3Glutamate and aspartate alleviate testicular/epididymal oxidative stress by supporting antioxidant enzymes and immune defense systems in boars显示文摘Several potential oxidative agents have damaging effects on mammalian reproductive systems.This study was conducted to investigate the effects of glutamate(Glu)and aspartate(Asp)supplementation on antioxidant enzymes and immune defense systems in the outer scrotum of boars injected with H2O2.A total of 24 healthy boars were randomly divided into 4 treatment groups:control(basal diet,saline-treated),H2O2(basal diet,H2O2-challenged outer scrotum(1 m L kg^–1 BW)),Glu(basal diet+2%Glu,H2O2-challenged),and Asp(basal diet+2%Asp,H2O2-challenged).Our results showed that both Glu and Asp supplementation improved testicular morphology and decreased the genital index in the H2O2-treated boars.Glu and Asp administration increased the antioxidant enzyme activities and affected the testicular inflammatory cytokine secretion but had no effect on sex hormone levels.Furthermore,the m RNA expression of CAT,Cu Zn SOD,and GPx4 was altered in the testes and epididymis of boars treated with Asp and Glu.Glu and Asp supplementation also modulated the expression of TGF-β1,IL-10,TNF-α,IL-6 and IL-1βin the testis and epididymis.These results indicate that dietary Glu and Asp supplementation might enhance antioxidant capacity and regulate the secretion and expression of inflammatory cytokines to protect the testes and epididymis of boars against oxidative stress.Wenjie Tang Jian Wu Shunshun Jin Liuqin He Qinlu Lin Feijun Luo Xingguo He Yanzhong Feng Binsheng He Pingping Bing Tiejun Li Yulong Yin 2020Science China(Life Sciences)2020,63,1:9
4Odontoblast β-catenin signaling regulates fenestration of mouse Hertwig's epithelial root sheath显示文摘The interaction between Hertwig's epithelial root sheath(HERS) and the adjacent mesenchyme is vitally important in mouse tooth root development. We previously generated odontoblast-specific Ctnnb1(encoding β-catenin) deletion mice, and demonstrated that odontoblast β-catenin signaling regulates odontoblast proliferation and differentiation. However, the role of odontoblast β-catenin signaling in regulation of HERS behavior has not been fully investigated. Here, using the same odontoblast-specific Ctnnb1 deletion mice, we found that ablation of β-catenin signaling in odontoblasts led to aberrant HERS formation. Mechanistically, odontoblast-specific Ctnnb1 deletion resulted in elevated bone morphogenetic protein 7(Bmp7) expression and reduced expression of noggin and follistatin, both of which encode extracellular inhibitors of BMPs. Furthermore, the levels of phosphorylated Smad1/5/8 were increased in HERS cells. In vitro tissue culture confirmed that BMP7 treatment disrupted the HERS structure. 5Advances in direct production of value-added chemicals via syngas conversion显示文摘Syngas conversion to fuels and chemicals is one of the most challenging subjects in the field of C1 chemistry. It is considered as an attractive alternative non-petroleum-based production route. The direct synthesis of olefins and alcohols as high value-added chemicals from syngas has drawn particular attention due to its process simplicity, low energy consumption and clean utilization of carbon resource, which conforms to the principles of green carbon science. This review describes the recent advances for the direct production of lower olefins and higher alcohols via syngas conversion. Recent progress in the development of new catalyst systems for enhanced catalytic performance is highlighted. 6Effect of Ermiao Fang with Xixin(Herba Asari Mandshurici) on bone marrow stem cell directional homing to a focal zone in an osteoarthritis rat model显示文摘OBJECTIVE: To investigate the effects of Ermiao Fang(EM) with medical guide Xixin(Herba Asari Mandshurici)(HAM) on bone marrow stem cell migration to a focal zone in osteoarthritis(OA) rats.METHODS: OA rats were induced by arthrectomy and assigned to sham-operated, model, EM, or EM plus HAM groups.All rats were injected with recombinant human granulocyte colony-stimulating factor 30μg·kg-1·d-1for7 days and treated with EMor EM plus HAM at 1.6 or 1.9 g·kg-1·d-1 for 3 or 6 weeks, respectively. Chondrocyte apoptosis and cartilage matrix components were tested by transferase-mediated deoxyuridine triphosphate-biotin nick end labeling assay and special staining. Levels of interleukin-1 beta(IL-1β) tumor necrosis factor alpha(TNF-α) nitric oxide(NO), and inducible nitric oxide synthase(iNOS) in serum were detected by enzyme-linked immunosorbent assay or radioimmunoassay. Matrix metalloproteinases(MMPs)-13,tissue inhibitors of metalloproteinases(TIMPs)-1,Bromodeoxyuridine(BrdU), cluster of differentiation 34(CD34), and stromal cell-derived factor 1(SDF-1) were measured by immunohistochemical assay.RESULTS:The EM and EM plus HAM groups had significantly less cartilage damage and synovium inflammation the model group. Moreover, the EM and EM plus HAM groups had less chondrocyte apoptosis and more proteoglycan and collagen content than the model group.The EM and EMplus HAM groups had obviously higher MMPs-13 and TIMPs-1 expression in the cartilage than the model group. Moreover, the two formula groups had less release of IL-1β, TNF-α, NO, and iNOS than model group. Importantly, the expressions of BrdU, CD34,and SDF-1 in cartilage were significantly higher in the EM and EM plus HAM-Medtreated rats than model group. 12Genome Assembly of Alfalfa Cultivar Zhongmu-4 and Identification of SNPs Associated with Agronomic Traits显示文摘Alfalfa(Medicago sativa L.)is the most important legume forage crop worldwide with high nutritional value and yield.For a long time,the breeding of alfalfa was hampered by lacking reliable information on the autotetraploid genome and molecular markers linked to important agronomic traits.We herein reported the de novo assembly of the allele-aware chromosome-level genome of Zhongmu-4,a cultivar widely cultivated in China,and a comprehensive database of genomic variations based on resequencing of 220 germplasms.Approximate 2.74 Gb contigs(N50 of 2.06 Mb),accounting for 88.39%of the estimated genome,were assembled,and 2.56 Gb contigs were anchored to 32 pseudo-chromosomes.A total of 34,922 allelic genes were identified from the allele-aware genome.We observed the expansion of gene families,especially those related to the nitrogen metabolism,and the increase of repetitive elements including transposable elements,which probably resulted in the increase of Zhongmu-4 genome compared with Medicago truncatula.Population structure analysis revealed that the accessions from Asia and South America had relatively lower genetic diversity than those from Europe,suggesting that geography may influence alfalfa genetic divergence during local adaption.Genome-wide association studies identified 101 single nucleotide polymorphisms(SNPs)associated with 27 agronomic traits.Two candidate genes were predicted to be correlated with fall dormancy and salt response.We believe that the alleleaware chromosome-level genome sequence of Zhongmu-4 combined with the resequencing data of the diverse alfalfa germplasms will facilitate genetic research and genomics-assisted breeding in variety improvement of alfalfa.Ruicai Long Fan Zhang Zhiwu Zhang Mingna Li Lin Chen Xue Wang Wenwen Liu Tiejun Zhang Long-Xi Yu Fei He Xueqian Jiang Xijiang Yang Changfu Yang Zhen Wang Junmei Kang Qingchuan Yang 2022Genomics, Proteomics & Bioinformatics2022,20,1:1
13Survival after radical cystectomy for bladder cancer:Multicenter comparison between minimally invasive and open approaches显示文摘Objective:To investigate oncological outcomes in patients with bladder cancer who underwent minimally invasive radical cystectomy(MIRC)or open radical cystectomy(ORC).Methods:We identified patients with bladder cancer who underwent radical cystectomy(RC)in 13 centers of the Chinese Bladder Cancer Consortium(CBCC).Perioperative outcomes were compared between MIRC and ORC.The influence of surgical approaches on overall survival(OS)and cancer-specific survival(CSS)in the entire study group and subgroups classified according to pathologic stage or lymph node(LN)status was assessed with the log-rank test.Multivariable Cox proportional hazard models were used to evaluate the association among OS,CSS and risk factors of interest.Results:Of 2098 patients who underwent RC,1243 patients underwent MIRC(1087 laparoscopic RC and 156 robotic-assisted RC,respectively),while 855 patients underwent ORC.No significant differences were noted in positive surgical margin rate and 90-day postoperative mortality rate.MIRC was associated with less estimated blood loss,more LN yield,higher rate of neobladder diversion,longer operative time,and longer length of hospital stay.There was no significant difference in OS and CSS according to surgical approaches(pZ0.653,and 0.816,respectively).Subgroup analysis revealed that OS and CSS were not significantly different regardless of the status of extravesical involvement or LN involvement.Multivariable Cox regression analyses showed that the surgical approach was not a significant predictor of OS and CSS.Conclusions:Our study showed that MIRC was comparable to conventional ORC in terms of OS and CSS.Weibin Xie Junming Bi Qiang Wei Ping Han Dongkui Song Lei Shi Dingwei Ye Yijun Shen Xin Gou Weiyang He Shaogang Wang Zheng Liu Jinhai Fan Kaijie Wu Zhiwen Chen Xiaozhou Zhou Chuize Kong Yang Liu Chunxiao Liu Abai Xu Baiye Jin Guanghou Fu Wei Xue Haige Chen Tiejun Pan Zhong Tu Tianxin Lin Jian Huang 2020Asian Journal of Urology2020,7,3:1

15Preparation of BiFeO_3 solution and its movement effects controlled by electrical field显示文摘This paper demonstrated our discovery of the movement effect of BiFeO_3 nanoparticles in solution by strong electrical polarization. First, BiFeO_3 nanoparticles were prepared by ball milling and characterized by SEM. Then, the preparation of BiFeO_3 solution was finished. BiFeO_3 nanoparticles are found to move in solution due to electrical field. The movement velocity of BiFeO_3 nanoparticles, which is about 10 μm·s^-1 , is influenced and controlled by the applied electrical field. The concentration of BiFeO_3 solution and the nanoparticle sizes also have effects on its movement properties. 17Micheliolide exerts effects in myeloproliferative neoplasms through inhibiting STAT3/5 phosphorylation via covalent binding to STAT3/5 proteins显示文摘Ruxolitinib is a cornerstone of management for some subsets of myeloproliferative neoplasms(MPNs);however,a considerable number of patients respond suboptimally.Here,we evaluated the efficacy of micheliolide(MCL),a natural guaianolide sesquiterpene lactone,alone or in combination with ruxolitinib in samples from patients with MPNs,JAK2V617F-mutated MPN cell lines,and a Jak2V617F knock-in mouse model.MCL effectively suppressed colony formation of hematopoietic progenitors in samples from patients with MPNs and inhibited cell growth and survival of MPN cell lines in vitro.Co-treatment with MCL and ruxolitinib resulted in greater inhibitory effects compared with treatment with ruxolitinib alone.Moreover,dimethylaminomicheliolide(DMAMCL),an orally available derivative of MCL,significantly increased the efficacy of ruxolitinib in reducing splenomegaly and cytokine production in Jak2V617F knock-in mice without evident effects on normal hematopoiesis.Importantly,MCL could target the Jak2V617F clone and reduce mutant allele burden in vivo.Mechanistically,MCL can form a stable covalent bond with cysteine residues of STAT3/5 to suppress their phosphorylation,thus inhibiting JAK/STAT signaling.Overall,these findings suggest that MCL is a promising drug in combination with ruxolitinib in the setting of suboptimal response to ruxolitinib.Huijun Huang Jinqin Liu Lin Yang Yiru Yan Meng Chen Bing Li Zefeng Xu Tiejun Qin Shiqiang Qu Liang Wang Gang Huang Yue Chen Zhijian Xiao 2023Blood Science2023,5,4:0
18Partially Linear Single-Index Model in the Presence of Measurement Error显示文摘The partially linear single-index model(PLSIM) is a flexible and powerful model for analyzing the relationship between the response and the multivariate covariates. This paper considers the PLSIM with measurement error possibly in all the variables. The authors propose a new efficient estimation procedure based on the local linear smoothing and the simulation-extrapolation method,and further establish the asymptotic normality of the proposed estimators for both the index parameter and nonparametric link function. 19An improved area-based approach for estimating plot-level tree DBH from airborne LiDAR data显示文摘The diameter at breast height(DBH) of trees and stands is not only a widely used plant functional trait in ecology and biodiversity but also one of the most fundamental measurements in managing forests. However, systematically measuring the DBH of individual trees over large areas using conventional ground-based approaches is labour-intensive and costly. Here, we present an improved area-based approach to estimate plot-level tree DBH from airborne Li DAR data using the relationship between tree height and DBH, which is widely available for most forest types and many individual tree species. We first determined optimal functional forms for modelling heightDBH relationships using field-measured tree height and DBH. Then we estimated plot-level mean DBH by inverting the height-DBH relationships using the tree height predicted by Li DAR. Finally, we compared the predictive performance of our approach with a classical area-based method of DBH. The results showed that our approach significantly improved the prediction accuracy of tree DBH(R^(2)=0.85–0.90, rRMSE=9.57%–11.26%)compared to the classical area-based approach(R^(2)=0.80–0.83, rRMSE=11.98%–14.97%). 20Study on the fluid flow rule of five-cylinder plunger pump hydraulic end显示文摘The change of velocity and pressure of flow field in suction and discharge chamber of five-cylinder plunger pump obtained by CFD under maximum and minimum stroke,the results show that:when the stroke is 79 rpm,the internal of pump head will produce pressure holding,and the internal organization of pump head body will be in a fatigue state of high pressure for a long time.When the rotate angle is small,the peak velocity at the gap of valve disc and valve seat reaches 9.60m/s,which is the orifice jet phenomenon.When the stroke is 299 rpm,the overall velocity curve is relatively stable,but the velocity of fluid flow through the pump head body is larger,and the maximum velocity reaches 18.72 m/s at the bottom corner of valve,it will produce the circumfluence and vortex discharge chamber at the same time,which will cause the increase of vibration of pump head body.So it should use proper punching gear in order to conducive to overall working life of pump.Zhanghua Lian Yang Liu Tiejun Lin Li Li Zhongqing Lei Chuanjun Han 2018Petroleum2018,4,4:0
