1The mortality of lung cancer attributable to smoking among adults in China and the United States during 1990–2017显示文摘Background:Statistical data on the burden and relevant risk factors of lung cancer are valuable for policy-making.This study aimed to compare the mortality of lung cancer attributable to smoking stratified by sex and age among adults in China and the United States(US).Methods:We extracted age-standardized mortality rates of lung cancer during 1990-2017 using the comparative risk assessment framework of the 2017 Global Burden of Disease study.We performed an age-period-cohort analysis to estimate time trend of lung cancer mortality attributable to smoking.Results:During 1990-2017,the age-standardized mortality rate of lung cancer was increasing in China but decreasing in the US for both sexes.The mortality attributable to smoking in China showed a generally increasing trend,while a continuous decrease was observed in the US.The age-period-cohort analysis showed a similar trend of age effect among adults between China and the US:the mortality substantially increased from the 30-34 to 80-84 age group and subsequently decreased in the 90-94 age group.However,the period effect rapidly increased in Chinese adults during 1990-2017,while it tended to be stable in the US although it was still slightly increasing in women.The cohort effect generally peaked in the earlier cohort born in 1902-1906 in the two countries.Conclusions:During 1990-2017,the lung cancer mortality attributable to smoking and the period effect are generally increasing in Chinese adults;the mortality attributable to smoking is decreasing in the US adults,but the period effect tends to be stable.The rapid aging and prevalence of smoking may intensify the increasing mortality of lung cancer in China.Xiaoxue Liu Yong Yu Minsheng Wang Sumaira Mubarik Fang Wang Yafeng Wang Runtang Meng Chuanhua Yu 2020Cancer Communications2020,40,11:17
2CRISPR-Cas13a mediated nanosystem for attomolar detection of canine parvovirus type 2显示文摘Canine parvovirus type 2(CPV-2) infection is the most lethal disease of dogs with higher mortality in puppies worldwide.In today’s world,dogs are an integral part of our communities as well as dogs breeding and rearing has become a lucrative business.Therefore,a fast,accurate,portable,and costeffective CPV-2 detection method with the ability for on-site detection is highly desired.In this study,we for the first time proposed a nanosystem for CPV-2 DNA detection with RNA-guided RNA endonuclease Cas13 a,which upon activation results in collateral RNA degradation.We expressed LwCasl3 a in prokaryotic expression system and purified it through nickel column.Activity of Cas13 a was verified by RNA-bound fluorescent group while using a quenched fluorescent probe as signals.Further Cas13 a was combined with Recombinase polymerase amplification(RPA) and T7 transcription to establish molecular detection system termed specific high-sensitivity enzymatic reporter un-locking(SHERLOCK) for sensitive detection of CPV-2 DNA.This nanosystem can detect 100 amol/L CPV-2 DNA within 30 min.The proposed nanosystem exhibited high specificity when tested for CPV-2 and other dog viruses.This CRISPR-Cas13 a mediated sensitive detection approach can be of formidable advantage during CPV-2 outbreaks because it is time-efficient,less laborious and does not involve the use of sophisticated instruments.Haroon Khan Adeel Khan Yufeng Liu Su Wang Sumaira Bibi Hongpan Xu Yuan Liu Samran Durrani Lian Jin Nongyue He Tao Xiong 2019Chinese Chemical Letters2019,30,12:8
3Pathogenesis-related protein genes involved in race-specific allstage resistance and non-race specific high-temperature adultplant resistance to Puccinia striiformis f. sp. tritici in wheat显示文摘Interactions of the stripe rust pathogen(Puccinia striiformis f. sp. tritici) with wheat plants activate a wide range of host responses. Among various genes involved in the plant-pathogen interactions, the expressions of particular pathogenesisrelated(PR) protein genes determine different defense responses. Different types of resistance have been recognized and utilized for developing wheat cultivars for resistance to stripe rust. All-stage resistance can be detected in seedling stage and remains at high levels throughout the plant growth stages. This type of resistance is race-specific and not durable. In contrast, plants with only high-temperature adult-plant(HTAP) resistance are susceptible in seedling stage, but become resistant when plants grow older and the weather becomes warmer. HTAP resistance controlled by a single gene is partial, but usually non-race specific and durable. The objective of this study was to analyze the expression of PR protein genes involved in different types of wheat resistance to stripe rust. The expression levels of 8 PR protein genes(PR1, PR1.2, PR2, PR3, PR4, PR5, PR9 and PR10) were quantitatively evaluated at 0, 1, 2, 7 and 14 days after inoculation in single resistance gene lines of wheat with all-stage resistance genes YrTr1, Yr76, YrSP and YrExp2 and lines carrying HTAP resistance genes Yr52, Yr59, Yr62 and Yr7 B. Races PSTv-4 and PSTv-37 for compatible and incompatible interactions were used in evaluation of PR protein gene expression in wheat lines carrying all-stage resistance genes in the seedlingstage experiment while PSTv-37 was used in the HTAP experiment. Analysis of quantitative real-time polymerase chain reaction(qRT-PCR) revealed that all of the PR protein genes were involved in the different types of resistance controlled by different Yr genes. However, these genes were upregulated at different time points and at different levels during the infection process among the wheat lines with different Yr genes for either all-stage resistance or HTAP resistance. Some of the genes were also induced in compatible interactions, but the levels were almost always higher in the incompatible interaction than in the compatible interaction at the same time point for each Yr gene. These results indicate that bothsalicylic acid and jasmonate signaling pathways are involved in both race-specific all-stage resistance and non-race specific HTAP resistance. 4High HbA1c is associated with higher risk of ischaemic stroke in Pakistani population without diabetes显示文摘Context:The role of glycated haemoglobin(HbA1c)in the prediction of ischaemic stroke in individuals without diabetes is underestimated.Aims:We performed a study to analyse the role of HbA1c in the risk prediction of ischaemic stroke in Pakistani population without diabetes.We further studied the difference between HbA1c values of individuals with diabetes and without diabetes with stroke.Settings and design:Single centre,case–control.Materials and methods:In phase I,a total of 233 patients without diabetes with ischaemic stroke and 245 as controls were enrolled.Association of HbA1c levels,lipid profiles and blood pressure recordings with ischaemic stroke was analysed.In phase II,comparison was done between diabetics and nondiabetics with stroke.Statistical analysis:Comparison of the mean variables was performed with Student’s t-tests.Logistic regression analysis with ischaemic stroke as the dependent variable was performed for phase I.Results:In phase I,the ischaemic stroke group had significantly higher HbA1c levels(5.9±2.9%vs 5.5±1.6%)compared with controls(p<0.05).Triglyceride cholesterol,high-density lipoprotein cholesterol,systolic blood pressure,diastolic blood pressure and HbA1c were the significant determinants of stroke(p<0.05).In phase II,mean HbA1c values were significantly higher in the diabetes group(7.6±2.1 vs 6.1±2.3)(p<0.05)but other parameters were not statistically significantly different(p>0.05).Conclusions:Higher HbA1c indicated a significantly increased risk for ischaemic stroke.An HbA1c value above 5.6%(prediabetic range)predicted future risk of stroke and efforts to maintain glucose level within the normal range(≤5.6%)in individuals with high cardiovascular risk are important.Ali Zohair Nomani Sumaira Nabi Shahzad Ahmed Mansoor Iqbal Haris Majid Rajput Suhail Rao 2016Stroke & Vascular Neurology2016,1,3:2

6Overview of priorities,threats,and challenges to biodiversity conservation in the southern Philippines显示文摘Human activities have severely impacted on many species and ecosystems.Thus,understanding the local biodiversity situation is crucial for implementing effective biodiversity conservation interventions.Mindanao in the southern Philippines is home to various unique species,particularly in its pristine ecosystems.However,the available biodiversity data for many terrestrial vertebrates and key areas remain incomplete.To address this issue,we synthesized published literature related to biodiversity from 2000 to 2022 in Mindanao.Moreover,this analysis used four key terrestrial vertebrates(amphibians,reptiles,birds,and mammals)in Mindanao as research objects.According to our findings,there was a significant and positive correlation between the number of biodiversity studies and species recorded.In terms of species richness,birds were the most recorded group(n=334 spp.),followed by reptiles(n=108 spp.),mammals(n=70 spp.),and amphibians(n=52 spp.).We also found that the number of endemic and threatened species varies geographically and across taxonomic groups.Yet,we discovered a significant disparity in the information available on biodiversity in different provinces of Mindanao.For example,the western provinces of Mindanao have had no record of biodiversity for more than two decades.Furthermore,we found that the changes in tree cover loss were consistent with biodiversity records,but this correlation is only significant for birds.Finally,we highlighted some critical threats and challenges to biodiversity,including deforestation,agricultural expansion,mining,and their impact on biodiversity conservation in Mindanao.Our findings suggested that biodiversity conservation should focus not only on areas with high levels of biodiversity but also on areas lacking biodiversity information.To do this,we call for strengthening collaboration among various institutions and digitizing and centralizing of information related to biodiversity.By gaining a deeper understanding of biodiversity in Mindanao,we can better and sustainably protect critical ecosystems in this region from the increasing threats posed by human activities.Angelo Rellama AGDUMA Francisco Gil GARCIA Ma.Teodora CABASAN Jonald PIMENTEL Renee Jane ELE Meriam RUBIO Sedra MURRAY Bona Abigail HILARIO-HUSAIN Kier Celestial Dela CRUZ Sumaira ABDULLAH Shiela Mae BALASE Krizler Cejuela TANALGO 2023Regional Sustainability2023,4,2:1
7Knowledge,attitudes and practices related to cystic echinococcosis endemicity in Pakistan显示文摘Background:Cystic echinococcosis(CE)is a human and animal health problem in many endemic areas worldwide.It is considered a neglected zoonotic disease caused by the larval form(hydatid cyst)of Echinococcus spp.tapeworm.There are limited studies on echinococcosis in Pakistan.Methods:A cross-sectional survey was conducted to find out recent knowledge,attitudes and practices on the occurrence of cystic echinococcosis in butchers and dog owners in both urban and rural areas of Rawalpindi/Islamabad regions,Pakistan.The quantitative data was collected in the form of questionnaires to investigate the knowledge and awareness of CE among community members and their routine practices that were behind the factors involved in hydatid cyst infection.The practices and infrastructure of abattoirs/butcher shops and their role in transmission of cystic echinococcosis were also evaluated in the present study.Results:The participants involved in the study were dog owners and people who kept animals.A total of 400 people were interviewed and 289 questionnaires were received.The results showed that only 4.1%of people have heard about the disease,and 58.1%were closely associated with dogs.Sixty-three percent of dogs in study area were consuming uncooked organs(e.g.liver,lung,etc.)of slaughtered animals,while 100%of dogs at butcher shops were consuming uncooked organs.Home slaughtering was common in 20.06%.Among butchers,32.3%had heard about zoonoses and 7.61%knew about CE.The statistical analysis showed that there was highly significant difference(P<0.05)among most of the practices that were associated with the prevalence of CE.Conclusions:It was concluded from the present study that,the knowledge and awareness of CE among people of Rawalpindi/Islamabad were low.Because of dogs and poor knowledge of CE among community members and butchers,the transmission of echinococcosis is facilitated.Therefore,there is urgent need to strengthen awareness and health education among people,as well as proper practices related to the CE not only in the study area,but also in other areas of Pakistan.Aisha Khan Kashf Naz Haroon Ahmed Sami Simsek Muhammad Sohail Afzal Waseem Haider Sheikh Saeed Ahmad Sumaira Farrakh Wu Weiping Guan Yayi 2018Infectious Diseases of Poverty2018,7,1:1

14Review of acute ischaemic stroke in Pakistan:progress in management and future perspectives显示文摘Ischaemic stroke is a major cause of neurological morbidity and mortality.The objective of this review article is to summarise facts pertaining to acute ischaemic stroke and its various aspects in a developing country like Pakistan,where resources are limited and the healthcare system is underdeveloped.No large-scale epidemiological studies are available to determine the true incidence of stroke in Pakistan.We reviewed the available literature on stroke from Pakistan and through this article we primarily aim to present the current acute ischaemic stroke management in Pakistan in juxtaposition to that of the developed world.We also intend to highlight areas for future development and improvement in management.The routine practice in Pakistan is that of using stat dose of aspirin in emergency(ER)at large with only a handful of centres offering thrombolytic therapy with recombinant tissue plasminogen activator for acute ischaemic stroke.This too is faced with the problem of long window periods before the patient reaches a proper stroke care centre.The facilities of interventional therapies like arterial thrombolysis and endovascular surgery are non-existent and rehabilitation facilities limited.The opportunities for training physicians in acute stroke are also scarce.Stroke in children is underdiagnosed and that in women not availing facilities at stroke care centres.While basic research has gained pace regarding local demographic data,advanced research and genetic studies are extremely limited.The field of stroke neurology needs to grow at a substantial pace in Pakistan to be at par with the developed world.Ali Zohair Nomani Sumaira Nabi Mazhar Badshah Shahzad Ahmed 2017Stroke & Vascular Neurology2017,2,1:1

20Human papillomavirus, tobacco, and poor oral hygiene can act synergetically, modulate the expression of the nuclear factor kappa B signaling pathway for the development and progression of head and neck cancer in the Pakistani population显示文摘Background: Head and neck cancers (HNCs) are a heterogeneous group of tumors that progress owing to varied enviromental and genetic risk factors. Viral infections are threatening and adept at altering the expression of cellular transcription factors such as nuclear factor kappa B (NF-κB) and deregulation of other cellular proteins like NF kappa B inhibitor alpha (IκBα). The present study was conducted to detect high-risk genotypes of human papillomavirus (HPV) and protein expression of NF-κB signaling pathway in HNC patients with HPV infection.Methods: For HPV detection, genomic DNA from 152 HNC tumors was extracted formalin-fixed paraffin-embedded tissue DNA kit. For genotyping, polymerase chain reaction (PCR) using a general primer, HPV type-specific primers and agarose gel electrophoresis were performed. Immunohistochemistry (IHC) was also performed on 4-μm thick tissue sections using HPV E6 monoclonal antibody. Protein expression analysis of NF-κB signaling pathway including p50, p65, and IκBα was performed using IHC.Results: PCR analysis showed that 24.3% (37/152) of HNC cases were HPV positive. Among HPV positive, 86.5% (32/37) were tobacco users, while among HPV negative, 66.9% (77/115) were tobacco users. A significant association of HPV positivity and tobacco user was observed by univariate analysis [P < 0.01;odds ratio (OR): 0.310, 95% confidence interval (CI): 0.110 to 0.870]. More HPV positive patients were with poor oral hygiene (78.3%) when compared with patients with good oral hygiene (21.6%) [P < 0.03, OR: 2.440, 95% CI: 1.650 to 3.600]. The results of the logistic regression analysis showed that age, tobacco use and oral hygiene are significant predictors (P < 0.02). PCR and IHC staining results confirmed that HPV16 was predominant among HNC cases (64.8%) when compared with HPV18 (35.2%). Expression of NF-κB proteins (p50, p65, and IκBα inhibitor) were also observed in HPV and non-HPV infected HNC tissues. IHC expression of p50, and p65 showed nuclear staining, while IκBα inhibitor showed cytoplasmic staining.
