| 1 | Hepatitis C virus core protein modulates several signaling pathways involved in hepatocellular carcinoma显示文摘Hepatocellular carcinoma(HCC) is the fifth most common cancer, and hepatitis C virus(HCV) infection plays a major role in HCC development. The molecular mechanisms by which HCV infection leads to HCC are varied. HCV core protein is an important risk factor in HCV-associated liver pathogenesis and can modulate several signaling pathways involved in cell cycle regulation, cell growth promotion, cell proliferation, apoptosis, oxidative stress and lipid metabolism. The dysregulation of signaling pathways such as transforming growth factor β(TGF-β), vascular endothelial growth factor(VEGF), Wnt/β-catenin(WNT), cyclooxygenase-2(COX-2) and peroxisome proliferator-activated receptor α(PPARα) by HCV core protein is implicated in the development of HCC. Therefore, it has been suggested that this protein be considered a favorable target for further studies in the development of HCC. In addition, considering the axial role of these signaling pathways in HCC, they are considered druggable targets for cancer therapy. | 2 | Immunomodulatory effects of astragalus polysaccharide on Human peripheral blood mononuclear cells co-cultured with cervical cancer cell lineImmunomodulatory effects of astragalus polysaccharide on Human peripheral blood mononuclear cells co-cultured with cervical cancer cell line显示文摘OBJECTIVE:To investigate immunomodulatory effects of Astragalus polysaccharides(APS)on the co-culture of peripheral blood mononuclear cells(PBMCs)with He La cervical cancer cell line.METHODS:To assess the proliferation of PBMCs,carboxyfluorescein succinimidyl ester(CFSE)-labeled PBMCs were co-cultured with He La cells and treated with different concentrations of APS.Supernatants of cell culture were collected for cytokines assay via enzyme-linked immunosorbent assay(ELISA).The impact of APS on the proliferation of PBMCs,induction of regulatory T cells(Tregs),and myeloid-derived suppressor cells(MDSCs)was carried out by flow cytometry.RESULTS:It was observed that APS could increase the proliferation of PBMCs co-cultured with He La cells(P<0.05).However,APS had no significant effects on the induction of Tregs and MDSCs in the co-culture assay(P>0.05).Furthermore,ELISA results demonstrated that APS could decrease IL-10 and TGF-βconcentration(P<0.05).CONCLUSION:The above-mentioned characteristics showed that APS might be able to modulate immune responses and improve anti-tumor effects through increasing the proliferation of PBMCs and decreasing inhibitory cytokines secretion as critical mediators of immune suppression in the tumor microenvironment. | Elham Shokati Mohammad-Reza Shokri Kobra Entezami Samaneh Khorrami Mona Amani Morteza Motallebnezhad Elahe Safari | 2021 | Journal of Traditional Chinese Medicine2021,41,5: | 5 |
| 3 | Differential reactivity of mouse monoclonal anti-HBs antibodies with recombinant mutant HBs antigens显示文摘AIM: To investigate the reactivity of a panel of 8 mouse anti-hepatitis B surface antigen (HBsAg) monoclonal antibodies (mAbs) using a collection of 9 recombinant HBsAg mutants with a variety of amino acid substitutions mostly located within the “a” region.METHODS: The entire HBs genes previously cloned into a mammalian expression vector were transiently transfected into COS7 cells. Two standard unmutated sequences of the ayw and adw subtypes served as controls. Secreted mutant proteins were collected and measured by three commercial diagnostic immunoassays to assess transfection efficiency. Reactivity of anti-HBs mAbs with mutated HBsAgs was determined by sandwich enzyme-linked immunosorbent assay (ELISA).RESULTS: Reactivity of anti-HBs mAbs with mutated HBsAgs revealed different patterns. While three mutants reacted strongly with all mAbs, two mutants reacted weakly with only two mAbs and the remaining proteins displayed variable degrees of reactivity towards different mAbs. Accordingly, four groups of mAbs with different but overlapping reactivity patterns could be envisaged. One group consisting of two mAbs (37C5-S7 and 35C6-S11) was found to recognize stable linear epitopes conserved in all mutants. | 5 | Bilateral same-session intravitreal injections of antivascular endothelial growth factors显示文摘AIM: To document the indications, safety and possible complications of bilateral same-session intravitreal antivascular endothelial growth factor(VEGF) injections performed in the ophthalmic operating room.METHODS: A retrospective case series study.Consecutive records of seventy four patients receiving simultaneous bilateral intravitreal injections of either ranibizumab or bevacizumab, between September 2010 and September 2013, were reviewed and the outcomes were assessed. Data collected included number of injections, indications for injections, pre-injection and post-injection visual acuity(VA), pre-injection and postinjection intraocular pressure and ocular and systemic complications/complaints after each injection.RESULTS: A total of 342 injections were administered to 74 patients, with a mean of 4.62 injections per patient.Seventy-three patients received bevacizumab(Avastin;Genentech Inc., South San Francisco, California, USA)alone, and only one patient received both bevacizumab and ranibizumab(Lucentis; Genentech Inc.) distributed between the injections. Pre- and post-injection VA follow-up measurements were available for 65 patients.Mean follow up period was 22 mo. The indications for initiating therapy were choroidal neovascular membrane from age-related macular degeneration(3 patients) and diabetic macular edema(71 patients). The mean Snellen VA before each injection was 6/22. The next post-injection follow-up mean Snellen VA was 6/20. One patient had a painful, culture-positive endophthalmitis in one eye 3d after bilateral bevacizumab. Another patient had a painless subconjunctival hemorrhage in one eye.No other ocular or systemic adverse side effects/complaints have been registered in this study group.CONCLUSION: Bilateral same-session intravitreal injections using a separate povidone-iodine preparation,speculum, needle, and syringe for each eye are well-tolerated. None of the subjects in this study requested to switch to alternating unilateral injections. | 7 | Improvement of the prediction performance of a soft sensor model based on support vector regression for production of ultra-low sulfur diesel显示文摘A novel data-driven, soft sensor based on support vector regression(SVR) integrated with a data compression technique was developed to predict the product quality for the hydrodesulfurization(HDS) process. A wide range of experimental data was taken from a HDS setup to train and test the SVR model. Hyper-parameter tuning is one of the main challenges to improve predictive accuracy of the SVR model.Therefore, a hybrid approach using a combination of genetic algorithm(GA) and sequential quadratic programming(SQP)methods(GA–SQP) was developed. Performance of different optimization algorithms including GA–SQP, GA, pattern search(PS), and grid search(GS) indicated that the best average absolute relative error(AARE), squared correlation coefficient(R2), and computation time(CT)(AARE = 0.0745, R2= 0.997 and CT = 56 s) was accomplished by the hybrid algorithm. Moreover, to reduce the CT and improve the accuracy of the SVR model, the vector quantization(VQ) technique was used. The results also showed that the VQ technique can decrease the training time and improve prediction performance of the SVR model. | 8 | Soft sensor design for hydrodesulfurization process using support vector regression based on WT and PCA显示文摘A novel method for developing a reliable data driven soft sensor to improve the prediction accuracy of sulfur content in hydrodesulfurization(HDS) process was proposed. Therefore, an integrated approach using support vector regression(SVR) based on wavelet transform(WT) and principal component analysis(PCA) was used. Experimental data from the HDS setup were employed to validate the proposed model. The results reveal that the integrated WT-PCA with SVR model was able to increase the prediction accuracy of SVR model. Implementation of the proposed model delivers the best satisfactory predicting performance(EAARE=0.058 and R2=0.97) in comparison with SVR.
