Application of next-generation sequencing technology to precision medicine in cancer: joint consensus of the Tumor Biomarker Committee of the Chinese Society of Clinical Oncology显示文摘Next-generation sequencing(NGS) technology is capable of sequencing millions or billions of DNA molecules simultaneously. Therefore, it represents a promising tool for the analysis of molecular targets for the initial diagnosis of disease, monitoring of disease progression, and identifying the mechanism of drug resistance. On behalf of the Tumor Biomarker Committee of the Chinese Society of Clinical Oncology(CSCO) and the China Actionable Genome Consortium(CAGC), the present expert group hereby proposes advisory guidelines on clinical applications of NGS technology for the analysis of cancer driver genes for precision cancer therapy. This group comprises an assembly of laboratory cancer geneticists, clinical oncologists, bioinformaticians, pathologists, and other professionals. After multiple rounds of discussions and revisions, the expert group has reached a preliminary consensus on the need of NGS in clinical diagnosis, its regulation, and compliance standards in clinical sample collection. Moreover, it has prepared NGS criteria, the sequencing standard operation procedure(SOP), data analysis, report, and NGS platform certification and validation. | Xuchao Zhang Zhiyong Liang Shengyue Wang Shun Lu Yong Song Ying Cheng Jianming Ying Weiping Liu Yingyong Hou Yangqiu Li Yi Liu Jun Hou Xiufeng Liu Jianyong Shao Yanhong Tai Zheng Wang Li Fu Hui Li Xiaojun Zhou Hua Bai Mengzhao Wang You Lu Jinji Yang Wenzhao Zhong Qing Zhou Xuening Yang Jie Wang Cheng Huang Xiaoqing Liu Xiaoyan Zhou Shirong Zhang Hongxia Tian Yu Chen Ruibao Ren Ning Liao Chunyan Wu Zhongzheng Zhu Hongming Pan Yanhong Gu Liwei Wang Yunpeng Liu Suzhan Zhang Tianshu Liu Gong Chen Zhimin Shao Binghe Xu Qingyuan Zhang Ruihua Xu Lin Shen Yilong Wu | 2019 | Cancer Biology & Medicine2019,16,1: | 14
The precise regulation of different COR genes by individual CBF transcription factors in Arabidopsis thaliana显示文摘The transcription factors CBF1/2/3 are reported to play a dominant role in the cold responsive network of Arabidopsis by directly regulating the expression levels of cold responsive(COR) genes. In this study,we obtained CRISPR/Cas9-mediated loss-of-function mutants of cbf1$3.Over 3,000 COR genes identified by RNA-seq analysis showed a slight but significant change in their expression levels in the mutants compared to the wildtype plants after being treated at 4 °C for 12 h.The C-repeat(CRT) motif(50-CCGAC-30) was enriched in promoters of genes that were up-regulated by CBF2 and CBF3 but not in promoters of genes up-regulated by CBF1.These data suggest that CBF2 and CBF3 play a more important role in directing the cold response by regulating different sets of downstream COR genes.More than 2/3 of COR genes were co-regulated by two or three CBFs and were involved mainly in cellular signal transduction and metabolic processes;less than 1/3 of the genes were regulated by one CBF,and those genes up-regulated were enriched in cold-related abiotic stress responses. Our results indicate that CBFs play an important role in the trade-off between cold tolerance and plant growth through the precise regulation of COR genes in the complicated transcriptional network. | Yihao Shi Jiaying Huang Tianshu Sun Xuefei Wang Chenqi Zhu Yuxi Ai Hongya Gu | 2017 | Journal of Integrative Plant Biology2017,59,2: | 7
Transient acquisition of cross-species infectivity during the evolution of SARS-CoV-2显示文摘Since the coronavirus disease 2019(COVID-19)pandemic began, its causative agent, severe acute respiratory syndrome coronavirus 2(SARS-CoV-2),has spread worldwide.During the global transmission of SARS-CoV-2,mutations in the viral genome have gradually accumulated and have led to the emergence of variants. These emerging variants;including 501Y.V1,501Y.V2 and 501Y.V3(also called the alpha, beta and gamma variants, respectively),rapidly became the predominant epidemic strains and subsequently spread worldwide. | Qi Chen Xing-Yao Huang Meng-Xu Sun Rui-Ting Li Hongjing Gu Ying Tian Rong-Rong Zhang Dan Luo Chao Zhou Yifei Zhang Tianshu Cao Na-Na Zhang Yong-Qiang Deng Xiao-Feng Li Cheng-Feng Qin | 2021 | National Science Review2021,8,11: | 3
Mode transition induced by back-breakdown of the gliding arc and its influence factors显示文摘Gliding arc is a mostly used non-equilibrium plasma generation method whose characteristic is affected by the backbreakdown phenomenon.Mode transition induced by back-breakdown of the gliding arc is studied in this work and effects of gas flow rate, applied voltage, electrode parameters on mode transition were studied.Experimental results show that there are two typical modes during the development of the gliding arc.Mode A comprises periodicitical development of the arc from the minimum gap breakdown to the longest extinguishment, while the arc in mode B continues to produce a back-breakdown at the ends of the electrodes. As the flow rate descends, the gliding arc gradually changes from mode A to B.It is the decrease of the arc velocity caused by lower flow rate leads to the occurrence of back-breakdown,which generates mode B.Smaller electrode opening angle, shorter length and wider minimum gap reduce the gliding speed,so that arc is more likely to enter mode B.As the applied voltage is increased, enhancing of the electric field strength on the breakdown path of the back-breakdown and thickening of the arc's diameter allow the gliding arc to enter mode B at a higher arc speed. | Ruobing Zhang Haochen Huang Tianshu Yang | 2020 | High Voltage2020,5,3: | 3
Genomic profiling and the impact of MUC19 mutation in hepatoid adenocarcinoma of the stomach显示文摘Dear Editor,Hepatoid adenocarcinoma(HAC)is a rare pathological subtype of extrahepatic tumor, featured by hepatoid differentiation andα-fetoprotein(AFP)-production[1,2].Hepatoid adenocarcinoma of the stomach(HAS),accounting for 0.3%to 1.0%of all gastric cancers(GCs),has attracted increasing attention due to its high degree of malignancy[3].Compared with classic GC,HAS showed a higher rate of vascular invasion, lymph node metastasis, and liver metastasis, with only 9.0%survival rate at 5 years[4].Currently,there is no effective treatment for HAS,and little is known about its pathogenesis.Herein,we investigated the molecular features of HAS and identified potential therapeutic targets for HAS. | Mengxuan Zhu Erbao Chen Shan Yu Chen Xu Yiyi Yu Xin Cao Wei Li Pengfei Zhang Yan Wang Baofeng Lian Shuirong Zhang Yueting Qu Lujia Huang Weiwei Shi Yuehong Cui Li Qian Tianshu Liu | 2022 | Cancer Communications2022,42,10: | 3

Toripalimab in advanced biliary tract cancer显示文摘Gemcitabine combined with platinum/fluorouracil drugs is the standard firstline treatment for advanced biliary tract cancers(BTCs).We explored the safety and efficacy of toripalimab plus gemcitabine and S-1(GS)as the first-line treatment for advanced BTCs.At a one-sided significance level of 0.025,a total of 50 patients could provide 80%power to show the efficacy at targeted progression-free survival(PFS)rate at 6 months of 70%versus 40%for the combined treatment. This single-arm,phase II study enrolled 50 patients with advanced BTCs who previously received no systemic treatment. The regimen was as follows:toripalimab(240 mg,i.v.,d1),gemcitabine(1,000 mg/m2,i.v.,d1 and d8),and S-1(40–60 mg bid p.o.,d1–14,Q21d).The primary endpoint was progression-free survival. The secondary endpoints included overall survival(OS),objective response rate(ORR),duration of response(DOR),and safety. The associations between response with PDL1 expression, tumor mutational burden(TMB),and genetic variations were explored.Patients were enrolled from January 2019 to August 2020,with a median follow-up time of 24.0 months(IQR:4.3–31.0 months). | Wei Li Yueqi Wang Yiyi Yu Qian Li Yan Wang Chenlu Zhang Xiaojing Xu Xi Guo Yu Dong Yuehong Cui Qing Hao Lujia Huang Houbao Liu Tianshu Liu | 2022 | The Innovation2022,3,4: | 1
GCN-LSTM spatiotemporal-network-based method for post-disturbance frequency prediction of power systems显示文摘Owing to the expansion of the grid interconnection scale, the spatiotemporal distribution characteristics of the frequency response of power systems after the occurrence of disturbances have become increasingly important. These characteristics can provide effective support in coordinated security control. However, traditional model-based frequencyprediction methods cannot satisfactorily meet the requirements of online applications owing to the long calculation time and accurate power-system models. Therefore, this study presents a rolling frequency-prediction model based on a graph convolutional network(GCN)and a long short-term memory(LSTM)spatiotemporal network and named as STGCN-LSTM.In the proposed method, the measurement data from phasor measurement units after the occurrence of disturbances are used to construct the spatiotemporal input. An improved GCN embedded with topology information is used to extract the spatial features, while the LSTM network is used to extract the temporal features. The spatiotemporal-network-regression model is further trained, and asynchronous-frequency-sequence prediction is realized by utilizing the rolling update of measurement information. The proposed spatiotemporal-network-based prediction model can achieve accurate frequency prediction by considering the spatiotemporal distribution characteristics of the frequency response. The noise immunity and robustness of the proposed method are verified on the IEEE 39-bus and IEEE 118-bus systems. | Dengyi Huang Hao Liu Tianshu Bi Qixun Yang | 2022 | Global Energy Interconnection2022,5,1: | 1

Effects of cavity-dispersion noncoaxiality on the generation of ultrabroadband femtosecond pulses显示文摘The effects of cavity-dispersion noncoaxiality (CDN) on the generation of ultrabroadband femtosecond pulses in KLM Ti:sapphire laser were investigated theoretically and experimentally. It was predicted that when the laser sub-cavity works near the coaxial operation point, the limitation of CDN on the bandwidth broadening is minimum, which is favorable for ultrabroadband pulse generation. On the basis of this prediction, femtosecond pulses with bandwidth of 650 to 1000 nm were directly generated from a home built KLM Ti:sapphire laser. To our knowledge, they are the broadest bandwidth pulses produced from KLM Ti:sapphire laser with similar oscillator configuration and gain crystal length of 3 mm. | JIAO ZhongXing LEI Liang HUANG ZhiLing WEN dinHui LAI Tianshu LIN WeiZhu | 2008 | Chinese Science Bulletin2008,53,5: | 1

Large deformation analysis of a cantilever beam made of axially functionally graded material by homotopy analysis method显示文摘Large deformation of a cantilever axially functionally graded (AFG) beam subject to a tip load is analytically studied using the homotopy analysis method (HAM). It is assumed that its Young’s modulus varies along the longitudinal direction according to a power law. Taking the solution of the corresponding homogeneous beam as the initial guess and obtaining a convergence region by adjusting an auxiliary parameter, the analytical expressions for large deformation of the AFG beam are provided. Results obtained by the HAM are compared with those obtained by the finite element method and those in the previous works to verify its validity. Good agreement is observed. A detailed parametric study is carried out. The results show that the axial material variation can greatly change the deformed configuration, which provides an approach to control and manage the deformation of beams. By tailoring the axial material distribution, a desired deformed configuration can be obtained for a specific load. The analytical solution presented herein can be a helpful tool for this procedure. | Xin LIN Yixin HUANG Yang ZHAO Tianshu WANG | 2019 | Applied Mathematics and Mechanics(English Edition)2019,40,10: | 1
Reservoir micro structure of Da'anzhai Member of Jurassic and its petroleum significance in Central Sichuan Basin, SW China显示文摘Based on the qualitative study of microscopic reservoir features using core analysis, cast and fluorescence thin sections inspection, scanning electron microscope(SEM)and field emission scanning electron microscope(FESEM)and quantitative examination of pore size and geometry using mercury injection,nano-CT and nitrogen adsorption, reservoir rock of Da’anzhai Member were divided into 9 types, while storage spaces were divided into 4 types and 14 sub-types.The study shows that sparry coquina is the most promising reservoir type.Pores that smaller than 1μm in diameter contribute 91.27%of storage space volume. Most of them exhibit slot-like geometry with good connectivity.By building up storage space models,it was revealed that micron scale storage spaces mainly composed of fractures and nanometer scale pores and fractures form multi-scale dual porosity system.Low resource abundance, small single well controlled reserve, and low production are related to the nano-scale pore space in Da’anzhai Memer, whereas the dual-porosity system composed of pores and fractures makes for long-term oil yield.Due to the existence of abundant slot-like pore space and fractures, economic tight oil production was achieved without stimulations. | PANG Zhenglian TAO Shizhen ZHANG Qin YANG Jiajing ZHANG Tianshu YANG Xiaoping FAN Jianwei HUANG Dong WEI Tengqiang | 2018 | Petroleum Exploration and Development2018,45,1: | 0

Periocular Biometric Recognition for Masked Faces显示文摘Since the outbreak of Coronavirus Disease 2019(COVID-19),people are recommended to wear facial masks to limit the spread of the virus.Under the circumstances, traditional face recognition technologies cannot achieve satisfactory results. In this paper,we propose a face recognition algorithm that combines the traditional features and deep features of masked faces. For traditional features,we extract Local Binary Pattern(LBP),Scale-Invariant Feature Transform(SIFT)and Histogram of Oriented Gradient(HOG)features from the periocular region, and use the Support Vector Machines(SVM)classifier to perform personal identification. We also propose an improved Convolutional Neural Network(CNN)model Angular Visual Geometry Group Network(A-VGG)to learn deep features. Then we use the decision-level fusion to combine the four features.Comprehensive experiments were carried out on databases of real masked faces and simulated masked faces, including frontal and side faces taken at different angles.Images with motion blur were also tested to evaluate the robustness of the algorithm.Besides, the experiment of matching a masked face with the corresponding full face is accomplished. The experimental results show that the proposed algorithm has state-of-the-art performance in masked face recognition, and the periocular region has rich biological features and high discrimination. | HUANG Qiaoyue TANG Chaoying ZHANG Tianshu | 2023 | Wuhan University Journal of Natural Sciences2023,28,2: | 0
Study of Impact Resistance Based on Porcupine Quills Bionic Thin-walled Structure显示文摘Using an electron microscope to observe the microstructure of a porcupine quills cross-section and a bionic method,a new bionic structure was proposed. The performance of the structure in terms of energy absorption, maximum impact force withstood, and impact force efficiency was evaluated using Ansys finite element simulation software to simulate the structure's impact. To examine the impact of ribs on the structural performance of the bionic porcupine quills,a control structure was developed.According to the results of the finite element simulation, the presence of ribs in the Bionic porcupine quills structure can transfer stress uniformly to the overall structure and share stress for some of the rupture-prone regions.Ribs reduce stress concentration in specific areas and increase the impact force efficiency of the structure. The SEA and IFE values of bionic porcupine quills were 30.01 kJ/kg and 84.22%,respectively.The structure is then optimized for parameter design in order to find the optimal structure by response surface in order to improve the structure's SEA and decrease its MIF.In order to evaluate the precision of the response surface, the optimal structure predicted is validated using finite element simulation. | Tianshu Huang Zhengyu Mao Lijun Chang Xingyuan Huang Zhihua Cai | 2023 | Journal of Bionic Engineering2023,20,5: | 0
