2Effective simulation of flexible lateral boundaries in two-and three-dimensional DEM simulations显示文摘Discrete element method (DEM) models to simulate laboratory element tests play an important role in advancing our understanding of the mechanics of granular material response, including bonded or cemented, particulate materials. Comparisons of the macro-scale response observed in a real physical test and a 'virtual' DEM-simulated test can calibrate or validate DEM models. The detailed, particle scale information provided in the DEM simulation can then be used to develop our understanding of the material behaviour. It is important to accurately model the physical test boundary conditions in these DEM simulations. This paper specifically considers triaxial tests as these tests are commonly used in soil mechanics. In a triaxial test, the test specimen of granular material is enclosed within a flexible latex membrane that allows the material to deform freely during testing, while maintaining a specified stress condition. Triaxial tests can only be realistically simulated in 3D DEM codes, however analogue, 2D, biaxial DEM simulations are also often considered as it is easier to visualize particle interactions in two dimensions. This paper describes algorithms to simulate the lateral boundary conditions imposed by the latex membrane used in physical triaxial tests in both 2D and 3D DEM simulations. The importance of carefully considering the lateral boundary conditions in DEM simulations is illustrated by considering a 2D biaxial test on a specimen of frictional unbonded disks and a 3D triaxial test on a bonded (cemented) specimen of spheres. 3儿童早期发展综合干预策略对改善贫困农村地区0~35月龄儿童养育照护的效果显示文摘目的 分析儿童早期发展综合干预策略对改善贫困地区0~35月龄儿童早期家庭养育照护的效果.方法 儿童早期发展综合干预项目(IECD)于2014年开始在4个贫困县农村地区试点开展,通过多种途径为项目地区0~35月龄儿童及其看护人提供儿童养育照护多维度综合干预.采用人群干预对照设计,评价干预对看护人健康与心理、儿童营养、早期学习、儿童保护和社会保障等方面的改善效果,分别于2013年和2016年开展基线和中期调查,采用双重差分模型估计干预效果,并控制社会人口学特征等混杂因素.结果 基线调查纳入0~35月龄儿童干预组1 468名、对照组1 485名,中期调查分别纳入1 384名和1 361名.干预组通过2年的干预,看护人抑郁患病率下降9.1%[中期34.8% (479/1 377)比基线43.9% (621/1 414)],对照组下降1.6%[34.3% (464/1 353)比35.9%(509/1 419)],双重差分模型控制因素混杂后,结果显示干预组下降幅度较同期对照组高7.0%(P=0.008);6~23月龄儿童辅食添加频次合格的比例上升10.4%[69.0% (532/771)比58.6%(481/821)],对照组上升幅度2.9%[66.4% (469/706)比63.5%(508/800)],双重差分模型控制混杂因素后,干预组上升幅度比对照组高8.2%(P=0.021);干预组儿童遭受暴力管教的比例下降6.2% [49.1% (478/973)比55.3%(554/1 001)],同期对照组上升4.5%[58.4% (560/959)比53.9%(558/1 036)],双重差分模型控制混杂因素后,两组变化幅度相差11.0%(P=0.001);干预组家庭拥有儿童图书≥3本的比例上升12.7%[42.7% (588/1 378)比30.0% (432/1 440)],同期对照组上升幅度4.2%[25.7% (349/1 357)比21.5%(298/1 388)]双重差分模型控制混杂因素后,干预组比对照组幅度高6.1%(P=0.007).结论 多部门合作儿童早期发展综合干预策略可以有效改善看护人心理状况、促进儿童喂养、支持儿童早期学习,并减少暴力管教等不恰当的养育行为,从而为儿童早期健康发展创造良好养育照护环境.石慧峰 张敬旭 王晓莉 徐媛媛 董胜利 赵春霞 黄小娜 赵颀 陈学锋 周烨 Mayo O'Sullivan Ron Pouwels Robert W.Scherpbier 2018中华儿科杂志2018,56,2:17

5Effect of sample size on the response of DEM samples with a realistic grading显示文摘This paper shows that for DEM simulations of triaxial tests using samples with a grading that is representative of a real soil,the sample size significantly influences the observed material response.Four DEM samples with identical initial states were produced:three cylindrical samples bounded by rigid walls and one bounded by a cubical periodic cell.When subjected to triaxial loading,the samples with rigid boundaries were more dilative,stiffer and reached a higher peak stress ratio than the sample enclosed by periodic boundaries.For the rigid-wall samples,dilatancy increased and stiffness decreased with increasing sample size.The periodic sample was effectively homogeneous.The void ratio increased and the contact density decreased close to the rigid walls.This heterogeneity reduced with increasing sample size.The positions of the critical state lines(CSLs) of the overall response in e-log p' space were sensitive to the sample size,although no difference was observed between their slopes.The critical states of the interior regions of the rigid-wall-bounded samples approached that of the homogeneous periodic sample with increasing sample size.The ultimate strength of the material at the critical state is independent of sample size.Xin Huang Kevin J.Hanley Catherine O'Sullivan Fiona C.Y.Kwok 2014Particuology2014,12,4:12
6Imaging of bone metastasis: An update显示文摘Early detection of skeletal metastasis is critical for accurate staging and optimal treatment. This paper briefly reviews our current understanding of the biological mechanisms through which tumours metastasise to bone and describes the available imaging methods to diagnose bone metastasis and monitor response to treatment. 7Influence of heterogeneity on rock strength and stiffness using discrete element method and parallel bond model显示文摘The particulate discrete element method(DEM) can be employed to capture the response of rock,provided that appropriate bonding models are used to cement the particles to each other.Simulations of laboratory tests are important to establish the extent to which those models can capture realistic rock behaviors.Hitherto the focus in such comparison studies has either been on homogeneous specimens or use of two-dimensional(2D) models.In situ rock formations are often heterogeneous,thus exploring the ability of this type of models to capture heterogeneous material behavior is important to facilitate their use in design analysis.In situ stress states are basically three-dimensional(3D),and therefore it is important to develop 3D models for this purpose.This paper revisits an earlier experimental study on heterogeneous specimens,of which the relative proportions of weaker material(siltstone) and stronger,harder material(sandstone) were varied in a controlled manner.Using a 3D DEM model with the parallel bond model,virtual heterogeneous specimens were created.The overall responses in terms of variations in strength and stiffness with different percentages of weaker material(siltstone) were shown to agree with the experimental observations.There was also a good qualitative agreement in the failure patterns observed in the experiments and the simulations,suggesting that the DEM data enabled analysis of the initiation of localizations and micro fractures in the specimens.Spyridon Liakas Catherine O’Sullivan Charalampos Saroglou 2017Journal of Rock Mechanics and Geotechnical Engineering2017,9,4:7
8Cancer of the uncinate process of the pancreas:surgical anatomy and clinicopathological features显示文摘BACKGROUND:The clinicopathological features of uncinate process pancreatic cancer(UPPC) are poorly described.Furthermore the anatomy of the uncinate process and its division during surgery are central to pancreaticoduodenectomy for UPPC.We set out to describe the embryology and anatomy of the uncinate process and the clinicopathological features of UPPC.DATA SOURCES:All published case series of UPPC were reviewed and included in this review.RESULTS:The true incidence of UPPC is difficult to quantify,with the reported incidence ranging from 2.5% to 10.7% of pancreatic cancer.There are 5 published series of UPPC including 117 patients,72 males and 45 females,aged from 45-53 years to 61-84 years.The median survival was 5 or 5.5 months in 3 of the series,12.1 months in another based only on potentially resectable lesions and 17 months in another based only on resected cases.CONCLUSIONS:The number of reported series of UPPC is limited,with vague symptoms as the predominant presenting features of the disease.The prognosis is poor with synchronous venous resection demonstrating a survival advantage.Adrian W. 11Characterization of artificial spherical particles for DEM validation studies显示文摘This paper describes a study in which advanced particle-scale characterization was carried out on spherical particles that can be used in experimental tests to validate discrete element method (DEM) simulations. Two types of particle, alkaline and borosilicate glass beads, made from two different materials, were considered. The particle shape, stiffness, contact friction properties and surface roughness were measured. The influences of hardness and roughness on the mechanical response of the particles were carefully considered. Compared to the alkaline beads, the borosilicate beads were more spherical and more uniform in size, and they exhibited mechanical characteristics closer to natural quartz sand. 14Novel en-bloc resection of locally advanced hilar cholangiocarcinoma: the Rex recess approach显示文摘Loco-regional recurrence after potentially curative resection remains a problem in hilar cholangiocarcinoma. Hilar dissection risks local spillage of tumor cells leading to suboptimal disease free survival. We have developed a new technique of radical resection for hilar cholangiocarcinoma based on the distinctive anatomy of the Rex recess of the liver, which has been assessed in two patients with locally advanced hilar cholangiocarcinoma. This technique included a right hepatectomy with en-bloc resection of the hepatoduodenal ligament and portal venous reconstruction to the left portal vein at the Rex recess. 15Suppression of monosodium urate crystal-induced inflammation by inhibiting TGF-β-activated kinase 1-dependent signaling:role of the ubiquitin proteasome system显示文摘Monosodium urate(MSU)crystals activate inflammatory pathways that overlap with interleukin-1β(IL-1β)signaling.However,the post-translational mechanisms involved and the role of signaling proteins in this activation are unknown.In the present study,we investigated the intracellular signaling mechanisms involved in MSU-induced activation of THP-1 macrophages and human nondiseased synovial fibroblasts(NLSFs)and the in vivo efficacy of an inhibitor of tumor grovArth factor-β(TGF-β)-activated kinase 1(TAK1),5Z-7-oxozeaenoI,in MSU-induced paw inflammation in C57BL76 mice.THP-1 macrophage activation with MSU crystals(25-200 ng/ml)resulted in the rapid and sustained phosphorylation of interleukin-1 receptor-activated kinase 1(IRAK1 Thr^(209))and TAK1(Thr^(184/187))and their association with the E3 ubiquitin ligase TRAF6.At the cellular level,MSU inhibited the deubiquitinases A20 and UCHL2 and increased 20s proteasomal activity,leading to a global decrease in K^(63)-linked ubiquitination and increase in K^(48)-linked ubiquitination in THP-1 macrophages.While MSU did not stimulate cytokine produaion in NLSFs,it significantly amplified IL-1β-induced IL-6,IL-8,and ENA-78/CXCL5 production.Docking studies and MD simulations followed by TAK1 in vitro kinase assays revealed that uric acid molecules are capable of arresting TAK1 in an active-state conformation,resulting in sustained TAK1 kinase activation.Importantly,MSU-induced proinflammatory cytokine production was completely inhibited by 5Z-7-oxozeaenol but not IRAK1/4 or TRAF6 inhibitors.Administration of 5Z-7-oxozeaenol(5 or 15 mg/kg;orally)significantly inhibited MSU-induced paw inflammation in C57BL/6 mice.Our study identifies a novel post-translational mechanism of TAK1 activation by MSU and suggests the therapeutic potential of TAK1 in regulating MSU-induced Inflammation.Anil K.Singh Mahamudul Haque Kayla O'Sullivan Mukesh Chourasia Madhu M.Ouseph Salahuddin Ahmed 2021Cellular & Molecular Immunology2021,18,1:2
16BRAF inhibitor treatment of melanoma causing colonic polyps:An alternative hypothesis显示文摘Colonic polyps may arise from BRAF inhibitor treatment of melanoma, possibly due paradoxical activation of the mitogen-activated protein(MAP)-kinase pathway. In an alternative evidence based scenario, tubular colonic adenomas with APC gene mutations have also been identified in the context of BRAF inhibitor treatment, in the absence of mutations of MAPK genes. A minority of colorectal cancers develop by an alternative 'serrated polyp pathway'. This article postulates a novel hypothesis, that the established phenotypic and molecular characteristics of serrated colonic polyps/CRC offer an intriguing insight into the pathobiology of BRAF inhibitor induced colonic polyps. Serrated polyps are characterized by a Cp G island methylation phenotype, MLH1 silencing and cellular senescence. They also have BRAF mutations.
