| 5 | Computational Prediction and Identification of Epstein-Barr Virus Latent Membrane Protein 2A Antigen-Specific CD8^+ T-Cell Epitopes显示文摘Epstein-Barr virus (EBV) associated nasopharyngeal carcinoma (NPC) is a high incidence tumor in Southeast Asia. Among EBV encoded proteins, latent membrane protein 2A (LMP2A) is an important antigen for T cell therapy of EBV. In this study, we predicted six HLA-A2 restricted CTL candidate epitopes of LMP2A by SYFPEITHI, NetMHC and MHCPred methods combined with the polynomial method. Subsequently, biological functions of these peptides were tested by experiments in vitro. In ELISPOT assay, the positive response of the LMP2A specific CTL stimulated by three (LMP2A264-272, LMP2A426-434 and LMP2A356-364) of six peptides respectively showed that the numbers of spots forming cells (SFC) ranged from 55.7 to 80.6 SFC/5 × 104 CD8+ T cells and the responding index (RI) ranged from 5.4 to 7. These three epitope-specific CTLs could effectively kill specific HLA-A2-expressing target cells. As a result, LMP2A264-272 (QLSPLLGAV), LMP2A426-434 (CLGGLLTMV) and LMP2A356-364 (FLYALALLL) were identified as LMP2A-specific CD8+ T-cell epitopes. | 6 | Preparation of Li_4Ti_5O_(12) submicrospheres and their application as anode materials of rechargeable lithium-ion batteries显示文摘We report on the preparation of spinel Li4Ti5O12 submicrospheres and their application as anode materials of rechargeable lithium-ion batteries. The spinel Li4Ti5O12 submicrospheres are synthesized with three steps of the hydrolysis of TiCl4 to form rutile TiO2, the hydrothermal treatment of rutile TiO2 with LiOH to prepare an intermediate phase of LiTi2O4+δ, and the calcinations of LiTi2O4+δ to obtain spinel Li4Ti5O12. The as-prepared products are investigated by X-ray powder diffraction (XRD), scanning electron microscopy (SEM), and transmission electron microscopy (TEM). The diameters of Li4Ti5O12 submicrospheres with novel hierarchical microstructures are about 200–300 nm with the assembly of 20–30 nm nanoparticles. The electrochemical properties of Li4Ti5O12 submicrospheres are measured by galvanostatical discharge/charge test and cyclic voltammetry (CV). The as-prepared Li4Ti5O12 display excellent discharge/charge rate and cycling capability. A high discharge capacity of 174.3 mAh/g is obtained in the first discharge at 1 C rate. Meanwhile, there is only tiny capacity fading with nearly 100% columbic efficiency in the sequential 5–50 cycles. Moreover, lithium-ion diffusion coefficient in Li4Ti5O12 is calculated to be 1.03 × 10-7 cm2/s. We propose a reaction mechanism based on phase transition from orthorhombic V2O5 to zinc pyrovanadate on first discharging and reversible Zn^2+ (de)intercalation in the open-structured hosts during subsequent cycling. This open and stable architecture enables a high reversible capacity of 319 mAh g^-1 at 20 mAg^-1 and a capacity retention of 81% over 500 cycles. | 9 | Low-temperature sintering and microwave dielectric properties of CaMg1−xLi2xSi2O6(x=0−0.3)ceramics显示文摘In this study, low-temperature fired CaMg1−xLi2xSi2O6 microwave dielectric ceramics were prepared via the traditional solid-state reaction method. In this process, 0.4 wt% Li2CO3-B2O3-SiO2-CaCO3-Al2O3 (LBSCA) glass was added as a sintering aid. The results showed that ceramics consisted of CaMgSi2O6 as the main phase. The second phases were CaSiO3 always existing and Li2SiO3 occurring at substitution content x > 0.05. Li+ substitution effectively lowered sintering temperature due to 0.4 wt% LBSCA and contributed to grain densification, and the most homogeneous morphology could be observed at x = 0.05. The effects of relative density, the second phase, and ionic polarizability on dielectric constant (εr) were investigated. The quality factor (Q × f) varied with packing fraction that concerned the second phase. Moreover, the temperature coefficient of the resonant frequency (τf) was influenced by MgO6 octahedral distortion and bond valence. Excellent dielectric properties of the CaMg1−xLi2xSi2O6 ceramic was exhibited at x = 0.05 with εr = 7.44, Q × f = 41,017 GHz (f = 15.1638 GHz), and τf = −59.3 ppm/°C when sintered at 900 °C. The coated materials are synthesized via a simple dry coating method of NCA with Zr(OH)_4 powders, and then characterized with scanning electron microscopy(SEM), transmission electron microscopy(TEM) and X-ray photoelectron spectroscopy(XPS). Experimental results show that amorphous Zr(OH)_4 powders have been successfully coated on the surface of spherical NCA particles, exhibiting improved electrochemical performance. 0.50 wt% Zr(OH)_4 coated NCA delivers a capacity of 197.6 mAh/g at the first cycle and 154.3 mAh/g after 100 cycles with a capacity retention of 78.1% at 1 C rate. In comparison, the pure NCA shows a capacity of 194.6 mAh/g at the first cycle and 142.5 mAh/g after 100 cycles with a capacity retention of 73.2% at 1 C rate. Electrochemical impedance spectroscopy(EIS) results show that the coated material exhibits a lower resistance, indicating that the coating layer can efficiently suppress transition metals dissolution and decrease the side reactions at the surface between the electrode and electrolyte. | 15 | A quantum-chemical study on the discharge reaction mechanism of lithium-sulfur batteries显示文摘Lithium-sulfur batteries have attracted a great interest in electrochemical energy conversion and storage, but their discharge mechanism remains not well understood up to now. Here, we report density functional theory (DFT) calculation study of the discharge mechanism for lithium-sulfur batteries which are based on the structure of S8 and Li2S x (1≤x≤8) clusters. The results show that for Li2S x (1≤x≤8) clusters, the most stable geometry is chainlike when x = 1 and 6, while the minimal-energy structure is found to be cyclic when x = 2-5, 7,8. The stability of Li2S x (1≤x≤8) clusters increases with the decreasing x value, indicating a favorable thermodynamic tendency of transition from S8 to Li2S. A three-step reaction route has been proposed during the discharge process, that is, S8 →Li2S4 at about 2.30 V, Li2S4 →Li2S2 at around 2.22 V, and Li2S2 → Li2S at 2.18 V. Furthermore, the effect of the electrolyte on the potential platform has been also investigated. The discharge potential is found to increase with the decrease of dielectric constant of the electrolyte. Here, we report that dEAAT2 is required for the larval startle response to sound stimuli. d EAAT2 was found to be enriched in the distal region of chordotonal neurons where sound transduction occurs. The Ca^(2+) imaging and electrophysiological results showed that disrupted dEAAT2 expression significantly reduced the response of chordotonal neurons to sound.More importantly, expressing d EAAT2 in the chordotonal neurons rescued these mutant phenotypes. | 20 | Cholecystectomy is associated with higher risk of recurrence after microwave ablation of hepatocellular carcinoma:a propensity score matching analysis显示文摘Objective:To explore the association between cholecystectomy and the prognostic outcomes of patients with hepatocellular carcinoma(H CC)who underwent microwave ablation(MWA).Methods:Patients with HCC(«=921)who underwent MWA were included and divided into cholecystectomy(n=114)and non-cholecystectomy groups(n=807).After propensity score matching(PSM)at a 1:2 ratio,overall survival(OS)and disease-free survival(DFS)rates were analyzed to compare prognostic outcomes between the cholecystectomy(«=114)and non-cholecystectomy groups(n=228).Univariate and multivariate Cox analyses were performed to assess potential risk factors for OS and DFS.Major complications were also compared between the groups.Results:After matching,no significant differences between groups were observed in baseline characteristics.The 1-,3-,and 5-year OS rates were 96.5%,82.1%,and 67.1%in the cholecystectomy group,and 97.4%,85.2%,and 74.4%in the non-cholecystectomy group(P=0.396);the 1-,3-,and 5-year DFS rates were 58.4%,34.5%,and 26.6%in the cholecystectomy group,and 73.6%,44.7%,and 32.2%in the non-cholecystectomy group(P=0.026),respectively.The intrahepatic distant recurrence rate in the cholecystectomy group was significantly higher than that in the non-cholecystectomy group(P=0.026),and the local tumor recurrence and extrahepatic recurrence rates did not significantly differ between the groups(P=0.609 and P=0.879).Multivariate analysis revealed that cholecystectomy(HR=1.364,95%Cl 1.023-1.819,P=0.035),number of tumors(2 vs.1:HR=2.744,95%Cl 1.925-3.912,P<0.001;3 vs.1:HR=3.411,95%Cl 2.021-5.759,P<0.001),and y-GT levels(HR=1.003,95%Cl 1.000-1.006,P<0.024)were independent risk factors for DFS.The best y-GT level cut-off value for predicting median DFS was 39.6 U/L(area under the curve=0.600,P<0.05).A positive correlation was observed between cholecystectomy and y-GT level(r=0.108,95%Cl-0.001-0.214,P=0.047).Subgroup analysis showed that the DFS rates were significantly higher in the non-cholecystectomy group than the cholecystectomy group when Y-GT>39.6 U/L(i3=0.044).The 5-,10-,15-,20-,and 25-year recurrence rates from the time of cholecystectomy were 2.63%,21.93%,42.11%,58.77%,and 65.79%,respectively.A significant positive correlation was observed between cholecystectomy and the time from cholecystectomy to recurrence(r=0.205,95%Cl 0.016-0.379,P=0.029).There were no significant differences in complications between groups(P=0.685).Conclusions:Patients with HCC who underwent cholecystectomy were more likely to develop intrahepatic distant recurrence after MWA,an outcome probably associated with increased y-GT levels.Moreover,the recurrence rates increased with time. | Hongcai Yang Yi Yang Jianping Dou Rui Cui Zhigang Cheng Zhiyu Han Fangyi Liu Xiaoling Yu Xiang Zhou Jie Yu Ping Liang | 2020 | Cancer Biology & Medicine2020,17,2: | 3 |
