| 1 | Efficacy and safety of a novel anti-HER2 therapeutic antibody RC48 in patients with HER2-overexpressing,locally advanced or metastatic gastric or gastroesophageal junction cancer:a single-arm phase II study显示文摘Background:Current treatment options for human epidermal growth factor receptor 2(HER2)-overexpressing gastric cancer at third-line have shown limited clinical benefit.Further,there is no specific treatment for HER2 immunohistochemistry(IHC)2+and fluorescence in-situ hybridization-negative patients.Here,we report the efficacy and safety of a novel anti-HER2 antibody RC48 for patients with HER2-overexpressing,advanced gastric or gastroesophageal junction cancer.Methods:Patients with HER2-overexpressing(IHC 2+or 3+),locally advanced or metastatic gastric or gastroesophageal junction cancer who were under at least second-line therapy were eligible and received RC482.5 mg/kg alone every 2 weeks.The primary endpoint was the objective response rate(ORR)assessed by an independent review committee.Secondary endpoints included progressionfree survival(PFS),overall survival(OS),duration of response,time to progression,disease control rate,and safety.Results:Of 179 patients screened,125 were eligible and received RC48 treatment.The ORR was 24.8%(95%confidence interval[CI]:17.5%-33.3%).The median PFS and OS were 4.1 months(95%CI:3.7-4.9 months)and 7.9 months(95%CI:6.7-9.9 months),respectively.The most frequently reported adverse events were decreased white blood cell count(53.6%),asthenia(53.6%),hair loss(53.6%),decreased neutrophil count(52.0%),anemia(49.6%),and increased aspartate aminotransferase level(43.2%).Serious adverse events(SAEs)occurred in 45(36.0%)patients,and RC48-related SAEs were mainly decreased neutrophil count(3.2%).Seven patients had adverse events that led to death were not RC48-related.Conclusions:RC48 showed promising activity with manageable safety,suggesting potential application in patients with HER2-overexpressing,advanced gastric or gastroesophageal junction cancer who have previously received at least two lines of chemotherapy. | Zhi Peng Tianshu Liu Jia Wei Airong Wang Yifu He Liuzhong Yang Xizhi Zhang Nanfeng Fan Suxia Luo Zhen Li Kangsheng Gu Jianwei Lu Jianming Xu Qingxia Fan Ruihua Xu Liangming Zhang Enxiao Li Yuping Sun Guohua Yu Chunmei Bai Yong Liu Jiangzheng Zeng Jieer Ying Xinjun Liang Nong Xu Chao Gao Yongqian Shu Dong Ma Guanghai Dai Shengmian Li Ting Deng Yuehong Cui Jianmin Fang Yi Ba Lin Shen | 2021 | Cancer Communications2021,41,11: | 29 |
| 2 | Effect of hydrophilic silica nanoparticles on hydrate formation: Insight from the experimental study显示文摘Invasion of drilling fluid into natural gas hydrate deposits during drilling might damage the reservoir,induce hydrate dissociation and then cause wellbore instability and distortion of the data from well logging. Adding nanoparticles into drilling fluid is an effective method in reducing the invasion of drilling fluid and enhancing borehole stability. However, the addition of nanoparticles might also introduce hydrate formation risk in borehole because they can act as the 'seeds' for hydrate nucleation. This paper presents an experimental study of the effect of hydrophilic silica nanoparticle on gas hydrate formation in a dynamic methane/liquid-water system. In the experiment, the ultrapure water with and without1.0 wt%–6.0 wt% concentrations of silica nanoparticles, grain sizes of 20 and 50 nm, were pressurized by methane gas under varied conditions of temperature and pressure. The induction time, the gas consumption, and the average rate of gas consumption in the system were measured and compared to those in ultrapure water. The results show that a concentration of 4.0 wt% hydrophilic SiO_2 particles with a grain size of 50 nm has a relatively strong inhibition effect on hydrate formation when the initial experimental condition is 5.0 °C and 5.0 MPa. Compared to ultrapure water, the hydrophilic nano-SiO_2 fluid increases the induction time for hydrate formation by 194% and decreases the amount and average rate of hydrate formation by 10% and 17%, respectively. This inhibition effect may be attributed to the hydrophilicity,amount and aggregation of silica nanoparticle according to the results of water activity and zeta potential measurements. | 3 | The precise regulation of different COR genes by individual CBF transcription factors in Arabidopsis thaliana显示文摘The transcription factors CBF1/2/3 are reported to play a dominant role in the cold responsive network of Arabidopsis by directly regulating the expression levels of cold responsive(COR) genes.In this study,we obtained CRISPR/Cas9-mediated loss-of-function mutants of cbf1$3.Over 3,000 COR genes identified by RNA-seq analysis showed a slight but significant change in their expression levels in the mutants compared to the wildtype plants after being treated at 4 °C for 12 h.The C-repeat(CRT) motif(50-CCGAC-30) was enriched in promoters of genes that were up-regulated by CBF2 and CBF3 but not in promoters of genes up-regulated by CBF1.These data suggest that CBF2 and CBF3 play a more important role in directing the cold response by regulating different sets of downstream COR genes.More than 2/3 of COR genes were co-regulated by two or three CBFs and were involved mainly in cellular signal transduction and metabolic processes;less than 1/3 of the genes were regulated by one CBF,and those genes up-regulated were enriched in cold-related abiotic stress responses.Our results indicate that CBFs play an important role in the trade-off between cold tolerance and plant growth through the precise regulation of COR genes in the complicated transcriptional network. | Yihao Shi Jiaying Huang Tianshu Sun Xuefei Wang Chenqi Zhu Yuxi Ai Hongya Gu | 2017 | Journal of Integrative Plant Biology2017,59,2: | 7 |
| 4 | Neural Mechanisms of Mental Fatigue Revisited: New Insights from the Brain Connectome显示文摘Maintaining sustained attention during a prolonged cognitive task often comes at a cost: high levels of mental fatigue. Heuristically, mental fatigue refers to a feeling of tiredness or exhaustion, and a disengagement from the task at hand;it manifests as impaired cognitive and behavioral performance. In order to effectively reduce the undesirable yet preventable consequences of mental fatigue in many real-world workspaces, a better understanding of the underlying neural mechanisms is needed, and continuous efforts have been devoted to this topic. In comparison with conventional univariate approaches, which are widely utilized in fatigue studies, convergent evidence has shown that multivariate functional connectivity analysis may lead to richer information about mental fatigue. In fact, mental fatigue is increasingly thought to be related to the deviated reorganization of functional connectivity among brain regions in recent studies. In addition, graph theoretical analysis has shed new light on quantitatively assessing the reorganization of the brain functional networks that are modulated by mental fatigue. This review article begins with a brief introduction to neuroimaging studies on mental fatigue and the brain connectome, followed by a thorough overview of connectome studies on mental fatigue. | 6 | Optimal Operation of Integrated Heat and Electricity Systems:A Tightening McCormick Approach显示文摘Combined heat and electricity operation with variable mass flow rates promotes flexibility,economy,and sustainability through synergies between electric power systems(EPSs)and district heating systems(DHSs).Such combined operation presents a highly nonlinear and nonconvex optimization problem,mainly due to the bilinear terms in the heat flow model—that is,the product of the mass flow rate and the nodal temperature.Existing methods,such as nonlinear optimization,generalized Benders decomposition,and convex relaxation,still present challenges in achieving a satisfactory performance in terms of solution quality and computational efficiency.To resolve this problem,we herein first reformulate the district heating network model through an equivalent transformation and variable substitution.The reformulated model has only one set of nonconvex constraints with reduced bilinear terms,and the remaining constraints are linear.Such a reformulation not only ensures optimality,but also accelerates the solving process.To relax the remaining bilinear constraints,we then apply McCormick envelopes and obtain an objective lower bound of the reformulated model.To improve the quality of the McCormick relaxation,we employ a piecewise McCormick technique that partitions the domain of one of the variables of the bilinear terms into several disjoint regions in order to derive strengthened lower and upper bounds of the partitioned variables.We propose a heuristic tightening method to further constrict the strengthened bounds derived from the piecewise McCormick technique and recover a nearby feasible solution.Case studies show that,compared with the interior point method and the method implemented in a global bilinear solver,the proposed tightening McCormick method quickly solves the heat–electricity operation problem with an acceptable feasibility check and optimality. | Lirong Deng Hongbin Sun Baoju Li Yong Sun Tianshu Yang Xuan Zhang | 2021 | Engineering2021,7,8: | 3 |

Here we prepared solutionized, peak-and over-aged ZK60 alloys with and without microalloying Ca(0.26 wt.%) to explore the effects of nanoscale precipitates on their corrosion behavior in detail via experimental analyses and theoretical calculations. The results suggested the peak-aged ZK60 alloy with Ca addition showed improved corrosion resistance in comparison with the alloys without Ca,owing to the contribution of Ca on the refinement of precipitates and increase in their number density. Although the precipitates and Mg matrix formed micro-galvanic couples leading to dissolution, the fine and dense precipitates could generate “in-situ pinning” effect on the corrosion products, forming a spider-web-like structure and improving the corrosion inhibition ability accordingly. The pinning effect was closely related to the size and number density of precipitates. | 14 | Optimization of Animal-Glue Binders for Casting Applications显示文摘In typical metal foundry applications,sand casting is still the most used technology.The related binder plays a very important role as its performances can directly influence the quality of castings.Among many binders,glues of animal origin have attracted much attention in recent years due to their reduced environmental impact.How-ever,they display some drawbacks such as the tendency to coagulate easily at room temperature and a relatively low strength.In this study,a novel gas-hardening casting binder was prepared using an animal glue and anhy-drous potassium carbonate as a hydrolyzing agent to avoid undesired agglomeration.Moreover,sodium pyropho-sphate and furfuryl alcohol were exploited as modifiers to obtain a binder with a high compressive strength.The best modification conditions,determined by means of an orthogonal design matrix approach,were 4 g of Na2CO3,sodium pyrophosphate,furfuryl alcohol and animal glue with a ratio of 4:12:100,at 85°C and with a duration of 115 min,respectively.The viscosity of the mixture obtained under these optimized conditions was 1250 mPa⋅s.The compressive strength of the binder,hardened by CO_(2) gas,was 4.00 MPa.Its gas evolution at 850°C was 15 ml⋅g-1,and its residual strength after 10 min calculation at 800°C was 0.01 MPa,which is high enough to meet the requirement of core-making in foundry.Moreover,after hydrolysis and further modification,animal glue and modifiers displayed a grafting reaction and an esterification reaction,respectively,which made the adhesive network denser and improved its thermal stability. | Tianshu Wang Guoqing Sun Shaojun Zhang Mingyu Wang Jixin Wang Yunwu Han | 2023 | Fluid Dynamics & Materials Processing2023,19,11: | 0 |

| 19 | Negative correlation between acetyl-CoA acyltransferase 2 and cetuximab resistance in colorectal cancer显示文摘The emergence of anti-EGFR therapy has revolutionized the treatment of colorectal cancer(CRC).However,not all patients respond consistently well.Therefore,it is imperative to conduct further research to identify the molecular mechanisms underlying the development of cetuximab resistance in CRC.In this study,we find that the expressions of many metabolism-related genes are downregulated in cetuximab-resistant CRC cells compared to their sensitive counterparts.Specifically,acetyl-CoA acyltransferase 2(ACAA2),a key enzyme in fatty acid metabolism,is downregulated during the development of cetuximab resistance.Silencing of ACAA2 promotes proliferation and increases cetuximab tolerance in CRC cells,while overexpression of ACAA2 exerts the opposite effect.RTK-Kras signaling might contribute to the downregulation of ACAA2 expression in CRC,and ACAA2 predicts CRC prognosis in patients with Kras mutations.Collectively,our data suggest that modulating ACAA2 expression contributes to secondary cetuximab resistance in Kras wild-type CRC patients.ACAA2 expression is related to Kras mutation and demonstrates a prognostic role in CRC patients with Kras mutation.Thus,ACAA2 is a potential target in CRC with Kras mutation. | Yitao Yuan Xun Sun Mengling Liu Suyao Li Yu Dong Keshu Hu Jiayu Zhang Bei Xu Sining Ma Hesheng Jiang Pengcong Hou Yufu Lin Lu Gan Tianshu Liu | 2023 | Acta Biochimica et Biophysica Sinica2023,55,9: | 0 |
