, it seems evident that the process of kelo recruitment was continuously in action in the studied landscape.Conclusions: Kelo trees are an omnipresent feature in natural pine-dominated forest landscapes with important contribution to forest structural and substrate diversity. 16Scalable integration of nano-,and microfluidics with hybrid two-photon lithography显示文摘Nanofluidic devices have great potential for applications in areas ranging from renewable energy to human health.A crucial requirement for the successful operation of nanofluidic devices is the ability to interface them in a scalable manner with the outside world.Here,we demonstrate a hybrid two photon nanolithography approach interfaced with conventional mask whole-wafer UV-photolithography to generate master wafers for the fabrication of integrated micro and nanofluidic devices.Using this approach we demonstrate the fabrication of molds from SU-8 photoresist with nanofluidic features down to 230 nm lateral width and channel heights from micron to sub-100 nm.Scanning electron microscopy and atomic force microscopy were used to characterize the printing capabilities of the system and show the integration of nanofluidic channels into an existing microfluidic chip design.The functionality of the devices was demonstrated through super-resolution microscopy,allowing the observation of features below the diffraction limit of light produced using our approach.Single molecule localization of diffusing dye molecules verified the successful imprint of nanochannels and the spatial confinement of molecules to 200 nm across the nanochannel molded from the master wafer.This approach integrates readily with current microfluidic fabrication methods and allows the combination of microfluidic devices with locally two-photon-written nano-sized functionalities,enabling rapid nanofluidic device fabrication and enhancement of existing microfluidic device architectures with nanofluidic features.Oliver Vanderpoorten Quentin Peter Pavan K.Challa Ulrich F.Keyser Jeremy Baumberg Clemens F.Kaminski Tuomas P.J.Knowles 2019Microsystems & Nanoengineering2019,5,1:2
